염주괴불주머니 (Corydalis heterocarpa) 의 항산화 활성 (Antioxidant Activity of the Salt Marsh Plant Corydalis heterocarpa)

  • The antioxidant activities of two crude extracts ($CH_2Cl_2$ and MeOH) and their solvent fractions (n-hexane, 85% aq. MeOH, n-BuOH, and $H_2O$ fractions) from Corydalis heterocarpa were determined by evaluating authentic $ONOO^-$ and $ONOO^-$ generated from SIN-1 (3-morpholinsydnonimine) in vitro as well as by measuring the degree of occurrence of intracellular reactive oxygen species (ROS) and nitric oxide (NO). Scavenging activities of solvent fractions on authentic $ONOO^-$ increased in the order of n-BuOH > 85% aq. MeOH > $H_2O$ > n-hexane fractions, while those on $ONOO^-$ generated from SIN-1 increased in the order of n-BuOH > 85% aq. MeOH > $H_2O$ > n-hexane fractions. In addition, all solvent fractions effectively inhibited the intracellular ROS and NO levels. The n-BuOH fraction especially exhibited the strongest ROS scavenging effect. Further purification of n-BuOH fraction led to the isolation of cnidimoside A, which presented the potent ROS scavenging effect at $10\;{\mu}M$. From these results, extracts of C. heterocarpa and its component, cnidimoside A, were predicted to be potentially useful as ingredients for protecting against oxidation.

Inhibition of MMP-2 and MMP-9 activities by solvent-partitioned Sargassum horneri extracts

  • Background: Matrix metalloproteinases (MMPs) are linked with several complications such as metastasis of cancer progression, oxidative stress, and hepatic fibrosis. Brown seaweeds are being extensively studied for their bioactive molecule content against cancer progression. In this context, Sargassum horneri was reported to possess various bioactivities including antiviral, antimicrobial, and anti-inflammatory partly due to its phenolic compound content. Methods: In this study, potential of S. horneri was evaluated through anti-MMP effect in HT1080 fibrosarcoma cells. S. horneri crude extract was fractionated with organic solvents, namely, water ($H_2O$), n-buthanol (n-BuOH), 85% aqueous methanol (85% aq. MeOH), and n-hexane. The non-toxicity of fraction samples (Sargassum horneri solvent-partitioned extracts (SHEs)) was confirmed by cell-viability assay. SHEs were tested for their ability to inhibit MMP enzymatic activity through gelatin digestion evaluation and cell migration assay. Expressions of MMP-2 and MMP-9 and tissue inhibitors of MMP (TIMPs) were evaluated by reverse transcription and Western blotting. Results: All fractions inhibited the enzymatic activities of MMP-2 and MMP-9 according to gelatin zymography. Except $H_2O$ fraction, fractions hindered the cell migration significantly. All tested fractions suppressed both mRNA and protein levels of MMP-2, MMP-9, TIMP-1, and TIMP-2. Conclusion: Overall, current results suggested that S. horneri has potential to be a good source for anti-MMP agents, and further investigations are underway for better understanding of the action mechanism and isolation and elucidation of the bioactive molecules.

Antimicrobial Activity of Medicinal Plants Against Bacillus subtilis Spore

  • Bacterial endospores, especially those of Bacillus and Clostridium genera, are the target of sterilization in various foods. We used Bacillus subtilis ATCC 6633 spores to screen novel antimicrobial substances against spores from medicinal plants. We collected 79 types of plant samples, comprising 42 types of herbs and spices and 37 types of medicinal plants used in traditional medicine in Korea and China. At a concentration of 1%(w/v), only 14 of the ethanol extracts exhibited antimicrobial activity against B. subtilis spores of at least 90%. Crude extracts of Torilis japonica, Gardenia jasminoides, Plantago asiatica, Fritllaria, and Arctium lappa showed particularly high sporicidal activities, reducing the spore count by about 99%. Consideration of several factors, including antimicrobial activity, extraction yields, and costs of raw materials, resulted in the selection of T. japonica, G. jasminoides, A. lappa, and Coriandrum sativum for the final screening of novel antimicrobial substances. Verification tests repeated 10 times over a 4-month period showed that the ethanol extract of T. japonica fruit reduced aerobic plate counts of B. subtilis spores the most, from $10^7$ to $10^4\;CFU/mL$ (99.9%) and with a standard deviation of 0.21%, indicating that this fruit is the most suitable for developing a novel antimicrobial substance for inactivating B. subtilis spores.

  • The roots of Cichorium intybus L. (chicory) have many nutritional qualities and also have been used for health promotion as traditional medicine. The antimicrobial activity of Cichorium intybus L.'s (chicory) ethanol extract on Bacillus cereus ATCC 10876 were investigated using spot on lawn method in this study. Ethanol extract exhibited antimicrobial activities against Bacillus cereus ATCC 10876 with significance. Also, the results produced the larger zones of inhibition against Bacillus cereus ATCC 10876 tested, with increasing concentration of Cichorium intybus L.'s (chicory) ethanol extracts,. Hence, Cichorium intybus L. (chicory) could be served for keeping the safety of various dairy products as natural antimicrobial agents.

오배자, 우슬, 가자 및 감초를 포함하는 혼합추출물의 항산화 및 항균활성 (Antioxidant and Antimicrobial Activities of Combined Extracts of Galla rhois, Achyranthes japonica Nakai, Terminalia chebula Retz and Glycyrrhiza uralensis)

  • In this study, antioxidant and antimicrobial activities of ethanolic crude extract and its five different solvent subfractions (namely, ethyl acetate fraction, n-butanol fraction, chloroform fraction, n-hexane fraction and the aqueous fraction) from a mixture of four different medicinal herbs (Galla rhois, Achyranthes japonica Nakai, Terminalia chebula Retz and Glycyrrhiza uralensis) were investigated. Among all the tested mixture combination of four medicinal herbs, 5:3:1:1 ratio of Galla:Achyranthes : Terminalia : Glycyrrhiza had the best antimicrobial effects against four strains of microorganisms (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa and Escherichia coli) and exhibited the highest DPPH radical-scavenging activity. Further sub-fractions with solvents were screened for antioxidant and antimicrobial activities. Antioxidant activity in order was ethyl acetate fraction > n-butanol fraction > chloroform fraction > nhexane fraction > aqueous fraction. The n-butanol extracted fraction showed the highest level of antimicrobial activity in com- parison to other fractions. In addition, all those fractions did not show any cytotoxicity against human skin cell CCD-986sk. These results suggest that 5:3:1:1 combination extracts of medicinal herbs (Galla : Achyranthes : Terminalia : Glycyrrhiza) may be potentially used as a safe natural antimicrobial preservative.

  • 소나무를 95% 에탄올로 추출하여 얻은 추출물을 40 mg/mL 첨가하였을때 Listeria monocytogenes Scott A, Listeria monocytogenes Brie I. isteria monocytogenes ATCC 19111 3균주의 성장이 억제되었으며 이를 다시 용매별로 순차 분획한 후 3균주에 대한 증식저해효과는 특히 ether 분획 물이 높게 나타났다. 소나무 추출물 40 mg/mL 첨가에 의해 Listeria monocytogenes 3균주 모두 균체의 성장 단계 중 유도기인 4시간째 회수한 균체에서 높은 생균수 감소 현상이 확인되었다. 전자현미경으로 확인한 균체의 형태는 대조구의 균체표면은 매끈하고 손상의 흔적이 없으나 처리구의 균체의 표면은 손상되었거나 형태가 이상을 일으킨 모습을 볼 수 있었다.

인진호 추출물의 식중독 세균들에 대한 생육억제 효과 (Antimicrobial Activity of the Aerial Part of Artemisia capillaris Extracts on the Food-Borne Pathogens)

  • 천연 보존료의 개발을 위한 식품학적 기초자료로 활용하기 위해 인진호 추출물을 이용하여 9종의 병원균과 식중독균에 대한 항균력을 검토한 결과는 다음과 같다. Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Vibrio prahemolyticus에 대해 각 용매 추출물에서 고루 항균성을 나타내었고, 물 추출물은 Vibrio prahaemolyticus에 대해서만 항균활성을 나타내었으며, 아세톤 추출물의 항균력이 가장 높았다. 인진호 아세톤 추출물의 최소저해농도(MIC)는 Staphylococcus aureus와 Bacillus subtilis에서 20 mg/mL, Vibrioparahaemolyticus에서 40 mg/mL 그리고 Salmonella typhimurium에서는 80 mg/mL로 나타났으며, 측정된 48시간 동안까지 최소저해농도 이상에서는 균이 거의 생육하지 못하였다. 인진호 아세톤 추출물의 순차 분획물의 항균활성은 에틸아세테이트와 부탄올 분획물이 Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Bibrio prahemolyticus에서, 클로로포름 분획물은 Staphylococcus aureus, Bacillus subtilis, Salmuneila typhimurium에서 항균활성을 나타내었으며, 헥산 분획물은 Bacillus subtilis에서만 항균활성을 나타내었다. 이같은 사실은 인진호 추출물의 항균성 물질은 특정 용매에만 용해되지 않고 다른 용매에도 용해되는 성분으로 단일 물질보다는 여러 성분일 것으로 생각된다 이상의 결과에서 천연물에 따라 항균력을 가지는 물질이 다양하여 추출용매의 선정이 항균성을 나타내는 성분을 찾아내는데 중요하다고 생각되며, 또한 인진호에서 추출된 항균성 물질이 Staphylococcus aureus나 Vibrio parahaemolyticus 등 식중독균의 생육억제에 효과가 있을 것으로 사료된다.

함초의 항산화성 및 함초를 첨가한 족발의 품질 특성 (Antioxidant Effects of Hamcho (Salicornia herbacea L.) and Quality Characteristics of Pettitoes (jokbal) added with Hamcho)

  • This study was conducted to ascertain the antioxidant properties of Hamcho (Salicornia herbacea L.) and as well as manufacture pettitoes using Hamcho as a natural antioxidant material for diverse applications. The DPPH radical scavenging activities of 25, 50, 100 mg/mL in methanol extracts from Salicornia herbacea L. increased significantly (p<0.05) to 12.8%, 21.6% and 39.1%, respectively. The total phenolic content was $38.58{\pm}4.25{\mu}M$ as tannic acid equivalent for 100 mg/mL of methanol extracts from Hamcho. FRAP value was $448.58{\pm}14.01{\mu}M$ as ascorbic acid equivalent in reducing power. To apply Hamcho to food as a natural antioxidant, pettitoes were prepared with added Hamcho. To manufacture pettitoes added with Hamcho, general quality characteristics of pettitoes added with 0%, 2%, 4% and 6% based on 100 g of pettitoes were determined. In conclusion, mineral components increased according to the added amount of Salicornia herbacea L. Crude protein showed the highest value with 2% added Hamcho. Overall, the addition of Hamcho led to hardening of the meat texture. As a result of measuring color in terms of brightness, redness, and yellowness with chromaticity, the L value (brightness) increased with added amount of Hamcho. There was no significant difference in redness or yellowness. As a result of the sensory evaluation of pettitoes according to addition of Hamcho, the group with 4% added Hamcho showed enhanced overall preference with improved flavor, taste and textural properties.

효소처리에 의한 백삼 저분자 화합물의 V79-4 세포주에 대한 항산화 활성 (Antioxidant activity of white ginseng extracts prepared by enzyme treatment on V79-4 cells induced by oxidative stress)

  • 본 연구는 효소분해를 통하여 인삼의 저분자 화합물을 추출하고 추출수율, 총페놀 함량, V79-4 세포주를 이용한 항산화 활성을 시험하였다. 조추출물의 추출 수율은 29.5-76%였으며, 총페놀 함량은 0.45-2.21%로 처리 효소에 따라 차이를 보였으며, pectinase 처리구가 추출 수율과 총페놀 함량이 가장 높았다. V79-4 세포주에 대해서는 pectinase 처리구와 ${\alpha}$-amylase 처리구에서 50% 이상의 세포 보호효과를 나타내었다. 세포내 항산화 관련 효소계의 활성을 시험한 결과 SOD활성은 처리 효소에 따른 유의적인 차이가 없었으나, CAT와 GPx 활성은 pectinase 처리구에서 유의적으로 높은 활성을 보였다. 또한 지질과산화 활성을 시험한 결과에서도 pectinase 처리구와 protease 처리구에서 50% 이상의 MDA 생성 억제를 보였다.

노루궁뎅이 버섯 및 추출물의 특성 (Characteristics of Hericium erinaceus and its Extracts)

  • 노루궁뎅이 버섯 및 그 추출물의 일반적인 식품의 성분과 기능적 특성을 조사하였다. 그 결과 노루궁뎅이 버섯의 조단백질은 8.01%로 나타나 단백질의 함량이 높은 것으로 나타났으며, 유리당은 glucose 47.09 mg%, fructose 34.65 mg%의 순으로 나타났으며 sucrose와 maltose는 나타나지 않았다. 유리아미노산은 glutamic acid alanine, threonine과 serine의 순으로 많은 함량을 나타내었다. 가용성 고형분 함량은 용매에 따른 차이가 없는 것으로 나타났으며, 무기질 함량은 칼륨을 제외하고는 물 추출물이 에탄올 추출물보다 높은 함량을 나타내었으며, superoxide 라디칼 소거능에서도 유사한 경향을 나타내었다. 페놀함량에서는 오히려 에탄을 추출물이 조금 높게 나타났으며 전자공여작용은 물 추출물(59.1 unit)이 에탄올 추출물(33.8 unit)보다 훨씬 높게 나타났다. 이상의 결과에서 용매에 따른 추출에서 물 추출물이 용매 추출물보다 전반적으로 기능적 특성을 가진 성분이 많이 추출되는 것으로 사료된다.
