  • Water-soluble crude polysaccharide (TM-1) prepared from the leaves of Teucrium viscidum var. miquelianum was fractionated into three polysaccharide fractions, TM-2, TM-3 and TM-4 by the addition of cetyltrimethylammonium bromide. The major polysaccharide fraction, TM-2, consisted of glucose, galactose, rhamnose, mannose, glucuronic acid and galacturonic acid, and all fractions contained galactose and glutose as the major neutral sugars. TM-4 showed the highest anti-complementary activity. When TM-4 was futher fractionated by anion exchange chromatography, TM4- II a and TM4-II b showed the most Potent anti-complementary activity. TM4- II a was composed mainly of galactose, arabinose and glucose in the molar ratios of 2.13 : 0.94 : 1.00 respectively, and contained a small amount of galacturonic acid and glucuronic acid.

A comparative study of the immuno-modulatory activities of ethanol extracts and crude polysaccharide fractions from Annona muricata L. (가시여지잎(Annona muricata L.) 에탄올 추출물과 조다당 분획분의 면역활성 비교)

  • This study compared the immuno-modulatory effects of ethanol extracts (A. muricata L. ethanol extracts, ALE) and crude polysaccharide fraction (A. muricata L. crude polysaccharide fraction, ALP) from Annona muricata L. in macrophages. Immuno-modulatory activity was determined by assessing cell viability, nitric oxide (NO) production, inducible NO synthase (iNOS) expression and cytokine production in RAW 264.7 a macrophage cell line. Both ALE and ALP treatment did not affect cytotoxicity, and ALP treatment significantly increased NO production. Additionally, cytokine production [tumor necrosis factor ($TNF-{\alpha}$; $2909.04{\pm}4.1pg/mL$), interleukin (IL)-6; $662.84{\pm}5.3pg/mL$, and $IL-1{\beta}$; $852.37{\pm}2.2pg/mL$), was highly increased in the ALP ($250{\mu}g/mL$) treated group compared to the ALE ($250{\mu}g/mL$) treated group ($TNF-{\alpha}$; $1564.50{\pm}6.1pg/mL$, IL-6; $517.24{\pm}4.1pg/mL$ and $IL-1{\beta}$; $237.23{\pm}1.8pg/mL$). Moreover, ALP treatment considerably increased the expression of mitogen-activated protein kinase (MAPKs) and nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) in the macrophages. Therefore, ALP can induce macrophage activation through MAPK and $NF-{\kappa}B$ signaling and this can be a potential candidate for development of nutraceuticals.

Chemical Properties and Assessment of Immunomodulatory Activities of Extracts isolated from Broccoli (브로콜리로부터 분리한 추출물의 In vitro 면역증진 활성평가 및 화학적 특성)

  • For the purpose of developing new immunomodulatory agents from broccoli, ethanol extract (BCEE), hot water extract (BCHW), and crude polysaccharide (BCCP) were isolated from broccoli, and their immunomodulatory activities and chemical properties were examined. In the in vitro cytotoxicity analysis, BCHW and BCCP did not affect the growth of tumor cells and normal cells. Murine peritoneal macrophages stimulated with BCCP showed higher production of IL-6, IL-12, and $TNF-{\alpha}$ cytokines than those stimulated with BCHW. Also, BCHW and BCCP did not show proliferation of splenic lymphocytes. In the in vitro assay for intestinal immunomodulatory activities, only BCCP enhanced GM-CSF secretion and the bone marrow cell-proliferating activity via cells in Peyer's patches at $1,000{\mu}g/mL$. Also, BCHW mainly contained 33.7% neutral sugars, such as arabinose, glucose, and galactose, and 30.7% uronic acid, and BCCP consisted of 42.6% neutral sugars, including arabinose, galactose, and glucose, and 50.5% uronic acid. The above results lead us to conclude that crude polysaccharide (BCCP) isolated from broccoli causes considerably high cytokine production in peritoneal macrophages and bone marrow cell proliferation, and the polysaccharide extraction process is indispensable for separation of new immunomodulatory agents from broccoli.

Extraction Characteristics of Polysaccharide from Fomitopsis pinicola Jeseng Mushroom (소나무 재생버섯(Fomitopsis pinicola Jeseng) 다당류의 추출 특성)

  • This study was conducted to investigate the extraction characteristics of the polysaccharide from Jeseng mushroom (Fomitopsis pinicola Jeseng). Yields of the polysaccharide extracted from powdered mushroom by autoclaving(120, 30 min) with water at different pH and salt concentration were 8.2~9.2% in pH 5~11, 4.7~5.5% in 1~5% salt solution, respectively. The yield by the 0.05~1.0 N KOH-extraction was ranged 3.45~13.20%, while that by HAS-extraction(homogenizing after KOH swelling) using 1~2.5 N KOH 73.6~78.4%. Content of carbohydrate, protein, lipid and ash of the crude polysaccharide extracted from fruits body and its cultured mycelium by method of water extraction, KOH extraction(0.005~1N) and HAS-extraction were ranged 86.5~92.6%, 2.3~13.1%, 0.1~4.2% and 0.1~1.7%, respectively. The polysaccharide were composed of 62.0~77.8 g/g of pentose, 138.0~187.8 g/g of hexose and 21.2~117.3 mg/g of protein. From these results, the polysaccharide extracted was supposed to be a protein-bound polysaccharide.

  • The enzyme-linked immunosorbent assay (ELISA)-inhibition test using a Clonorchis sinensis species-specific mouse monoclonal antibody (MAb) , CsHyb 0605-23, showed increased specificity over the conventional ELISA used for serodiagnosis of clonorchiasis. To characterize the corresponding antigen further, the MAb was tested against polysaccharide, protein and glycolipid fractions obtained from a crude extract of C. slnensis adult worms, using chloroform, methanol and phenol extractions. Only the polysaccharide fraction was recognized by the mb among those fractions. Mild oxidation of the antigen with sodium periodate showed decreased reactivity against the MAb. We concluded that the antigen and antigenic determinants recognized by the MAb are carbohydrates.

  • In order to investigate the antitumor components of Korean wild higher fungi, the carpo-phores of Pholiota squarrosa collected in Kaung Nung area were extracted with water by refluxing for eight hours at 80-90.deg. C. The extracts were dialized for six days against distilled water and lyophilized to produce crude polysaccharide powders. They were found to have the antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Pholiota squarrosa was 78.7% in the dose of 20mg/kg/day for the period of ten days. The tumor in two of the ten mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The total polysaccharide was 42% by Anthrone method and the protein was 55% by Lowry-Folin method in the antitumor component of Pholiota squarrosa.

  • An edible marine red alga, Grateloupia filicina, collected from Jeju Island of Korea was hydrolyzed by cheap food-grade carbohydrases (Viscozyme, Celuclast, AMC, Termamyl, and Ultraflo) to investigate its anticoagulant activity. Among the tested enzymatic extracts of G. filicina, a Termamyl extract showed the highest anticoagulant activity. Anion-exchange chromatography on DEAE-cellulose and gel-permeation chromatography on Sepharose-4B were used to purify the active polysaccharide from the crude polysaccharide fraction of G. filicina. The purified sulfated polysaccharide (0.42 sulfate/total sugar) showed ${\sim}1,357kDa$ molecular mass and was comprised mainly of galactose(98%) and 1-2% of glucose. The sample showed potential anticoagulant activity on activated partial thromboplastin time (APTT) thrombin time (TT) assays. The purified G. filicina anticoagulant (GFA) inhibited the coagulation factor X (92%), factor II (82%), and factor VII (68%) of the coagulation cascade, and the molecular interaction (protein-polysaccharide) was highly enhanced in the presence of ATIII (antithrombin III). The dissociation constant of polysaccharide towards serine proteins decreased in the order of FXa (58.9 nM) >FIIa (74.6 nM) >FVII (109.3 nM). The low/less cytotoxicity of the polysaccharide benefits its use in the pharmaceutical industry; however, further studies that would help us to elucidate the mechanism of its activity are needed.

Proximate Analysis and Antioxidant Activity of Cultivated Wild Panax ginseng (연근별 산양삼의 일반성분 및 항산화 효과)

  • Proximate analysis and antioxidant activity of cultivated wild Panax ginseng (CWPG) were investigated to provide fundamental information of CWPG with different ages and to increase its industrial application. Proximate analyses of CWPG with different ages were performed. Extraction of CWPG with different ages was carried out using heat-reflux extraction, and their extraction yield, crude saponin content, ginsenoside content, and antioxidant activity were analyzed. Moisture content decreased, but crude fat and crude protein were increased with aging. Extraction yield and crude saponin contents did not show a specific pattern while 5-year-old CWPG revealed the highest extraction yield and crude saponin content. All CWPGs showed typical ginsenoside profiles containing C-K and Rh2 ginsenosides, which are not found in ginseng. The 3-year-old CWPG showed the highest antioxidant activity including total phenolic content, total flavonoid content, and DPPH and ABTS radical scavenging activities. Moreover, 3-year-old CWPG also revealed the highest acidic polysaccharide content. Therefore, these results suggested that 3-year-old CWPG, which is the cheapest, can be used in industrial application due to its high antioxidant activity and acidic polysaccharide content with similar ginsenoside profile compared to 5- and 7-year-old CWPGs.

Carbon Source Affects Synthesis, Structures, and Activities of Mycelial Polysaccharides from Medicinal Fungus Inonotus obliquus

  • The effects of various carbon sources on mycelial growth and polysaccharide synthesis of the medicinal fungus Inonotus obliquus in liquid fermentation were investigated. After 12-d fermentation, mycelial biomass, polysaccharide yield, and polysaccharide content were significantly higher in Glc+Lac group (glucose and lactose used as combined carbon source) than in other groups. Crude polysaccharides (CIOPs) and the derivative neutral polysaccharides (NIOPs) were obtained from mycelia fermented using Glc, fructose (Fru), Lac, or Glc+Lac as carbon source. Molecular weights of four NIOPs (termed as NIOPG, NIOPF, NIOPL, and NIOPGL) were respectively 780.90, 1105.00, 25.32, and 10.28 kDa. Monosaccharide composition analyses revealed that NIOPs were composed of Glc, Man, and Gal at different molar ratios. The NIOPs were classified as α-type heteropolysaccharides with 1→2, 1→3, 1→4, 1→6 linkages in differing proportions. In in vitro cell proliferation assays, viability of RAW264.7 macrophages was more strongly enhanced by NIOPL or NIOPGL than by NIOPG or NIOPF, and proliferation of HeLa or S180 tumor cells was more strongly inhibited by NIOPG or NIOPGL than by NIOPF or NIOPL, indicating that immune-enhancing and anti-tumor activities of NIOPs were substantially affected by carbon source. qRT-PCR analysis revealed that expression levels of phosphoglucose isomerase (PGI) and UDP-Glc 4-epimerase (UGE), two key genes involved in polysaccharide synthesis, varied depending on carbon source. Our findings, taken together, clearly demonstrate that carbon source plays an essential role in determining structure and activities of I. obliquus polysaccharides by regulating expression of key genes in polysaccharide biosynthetic pathway.

Antioxidant and neuroprotective effects of crude polysaccharide fractions from Cudrania tricuspidata fruits (꾸지뽕 열매 조다당류 분획물의 산화방지 활성 및 신경세포 보호 효과)

  • The current study examined antioxidant and neuronal cell protective effects of the crude polysaccharide fraction in Cudrania tricuspidata fruits (CTP). The radical scavenging activities of (1,1-diphenyl-picrylhydrazyl and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)) and reducing power and FRAP of CTP were increased dose-dependently. In addition, the expression of neuroprotective effect of CTP was tested in HT22 mouse hippocampal cells. CTP treatment exhibited non cytotoxicity at dose levels below $500{\mu}g/mL$. Within this optimal concentration range, CTP treatment significantly increased cell viability in $H_2O_2-treated$ HT22 cells. Furthermore, CTP treatment increased superoxide dismutase (SOD) activity and decreased malonaldehyde (MDA) levels in HT22 cells. Therefore, these results indicate that the crude polysaccharide fraction from Cudrania tricuspidata fruits (CTP) possesses antioxidant activities and displays therapeutic potential as a useful source material in the development of brain disorder treatments targeting oxidative stress in neuronal cells.
