Estimation of environmental effects and genetic parameters for somatic cell score, stress and immunological traits in Holstein cattle (젖소에 있어서 원유 중 체세포수, 스트레스 및 면역물질에 대한 환경효과와 유전모수 추정)

  • Milk yield and its quality traits determine the dairy enterprise profitability and sustainability. Milk quality traits including somatic cell counts (SCC) is an upcoming economic challenge for dairy farming community in Korea. This study estimated the effect of parity, stage of lactation (early, mid and late lactation) on SCC, stress (blood cortisol) and immunity (blood IgG, lymphocyte and neutrophil) traits, their heritabilities and genetic correlations between them. SCS and blood neutrophil count were significantly affected by both parity and stage of lactation, however; IgG was affected by only stage of lactation, and blood cortisol and lymphocyte were not affected by both factors. The SCS has shown increasing trend with the parity, however; the difference between first and second parity, second and third parity were not significant. The SCS in early (≤90 days) and late lactation (181≤days) were higher than that of mid lactation (91~180 days). Cortisol concentration in blood was lowest in fourth parity, however; the differences among the first three parties were not significant. The IgG was higher in fourth parity compare with first parity however; all other comparisons were noted non-significant. The IgG concentration was significantly higher in early lactation than those of mid and late lactation. The blood lymphocytes were decreased with increasing parity however the differences beyond second parity were not significant. The neutrophils were increased with the increasing lactation stage however; the difference between early and mid lactation was not significant. Although heritability of SCS was still lower, but it was meaningful value (0.09) and may be considered to improve milk quality. The genetic correlations between SCS and cortisol (-0.96), and lymphocyte (-0.76) were highly negative. Heritability of cortisol was low, however genetic correlations between cortisol and lymphocyte (0.79) was highly positive. IgG with medium heritability was correlated negatively with lymphocyte (-0.88) and neutrophil (-0.98). Lymphocyte was lowly heritable and highly correlated with neutrophil concentration (0.87).This study suggested that cortisol, IgG, lymphocyte and neutrophil being positively genetically correlation with somatic cell score could be used as alternative traits to enhance milk quality in Holstein cattle. Further studies are warranted to estimate genetic relationships between immunological and production traits to increase the genetic merit of Holstein cows for milk yield, to improve animal health and economic viability under intensive management system.

The Study on the Relationship between Changes of Rumen Microflora and Bloat in Jersey Cow (저지종 젖소의 반추위 내 미생물 균총 변화와 고창증 발병간의 상관관계 연구)

  • This study was conducted to investigate the relationship between changes of rumen microflora and bloat in Jersey cow. Jersey cows (control age: 42 months, control weight: 558kg; treatment age: 29 months, treatment weight 507kg) were fed on the basis of dairy feeding management at dairy science division in National Institute of Animal Science. The change of microbial population in rumen was analyzed by using next generation sequencing (NGS) technologies due to metabolic disease. The diversity of Ruminococcus bromii, Bifidobacterium pseudolongum, Bifidobacterium merycicum and Butyrivibrio fibrisolvens known as major starch fermenting bacteria was increased more than 36-fold in bloated Jersey, while cellulolytic bacteria community such as Fibrobacter succinogenes, Ruminococcus albus and Ruminococcus flavefaciens was increased more than 12-fold in non-bloated Jersey. The proportion of bacteroidetes and firmicutes was 33.4% and 39.6% in non-bloated Jersey's rumen, while bacteroidetes and firmicutes were 24.9% and 55.1% in bloated Jersey's. In conclusion, the change of rumen microbial community, in particular the increase in starch fermenting bacteria, might have an effect to occur the bloat in Jersey cow.

Estimation of Influence of Milking System Type on Milking Center Effluent Amount and its Characteristics (착유시스템 유형별 세척수의 발생량과 특성)

  • The purpose of this study was to determine the effect of milking system type on milking center effluent production through the four seasons. Four different types of milking systems (Bucket, Pipeline, Tandem and Herringbone) were estimated, in duplicate, through the different seasons. The following conclusions can be drawn from this study. 1. The quantity of wastewater produced from Tandem and Herringbone milking systems were significantly larger than Bucket milking system (p<0.05). 2. The main wastewater production was from the washing of milking apparatus. Tandem and Herringbone milking systems produced 398.8 and $407.7{\ell}$/day of wastewater, respectively, for apparatus washing. These values were significantly higher than the other milking systems during the summer (p<0.05). 3. The average wastewater production from the various milking systems was $15.4{\ell}$/head/day. The quantity of wastewater production during summer ($16.4{\ell}$/head/day) season was higher than of the other seasons. 4. The highest level of $BOD_5$ ($906.4mg/\ell$) was produced from the washing of the parlor floor and the lowest level of $BOD_5$ ($212.4mg/\ell$) was produced from the washing of the udders of the cows. 5. The pH of dairy wastewater was in the range of $7.3{\sim}8.2$ and the average levels of $BOD_5$, COD, SS, T-N, and T-P were 731.2, 479.0, 751.6, 79.1, $14.7mg/\ell$, respectively. Following conclusions can be drawn from this experiment. The quantities of wastewater production from Bucket, Pipeline, Tandem and Herringbone milking system were 143.9, 487.9, 914.0, and $856.7{\ell}$, respectively. The average wastewater produced from the milking systems was $15.4{\ell}$/head per day. In order to effectively manage on the wastewater from milking systems, dairy farms need to consider the milking system type and farm size when determining the optimum wastewater treatment system.

The Impact of Milk Production Level on Profit Traits of Holstein Dairy Cattle in Korea (국내 Holstein종 젖소의 생산수준이 젖소의 수익형질에 미치는 효과)

  • Data including 1,372,050 milk records pertaining to 438,019 cows from 1983 to 2011 collected during performance tests conducted by the National Livestock Cooperative Dairy Improvement Center were used to calculate milk income and profit of individuals and investigate the effects of production levels of early lactation (parity 1 and 2, respectively). Individuals with a moderate level of early lactation stayed longer in herds. Among parity 1, the 9,000 kg or higher group had a lower mean number of lactations than the overall mean of 3.13. The 7,000 kg or lower and 10,000 kg or higher groups had lower mean life time milking days than the overall mean of 1,076.8 days. Standard deviations of lifetime traits tended to decrease as production levels increased. For parity 2, the 11,000 kg or higher group had a lower mean number of lactation than the overall mean of 3.43. The lifetime milking days was highest in the 12,000 kg group (1,212.0 days), and generally smaller in the lower groups. Profit increased as the production level of groups increased for both parity 1 and 2. In groups with low production levels, profit of parity 1 was higher than that of parity 2, while the reverse was true in groups with high production levels. These results suggest that individuals in the low production groups had a greater likelihood to be culled due to reproductive or other problems. Furthermore, the accuracy of the prediction of lifetime profit of individuals with a milk yield of 305 days seems to be higher for parity 2 than parity 1; therefore, it is desirable to predict lifetime profit using the 305d milk yield of parity 2. In conclusion, breeding goals are based on many factors in functions for the estimation of profit; however, production levels during early lactation (parity 1 and 2) can be used as indicators of profit to extend profitability.

  • Bacteriostatic activity of secretory immumoglobulin A (SIgA) in human and bovine colostrums on enterotoxigenic type Salmonella was tested in the tissue culture medium. SIgA was used in $0.1{\sim}5.0mg/m{\ell}$ concentration with or without the addition of egg lysozyme tested for theirs bacteriostatic activites. 1. Bovine SIgA rich fraction with a large amount of $IgG_1$-dimer could be prepared from bovine colostrum of Holstein cows by anion exchange chromatography using DEAE-Sephadex A-50 and gel filtration on Sephadex G-200 and Sepharose 6B. 2. Human SIgA appeared to be the most bacteriostatic effect for all varieties of Salmonella in a range of $0.5{\sim}1.0mg/m{\ell}$ Bovine SIgA showed a marked bacteriostatic effect increased by increasing the concentration. Bovine IgG had not show bacteriostatic effect against both enterotoxigenic type Salmonella. Egg lysozyme as well as bovine SIgA also showed a marked bacteriostatic effect increased by increasing the concentration. 3. When the growth inhibition of human SIgA was tested by adding egg lysozyme with time interval, egg lysozyme showed bacteriostatic effect as compared with control. But human SIgA and adding with lysozyme showed slight the bacteriostatic effect. 4. When the growth inhibition of bovine SIgA was tested by adding egg lysozyme with time interval, all treatment against S. enteritidis showed bacteriostatic effect as compared with control In the case of S. typhinwrium, egg lysozyme showed a marked bacteriostatic effect as compared with control.

Decrease of Aflatoxin M1 Level in Raw Cow’s Milk using the Hazard Analysis and Critical Control Points (HACCP) System (HACCP 제도에 의한 우유의 아플라톡신 M1의 저감화)

  • Aflatoxin M1 can be produced in cow’s milk when cows eat contaminated produce. Milk is a major source of food for infants and for children who have a weak level of immunity, and the detection of Aflatoxin M1 for risk assessment is necessary in order to reduce the amount of it in milk. In this study, the Aflatoxin M1 level was monitored for one year in raw milk samples obtained from Chungnam Province, Korea. The milk samples were divided into three categories: 1. milk samples from a standard general farm, 2. milk samples from a HACCP controlled farm, and 3. milk samples from the supply of Aflatoxin M1 reduced fodder. The average concentrations of Aflatoxin M1 in milk were 0.023±0.005 ug/l for the standard general farm, 0.017±0.004 ug/l for the HACCP controlled farm, and 0.013±0.003 ug/l for the supply of Aflatoxin M1 reduction fodder. Milk collected from the supply of Aflatoxin M1 reduction fodder had the lowest level of Aflatoxin M1. However, when efficiency and economic aspects are considered the most effective way of reducting Aflatoxin M1, could be taking milk from the HACCP controlled farm and implementing good feed management. Institutional support from the government, careful management of dairy farming, and a strict farm sanitation program are required in order to lower the level of Aflatoxin M1 in milk.

Effect of Byproducts Supplementation by Partically Replacing Soybean Meal to a Total Mixed Ration on Rumen Fermentation Characteristics In Vitro (대두박 대체 부산물 위주의 TMR 사료가 반추위 내 미생물의 In Vitro 발효특성에 미치는 영향)

  • This study was performed to evaluate the effects of replacing basic total mixed ration (TMR) with fermented soybean curd, Artemisia princeps Pampanini cv. Sajabal, and spent coffee grounds by-product on rumen microbial fermentation in vitro. Soybean in the basic TMR diet (control) was replaced by the following 9 treatments (3 replicates): maximum amounts of soybean curd (SC); fermented SC (FSC); 3, 5, and 10% FSC + fermented A. princeps Pampanini cv. Sajabal (1:1, DM basis, FSCS); and 3, 5, 10% FSC + fermented coffee meal (1:1, DM basis, FSCC) of soybean. FSC, FSCS, and FSCC were fermented using Lactobacillus acidophilus ATCC 496, Lactobacillus fermentum ATCC 1493, Lactobacillus plantarum KCTC 1048, and Lactobacillus casei IFO 3533. Replacing dairy cow TMR with FSC treatment led to a pH value of 6 after 8 h of incubation-the lowest value measured (p<0.05), and FSCS and FSCC treatments were higher than SC and FSC treatment after 6 h (p<0.05). Gas production was higher in response to 3% FSC and FSCC treatments than the control after 4-10 h. Dry matter digestibility was increased 0-12 h after FSC treatment (p<0.05) and was the highest after 24 h of 10% FSCS treatment. $NH_3-N$ concentration was the lowest after 24 h of FSC treatment (p<0.05). Microbial protein content increased in response to treatments that had been fermented by the Lactobacillus spp. compared to control and SC treatments (p<0.05). The total concentration of volatile fatty acids (VFAs) was increased after 6-12 h of FSC treatment (p<0.05), while the highest acetate proportion was observed 24 h after 5% and 10% FSCS treatments. The FSC of propionate proportion was increased for 0-10 h compared with among treatments (p<0.05). The highest acetate in the propionate ration was observed after 12 h of SC treatment and the lowest with FSCS 3% treatment after 24 h. Methane ($CH_4$) emulsion was lower with A. princeps Pampanini cv. Sajabal and spent coffee grounds treatments than with the control, SC, and FSC treatments. These experiments were designed to replace the by-products of dairy cow TMR with SC, FSC, FSCS, and FSCC to improve TMR quality. Condensed tannins contained in FSCS and FSCC treatments, which reduced $CH_4$ emulsion in vitro, decreased rumen microbial fermentation during the early incubation time. Therefore, future experiments are required to develop a rumen continuous culture system and an in vivo test to optimize the percentages of FSC, FSCS, and FSCC in the TMR diet of the dairy cows.

A Therapeutic Effect of Ozonated Oil on Bovine Mastitis (젖소 유방염에 대한 Ozonated oil의 치료효과)

  • Forty- nine quarters from 24 lactating cows with chronic mastitis were selected. The cows were raised on dairy farms in Gongju, Jochiwon and Yeongi in Chungnam province, and Iksan in Jeonbuk province, Korea. The 49 quarters with bovine mastitis were divided into control (7 quarters) and experimental (42 quarters) groups. The experimental quarters were assigned to experimental group A (10 quarters, somatic cell count: $50-100\times10^4/ml)$, experimental group B (14 quarters, somatic cells count: $100-300{\times}10^4/ml)$, and experimental group C (18 quarters, somatic cells count: $>300\times10^4/ml$), according to the number of the somatic cells in their milk. The quarters of control group were treated with norfloxacin ointment (10 g/tube) based on the result of sensitivity, twice a day for 3 days. The quarters or experimental groups were infused 10ml or ozonated oils twice a day for 3 days. After treatment, the milk of the control group contained non-significantly lower numbers of somatic cells and bacteria on day 7, compared with pretreatment levels. Experimental groups A, B and C had lower somatic and bacterial cells in their milk on day 7, compared with pretreatment levels. Experimental group B and C had significantly lower numbers of somatic cells in their milk ell day 7 than before treatment (p<0.01). However, no significant difference in somatic cell numbers was detected between the control alld experimental groups. It was concluded that ozone therapy with ozonated oil applied on bovine mastitis might be effective.

Effect of Claw Trimming on Milk Yield and Its Composition in Lactating Dairy Cows (젖소에 있어서 삭제(削蹄)가 유량 및 유성분에 미치는 영향)

  • This study was carried out to investigate effect of claw trimming on milk yield and its composition in Holstein at different lactation stages. 1 . There was no difference in daily milk yield between control and claw trimming in early, mid and late lactating Holsteins. 2. Somatic cell count (SCC) was lower in early lactation and it was higher in late lactation when claws were trimmed in Holstein. However, claw trimming did not affect SCC during mid lactation in Holstein. 3. Milk fat, protein and total solids were decreased during late lactation in Holstein after claw trimming. However, milk composition was not affected by claw trimming in early and mid lactating Holsteins.

Genetic Parameters for Milk Production and Somatic Cell Score of First Lactation in Holstein Cattle with Random Regression Test-Day Models (임의회귀 검정일 모형을 이용한 홀스타인 젖소의 1산차 산유형질 및 체세포지수에 대한 유전모수)

  • The objective of this study was to estimate genetic parameters for test-day milk production and somatic cell score using field data collected by dairy herd improvement program in Korea. Random regression animal models were applied to estimate genetic variances for milk production and somatic cell score. Heritabilities for milk yields, fat percentage, protein percentage, solid-not-fat percentage, and somatic cell score from test day records of 5,796 first lactation Holstein cows were estimated by REML algorithm in single trait random regression test-day animal models. For these analyses, Legendre polynomial covariate function was applied to model the fixed effect of age-season, the additive genetic effect and the permanent environment effect as random. Homogeneous residual variance was assumed to be equal throughout lactation. Heritabilities as a function of time were calculated from the estimated curve parameters from univariate analyses. Heritability estimates for milk yields were in range of 0.13 to 0.29 throughout first lactation. Heritability estimates for fat percentage, protein percentage and solid-not-fat percentage were within 0.09 to 0.11, 0.12 to 0.19 and 0.17 to 0.23, respectively. For somatic cell score, heritabilities were within 0.02 to 0.04. Heritabilities for milk productions and somatic cell score were fluctuated by days in milk with comparing 305d milk production.
