Hepatoprotective Effects of Oyster Hydrolysate on Lipopolysaccharide/D-Galactosamine-Induced Acute Liver Injury in Mice (Lipopolysaccharide/D-Galactosamine에 의해 유도된 급성 간 손상 생쥐모델에서 굴가수분해물의 간 보호 효과)

  • Oxidative stress and inflammation are key factors responsible for progression of liver injury. A variety of functions of oyster hydrolysate (OH) are affected by their antioxidant and anti-inflammatory activities. However, little is known regarding the effects of OH on a liver injury model. This study was performed to evaluate the effects of OH on acute liver injury induced by lipopolysaccharide/D-galactosamine (LPS/D-GalN) in mice. Experimental groups were divided into six groups as follows (each group, n=10): control (saline), LPS/D-GalN, LPS/D-GalN+OH (100 mg/kg), LPS/D-GalN+OH (200 mg/kg), LPS/D-GalN+OH (400 mg/kg), and LPS/D-GalN+silymarin (25 mg/kg, positive control). The experimental acute liver injury model was induced with LPS ($1{\mu}g/kg$) and D-GalN (400 mg/kg). We first analyzed antioxidant and anti-inflammatory activities in OH. OH showed high DPPH and ABTS radical scavenging activities and reduced ROS generation in Chang cells in a dose-dependent manner. In addition, OH showed anti-inflammatory activities, such as inhibition of cyclooxygenase-2 and 5-lipooxygenase. Treatment with OH down-regulated tumor necrosis factor $(TNF)-{\alpha}$, interleukin (IL)-6, and $IL-1{\alpha}$ expression levels in LPS-stimulated RAW264.7 cells. OH significantly reduced LPS/D-GalN-induced increases in the concentrations of alanine transaminase and aspartate aminotransferase in serum. In the LPS/D-GalN group, liver tissues exhibited apoptosis of hepatocytes with hemorrhages. These pathological alterations were ameliorated by OH treatment. Consistently, hepatic catalase activity was low in the LPS/D-GalN group compared to the control group, and catalase activity was significantly restored by OH treatment (P<0.05). Furthermore, OH markedly reduced the LPS/D-GalN-induced increase in $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 levels in liver tissue. Taken together, these results show that OH has hepatoprotective effects on LPS/D-GalN-induced acute liver injury via inhibition of oxidative stress and inflammation, suggesting that OH could be used as a health functional food and potential therapeutic agent for acute liver injury.

Effects of Different Dietary Oil and d-Limonene on Histopathological and Biochemical Changes in Experimental Hepatocarcinogenesis (식이지방의 종류 및 d-Limonene 투여가 간 발암과정에 미치는 영향)

  • The purpose of this study was to investigate the effcts of n-3, n-6 fatty arid and d-limonene on histopathological and biochemical changes in experimental rat hepatocarcinogenesis. To attain the above objectives, weanling Sprague-Dawley female rats were intraperitoneally injected twice with a dose of diethylnitrosamine(DEN, 50mg/kg body weight) and after 1 week 0.05% phenobarbital was provided with water. Sardine oil rich in n-3 fatty acids and corn oil rich in n-6 fatty acids were fed at 15% by weight and 5% d-limonene was added to the diet in each group. Ten weeks or 20 weeks after DEN treatment, rats were sacrifirced. The formation of glutathione S-transferase placental form positive(GST-P$\^$+/) foci was significantly decreased by the treatment of either sardine oil or d-limonene HMG-CoA reductase activity was not affected by dietary oils and d-limonene. Protein kinase C (PKC) activity was decreased by either sardine oil or d-limonene. Particularly d-limonene decreased the membrane PKC activity. Membrane Cholesterol/Phospholipid(Chol/PL) ratio was significantly decreased by d-limonene in sardine oil group. The data showed that GST-P$\^$+/ foci number was positively correlated with membrane PKC activity and serum cholesterol and negatively correlated with liver cholesterol level. These results suggest informations about the correlation between histopathological and biochemical changes such as cholesterol metabolism and PKC activity in experimental hepatocarcinogenesis and thereby can elucidate the possible mechanism related to the cancer inhibition.(Korean J Nutrition 33(1) : 23-32, 2000)

  • As technology advances, there is a demand for development of hard, solid lubricant coating. (Ti,Cr)N-$MoS_2$, films were deposited on SKD 11 tool steel substrate by co-deposition of $MoS_2$, with (Ti,Cr)N using a D.C. magnetron sputtering process. The influence of the $N_2/Ar$ gas ratio, the deposition temperature, the amount of $MoS_2$ in the films, and the bias voltage on the mechanical and the structural properties of the films were investigated. Wear tests were performed on the films deposited in various conditions.

Effects of Restricted Feeding on Intake, Digestion, Nitrogen Balance and Metabolizable Energy in Small and Large Body Sized Sheep Breeds

  • Ninety six intact male sheep (12 months old with mean live weight of about 35 kg) were used to assess the effects of restricted feeding on intake, digestion, nitrogen balance and metabolizable energy (ME). The animals were selected from two known Iranian small and large body size breeds: 48 Sangsari (S) and 48 Afshari (A), and were divided into two equal groups: restricted (R) and a control (C). Each group had 48 sheep (24 each breed). The experiment had a duration of 15 and 75 days adaptation and treatment periods, respectively. The animals were individually placed in metabolism cages and fed a diet based on pelleted concentrate mixture consisting of alfalfa, barley grain, cottonseed meal and barley straw. The animals in group C were fed ad libitum, while animals in group R were fed at maintenance level and maintained a relatively constant live weight. During the experiment, the average daily weight gain (ADG) of S and A animals in R group was 0.34 and -0.25 g/d (0.02 and -0.02 $g/kg^{0.75}/d$), respectively. While that of S and A animals in C group was 174.4 and 194.4 g/d (10.16 and 11.48 $g/kg^{0.75}/d$), respectively. Nitrogen (N) was determined by both measured and regression methods. Animals of R group stayed at about zero N balance (0.01 and -0.00 g $N/kg^{0.75}/d$ for S and A animals, respectively). The N retention of animals of both S and A breeds in C group were similar (0.45 and 0.46 g $N/kg^{0.75}/d$, respectively). Digestible organic matter intake (DOMI) and ME requirement for maintenance (MEm) were measured by both constant weight technique and regression method by regressing N balance on DOMI and ME intake on ADG. The measured DOMI during constant weight was 24.61 and 24.27 g $DOMI/kg^{0.75}/d$ and the calculated DOMI from regression equation was 24.24 and 24.22 g $DOMI/kg^{0.75}/d$, for S and A animals, respectively. The measured MEm was 402 and 401 kJ $ME/kg^{0.75}/d$ and the calculated MEm from regression analysis was 398 and 400 kJ $ME/kg^{0.75}/d$ for S and A breeds, respectively. There were no significant differences between both measured and regression techniques. There was no significant difference between S and A breeds for DOMI, N retention, MEm, digestibility and metabolizability values. Digestibility values for OM, GE and CP and metabolizability were significantly (p<0.05) higher in restricted feeding sheep compared with that of sheep fed ad libitum.

  • In halobacterial membrane, pharaonis phoborhodopsin (or pharaonis sensory rhdopsin II, psRII) forms a complex with its transducer pHtrII. Flash-photolyis of D75N mutant did not yield M-intermediate but an O-like intermediate is observed. We examined the interaction between D75N of ppR and t-Htr (truncated pHtrII). These formed a complex in the presence of n-dodecyl-$\beta$-D-maltoside, and the association accelerated the decay of the 0 of D75N from 15 to 56 s$\^$-1/. From the decay time constants under varying ratios of D75N and t-Htr, n, the molar ratio of D75N/t-Htr in the complex, and K$\_$D/, the dissociation constant, were estimated. The value of n was unity and K$\_$D/ was estimated to 146 nM. This K$\_$D/ value can be considered as the association between the photo-intermediate and t-Htr, which is deduced by the method of estimation. Previously we (Photochem. Photobiol. 74, 489-494 (2001)) reported K$\_$D/ of 15 $\mu$M for the interaction between the wild-type and t-Htr by means of the change of M-decay rates. Therefore, this value should be the K$\_$D/ value for the interaction between M of the wild-type and t-Htr.

Development of Marine Casualty Forecasting System (I). Construction and Analysis of Marine Casualty Numerical D/B (해양사고 예보 시스템 개발(I): 해양사고 수량화 D/B구축과 분석)

  • The paper describes on the construction and analysis of marine casualty numerical D/B (N-D/B) to implement Korean MArine Casualty Forecasting System (K-MACFOS). The main target of K-MACFOS is to broadcast the prediction number and risk level of marine casualties as like daily weather forecasting. The data relating to a total of 724 ship casualties in the west-southern sea area (33oN∼35oN, 124oE∼127oE) of Korean peninsula for 11 years (1990∼2000) have been compiled and converted into quantitative data with 14 numeric conversion scales. Through the statistical analysis using contour-map visualization, the usability of N-D/B and the casualty features of the target sea areas are discussed. In addition, the optimum year-band selection method is also proposed to provide correct N-D/B analysis and precise prediction of the number of marine casualties.

The Design of a Wideband 3 dB Quadrature Coupler using N-Section Parallel-Coupled Lines (N단 평행 결합 선로를 이용한 90° 광대역 3 dB 결합기 설계)

  • In this paper, we proposed a 3 dB coupler using N-section parallel-coupled lines and designed a very compact one based on the analysis results. The coupled line has been analyzed by spectral domain method. After we obtain the s-parameters of N-section parallel-coupled lines by using port reduction method 4-port s-parameters are derived. The 3 dB couplers, which were fabricated, are not necessary to implement high impedance lines and tight coupling gaps as Lange Couplers because loose coupling is used. To realize a minimum section, we used the PCB that has high a dielectric constant and a thickness. The experimental results show that it has wide bandwidth of about 42 %(0.5 dB unbalance) from 3.6 GHz to 5.5 GHz and phase difference within 1 degree. Also, The isolation characteristics about 15 dB at its pass-band are obtained.

  • Vanilloid receptor I (VR1) is a nonselective cation ion channel placed in the plasma membrane of peripheral sensory neurons that is potential target for analgesia A series of N-4-substituted-benzyl-N'-tert-butylbenzyl thioureas were prepared for the study of their agonistic/antagonistic activities to the vanilloid receptor in rat DRG. Their structure-activity relationship in reveals that not only the two oxygens and amide hydrogen of sulfonamido group but also the optimal size of methyl in methanesulfonamido group play an integral role for the antagonistic activity on vanilloid receptor.

  • In [2] and [3], the Shinwon polynomial S$_{n}$(x) of order n is defined and studied in some details. In this paper we will define the general Shinwon polynomial S$_{n}$(a,x) and the Dickson polynomial D$_{n}$(a,x) of the second kind of order n which is a slightly changed form of the Dickson polynomial g$_{n}$(a,x), and show that D$_{n}$(a,x) is closely related to S$_{n}$(a,x).EX> n/(a,x).
