The Biomechanical Characteristics of Isoflavone-Treated Ovariectomized Rat's Femur (이소플라본을 투여한 난소절제 흰쥐 대퇴골의 생체역학적 분석)

  • This study was to investigate the biomechanical characteristics of isoflavone-treated ovariectomized growing rat's femur. 4-week-old Wistar (female rats were randomly divided into 5 groups and assigned to sham(SH) group, and four ovariectomized groups consisting of one ovariectomized(OVX) group and genisetin(10 mg/kg b.w. )(OVX+G), daidzein(10 mg/kg b.w.)(OVX+D), and 17 $\beta$-estradiol(10 $\mu$g/kg b.w.)(OVX+ES) in AIN-76 modified diet for 8 weeks. The OVX groups gained more body weight than the SH group. Femoral weight was increased by feeding genistein and estradiol, whereas femoral length among groups was not significantly different. The breaking farce, stiffness, deformation, and energy values of the OVX group were lower than those of other groups. The stress, strain, and Young's modulus values of the OVX group were higher than those of other groups.

  • Soy is an important food in Asia and many studies have suggested that the low incidences of chronic diseases in Asian countries are associated with diets that are rich in soy. Soy contains many kinds of phytochemicals, and soy isoflavones and soyasaponins have received considerable attention. Twelve isoflavone components have been isolated from soy: three aglycones (daidzein, genistein, and glycitein), and their respective nine glucosidic conjugates. Soy isoflavones are similar in structure to estrogen and exhibit both estrogenic and antiestrogenic activities. Soy isoflavones exhibit anticancer activity, can reduce the risk of cardiovascular disease, and are beneficial to brain and bone health. Soyasaponins are divided into three groups (A, B, and E saponins), and they exhibit hypocholesterolemic, anticancer, hepatoprotective, antioxidative, and anti-human-immunodeficiency-virus effects. Despite the abundant literature suggesting that soy isoflavones and soyasaponins have potential applications in preventive medicine, further research is needed to standardize dosages and ensure their efficacy.

  • Soybeans contain the phytoestrogens genistein and daidzein, their glucosides genistin and daidziin and coumesterol. These isflavonoid compounds are capable of producing an estrogenic response in a number of diverse species. This study determined optimum conditions for extraction of isoflavones in defatted soybean meal. Extraction of isoflavones was conducted at various conditions such as difference concentrations of extraction solvent, temperature, time and pH to extact. The most optimum extraction conditions was achieved at 75% ethanol, $80^{\circ}C$, pH4 and extract for three hours. In addition. isoflavones with high purity were separated by adding up to 4%(w/v) of calcium chloride dihydrate.

Effects of Germinated Soybean Powder Addition on Isoflavone Contents and Characteristics of Injulmi (발아콩가루 첨가가 인절미의 이소플라본 함량 및 특성에 미치는 영향)

  • The effects of the addition of germinated soybean powder on Injulmi characteristics were investigated in this study. Injulmi was prepared with five different levels of germinated soybean powder (0%, 4%, 8%, 12% and 16%)and the physical properties were examined. The moisture content decreased, while the protein, fat and ash contents increased With increasing germinated soybean powder content. Total isoflavone contents, 3.39${\sim}$15.35 mg% before cooking, slightly increased to 3.94${\sim}$15.96 mg% after cooking. Genistin, genistein, daidzin and daidzein, which are blown to be the major functional isoflavones, accounted for approximately 93% of total isoflavone contents. The color of Injulmi darkened slightly and became greenish yellow with the addition of germinated soybean powder. Textural profile analysis showed that hardness and gumminess increased, but cohesiveness and springiness decreased with increasing germinated soybean powder level. Sensory test showed that Injulmi prepared with up to 8% added germinated soybean powder was evaluated as being equally acceptable as Injulmi without germinated soybean powder.

Composition and Antioxidative Characteristics of Phenolic Fraction Isolated from Soybean Fermented Food (콩 발효식품으로부터 분리한 페놀물질획분의 성분분석과 항산화 작용 특성)

  • Doenjang(fermented soybean paste) was prepared by the series of processes including soaking, cooking, first fermentation(3 days at 30$\pm$2$^{\circ}C$) for the preparation of meju(soybean koji) after inoclulation of Aspergillus oryzae, and further fermentation (60 days at 30$\pm$2$^{\circ}C$) for the ripening after addition of salt 13% to meju. the crude phenolics extracted from defatted soybean and doenjang were fractionated onto the neutral phenolics(isofavonoids) and acidic phenolics(phenolic acids), respectively. Composition and antioxidative characteristics of phenolic fractions were determined. The neutral phenolic fractions contained genistin, genistein and daidzein; on the other hand, acidic phenolic fractions had syringic acid and seven other components. The content of genistin in doenjang dramatically decreased at the early stage of fermentation, whereas the content of genistein rather increased. In addition, the content of syringic acid of acidic phenolic fractions were increased during fermentation. These changes in individual phenolic components affected the antioxidative activity of neutral phenolics or acidic phenolics. antioxidative activity of phenolic compounds were evaluated during soybean fermentation. The antioxidative and free radical scavenging activity of neutral phenolic fractions and acidic phenolic fractions on linoleic acid autoxidation were also investigated.

Estimated dietary isoflavone intake among Korean adults

  • This study estimated the isoflavone intake level in Koreans using Food Frequency Questionnaire and analyzed related variables. The results showed that the average daily intake of isoflavone in adults was shown as 23.1 mg. The isoflavone intake level at 50 percentile was 16.9 mg ($0{\sim}90\;mg$), and 10% of adults took almost 50 mg of isoflavone a day and 10% took about 5 mg a day. The major food sources for isoflavone in Koreans were in the order of soybean, soybean paste, soy milk, soybean curd (tofu), and bean sprouts; the intake was different depending on age, educational background, occupation, economic standard, and family type. The result showed higher isoflavone intake levels in the group over 30 years old and the highest isoflavone intake in subjects working in farming/fishery, followed by housemakers. According to the differences by families the families with elderly members showed 50% higher isoflavone intake than young families with friends or siblings. Depending on related ecological variables, therefore, various nutrition education programs should be developed for a variety of intakes of soybean foods, along with easy and simple cooking methods as parts of continuous research.

Antiproliferative Constituents from the Vinegar Treated Small Black Soybean (Glycine max Merr.)

  • The chloroform-soluble fraction of the vinegar treated small black soybean [Glycine max (Leguminosae)] showed antiproliferative activity against human myeloid leukemia HL-60 cells, in terms of inhibition of proliferation and induction of apoptosis. Bioassay-guided chromatography of the chloroform-soluble fraction resulted in the isolation of two isoflavonoid compounds, genistein and daidzein, as active principles. Genistein showed more potent antiproliferative effects against HL-60 cells. Treatment of HL-60 cells with genistein induced apoptosis in a dose dependent manner. Apoptosis was judged by the detection of DNA fragmentation by a flow cytometry and the degree of apoptosis was assayed by RT-PCR.

Quantitative Analysis of Isoflavones and Lignans in Sea Vegetables Consumed in Korea Using Isotope Dilution Gas Chromatography-Mass Spectrometry

  • The phytoestrogens including isoflavones (genistein, daidzein, biochanin A, formononetin, and glycitein), coumestrol, and lignans (secoisolariciresinol, matairesinol, and anhydrosecoisolariciresinol) were quantified in edible sea vegetables from Korea. Sea vegetable samples were collected based on domestic consumption data. After hydrolysis of phytoestrogen glycosides in prepared samples, aglycones of phytoestrogens were extracted with diethyl ether and analyzed with isotope dilution gas chromatography-mass spectrometry in selected ion monitoring mode (ID-GC-MS-SIM). Total samples included 19 samples representing eight species. Most of the samples showed rather low concentrations, ranging from not determinated to $79.2\;{\mu}g/kg$ for isoflavones and from 106.4 to $694.8\;{\mu}g/kg$ for lignans. The daily intake of phytoestrogen from sea vegetables, estimated from the present data and domestic consumption data, was about $0.13\;{\mu}g/day$ for isoflavones and $2.0\;{\mu}g/day$ for lignans. When we compared these results with those from legumes, sea vegetables would not be considered the major source of phytoestrogens in the Korean diet.

Antimicrobial Compounds Profile During Cheonggukjang Fermentation Against Xanthomonas oryzae pv. oryzae (Xoo)

  • Xanthomonas oryzae causes rice bacterial blight, which has been reported as one of the most destructive diseases of rice. Metabolites were identified through cheonggukjang, a traditional Korean fermented soybean product fermented by the Bacillus spp., to control the bacteria. HPLC, MS, and UPLC-Q-TOF-MS analyses were performed to identify metabolites responsible for antimicrobial activity. In this analysis, the m/z values of 253.0498, 283.0600, 269.0455, 992.6287, and 1,006.6436 were identified as daidzein, glycitein, genistein, surfactin B, and surfactin A, respectively. The levels of surfactin B and surfactin A were found to be high at 24 h (4.35 ${\mu}g$/ml) and 36 h (3.43 ${\mu}g$/ml) of fermentation, respectively.

Antioxidant and Anti-Diabetic Activities of Soybean Fermented with Monascus (홍국발효 대두의 항산화 및 항당뇨 활성)

  • This study was performed to investigate the antioxidant and antidiabetic activities of soybean fermented with Monascus. Also, the changes in the content of isoflavones and Monacolin K were analyzed. It was observed that the glycoside forms of daidzin and genistin were converted to aglycones of daidzein and genistein within 6 days of fermentation. The product can be used as a health functional material that can increase bioavailability. Monacolin K production was found to increase significantly with the progression of fermentation with an increase to 0.04 mg/g and 0.44 mg/g on 6 and 12 days of fermentation, respectively. The DPPH radical scavenging activity of soybean fermented with Monascus was significantly increased compared to that of soybean. The protein expression of inflammation-related genes (TNF-α, IL-6, and COX-2) in the MIN cell was significantly increased in the presence of alloxan compared to the normal group, but a decrease was observed in the presence of soybean fermented with Monascus. In conclusion, soybean fermented with Monascus showed the highest antidiabetic and antioxidant effects. These results suggest that soybean fermented with Monascus has the potential to be used as a beneficial ingredient with antidiabetic and antioxidant effects.
