Genetic Polymorphisms in Plasmodium vivax Dihydrofolate Reductase and Dihydropteroate Synthase in Isolates from the Philippines, Bangladesh, and Nepal

  • Genetic polymorphisms of pvdhfr and pvdhps genes of Plasmodium vivax were investigated in 83 blood samples collected from patients in the Philippines, Bangladesh, and Nepal. The SNP-haplotypes of the pvdhfr gene at the amino acid positions 13, 33, 57, 58, 61, 117, and 173, and that of the pvdhps gene at the positions 383 and 553 were analyzed by nested PCR-RFLP. Results suggest diverse polymorphic patterns of pvdhfr alone as well as the combination patterns with pvdhps mutant alleles in P. vivax isolates collected from the 3 endemic countries in Asia. All samples carried mutant combination alleles of pvdhfr and pvdhps. The most prevalent combination alleles found in samples from the Philippines and Bangladesh were triple mutant pvdhfr combined with single mutant pvdhps allele and triple mutant pvdhfr combined with double wild-type pvdhps alleles, respectively. Those collected from Nepal were quadruple mutant pvdhfr combined with double wild-type pvdhps alleles. New alternative antifolate drugs which are effective against sulfadoxine-pyrimethamine (SP)-resistant P. vivax are required.

A Review on Clinical Studies of Herbal Treatment for Infertility Caused by Polycystic Ovary Syndrome -Focusing on Chinese Clinical Trials- (다낭성 난소증후군의 난임치료에 대한 한약임상연구 경향 분석 -중국의 임상연구를 중심으로-)

  • Objectives: The purpose of this study is to review the trend of chinese clinical studies of herbal treatment for infertility caused by Polycystic Ovary Syndrome (PCOS). Methods: We searched the clinical studies published from 1979 to 2013 through CAJ (Chinese Academic Journal) with keywords of "PCOS and 不孕" or "多囊卵 巢綜合證 and 不孕". We analyzed the intervention type of clinical studies. In addition, We researched randomized controlled trials (RCTs) published from 2009 to 2013, assessed the quality of RCTs using the Jadad Scale and reanalyzed the intervention type of RCTs. Results: One hundred and seventeen clinical studies were finally selected. Clinical studies of herbal treatment for infertility caused by PCOS increased remarkably after the 2000s in China. Combined herbal medicine and western medicine was used the most frequently. RCTs published from 2009 to 2013 were fifty. Average of Jadad score of RCTs was 1.2. Combined herbal medicine and western medicine was also the most frequently used as intervention in RCTs. Conclusions: Clinical studies on effect of combined herbal medicine and western medicine for infertility caused by PCOS were mostly published in China. But Quality of studies was generally low. Further research on effect of herbal treatment for infertility caused by PCOS is needed.

Application of the Artificial Coral Reef as a Coastal Erosion Prevention Method with Numerical-Physical Combined Analysis (Case Study: Cheonjin-Bongpo Beach, Kangwon Province, South Korea)

  • Artificial Coral Reefs (ACRs) have been introduced to help solve coastal erosion problems, but their feasibility has not been assessed with field data. This study conducted a feasibility analysis of ACRs on their erosion mitigation effects by performing a case study of Cheonjin-Bongpo beach, South Korea. A numerical-physical combined analysis was carried out using a SWAN model simulation and physical model test with a scale of 1/25 based on field observations of Cheonjin-Bongpo beach. Both Dean's parameter and the surf-scaling parameter were applied to comparative analysis between the absence and presence conditions of the ACR. The results for this combined method indicate that ACR attenuates the wave height significantly (59~71%). Furthermore, ACR helps decrease the mass flux (~50%), undertow (~80%), and maximum wave set up (~61%). The decreases in Dean's parameter (~66%) and the surf-scaling parameter suggest that the wave properties changed from the dissipative type to the reflective type even under high wave conditions. Consequently, an ACR can enhance shoreline stability.

Reconsideration on the Origination and Changes of Changbang-anchogongs in Joseon Period (조선 시대 창방안초공의 기원과 변천 재고찰)

  • The purpose of this study was to reconsider existing hypotheses on the orinination and changes of changbang-anchogongs and their types and suggest an alternative hypothesis. The earliest changbang-anchogongs in $17^{th}$ century is presumed to originate as imitations of chungbang ppaelmoks for ornamental and reinforcing parts for changbangs. They were framed whole sides of buildings equally, and were used in small numbers of extant royal buildings and Buddhist halls. From late $17^{th}$ century, enlarged dragon shape changbang-anchogongs applied to Buddhist halls maily to decorate and strengthen front side of buildings. As a result of exchange between changbang-anchogongs and Ikgong type brackets, combined changbang-anchogongs in Buddhist halls were invented for stronger connection with upper bracketing units and have been spread during $18^{th}$ century. Danpyeongbang anchogongs were originated by the connection between short pieces of pyeongbangs and early type of anchogongs similar to ppaelmoks. They belong to minority subtype, but were drawn in illustrations by mistake, as parts of main halls of royal palaces from $19^{th}$ century, because of painter's confusion about new techniques for changbang-anchogongs. Combined changbang-anchogongs for royal buildings were presumed to be invented in 1790s by stimulation from a Buddhist hall related to royal tomb. However, unlike Buddhist precedence, they were used to reinforce and decorate whole sides, even corners, of buildings, and their frameworks and shapes were imported from Daeryang-anchogongs at flanks of ritual halls in royal tombs and royal shrines from $17^{th}$ century.

  • This study has the purpose to adapt IT technology on Window Type Ventilation System for the energy saving and providing of user-centered comfortable environment. This is Derived a look at the case of the window type ventilation system and researched its IT technology for reducing energy applied to the Green Home. This indicates a solution for the established Window Type Ventilation System which can not be satisfied with user's requirement by proposing Window Type Ventilation System applied to IT technology that makes it control the intelligent, combined indoor environment system and providing information. Also, it shows energy saving efficiency of Window Type Ventilation System applied to IT technology based on the model study, analysing the performance of air-conditioning and ventilation energy saving through the experiment to compare with the established Window Type Ventilation System. The result of this study has the significance that it suggests an alternative for energy saving of housing space.

  • We observed the temperature-dependent evolution and behavior of P3HT type-II phase during a real time annealing process from a cryo-cooled low temperature in the absence and presence of an Al electrode. A poly (3-hexylthiophene) (P3HT) Type-II phase in the P3HT:PCBM films started to form near at $-10^{\circ}C$, regardless of Al layer presence. In the absence of an Al layer, type-II phase was extinct at $30^{\circ}C$. However, the extinction temperature was extended to $50^{\circ}C$ in the presence of the Al layer. Simultaneously, combined with the type-II phase, a 1:3 ordered P3HT type-II (1/3,0,0) super-lattice peak evolved. These type-II domains tended to be formed near the Al electrode layer with higher aligned status than host P3HT crystals.

A Study on the Spatial Configuration of Internal Functions of Public Area in City Hotels - Focusing on middle size first-class hotels in Korea and Japan - (도시 호텔 공용부문에 나타난 내부기능구성형식에 관한 연구 - 한국과 일본의 중규모 특급호텔을 중심으로 -)

  • The purpose of this study is focused on defining the types of spatial configuration of internal functions in city hotels by researching and analyzing city hotels in Korea and Japan. The scope of this study is limited to the period of early 1980's to the end of 1990's and 18 city hotels in Korea and Japan are selected as subject. Researching existing records and analyzing plans, surveys are used as the method of the study. The result of this study is as follow. First, block composition of floor plans that is used for hotel construction could be divided into three types (immanent type-6hotels, combined type-7hotels, adjacent type-4hotels). Second, the result of analysis about the internal functions and structural types of public area in city hotels is as follow. More Korean city hotels are taking concourse type (Korea 77.8%, Japan 55.6%) and hall type (Korea 22.2%, Japan 11.1%) than Japanese city hotels. However, the mixed type (concourse type +hall type) is only used in Japan (Korea 0%, Japan 33.3%). Third, as the result of the comparison of the size of public area between Korea and Japan, Korean hotels (36.70%) have larger space than Japanese hotels only in the service area. public (Korea 33.98% < Japan 34.71%), business (Korea 27.68% Japan 31.41%)and empty (Korea 1.64% < Japan 6.13%) area is taking larger space in Japanese city hotels than Korean.

Effect of DHEA Administration Alone or Exercise combined with DHEA before Steroid Treatment on Rat Hindlimb Muscles (스테로이드 치료 전 DHEA 단독투여와 DHEA 투여와 운동의 동시적용이 스테로이드에 의해 유발되는 쥐 뒷다리근의 위축 예방에 미치는 효과)

  • Purpose: The purpose of this study was to determine the effect of Dehydroepiandrosterone(DHEA) administration alone or exercise combined with DHEA before steroid treatment on rat hindlimb muscles. Methods: Male Sprague-Dawley rats were assigned to one of three groups: a steroid group(S, n=10) that had no treatment for 7 days before steroid treatment; a DHEA-steroid group(DS, n=8) that had 0.34 mmol/kg/day DHEA injection once a day for 7 days before steroid treatment and an exercise+DHEA-steroid group(EDS, n=9) that ran on the treadmill combined with 0.34 mmol/kg/day DHEA injection for 7 days before steroid treatment. At 15 days all rats were anesthetized and soleus, plantaris and gastrocnemius muscles were dissected. Body weight, food intake, muscle weight, myofibillar protein content and cross-sectional area of the dissected muscles were determined. Results: The DS group showed significant increases(p<.05) as compared to the steroid group in body weight, and muscle weight of gastrocnemius muscles. The EDS group showed significant increases(p<.05) as compared to the S group in body weight, muscle weight, myofibrillar protein content, and Type II fiber cross-sectional area of soleus, plantaris and gastrocnemius muscles. Conclusion: Exercise combined with DHEA administration before steroid treatment prevents steroid induced muscle atrophy, with exercise combined with DHEA administration being more effective than DHEA administration alone in preventing muscle atrophy.

Combined Treatment with Low-Level Laser and rhBMP-2 Promotes Differentiation and Mineralization of Osteoblastic Cells under Hypoxic Stress

  • BACKGROUND: The aim of this study was to evaluate the combined effect of low-level laser treatment (LLLT) and recombinant human bone morphological protein-2 (rhBMP-2) applied to hypoxic-cultured MC3T3-E1 osteoblastic cells and to determine possible signaling pathways underlying differentiation and mineralization of osteoblasts under hypoxia. METHODS: MC3T3-E1 cells were cultured under 1% oxygen tension for 72 h. Cell cultures were divided into four groups: normoxia control, low-level laser (LLL) alone, rhBMP-2 combined with LLLT, and rhBMP-2 under hypoxia. Laser irradiation was applied at 0, 24, and 48 h. Cells were treated with rhBMP-2 at 50 ng/mL. Alkaline phosphatase activity was measured at 3, 7, and 14 days to evaluate osteoblastic differentiation. Cell mineralization was determined with Alizarin red S staining at 7 and 14 days. Western blot assays were performed to evaluate whether p38/protein kinase D (PKD) signaling was involved. RESULTS: The results indicate that LLLT and rhBMP-2 synergistically increased alkaline phosphatase (ALP) activity and mineralization. Western blot analyses showed that expression of type I collagen, runt-related transcription factor 2 (RUNX2), and Osterix (Osx), increased and expression of hypoxia-inducible factor 1-alpha ($HIF-1{\alpha}$), decreased more in the LLLT and rhBMP-2 combined group than in the rhBMP-2 or LLL alone groups. Moreover, LLLT and rhBMP-2 stimulated p38 phosphorylation and rhBMP-2 and LLLT increased Prkd1 phosphorylation. CONCLUSION: Combined treatment with rhBMP-2 and LLL induced differentiation and mineralization of hypoxic-cultured MC3T3-E1 osteoblasts by activating p38/PKD signaling in vitro.

Effects of lymphocyte DNA damage levels in Korean plant food groups and Korean diet regarding to glutathione S-transferase M1 and T1 polymorphisms (건강한 성인의 glutathione S-transferase M1과 T1 유전자 다형성에 따른 한식에서의 식물성 식품군과 한식의 DNA 손상 감소 효과)

  • Purpose: GST (glutathione S-transferase) M1 and T1 gene polymorphisms are known to affect antioxidant levels. This study was carried out to evaluate genetic susceptibility by measuring the effect of DNA damage reduction in the Korean diet by vegetable food according to GST gene polymorphisms using the ex vivo method with human lymphocytes. Methods: Vegetable foods in the Korean diet based the results of the KNHANES V-2 (2011) were classified into 10 food groups. A total of 84 foods, which constituted more than 1% of the total intake in each food group, were finally designated as a vegetable food in the Korean diet. The Korean diet applied in this study is the standard one-week meals for Koreans (2,000 Kcal/day) suggested by the 2010 Dietary Reference Intakes for Koreans. Ex vivo DNA damage in human lymphocytes was assessed using comet assay. Results: In the Korean food group, the DNA damage protective effect of GSTM1 and GSTT1 was found to be greater in mutant type and wild-type, respectively. and the DNA damage protective effect according to the combined genotype of GSTM1 and GSTT1 was different depending on the food group. On the other hand, in Korean Diet, the DNA damage protective effect appeared to be larger in GSTM1 wild-type than in mutant type and was found to not be affected by GSTT1 genotype. Conclusion: These results can be used as basic data to demonstrate the superiority of the antioxidant function of Korean dietary patterns and food groups. Furthermore, it may be a starting point to begin research on customized antioxidant nutrition according to individual genes.
