Effect of Sugar Combination in Tris-buffer on the Viability of Post-thaw Spermatozoa in Canine

  • The purpose of this study was to investigate the effect of kind and combination of sugars on the viability and acrosome damage of post-thaw spermatozoa in canine. The extender used was Tris-citric acid extender (Tris-buffer) supplemented with 20% Egg-yolk, 8% glycerol, 1% Equex STM paste, and 70 mM sugars such as monosaccharide (fructose and xylose) and disaccharide(trehalose). To evaluate of sugar combination, the sugars supplemented in Tris-buffer were combined such as single (fructose, xylose, trehalose), two combinations (Fruc+Tre, Fruc+xyl, Tre+xyl) and three combinations (Fruc+Tre+Xyl). The concentration of sperm collected were adjusted of 50${\times}$10$\^$6/ per straw for freezing. The frozen spermatozoa were thawed at 37$^{\circ}C$ for 1 min and then analysis for CASA program in Livestock Improvement Main Center, NACF. The motility of post-thaw spermatozoa in Fruc+Tre was higher than those in fructose, trehalose, xylose, Fru+xyl, Tre+xyl and Fru+Tre+Xyl (79% vs. 63, 66, 70, 71, 74 and 75%). The progressive motility after CASA analysis in Fuc+Tre group was also higher than those in fructose, trehalose, xylose, Fru+xyl, Tre+xyl and Fru+Tre+Xyl (67% vs. 53, 57, 60, 61, 62 and 64%). The acrosome damage of post-thaw spermatozoa stained was not significantly different among treatment groups such as fructose, trehalose, xylose, Fru+Tre, Fru+xyl, Tre+xyl and Freu+tre+xyl (17.7, 18.3, 28.0, 17.0, 19.7, 20.0 and 19.0%). The results indicated that the motility and progressive motility of post-thaw spermatozoa in Fru+Tre group was better, and acrosome normality was not different among all groups. The use of Tris-buffer supplemented with Fru+Tre as sugar for frezing of canine spermatosoa could be better and apply to semen banking and artificial insemination.

Review about the Impacts from Volcanic Ash Fall (화산재 강하로부터의 영향 고찰)

  • The materials generating from volcanic eruption are volcanic gases, lavas and pyroclastic materials. Volcanic ash which has small-grain size (< 2 mm in diameter) can be moved easily and disperse widely, thus it may affect to communities across hundreds of square kilometers. The impacts from volcanic ash fall on people, structures, equipments, plants and livestock largely depend on ash thickness. According to increasing ash thickness, the intensity and area of damage may increase and affect significant damages not to human health but also to infrastructures. To reduce the impacts from volcanic ash fall, we have to establish the guidances about the nature and extent of the hazard and prepare the actions to increase abilities of communities to manage hazard. Although we don't have any experience caused by volcanic ash fall during and after volcanic eruption, we need to prepare the impacts of volcanic ash fall for future eruption in the areas surrounding Korea.

  • Simulation using the virtual reality has been applied in various fields such as exercise, education, disaster simulation training, but there is a little research on disease caused by virus transmission. In this study, we conducted simulation studies and analysis of avian influenza disaster response training. The annual avian influenza virus is recurring every year in Korea, but there are still few solutions and preventive measures for the preventing the avian influenza. The avian influenza can cause a great deal of societal harm and enormous economic damage. Prophylaxis is important because livestock epidemics, such as avian influenza and foot-and-mouth disease, have a large impact on farm households. Therefore, we proposed and analyzed contents that can be avoided through simulation of avian influenza disaster response presented in this study.

  • This experiment was carried out to identify the main pathogens infected with Piper sudangrass and to evaluate the qualitative and quantitative damage of the plants infected with leaf blight (Helminthosporium turcicum PASS) under different levels of nitrogen fertilizer (20, 30 and 40 kg/10a). The experiment was design as a randomized block design with 4 replications at experimental field of Livestock Experiment Station in Suweon, 1984. The results obtained are summarized as follows: 1. The main fungi isolated from sudangrass were identified as Helminthoporium turcicum and Collectotricum graminicolum, but 1 species fungus was not identified. 2. Leaf blight was first found on June 20 and appeared extremely in the regrowth plants from July to September. The rate of attack was associated with increasing of nitrogen fertilization (P<0.01). 3. Fresh and dry matter yields were as much as 47 and 38 percentage lower in leaf blight infected plants (disease severity: V) than those of no visible infection. 4. Concentrations of crude fat and crude protein were decreased in the plants infected with Helminthosporium leaf blight, but lignin content was increased. Dry matter and organic matter digestibility of the plants were negative correlated with the infection of leaf blight(P<0.01).

Survey on Occurrence and Management of Disease and Pests in Organic Peach Orchards (유기재배 복숭아 과원의 관리현황 및 병해충 발생 실태)

  • The occurrence and management of disease and pests in six organic peach orchards were surveyed from March 2015 to March 2017. In this period, the number of certified organic and non-chemical peach farms increased to 65.5% and 31.7%, respectively. Certified organic peach farms were selected based on more than $4,000m^2$ of cultivation area and three tons of production, and their cultivation status was examined. All of the farms were either cultivated green manure crop or sod, and limited vegetation control to a minimum. For the management of soil nutrients, many farmers used livestock manure, oilcake and self-manufacturing liquid fertilizer. It was surveyed that bordeaux mixture, lime sulfur, pheromone for mating disruption of moths and plant extract were used for disease and pest control. The damage caused by the pests and diseases were 31.6% and 24.1%, respectively. The oriental fruit moth showed the highest damage rate (13.5%) in the organic peach orchards, followed by the brown rot (13.0%), peach fruit moth (7.3%) and bacterial shot hole (7.3%).

Individual Pig Detection Using Kinect Depth Information and Convolutional Neural Network (키넥트 깊이 정보와 컨볼루션 신경망을 이용한 개별 돼지의 탐지)

  • Aggression among pigs adversely affects economic returns and animal welfare in intensive pigsties. Recently, some studies have applied information technology to a livestock management system to minimize the damage resulting from such anomalies. Nonetheless, detecting each pig in a crowed pigsty is still challenging problem. In this paper, we propose a new Kinect camera and deep learning-based monitoring system for the detection of the individual pigs. The proposed system is characterized as follows. 1) The background subtraction method and depth-threshold are used to detect only standing-pigs in the Kinect-depth image. 2) The standing-pigs are detected by using YOLO (You Only Look Once) which is the fastest and most accurate model in deep learning algorithms. Our experimental results show that this method is effective for detecting individual pigs in real time in terms of both cost-effectiveness (using a low-cost Kinect depth sensor) and accuracy (average 99.40% detection accuracies).

Improvement of Egg Tray Design to Decrease Eggshell Damage (계란(鷄卵)의 난각파손(卵殼破損) 방지(防止)를 위한 난좌(卵座) 개발(開發)에 관한 연구(硏究))

  • This study was carried out to investigate the effects of shperical surface of eggs tray and eggshell on breaking strength of eggs for the development of a new egg tray design to prevent eggshell breakage. The results obtained were as follows: 1. According to the results obtained from investigation on the types of 22 egg trays collected from 9 countries(USA, Italy, France, Thailand, Japan, Netherland, Russia, Korea and Swiss), all egg trays except those from Korea were different in their types. 2. According to the determination of breaking strength for 4 pilot egg trays, type B and D were high in breaking strength. Trays B and D which would fit with the spherical surface of egg so that they could increase breaking strength were most recommendable. Breaking strength was not increased even though egg tray was strong. Thickness of eggshell of experimental eggs used in this experiment have no significance (P>0.05). 3. When eggs are packed and transported in egg tray, the breaking strength will depend on how much spherical surface of egg are covered by the spherical surface of the egg tray. Therefore, the inner spherical surface of egg tray must fit to the outer spherical surface of eggs when the egg tray is manufactured. 4. At present, there is only one type of egg tray being used in Korea despite the various egg sizes. In order to minimize the eggshell breakage, however, proper sizes or types of egg trays should be developed for each different categories of the egg size.

Prevalence of cryptosporidiosis and giardiasis in Korean native calves in Gimje area (김제 지역 한우 송아지의 크립토스포리듐, 지알디아 발생 현황)

  • This study was performed to identify the prevalence of cryptosporidiosis and giardiasis in diarrhea feces of Korean native calves raised in Gimje, Jeollabuk-do, with a rapid diagnostic kit. The prevalence of both diseases were classified according to the age of calf, season of sampling period, and sampled farm where diarrheic calf is commonly observed. In diarrhea feces of 431 calves, cryptosporidiosis had a high incidence rate between 1 and 3 weeks of age, whereas giardiasis showed a high incidence between 4 and 6 weeks of age. It is thought that the incidence of protozoan diseases is markedly influenced by the calf's postnatal age. Regarding with the sampling season, the prevalence of cryptosporidiosis and giardiasis in calves was significantly higher in spring and summer than in autumn and winter (P<0.05). Among the sampled farms where both diseases are commonly observed, cryptosporidiosis was more frequently detected than giardiasis in calves. Particularly, in farms where cryptosporidiosis is common, it tends to be a disease that is difficult to eradicate as it occurs continuously throughout the year. Hygiene management such as regular cleaning and disinfection of the livestock and the surrounding environments is considered necessary to reduce and prevent damage from these protozoal diseases.

A fundamental study on the installation methods of automatic identification buoy on coastal gill net (연안자망 부이에 어구자동식별 장치 설치방안에 관한 기초적 연구)

  • As a series of fundamental researches on the development of an automatic identification monitoring system for fishing gear. Firstly, the study on the installation method of automated identification buoy for the coastal improvement net fishing net with many loss problems on the west coast was carried out. Secondly, the study was conducted find out how to install an automatic identification buoy for coastal gill net which has the highest loss rate among the fisheries. GPS for fishing was used six times in the coastal waters around Seogwipo city in Jeju Island to determine the developmental status and underwater behavior to conduct a field survey. Next, a questionnaire was administered in parallel on the type of loss and the quantity and location of fishing gear to be developed and the water transmitter. In the field experiment, the data collection was possible from a minimum of 13 hours, ten minutes to a maximum of 20 hours and ten minutes using GPS, identifying the development status and underwater behavior of the coastal gillnet fishing gear. The result of the survey showed that the loss of coastal net fishing gear was in the following order: net (27.3%), full fishing gear (24.2%), buoys, and anchors (18.2%). The causes were active algae (50.0%), fish catches (33.3%) and natural disasters (12.5%). To solve this problem, the installation method is to attach one and two electronic buoys to top of each end of the fishing gear, and one underwater transmitter at both ends of the float line connected to the anchor. By identifying and managing abnormal conditions such as damage or loss of fishing gear due to external factors such as potent algae and cutting of fishing gear, loss of fishing gear can be reduced. If the lost fishing gear is found, it will be efficiently collected.

Improvement of Preservation Quality of Chilled Bull Semen Using ${\alpha}$-tocopherol as an Antioxidant

  • Alpha-tocopherol as an antioxidant acts in preservation of chilled semen by preserving cell membrane damage from lipid peroxidation. Optimum concentrations of ${\alpha}$-tocopherol in egg yolk-citrate (EYC) extender need to be studied in crossbred bull's semen. Different concentrations of ${\alpha}$-tocopherol viz. 0, 1, 2, 4 and 6mg per ml of extender were used. Semen was collected once a week from four bulls used to regular collection, aged 4 to 7 years, weighing 320 to 450 kg, and with body condition score 4 to 4.5 and scrotal circumference 23 to 32 cm. Semen was evaluated routinely and sperm morphology was viewed under light microscope at ${\times}1,000$ magnification after fixing with buffered formal saline. Over 90% had normal head, acrosome, mid-piece and tail. Semen was diluted with egg-yolk-citrate extender to produce $15{\times}10^6$ spermatozoa/ml and 0, 1, 2, 4 and 6 mg/ml ${\alpha}$-tocopherol were added. The semens amples were kept at $8^{\circ}C$. Sperm motility and viability were examined daily up to 5 days under light microscopy at ${\times}200$ magnification. Sperm viability was acceptable (${\geq}40%$) up to the $4^{th}$ day with all concentrations of ${\alpha}$-tocopherol and up to the $5^{th}$ day with 2 mg/ml ${\alpha}$-tocopherol. Sperm motility was acceptable (${\geq}40%$) up to the $3^{rd}$ day irrespective of ${\alpha}$-tocopherol concentration, and up to the $4^{th}$ day with 2 mg/ml ${\alpha}$-tocopherol. It is suggested that the lifespan of chilled semen may be extended up to 4 days by adding 2mg/ml ${\alpha}$-tocopherol.
