  • The crude extract of Streptomyces sp. strain KM86-9B, isolated from a marine sponge, displayed significant inhibition on topoisomerase I activity. Investigation of the ca usative components by bioactivity-directed fractionation resulted in the isolation of a series of iso- and anteiso-fatty acids.

  • Chemical investigation of the crude extract of the Egyptian marine tunicate Eudistoma laysani led to the isolation of a new nucleoside; 3-deazainosine and four known ones; inosine, 2'-deoxyuridine, adenosine and 2'-deoxyadenosine. Structural elucidation of the isolated compounds was based on intensive studies of their spectral data including 1D ($^{1}H$ and $^{13}C$) and 2D ($^{1}H-^{1}H$ COSY, HMQC, HMBC) NMR together with mass spectra. The antioxidant effects of the isolated compounds were determined using DPPH, where they exhibited significant activities.

  • A calcium-dependent sialic acid-binding lectin has been isolated by thyroglobulin-affinity chromatography from the coastal crab Macrophthalmus Japonicus. This lectin, Macrophthalmus Japonicus lectin (MJL), was eluted with 50mM Tris-HCl, 0.3 M NaCl, 10 mM EDTA, and the recovery yield from the crude protein extract was about 5.6%. The molecular weight of MJL was estimated as 65 kDa in SDS-PAGE both under reducing and non-reducing conditions. MJL induced an agglutination reaction in rabbit, rat, and mouse erythrocytes, but not in human ABO types. This activity was effectively inhibited by sialoglycoproteins such as fetuin, bovine submaxillary mucin, and thyroglobulin.

  • An effective purification method was developed for producing Homoharringtonine (HHT), to guarantee high purity and yield from Cephalotaxus koreana. This process was a simple and efficient procedure, for the isolation and purification of HHT form the biomass of Cephalotaxus koreana, consisting of extract, adsorbent treatment, precipitation and followed by a chromatography. The extraction, adsorbent treatment and precipitation in pre-purification process allows for rapid and efficient separation of HHT from many compound and dramatically increases the yield and purity of crude HHT for HPLC purification steps compared to alternative processes. This purification processes serves to minimize solvent usage, size, and complexity of the operations for HHT purification.

  • We examined crude drug that originated from Mulberry (Morus alba L., Family; Morceae) leaves containing many nutritional components, which are the best food source for silk worms.   • A eight weeks feeding experiment was conducted to investigate the effects of different dietary dimethyl-$\beta$-propiothetin (DMPT) levels on growth and chemical components in red sea bream (Pagrus major). The content of DMPT extracted from Ulva pertusa by auroclaving was 32.5mg%. Body weight gain, feed efficiency and feed intake were significantly increased with dietary DMPT level. After eight weeks of feeding trial, moisture content decreased with supplemented DMPT level, while crude protein and lipid contents increased with dietary DMPT level. Amino acid contents of muscle was elevated in the DMPT added groups compared to control group. Amino acid composition of muscle in fish fed the diets supplemented DMPT was not different to that of control group. Fatty acid contents of muscle in fish increased with dietary DMPT level.

  • To assess the quality characteristics of black-goat meat extracts and to develop their quality index, the ingredient ratio, manufacturing process and general composition of black-goat meat extracts were investigated. A partial meat of black-goat had high protein content(20.2∼21.6%) and low lipid content (1.9∼3.3%). The crude protein contents, 3.5∼6.1%, of black-goat meat extracts had a high correlation (r=0.88, p<0.01) with black-goat meat content and the soluble solid content had a high correlation (r=0.87. p<0.01) with the subsidiary material content. Two commercial products had total bacteria of 10$^1$∼10$^2$ cfu/ ml. showing the necessity of pressure sterilization of final process.

  • A strain of alkalophilic Bacillus sp. C-21 has been Isolated from sold. The strain was capable of producing large amount of cyclodextrin glycosyltransferase (CGTase) in the high alkaline pH medium. The preferable medium composition was determined to be as follows : 1.0% soluble starch, 1.0% peptone, 0.5% yeast extract, 0.1% K2HP04, 0.02% MgSO4.7H2O and 1.0% Na2CO3(pH 10.0) The highest enzyme production was observed after 30hours of cultivation at 33$^{\circ}C$. The optimum temperature and pH for the activity of crude enzyme were 6$0^{\circ}C$ and 6.0, respectively. The enzyme was stable between pH 6.0 and 9.6, and up to 55$^{\circ}C$.

Flavonoid Fraction Purified from Rhus verniciflua Stokes Actively Inhibits Cell Growth Via Induction of Apoptosis in Mouse Tumorigenic Hepatocytes

  • Dietary flavonoids are currently receiving considerable attention in developing novel cancer-preventive approaches because of their potential capacities to actively induce apoptosis of cancer cells. In our previous report, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein and named RCMF (RVS chloroform-methanol fraction), was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) that is traditionally used as food additive and herbal medicine. In this study, we evaluated the effects of the RCMF on cell proliferation and apoptosis using SV40-transformed tumorigenic hepatocytes, BNL SV A.8. Tritium uptake assay showing the proliferative capacity of the cells was strongly suppressed in the presence of RCMF. This anti-proliferative effect was further confirmed through trypan blue exclusion. RCMF-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. Collectively, this study demonstrated that RCMF can be approached as a potential agent that is capable of significantly inhibiting cell growth of hepatic cancer cells.

  • In order to investigate the effect of Kamiondam-tang on the cardiovascular system and sedative action, pharmacological studies have been carried out with water extract. The results of this investigation were summarized as follows; The sedative action was significantly recognized in mice. Vasodilative action in rabbit peripheral blood vessel and hypotension in anesthetized rabbit were remarkably recognized. Inhibitory effect of cardiac systolic action was recognized by Straub method in frogs.
