신광벼 유래의 벼 줄무늬잎마름병 저항성 주동 QTL qSTV11SG탐색 (Identification of a Major QTL, qSTV11SG, Associated with Resistance to Rice Stripe Virus Disease Originated from Shingwangbyeo in Rice (Oryza Sativa L.))

  • 벼 줄무늬잎마름병 저항성 유전자 및 연관 DNA 마커 탐색을 위하여 줄무늬잎마름병에 저항성인 통일형 품종인 신광 벼 이용 여교잡 집단을 육성하였다. 줄무늬잎마름병 저항성 유전자에 대한 QTL을 분석한 결과 11번 염색체에 위치하는 SSR 마커 RM6897이 탐색되었으며 전체 표현형 변이의 44.2%를 설명하였다. DNA 마커 RM6897은 여교잡 집단에서 생물검정과 유전자형이 일치하였다. 또한 자포니카 품종들에서 저항성 27품종과 감수성 23품종에 대해 구분이 가능하였다. 따라서 신광벼 유래의 줄무늬잎마름병 저항성원 및 분자마커는 자포니카 품종의 바이러스 저항성 향상에 효율적으로 활용될 것으로 기대된다.

  • In Korea, buckwheat has historically held a very important position because it has been used as a food and traditional medicine. The objectives of this report were to mainly review the up to present papers includings 75 references about buckwheat's rutin, functional ingredients and their variations. Buckwheat provides an abundance of such nutrition as protein, amino acids, vitamin($B_1,\;B_2$, E, P), minerals(Fe, Zn, Mn, Mg, P, Cu, Ca, Se) and dietary fiber. Buckwheat's essential amino acid, lysine, is notably higher amount than cereals. Rutin(vitamin P), one of a group of flavonoids, is abundant in buckwheat and noted for its beneficial function of reducing every kinds of radiation and vascular diseases, diabetes mellitis and retinal hemorrhage, etc. Rutin also acts as a pigment stabilizer and acetone extracts of buckwheat have a potent tyrosinase inhibitory activity.

작물에 함유된 Phytoestrogen의 특성과 생리활성 (Biological Activities of Phytoestrogens in Plant and Foodstuff)

  • Phytoestrogens are oestrogenic compounds found in plants and consist of isoflavones, lignans, and coumestans. The structural similarity of phytoestrogens to endogenous oestrogens has promoted the hypothesis that phytoestrogens exert hormonal or anti-hormonal effects relevant to the risk of hormone-dependent disease and/or their suitability as a dietary alternative to hormone replacement therapy. Epidemiological studies suggest that food stuffs containing phytoestrogens may have a beneficial role in protecting against a number of chronic disease and conditions. It is thought that these estrogen-like compounds may protect against chronic diseases, such as hormone-dependent cancers, cardiovascular disease and osteoporosis. Furthermore, phytoestrogens are used as a natural alternative to hormone replacement therapy and to reduce menopausal symptoms. Phytoestrogens are considered good candidates for use in natural therapies and as chemopreventive agents in adults. However safe and efficacious levels have yet to be established.

Antioxidant Effect of Flavonoids Isolated from the Root of Clematis trichotoma Nakai

  • The antioxidant effect of methanol extract (ME) and water extract (WE) from Clematis trichotoma was evaluated as primary study to scavenge stable 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH), inhibited iron-induce lipid peroxidation in linoleic acid emulsion, peroxidation of liposome induced by $Fe^{3+}/H_2O_2/ascorbie$ acid, and on $Fe^{2+}/H_2O_2$ induced the mitochondrial lipid peroxidation. In secondary study, five flavonoids as luteolin (1), quercetin (2), apigenin (3), hirsutrin (4), kaempferol-3-O-glucoside were isolated (5). Among them, compounds 1 and 2 showed good activities in all the model systems. Compound 3 exhibited moderate antioxidant activities in both radical scavenging and these lipid peroxidation systems tested. Compound 4 showed significant inhibitions in liposome peroxidation and compound 5 displayed weak inhibition in all four tested systems. All the results presented herein indicate that products of C. trichotoma maybe useful in inhibiting membrane lipid peroxidation and preventing free radical-linked diseases.

치자의 Geniposide 함량 기준 설정 (Standardization of Geniposide Content of Gardeniae Fructus)

  • Gardeniae Fructus (Gardenia jasmmoides Ellis) is widely used as a natural food colorant and as a traditional medicine for the treatment of hepatic and inflammatory diseases. The quality evaluation of Gardeniae Fructus was carried out by HPLC using geniposide as a marker. The geniposide was isolated from commercial Gardeniae Fructus on ODS column using a mixed solvent of acetonitrile-water (v/v 9:91) as mobile phase and a detection wavelength 240 mm. The content of geniposides in twenty different samples was in a range of 1.20% to 7.17% of total tissues and the average was 4.97%.

  • 유백피 물 추출물의 위궤양치료 효과를 확인하기 위하여 여러 비율의 농도로 rat에 대한 투여량을 조절하면서 Shay, aspirin 및 Indomethacin으로 유도된 위궤양에 대하여 실험하였던 바 유백피의 물 추출물을 각기 500 및 1000mg/kg을 투여하였을 때 항궤양 효과가 유의성 있게 농도 의존적으로 증가하였으며 Shay궤양이나 aspirin유발 궤양보다도 indomethacin유발 궤양의 경우에 더욱 항궤양 효과가 좋았다.

  • Crop Protection Products (CPPs) are a useful weapon in our fight against the pests, diseases and weeds that harm or destroy our food. To be effective CPPs need to exhibit some persistence. The consequence of this is that residues of the original material or its metabolites may remain on food and thus there is a potential for the intake of such residues. The issue of CPP residues in food is an emotional subject and is rarely debated rationally or supported by scientific facts. Media headlines like “Poison in our food” or “These days enough Pesticides (CPPs) are sprayed onto vegetables to turn you into one” are testimony to the high level of emotion that surrounds the subject of CPP residues in food. Recent surveys of consumers have indicated that more than 80 percent view CPP residues in food as a “serious hazard”. This significantly exceeds concerns over drugs, hormones in meat, nitrates in food, irradiated foods, food additives, or artificial colours.

  • ${\gamma}$-Aminobutyric acid (GABA) is a non-protein amino acid that is widely distributed in plant and animal kingdom. GABA is found in tissues of the central nervous system (CNS) in animals. GABA functions as a the major inhibitory neurotransmitter in the CNS by acting through the GABA receptors. Clinical studies have revealed the relationship between an increased intake of GABA or analogues with several health benefits, including lowering of blood pressure in mildly hypertensive animals and humans. Furthermore, GABA would also has an inhibitory effect on cancer cell proliferation, stimulates cancer cell apoptosis and plays a role in alcohol-associated diseases and schizophrenia. In plants, interest in the GABA emerged mainly from experimental observations that GABA is largely and rapidly produced in large amounts in response to biotic and abiotic stresses. In this study, we speculated the properties and metabolism of GABA in plant and functions in relation to the responses to environmental stresses.

짚신나물 추출물의 알레르기 반응 억제 효과 (The Inhibitory Effect of Agrimonia pilosa Ledeb Extract on Allergic Reaction)

  • Complementary and alternative medicines are considered as a promising research field to develop new therapies for various allergic diseases. In this study, we investigated the anti-allergic effect of Agrimonia pilosa Ledeb (AP) by using passive cutaneous anaphylaxis in mice and its mechanism of action in mast cells. The extract of AP reversibly inhibited degranulation in RBL-2H3 cells and bone marrow-derived mast cells (BMMCs). AP also suppressed the passive cutaneous anaphylaxis inducing by IgE and antigen (Ag) in a dose-dependent manner. In the study to find its mechanism of action, AP inhibited the phosphorylation of Syk kinase, a pivotal protein which is regulated by Src-family kinase for activation of mast cells. In addition, AP also suppressed activation of Akt and Erk1/2 that are critical for the production of cytokines in mast cells. The results strongly suggest that AP exerts anti-allergic activity in vitro and in vivo through the inhibition of activation of Syk in mast cells.

Constructing Reference Transcriptome Sets of Codonopsis lanceolate(Deodeok) and Ixeridium dentatum

  • As the aging population increases and interest in well-being increases, the importance of developing special crops increases. Natural medicine based on the special crops has been mainly used as an adjunct therapy for many diseases and symptoms based on culture, traditional medicine, and experience. In particular, it is attracting attention as a new resource to develop new drugs such as Artemisinin, a treatment for malaria. In order to efficiently use crops, it is essential to establish omics data such as genomes, transcriptomes, and metabolites of special-purpose crops. However, many special-purpose crops have large, heterogeneous and polyploid genomes that require high cost and long time to reference genome sequencing. Therefore, we built an inexpensive, fast, but very usefill reference transcriptome as the first step. We constructed high-quality reference transcriptom sets of Codonopsis lanceolata and Ixeridium dentatum with PacBio data. Our team will continue to construct reference transcriptoms of more special-purpose crops, and the data will be released by the National Agricultural Biotechnology Information Center (NABIC) in order to be widely used in agricultural as well as medical R&D.
