  • The content of two steroids-campesterol (1) and ${\beta}$-sitosterol (2), and four triterpenes-taraxetrone (5), ${\beta}$-amyrin (6), (-)-friedelin (7), glutinol (8) in the Orostachys japonicus A. Berger cultivated under various conditions was estimated and compared with those in wild one. The present investigation disclosed that there are no significant difference in their contents between cultivated Orostachys japonicus A. Berger and wild one. from viewpoint of the content of the steroids 1 and 2, and the triterpenes 5-8, the quality of cultivated Orostachys japonicus A. Berger is not inferior to the wild one.

Total Phenol Contents and Antioxidant Activity in Orostachys japonicus A. Berger Grown under various Cultivation Conditions (재배 조건에 따른 바위솔의 총페놀함량 및 항산화효과)

  • Changes in the contents of total phenolic compounds in as Orostachys japonicus well as their antioxidant capacity according to the control of night-break, day-length control, variation in luminosity and amount of fertilizer used were examined. The results were compared with those obtained from wild Orostachys japonicus. The contents of the total phenolic compounds in Orostachys japonicus were roughly proportional to the increasing level of irradiation and nitrogen fertilization. The total content of phenolic compounds in the wild Orostachys japonicus of was higher than cultivated under various conditions. The antioxidant capacities of the Orostachys japonicus were increased roughly with increasing level of irradiation and nitrogen fertilization. Overall, the total content of phenolic compounds of Orostachys japonicus and their antioxidant capacity were not significantly affected by the amount of phosphorus and potassium fertilizer used.

  • The herbs of Orostachys japonicus (Crassulaceae) have been used to treat gastric cancer, gastric ulcer or hemorrhage. Flavonoid glycosides, mainly kaempferol (Kp)- and quercetin (Qc) glycosides, have been isolated from O. japonicus; however, no quantitative information on those flavonol glycosides and no peroxynitritescavenging activity of the Orostachys extracts have been reported. In this study, Kp- and Qc glycosides were qualitatively and quantitatively analyzed by high-performance liquid chromatography (HPLC) in eight Orostachys and a Meterostachys species including O. japonicas, O. margaritifolius, O. chongsunensis, O. minuta, O. ramosus, O. malacophylla, O. latiellipticus, O. iwarenge, O. iwarenge for. magnus, and Meterostachys sikokiana distributed or cultivated in Korea. Distinctively, O. margaritifolius contained two flavonol 3,7-di-O-glycosides of Kp 3,7-di-O-glucoside and Kp 3-rhamnosyl-7-glucoside, but O. japonicus had two flavonol 3-O-rutinosides, Kp 3-rutinoside and Qc 3-rutinoside. The three species of O. margaritifolius (24.36 mg/g MeOH extract), O. japonicus (21.28 mg/g), and O. minuta (19.50 mg/g) showed relatively higher flavonoid contents. The flavonol glycosides were analyzed using eight standard compounds (Kp, Qc, Qc 3-O-rhamnoside, Qc 3-O-glucoside, Kp 3- O-rutinoside, Qc 3-O-rutinoside, Kp 3-O-rhamnosyl-7-O-glucoside, Kp 3,7-di-O-glucoside). The present HPLC method was validated to verify the linearity, precision, and accuracy. In addition, the peroxynitrite-scavenging activity was also discussed.

Effects of Growth Regulators on Shoot Regeneration and Polysaccharide Production of Orostachys japonicus Berger

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. O. japonicus was cultured in media containing various growth regulators and carbon sources. The highest regeneration rate was achieved in 1.0 and $3.0\;mg\;l\;^{-1}$ of 2,4-D concentration, while the lowest was obtained in $10.0\;mg\;l\;^{-1}$ 2,4-D concentration. When different carbone sources were added in the culture medium, plant growth was high in 3% sucrose treatment. The micropropagated shoots were successfully acclimatized in artificial soils and produced comparable amont of polysaccharide compred to parent cultivated plants.

Anticancer Effects of Cultivated Orostachys japonicus on Human Prostate Cancer Cells (인체 전립선 암세포에서 재배 와송의 항암효과)

  • This study was performed to determine the anticancer effects of cultivated Orostachys japonicus (COJ) and wild Orostachys japonicus (WOJ) on primary human prostate cancer cells (RC-58T/h/SA#4 cells). The morphology of cells treated with COJ and WOJ was distorted to shrunken cell masses. In addition, cell death induced by COJ and WOJ was associated with increased population of cells in sub-G1 phase as well as the formation of apoptotic bodies and nuclear condensation. COD and WOJ markedly reduced the number of viable prostate cancer cells in a dose-dependent manner, and cell numbers were lower than control cells. COJ and WOJ also inhibited increases in cell proliferation induced by environmental hormones such as dioxin and bisphenol A in charcoal-treated FBS (cFBS) medium. COJ and WOJ methanol extracts at the tested concentrations (150, 300, and 600 ${\mu}g/mL$) also dose-dependently inhibited cell proliferation induced by environmental hormones. These results indicate that COJ and WOJ exert anticancer effects on primary human prostate cancer cells.

Cultivated Orostachys japonicus Induces Apoptosis in Human Colon Cancer Cells (인체 대장암 세포주 SW480에서 재배 와송의 세포 사멸 유도 효과)

  • This study was performed to elucidate the anticancer activities and the mechanism of chloroform fractions from cultivated Orostachys japonicus (CFCOJ) in human colon cancer cells. CFCOJ markedly decreased viable cell numbers in both a dose-dependent and time-dependent manner within SW480 cells. Cell death induced by CFCOJ increased cell populations in the sub-G1 phase, as well as the formation of apoptotic bodies, nuclear condensation, and induced DNA fragmentation. CFCOJ-induced apoptosis was associated with the activation of initiator caspase-8 and -9, as well as the effector caspase-3. CFCOJ also stimulated Bid cleavage, indicating that the apoptotic action of caspase-8-mediated Bid cleavage leads to the activation of caspase-9. CFCOJ increased the expression of the proapoptotic protein, Bax, and decreased the expression of the antiapoptotic protein, Bcl-2. These results indicate that CFCOJ exert anticancer effects on human colon cancer SW480 cells through a caspase-dependent apoptotic pathway.

Effect of Long-day and Night-break Treatment on Growth and Anthesis of Orostachys japonicus A. Berger (장일과 암기중단 처리가 바위솔의 생장과 개화에 미치는 영향)

  • Orostachys japonicus, called Wasong and used as anti-tumor medicinal plant, was cultivated in plastic house. The experiment was done to clarify the effect of long-day and night-break treatment at the timing of bolting on its morphological characters, organ dry weight and flowering of florets. After grown in 15cm plastic boxes containing 2:1 soil:peat moss mixture for about 4 months, long-day of 16 hours and night-break of 2 hours around midnight were treated from Sept. 9. The plants were sampled 5 times at 2-week interval after the treatments. Long-day and night-break treatment delayed the growth of inflorescence and showed greater stem diameter on the last sampling and no. of leaves and bracts than the natural daylength. The treatments also had greater leaf and bract dry weight since 2 weeks, and the other fraction and total dry weights since 4 weeks but less floret dry weight from 4 to 6 weeks after the treatments than the natural daylength. The treatments, however, decreased no. of flowered florets and ratio of flowering plants although all the treatments showed nearly the same no. of total florets per plant until 6 weeks after the treatments, late October, which resulted in the modification of source to sink or vice versa. In the natural daylength, the florets were functioned as sink, while root, leaf and bract as source, but in the long-day and night-break treatments stem and florets were done as sink.

Anti-cancer Effects of Cultivated Orostachys japonicus on Human Colon Cancer Cell Line SW480 (인체대장암세포주 SW480에 대한 재배 와송의 항암효과 연구)

  • Orostachys japonicus (OJ) is a medicinal herb with immunoregulatory, anti-aging, anti-oxidative, and many other therapeutic properties. The purpose of this study was to elucidate the anti-cancer property of cultivated OJ. SW480 cell viability was significantly reduced by cumulative exposure to OJ extract. We also observed inhibitory effects of OJ after 72 hr through the growth and migration of SW480 cells using scratch assay. SW480 cells in OJ-free medium began to move into the scratch site at 24 hr; however, cells in medium containing OJ did not migrate into the scratch site until 48 hr. Male C57BL/6 mice (4 weeks old) were orally administered OJ extract for 31 days before injection of SW480 cells. At 7, 14, and 28 days after subcutaneous injection of SW480 cells, tumor weight and volume were analyzed. The body weight of the OJ-treated group was continuously increased during administration of the OJ extract relative to the control group. Injection of SW480 cells caused a reduction in body weight in all groups; however, the OJ-treated group exhibited a significant increase in body weight after 14 days. Tumor weight and volume were lower in the OJ-treated group than in the control group after 28 days. Although these results suggest that OJ suppresses the proliferation and migration of human colon cancer cells, additional studies are required to provide preclinical evidence before launching clinical trials evaluating OJ as an anti-cancer biohealth product.

Effect of Night-break Timing on Growth, Bolting and Anthesis of Orostachys japonicus (암기중단 처리시기에 따른 바위솔의 생장, 추대 및 개화)

  • Orostachys japonicus, Wasong as herb medicine, has been artificially cultivated as an anti-tumor medicinal. The experiment was carried out to examine the effect of natural daylength as control or night-break treatment (NB) imposed at June 20, July 18 or Aug. 15 on its growth, dry weights of leaf and bract, stem, floret and root, and morphological characters including bolting and floret flowering. After a plant was grown in a 15cm plastic pot containing a 2 : 1 soil : peat moss mixture on May 23, three treatments with above differing night-break had been imposed around midnight up to Nov. 7. The plants were sampled 3 times at the same day forced to night-break and then done 6 times by 2-week interval after the final NB. Plant height and inflorescence length of all the NB increased with delayed NB but declined in comparison with the natural daylength. No. of leaves including bracts showed similar response to plant height although NB given before July 18 showed less leaves and bracts. Stem diameters of NB were continuously increased to middle Sept. to middle Oct. while that of natural daylength decreased after middle Oct. Natural daylength or NB given on Aug. 15 had greater fraction, shoot and total dry weights resulting from increment of leaf and bract up to Aug. or of floret, stem and root after Sept. The earlier NB, the later formation of florets and the less number of flowering florets whereas in natural daylength florets on inflorescence begun to be formed from middle Sept. were sharply increased up to middle Oct. when all the plants were flowered. Bolting was not formed in the plant of the earliest NB of June 20, and thereby no anthesis of florets up to early Nov. It was concluded that year-round cultivation of Orostachys japonicus plants was possible through controlling the NB timing because its bolting and flowering of florets separately occurred.

  • Orostachys japonjcus, called Wasong in herb medicine, has been artificially cultivated as an anti-tumor medicinal. The experiment was done to examine the effect of night-break periods imposed immediately before its bolting time on its morphological, flowering-related characters and fraction dry weights. After a plant was grown in a 15cm plastic pot containing a 2:1 soil:Peat moss mixture for about 3 months, three different night-break periods (0.5, 1 and 2 hours) around midnight were treated from Aug. 24. to compare with the natural daylength. The plants were sampled 6 times by 2-week interval after the treatments. Plant height and inflorescence length of all the treatments inclined with time lapse after the treatment and were shorter in 2 hour night-break due to slow increment than in the other treatments, while stem diameter showed reverse result. All the treatments except 2 hour night -break were nearly same in fraction, shoot and total dry weights per plant; two hour night-break treatment had greater leaf and bract weight from 6 weeks, greater stem, shoot and total weights from 2 weeks and greater root weight from 4 weeks but did less floret weight after the treatment. Although florets on the inflorescence were formed in 2 hour night-break treatment, flowering florets and flowering plants never occurred. In the other treatments showed the similar response, however, more florets appeared from 2 weeks, flowering florets was sharply increased from 6 weeks and flowering plants were reached up to 100% from 6 to 8 weeks after the treatment. Inflorescence length or number of total florets per plant in 2 hour night-break was positive-correlated to all the fraction dry weights except that those of natural daylength was not done, meaning that its artificial cultivation should permit bolting to secure more shoot dry matter.
