Characterization of Benzoate Degradation via ortho-Cleavage by Streptomyces setonii

  • Streptomyces are widespread in nature and play a very important role in the biosynthesis as well as biodegradation of natural and unnatural aromatic compounds. Both qualitatively and quantitatively through TLC and UV spectrophotometric assays, it was observed that the thermophilic soil bacteria S. setonii (ATCC 39116), which can utilize a benzoate as a sole carbon and energy source in a minimal liquid culture, was not very sensitive to the benzoate concentation and to the culture conditions such as the pH and temperature. The in vitro conversion of a catechol to a cis, cis-muconic acid by a crude S. setonii lysate implies that the aromatic ring cleavage by S. setonii is initiated by a thermostable catechol-1,2-dioxygenase, the key enzyme in the ortho-cleavage pathway of aromatic compound biodegradation. Unlike non-degrading S. lividans, S.setonii was also highly resistant to other similar hazardous aromatic compounds, exhibiting almost no adverse effect on its growth in a complex liquid culture.

Sphingosine-1-Phosphate Decreases Melanin Synthesis via Sustained ERK Activation and Subsequent MITF Degradation

  • This study shows that sphingosine-1-phosphate (SPP) significantly inhibits melanin synthesis in a concentration-dependent manner, and that the activity of tyrosinase was also reduced in SPP-treated cells. In contrast. a specific extracellular signal-regulated protein kinase (ERK) pathway inhibitor, PD98059 increased tyrosinase activity and melanin production, and PD98059 restored the reduced tyrosinase activity and pigmentation induced by SPP. Role of the mammalian ATG8/LC3 family in autophagy: differential and compensatory roles in the spatiotemporal regulation of autophagy

  • Autophagy, an evolutionarily conserved cellular degradation pathway of the lysosome, is associated with many physiological and pathological processes. The hallmark of autophagy is the formation of the autophagosome that engulfs and degrades cytosolic components via its fusion with the lysosome, in either a selective or a non-selective manner. Autophagy is tightly regulated by proteins encoded by autophagy-related (atg) genes. Among these proteins, ATG8/LC3 is essential for autophagosome biogenesis/maturation and it also functions as an adaptor protein for selective autophagy. In mammalian cells, several homologs of yeast Atg8 such as MAP1LC3, GABARAP, and GABARAPL 1/2 have been identified. However, the biological relevance of this gene diversity in higher eukaryotes, and their specific roles, are largely unknown. In this review, we describe the mammalian ATG8/LC3 family and discuss recent advancements in understanding their roles in the autophagic process.

갯벌에서 분리한 유기염소계 살충제 Endosulfan 분해 세균의 특성 (Characterization of Organochlorine Insecticide Endosulfan-Degrading Bacterium Isolated from Seaside Sediment)

  • An endosulfan degrading bacterial strain, K-1321, was isolated by endosulfan-enrichment culture from a seaside sediment collected at Dadaepo Beach, Busan, Korea. The strain was identified as a Serratia sp. based on the results of morphological, biochemical and 16S rDNA homology analyses. Serratia sp. K-1321 was able to completely degrade 50 ppm endosulfan in culture media and soil within 6 weeks at $25^{\circ}C$. GC/MS analysis revealed that endosulfan diol was an intermediate of the bacterial endosulfan degradation. Considering the above results, we concluded that Serratia sp. K-1321 utilized endosulfan as a carbon source and metabolized endosulfan via a less toxic pathway, such as the formation of endosulfan diol as an intermediate.

Researches Using Radio-labelled Insecticides in Korea

  • The scientific articles using radio labelled insecticides performed by Korean researchers were reviewed. The research works were divided into 4 categories such as soil, plant, animal and insect. All researches used $^{14}C$-labelled chemicals, and the $^{14}C$-carbofuran was widely used among them. Fate of insecticides, bound-residues and metabolic process were staple concerning area in soil study. And the uptake and translocation, metabolism and metabolites also a major interests in plant study. As well as the degradation, metabolic pathway and metabolites, and distribution of chemicals in animal tissue were another point of consideration in animal study. And finally, the penetration ratio into body and resistant mechanism were the major concerning views of study with insects.

Melibiose로부터 M-5중간체 galactosylglycerol의 합성 (Synthesis of galactosylglycerol from Melibiose as M-5 Intermediate)

  • The galactolipid M-5, which showed anti-inflammatory activity is glycoglycerolipid isolated from the Okinawa marine sponge Phyllospongia foliascens. Glycolipids have been synthesized by various methods, especially it were generally known that synthetic method of M-5 analogue and synthetic method of various glycolipids by glycosidation after synthesis of glycerolipid part. The others, it was not suggested that synthetic method via glycosylglycerol obtained by degradation from diglycoside. This study was carried out to investigate the synthesis of galactosylglycerol from melibiose as M-5 intermediate. Synthesis of galactosylglycerol was accomplished by selective protection of hydroxy group of sugar and diol cleavage by Pb(OAc)$_4$. As a result, galactosylglycerol was synthesized by 8 steps pathway and their structures were elucidated by analysis instrument.

Comparison of tdcA Expression Between Escherichia coli and Salmonella enterica Serovar Typhimurium

  • Both Escherichia coli (E. coli) and Salmonella enterica serovar Typhimurium (S. Typhimurium) have a tdc operon that encodes enzymes involved in a metabolic pathway for the degradation of L-serine and L-threonine. However, S. Typhimurium does not have the tdcR gene, which is a positive regulator in E. coli. In the present study, transcriptional analysis revealed that tdcA expression in E. coli is higher under anaerobic than aerobic growth conditions, but the opposite is true in S. Typhimurium. Interestingly, a tdcR mutant strain of E. coli showed a similar expression pattern to that observed in S. Typhimurium and was also induced by anaerobic shock. These results suggest that the induction of tdcA expression by anaerobic conditions is observable when tdcA expression is low owing to the absence of TdcR.

Critical Roles of Deubiquitinating Enzymes in the Nervous System and Neurodegenerative Disorders

  • Post-translational modifications play major roles in the stability, function, and localization of target proteins involved in the nervous system. The ubiquitin-proteasome pathway uses small ubiquitin molecules to degrade neuronal proteins. Deubiquitinating enzymes (DUBs) reverse this degradation and thereby control neuronal cell fate, synaptic plasticity, axonal growth, and proper function of the nervous system. Moreover, mutations or downregulation of certain DUBs have been found in several neurodegenerative diseases, as well as gliomas and neuroblastomas. Based on emerging findings, DUBs represent an important target for therapeutic intervention in various neurological disorders. Here, we summarize advances in our understanding of the roles of DUBs related to neurobiology.

Polypyrrole Doped with Sulfonate Derivatives of Metalloporphyrin: Use in Cathodic Reduction of Oxygen in Acidic and Basic Solutions

  • Incorporation of metalloporphyrins into polypyrrole (PPy) film was achieved either by electropolymerization of pyrrole in the presence of metal-tetra(sulfonatophenyl)porphyrin anion (MTSPP, M=Co, Fe) or by metalizing hydrogenated tetra(4-sulfonatophenyl)porphyrin anion (H2TSPP) doped into PPy through ion-exchange. Electrochemical reduction of oxygen on the PPy doped with metallo porphyrin (PPy-MTSPP) was studied in acidic and basic solutions. Oxygen reduction on PPy-MTSPP electrodes appeared to proceed through a 4-electron pathway as well as a 2-electron path. In acidic solutions, the overpotential for O2 reduction on PPy-CoTSPP electrode was smaller than that on gold by about 0.2 V. In basic solutions the overpotential of the PPy-CoTSPP electrode in the activation range was close to those of Au and Pt. The limiting current was close to that of Au. However, polypyrrole doped with cobalt-tetra(sulfonatophenyl)porphyrin anion (PPy-CoTSPP) or with iron-tetra(sulfonatophenyl)porphyrin anion (PPy-FeTSPP) was found to have limited potential windows at high temperatures (above 50 ℃), and hence the electrode could not be held at the oxygen reduction potentials in basic solutions (pH 13) without degradation of the polymer.

  • This study shows that sphingosine-1-phosphate (S1P) significantly inhibits melanin synthesis in a concentration-dependent manner, and that the activity of tyrosinase was also reduced in S1P-treated cells. In contrast, a specific extracellular signal-regulated protein kinase (ERK) pathway inhibitor, PD98059 increased tyrosinase activity and melanin production, and PD98059 restored the reduced tyrosinase activity and pigmentation induced by SIP. We also found that S1P induces the sustained activation of ERK and the subsequent degradation of microphthalmia-associated transcription factor (MITF), which plays a key role in melanogenesis.
