Effects of Psoralen and Angelicin on Hepatic Drug-Metabolizing Enzyme Activities

  • The effects of psoralen and angelicin on hepatic microsomal drug-metabolizing enzyme (DME) activities were investigated to elucidate the mode of the interaction of furanocoumarins with DME system. A single administration (30 mg/kg,i. p.) of both coumarins to mice cased a significant prolonagation of hexobarbital-induced hypnosis as well as an increase in strychnine toxicity. The inhibitory potencies of both coumarins as measured by rat hepatic microsomal aminopyrine N-demethylase and hexobarbital hydroxylase activities in vitro were considerably weaker than those of other furanocoumarins which possess a side chain moiety. Both coumarins were found to have significant inducing effects of DME system, with repeated treatments of them. The activities of an angular coumarin were stronger than those of a linear coumarin.

  • Systematic fractionation of Angelica gigas roots led to the isolation of linear coumarins such as decursinol angelate, decursin and nodakenin. These compounds were tested for their effects on rat lens aldose reductase. Nodakenin was demonstrated to exhibit significant inhibition of rat lens aldose reductase activity with $IC_{50}$ value of $7.33\Mu\textrm{M}$.

  • Five coumarins and a pyrimidine were isolated from the roots of Angelica gigas. Their structures were elucidated as bergapten (1), decursinol angelate (2), decursin (3), nodakenetin (4), uracil (5) and nodakenin (6) by spectral analysis. Among them, bergapten (1) and uracil (5) were isolated for the first time from this plant part.

  • Peucedani Radix is the root of Angelica decursiva Franchet et Savatier (=Peucedanum decursivum Maximowicz) or Peucedanum praeruptorum Dunn in several Asian countries. The coumarins contained in Peucedani Radix were quantitatively analyzed using HPLC-DAD to develop a simultaneous determination for the quality control of A. decursiva and P. praeruptorum. For quantitative analysis, four major coumarins contained in these medicinal plants were assessed. Nodakenin (1), nodakenetin (2), praeruptorin A (3), and praeruptorin B (4) were separated with a Phenomenex Luna C18 column ($5{\mu}m$, $4.6{\times}250mm$) under the gradient conditions using distilled water with 0.1% phosphoric acid and acetonitrile with 0.1% phosphoric acid as the mobile phase, at a flow rate of 1.0 ml/min and a detection wavelength of 330 nm. This method was fully validated for linearity, accuracy, precision, recovery, and limit of detection and quantification. As a result, A. decursiva and P. praeruptorum were clearly classified by the quantification of four major coumarins in extracts. Also, the pattern recognition analysis based on HPLC indicates that all of the samples were largely clustered into two groups. Therefore, it is possible to distinguish between A. decursiva and P. praeruptorum and contribute to quality control.

Coumarins and Chromones from Angelica genuflexa

  • Thirteen compounds were isolated from the roots of Angelica genuflexa through repeated silica gel column chromatography. Nine coumarins, isoimperatorin (1), osthol (2), demethylsuberosin (3), oxypeucedanin (4), heraclenin (5), pabulenol (7), umbelliferone (8), oxypeucedanin hydrate (9) and marmesinin (11), and four chromones, hamaudol (6), cimifugin (10), sec-O-glucosylhamaudol (12) and prim-O-glucosylcimifugin (13), were identified by physicochemical and spectroscopic analysis. Among these, compounds 3, 5, 6, 8, 12, and 13 were isolated for the first time from the roots of Angelica genuflexa. These coumarins and chromones were examined for their anticomplement activity. Demethylsuberosin (3) showed a weak anticomplement activity with an $IC_{50}$ value of $390\;{\mu}M$.

Coumarins from the aerial parts of Artemisia iwayomogi Kitamura

  • Artemisia iwayomogi (Compositae), a perennial mugwort, is native to Korea and widely distributed in Japan, Russia, and China. A. iwayomogi and A. capillaris are similar in morphology and pharmacological activity and have been used for the same medicinal purposes in Korea. While various ingredients such as coumarins and flavonoids and their activity studies have been reported for A. capillaris, few studies have been conducted on the pharmacologically active components of A. iwayomogi. In Korea, A. capillaris is not economical because only young leaves are used as a medicinal material. Because of this, A. iwayomogi is frequently used in Korea, indicating the need to study its pharmacologically active components. Therefore, a phytochemical study was initiated to isolate active compounds from the aerial parts of A. iwayomogi. Finally, four coumarins, umbelliferone (1), esculetin (2), grevillone (3), and scoparone (4) were isolated for the first time from the aerial parts of A. iwayomogi in this study.

  • A series of novel pseudopeptides contained coumarin skeleton were synthesized through the Ugi-four-component reaction. The 3-substituted coumarin-3-carboxamides were formed through reaction of benzaldehyde derivatives, anilines, coumarin-3-carboxylic acid and isocyanides with high yields and high bond-forming efficiency at room temperature. These novel amidated coumarins exhibit brilliant fluorescence in range of 535-547 nm in chloroform.

  • 광독성 물질인 쿠마린과 푸로쿠마린의 광화학반응을 분광분석, 삼중상태 켄칭, 형광법을 써서 연구하였다. 크산토톡신과 티민 또는 DNA 수용액의 광화학반응에서 ${\beta}$-카로텐을 켄치로 썼으나 아무런 켄칭이 일어나지 않는 것으로 보아 이 반응은 수명이 긴 크산토톡신의 삼중상태보다 수명이 짧은 들뜬 단일상태에서 일어나는 것으로 생각된다.

  • Aluminum containing mesoporous molecular sieves Al-SBA-1 ($n_{Si}/n_{Al}$= 2, 5, 10, 15) was synthesized with a one-step method, and the catalyst was characterized with powder X-ray diffraction (XRD), nitrogen adsorption-desorption, thermogravimetric analysis (TGA), scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FT-IR). Finally, we studied the catalytic activities of Al-SBA-1 molecular sieves in Pechmann reaction of 3-methoxyphenol and Ethyl acetoacetate to get 7-methoxy-4-methyl coumarin. Four parameters reaction temperature, time, catalyst weight and reactants ratio were optimized.

  • From the root of Angelica decursiva-albiflora Yook, which has been used as a folk medicine for a sedative, analgesic and expectorant, four free coumarins, e.g., decursidin(I), decursin(II), umbelliferone(III) and nodakenetin(IV), and two coumarin glycosides, e.g., nodakenin(V) and decuroside I(VI) were isolate. The cytotoxicity of nodakenin(V) against L-1210 leukemia cells was less effective than cisplatin, but in the nephrotoxicity against rabbit kidney proximal tubular cell nodakenin(V) showed remarkably less nephrotoxicant than cisplatin.
