Effects of Mahwangsan(麻黃散) Extract and Its Constituent Drug on the Contraction of Isolated Guinea pig Trachea Smooth Muscle (마황산(麻黃散) 및 그 구성약물(構成藥物)이 GUINEAPIG의 기관지(氣管支) 평활근(平滑筋)에 미치는 영향(影響))

  • Mahwangsan is used in the treatment of asthma due to wind and cold(風寒喘). This study was carried out to investigate the effect of Mahwangsan extract and each drug composing it on the contractile force of the isolated guinea pig trachea muscle and elucidate its mechanism. The results were obtained as follows; 1. The isolated trachea smooth muscle of guinea pig was signifi-cantly relaxed by the administration of Mahwangsan extract. 2. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administration of Mahwangsan extract. 3. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administraction of Herba Ephedrae, Cortex Cinnamoni, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 4. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Mahwangsan extract. 5. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 6. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Mahwangsan extract. 7. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract.

  • In order to study the effects of Goosunsan known clinically for their effects of treatment for cough and asthmas the study was carried out to investigate the effect of Goosunsan extract on the contractile force of the isolated guinea pig's vaviovs kinds smooth muscles and elucidate its mechanism The result were obtained as follows: 1. The isolated trachea & ileum smooth muscles of guinea pig was suspended in the organ bath with oxygenated kreb's Henselsite bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with Isometric transducer (nacro F-60). The resting tension was approximately 0.5g. 2. The isolated trachea & ileum smooth muscles of guinea pig was remakably relaxed by the administration of Goosunsan. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 4. The contractile response of the ileum smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan extract. 6. The contractile response of the ileum smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan. 7. INFLUENCE OF TOTAL GINSENG SAPONIN ON VASOCONSTRICTORS -INDUCED CONTRACTILE RESPONSES IN THE RAT AORTA

  • The present study was designed to examine the effect of total ginseng saponin on contractile responses of vasoconstrictors in the rat aorta. Phenylephrine (an adrenergic ${\alpha}$$_1$-receptor agonist) and high potassium (a membrane depolarizing agent) caused greatly contractile responses in the rat aorta, respectively. However, in the presence of total ginseng saponin (600 $\mu\textrm{g}$/$m\ell$), the contractile responses of phenylephrine (10$\^$-5/ and 10$\^$-7/ M) and high potassium (3.5 ${\times}$ 10$\^$-2/ and 5.6 ${\times}$ 10$\^$-2/ M) were markedly potentiated whereas prostaglandin F$\sub$2${\alpha}$/ (5 ${\times}$ 10$\^$-6/ M)-induced contractile response was not affected. The contractile responses induced by phenylephrine (10$\^$-5/ M) and high potassium (3.5 ${\times}$ 10$\^$-2/ M) even in the presence of total ginseng saponin (600 $\mu\textrm{g}$/$m\ell$) were greatly inhibited by the pretreatment of nicardipine (10$\^$-6/ M), a calcium channel blocker. Taken together, these experimental results suggest that total ginseng saponin can enhance the contractile responses evoked by stimulation of adrenergic ${\alpha}$$_1$-receptor and the membrane depolarization in the rat aorta, which seems to be associated to calcium influx.

  • Vascular reactivity can be influenced by the vascular region, animal age, and pathologies present. Prostaglandins (produced by COX-1 and COX-2) play an important role in the contractile response to phenylephrine in the abdominal aorta of young rats. Although these COXs are found in many tissues, their distribution and role in vascular reactivity are not clear. At a vascular level, they take part in the homeostasis functions involved in many physiological and pathologic processes (e.g., arterial pressure and inflammatory processes). The aim of this study was to analyze changes in the contractile response to phenylephrine of thoracic/abdominal aorta and the coronary artery during aging in rats. Three groups of rats were formed and sacrificed at three distinct ages: prepubescent, young and old adult. The results suggest that there is a higher participation of prostanoids in the contractile effect of phenylephrine in pre-pubescent rats, and a lower participation of the same in old rats. Contrarily, there seems to be a higher participation of prostanoids in the contractile response of the coronary artery of older than pre-pubescent rats. Considering that the changes in the expression of COX-2 were similar for the three age groups and the two tissues tested, and that expression of COX-1 is apparently greater in older rats, COX-1 and COX-2 may lose functionality in relation to their corresponding receptors during aging in rats.

Characteristics of Diprophylline-Induced Bidirectional Modulation on Rat Jejunal Contractility

  • In this study, we propose that diprophylline exerts bidirectional modulation (BM) on the isolated rat jejunal segment depending on its contractile state. The results supported the hypothesis. Diprophylline ($20{\mu}M$) exerted stimulatory effects on the contractility of jejunal segment in six low contractile states while inhibitory effects in six high contractile states, showing the characteristics of BM. Diprophylline-induced stimulatory effect was significantly blocked by atropine, indicating the correlation with cholinergic activation. Diprophylline-induced inhibitory effect was partially blocked by phentolamine, propranolol, and L-N-Nitro-Arginine respectively, indicating their correlation with sympathetic activation and nitric oxide-mediated relaxing mechanisms. Diprophylline-induced BM was abolished by tetrodotoxin or in a $Ca^{2+}$ free condition or pretreated with tyrosine kinase inhibitor imatinib, suggesting that diprophylline-induced BM is $Ca^{2+}$ dependent, and that it requires the presence of enteric nervous system as well as pacemaker activity of interstitial cells of Cajal. Diprophylline significantly increased the reduced MLCK expression and myosin extent in constipation-prominent rats and significantly decreased the increased MLCK expression and myosin extent in diarrhea-prominent rats, suggesting that the change of MLCK expression may also be involved in diprophylline-induced BM on rat jejunal contractility. In summary, diprophylline-exerted BM depends on the contractile states of the jejunal segments, requires the presence of $Ca^{2+}$, enteric nervous system, pacemaker activity of interstitial cells of Cajal, and MLCK-correlated myosin phosphorylation. The results suggest the potential implication of diprophylline in relieving alternative hypo/hyper intestinal motility.

Comparision of Proximal and Distal Large Intestinal Motility in Rabbit (가토의 근위와 원위대장 평활근의 운동성 비교)

  • To validate the comparision of proximal and distal large intestinal motility, the amplitude and frequency of spontaneous motility, the effect of acetylcholine, the effect of atropine on the response of acetylcholine, the effect of histamine and the effect of pyrilamine and cimetidine on the response of histamine were investigated in rabbit. The results were summarized as follows: 1. The amplitude of spontaneous motility was more powerful on the proximal large intestine than that of the distal large intestine, but the frequency of spontaneous motility was similar on the both proximal and distal large intestine in rabbit. 2. Acetylcholine caused the contraction of proximal and distal large intestine, and the contractile response were increased between the concentration of acetylcholne $10^{-9}$ and $5{\times}10^{-6}M$ and $10^{-7}$ and $10^{-4}M$ on the proximal and distal large intestine, respectively, with dose-dependent manner in rabbit. 3. The contractile response induced by acetylcholine was completely blocked by the post-treatment with cholinergic receptor blocker, atropine $10^{-6}M$. 4. Histamine caused the contraction of proximal and distal large intestine and the contractile response were increased between the concentration of histamine $10^{-9}$ and $5{\times}10^{-5}M$ and $10^{-5}$ and $10^{-3}M$ on the proximal and distal large intestine, respectively, with dose-depend ent manner in rabbit. 5. The contractile response induced by histamine was completely blocked by the pretreatment with $H_1$-receptor blocker, pyrilamine $10^{-6}M$, but not blocked by the pretreatment with $H_2$-receptor blocker, cimetidine $10^{-6}M$.

Study on the Effects of Chukdamtanggamibang on blood pressure, regional cerebral blood flow(rCBF) and smooth muscle (척담탕가미방이 혈압, 뇌혈류량 및 평활근에 미치는 효능에 관한 연구)

  • This study was aimed to investigate the effect Chukdamtanggamibang on the vascular systems including changes in blood pressure and regional cerebral blood flow(rCBF) of male Sprague-Dawely rats, contractile force of guinea pig`s tracheal smooth muscle and abdominal aorta and femoral artery in rabbits. Blood pressure was not affected by Chukdamtanggamibang in rats. rCBF was significantly increased by Chukdamtanggamibang in a dose-dependent manner. Contractile force of isolated guinea pig`s tracheal smooth muscle evoked by His ({TEX}$ED_{50}${/TEX}) were inhibited significantly by Chukdamtanggamibang. Propranolol, indomethacin and methylene blue did not significantly alter the inhibitory effect of Chukdamtanggamibang. Contractile force of isolated rabbit`s abdominal aorta and femoral artery evoked by NE ({TEX}$ED_{50}${/TEX}) were inhibited significantly by Chukdamtanggamibang. ODQ and L-NNA significantly attenuated the inhibitory effects of Chukdamtanggamibang in abdominal aorta, whereas propranolol did not significantly alter the inhibitory effect of Chukdamtanggamibang. These results indicate that Chukdamtanggamibang can relax hitamine-induced contraction of guinea pig`s tracheal smooth muscle and that this inhibition involves, in part, the relation to the soluble guanylyl cyclase synthesis and nitric oxide (NO) synthesis.

  • The aims of this study were to investigate the effect of adenosine triphosphate (ATP), which has been known as the neurotransmitter of nonadrenergic, noncholinergic nerves, and the source of $Ca^{\sharp}$ in the effect of ATP on the isolated renal artery of pig. The results of this study were summarized as follows: 1. ATP caused the contraction and the contractile responses were increased in a dose-dependent manner between the concentration of ATP $2{\times}10^{-3}M$ and $10^{-2}M$ on the isolated renal artery of pig. 2. The contractile responses induced by ATP $(5{\times}10^{-3}M)$ were not blocked by pretreatment with cholinergic receptor blocker (atropine, $10^{-6}M$), $\alpha$-adrenergic recptor blocker(phentolamine, $10^{-6}M$) or $\beta$-adrenergic receptor blocker (propranolol, $10^{-6}M$), and $H_1$-receptor blocker (pyrilamine, $10^{-6}M$) or $H_2$-receptor blocker (cimetidine, $10^{-6}M$) on the isolated renal artery of pig. 3. The contractile responses induced by ATP $(5{\times}10^{-3}M)$ were not appeared in $Ca^{\sharp}$-free medium. As the concentration of $Ca^{\sharp}$ in $Ca^{\sharp}$-free medium was increased, the contractile responses induced by ATP $(5{\times}10^{-3}M)$ were enhanced but were completely inhibited by pretreatment with $Ca^{\sharp}$-channel blocker, papaverine $(5{\times}10^{-5}M)$ or verapamil $(5{\times}10^{-5}M)$ on the isolated renal artery of pig.
