Biocontrol Activity of Volatile-Producing Bacillus megaterium and Pseudomonas protegens against Aspergillus flavus and Aflatoxin Production on Stored Rice Grains

  • In our previous study, three bacterial strains, Bacillus megaterium KU143, Microbacterium testaceum KU313, and Pseudomonas protegens AS15, were selected as effective biocontrol agents against Aspergillus flavus on stored rice grains. In this study, we evaluated the inhibitory effects of the volatiles produced by the strains on A. flavus growth and aflatoxin production on stored rice grains. The three strains significantly reduced mycelial growth of A. flavus in dual-culture assays compared with the negative control strain, Sphingomonas aquatilis KU408, and an untreated control. Of these tested strains, volatiles produced by B. megaterium KU143 and P. protegens AS15 markedly inhibited mycelial growth, sporulation, and conidial germination of A. flavus on agar medium and suppressed the fungal populations in rice grains. Moreover, volatiles produced by these two strains significantly reduced aflatoxin production in the rice grains by A. flavus. To our knowledge, this is the first report of the suppression of A. flavus aflatoxin production in rice grains using B. megaterium and P. protegens volatiles.

MoRBP9 Encoding a Ran-Binding Protein Microtubule-Organizing Center Is Required for Asexual Reproduction and Infection in the Rice Blast Pathogen Magnaporthe oryzae

  • Like many fungal pathogens, the conidium and appressorium play key roles during polycyclic dissemination and infection of Magnaporthe oryzae. Ran-binding protein microtubule-organizing center (RanBPM) is a highly conserved nucleocytoplasmic protein. In animalia, RanBPM has been implicated in apoptosis, cell morphology, and transcription. However, the functional roles of RanBPM, encoded by MGG_00753 (named MoRBP9) in M. oryzae, have not been elucidated. Here, the deletion mutant ΔMorbp9 for MoRBP9 was generated via homologous recombination to investigate the functions of this gene. The ΔMorbp9 exhibited normal conidial germination and vegetative growth but dramatically reduced conidiation compared with the wild type, suggesting that MoRBP9 is involved in conidial production. ΔMorbp9 conidia failed to produce appressoria on hydrophobic surfaces, whereas ΔMorbp9 still developed aberrantly shaped appressorium-like structures at hyphal tips on the same surface, suggesting that MoRBP9 is involved in the morphology of appressorium-like structures from hyphal tips and is critical for development of appressorium from germ tubes. Taken together, our results indicated that MoRBP9 played a pleiotropic role in polycyclic dissemination and infection-related morphogenesis of M. oryzae.

  • The purpose of this study is to evaluate the efficacy of metabolites produced form Streptomyces rimosus in controlling the growth of sapwood - inhabiting fungi. In order to carry out this task, the following specific fungi were tested : sapstain fungi - Ceratocystis pilifera, Ceratocystis piceae, and Aureobasidium pullulans ; mold fungi - Trichoderma hazianum, Trichoderma viride, Penicillium cirtrinum, and Aspergillus niger. Based on the tests, the following observations can be drawn. 1. The conidial germination of sapstain and mold fungi was completely inhibited leaving a clear zone around the paper disc treated with metabolites. The best inhibition was observed in A. pullulans plate and the least in T. viride. 2. Concentration of SB medium for the production of metabolites from St, rimosus affected antifungal activity of metabolites against sapwood - inhabiting fungi. Metabolites prepared from 1/3${\times}$SB medium showed the best activity and the least activity was observed in metabolites form 1/4${\times}$medium. 3. in vivo and in vitro test using wood blocks, treatment of pine sapwood blocks with metabolites also inhibited conidial germination and thus prevented discoloration. 4. Treatment with metabolites did not change the macroscopic structure of wood and did not cause the discoloration of the surface of wood by pigments produced form St. rimosus. In conclusion the results of this study indicate that antifungal metabloites of St, rimosus could be used for the biological control of sapstain and mold fungi.

Weeding Efficacy of Melanized Formula with Epicoccosorus nematosporus on Eleocharis kuroguwai in the Field

  • The study was conducted to determine the cultural conditions and the effect of inert fillers for melanization and sporulation abilities of sodium alginate pellets, and the weeding efficacy of the formula in the field. Melanin production of E. nematosporus was affected by striking frequency. Percentage of melanized beads was increased to 80.6% at higher rpm up to 180. The melanized pellets produced more conidia with abundant mucilage than unmelanized pellets. Shaker culture of Epicoccosorus nematosporus with sodium alginate yielded a total of 55 mg per 100 pellets. Percentage of melanized pellets was highest with 81.0% and 83.3% of melanization, when wheat bran and rice polish were amended and produced the conidia with 65.4 and 68.4 mg per 100 pellets, respectively. When 1 L of conidial suspension of 6.0$\times$$10^5$ conidia per ml was applied on 30-day-old plants in a plot, 74.5% of the plants were killed within 20 days, whereas, its melanized sodium alginate pellets killed 57.8% of the plants in the same period. The number of tuber formation of Eleocharis kuroguwai in the untreated control plots was 128.5 per plot, but those of the plots treated with conidial suspension and melanized pellets were 22.1 and 39.7, respectively, at the end of the season. Results of this study showed that melanization of mycelia-mixed sodium alginate are an important sporulation factor in E. namatosporus as a mycoherbicide.

  • Thirtythree strains of the Aspergillus spp. isolated from foodstuffs were observed through some physiological characteristics for detection of identification key of Aspergillus spp. 1) Each strain of Aspergillus spp. had their specific characteristics and could be used for identification of species. 2) Excellent amylase-producing fungi were observed among the isolated strains of Aspergillus spp. 3) Amylase activities increased for one week incubation period. 4) In the tests of common characters of aflatoxin-producing fungi among the 33 strains of Aspergillus spp., for example, conidial size, presence of sclerotia, kojic acid, and pigment production, coloration of phenol, reduction of methylene blue, etc.

Leaf Blotch of Allium fistulosum Caused by Cladosporium allii-cepae (Cladosporium allii-cepae에 의한 파 누른무늬병)

  • Leaf blotch of Welsh onion was observed at a farmer's field at Kangnung in July 1990. Further collection o the diseased materials was made in 1993 and 1994. The symptom initially appeared as discolored spot and later yellowish blotch with the long axis parallel to the leaf veins, which turned to sooty colored concentric lesion due to heavy production of conidiophores and conidia. Cladosporium allii-cepae (Ranojevic) M. B. Ellis was repeatedly isolated from the leaf lesions. Healthy Welsh onion inoculated with conidial suspension of the fungus produced the typical symptom 10 days after inoculation. Koch's postulates were fulfilled by reisolation of the fungus. This is the first report of a Cladosporium leaf blotch of Welsh onion in Korea.

Enhanced Thermotolerance of Entomopathogenic Beauveria bassiana and Metarhizium anisopliae JEF-isolates by Substrate Modification

  • Entomopathogenic Beauveria bassiana and Metarhizium anisopliae are well-known biological control agents worldwide, and have high potential in industrialization. However, their thermo-susceptibility limits long-term storage under high temperature conditions and insecticidal activity after application in target pests. The virulences of M. anisopliae JEF-003 and JEF-004, and B. bassiana JEF-006 and JEF-007 against bean bug (Riptortus pedestris), and the thermotolerance of conidia produced on three kinds of grains, sorghum, millet and Italian millet as substrates for solid cultures were investigated. Of the three grains, Italian millet was the suitable grains in the production of thermotolerant conidia, and a significant relationship between conidial thermotolerance and the media was demonstrated. This work suggests that biological characteristics of entomopathogenic fungi are altered by culture media, and these results will be a chance to understand the factors in the media which make such changes.

Morphology and Phylogeny of Two Novel Species within the Class Dothideomycetes Collected from Soil in Korea

  • Two fungal strains (KNU-JJ-1827 and KNU-JJ-1829) belonging to the class Dothideomycetes were discovered from Jeju Island, Korea during this investigation of soil microfungi. Strain KNU-JJ-1827 showed fewer conidial septations, larger conidiogenous cells, and smaller conidia compared to the previously identified closest species of Didymocyrtis. Strain KNU-JJ-1829 revealed the similar characteristics of the nearest certain species of the genus Parathyridaria with the production of conidiogenous cells and conidia, because no asexual morphs were detected from the closest type strain Parathyridaria rosae. The novelty of the strains was also confirmed by analyzing molecular data using internal transcribed spacer regions and 28S rDNA. The molecular phylogeny also strongly support the detailed description and illustration for each proposed species as Didymocyrtis septata sp. nov. (KNU-JJ-1827) and Parathyridaria ellipsoidea sp. nov. (KNU-JJ-1829) isolated from soil in Korea.

Sporulation or Cercospora canescens Ellis & Martin in culture (녹두 갈색무늬병균(Cercospora canescens Ellis & Martin)의 분생포자 형성에 관한 연구)

  • This study was conducted to obtain a supply of conidia sufficient for screening mungbean mutant lines for a source of resistance to Cercospora leaf spot caused by Cercospora canescens Ellis and Martin. Abundant sporulation occurred in cultures on mungbean leaf decoction oatmeal agar(MOA) exposed to about 2,500 Lux of fluorescent light. but it did not occur in continuous darkness. The conditions that produced maximum number of conidia was not coincided with those for vegetative growth and pigmentation in culture medium. Removal of aerial mycelium in culture by brushing with sterile water so enhanced the conidial production that oatmeal agar medium(OA) could be useful for production of abundant conidia by the treatment.
