Cytotoxic Effects of Methanol Extract and Fractions from Echinacea angustifolia on Cancer Cells (암세포에 대한 Echinacea angustifolia 순차 용매 추출물의 세포독성 효과)

  • Echinacea is a North American native medicinal herb used traditionally for wounds, burns, snake or insect bites, colds, infections, and inflammation by indigenous Americans. We investigated the effects of the root and stem of fresh Korean-grown Echinacea angustifolia methanol extracts and fractionation extracts on the cytotoxicity against cancer cells (HL60, 3LL). The extracts were prepared by step-wise fractionation of methanol extracts of Echinacea angustifolia using hexane, chloroform, ethyl acetate, buthanol, and water. From the tests, root and stem parts of Echinacea showed the cytotoxic effect on cancer cells. The cytotoxie rate of the ethyl acetate fraction of the root parts showed 79% against HL60 cells at low concentration (0.125 mg/mL), and hexane fraction of the root and stem parts gradually increased as the concentration of samples increased, and the root parts showed 82% at 1.0 mg/mL concentration against HL60 cells, chloroform fraction of the root part showed 78.4% against HL60 cells and 68.4% on 3LL cells at 1.0 mg/mL concentration, water and butanol fraction of these results, it is considered that ethyl acetate fraction of the root and stem parts showed 60.1% to 77.1% against HL60 cells, after testing by MTT assay system. From these results, it is considered that ethyl acetate fraction of the Echinacea angustifolia root parts has stronger anticancer effects than any other fractions in vitro.

Extraction of Crude-BMP from Bovine Cortical Bone for Bone Grafts (골이식물로서의 소뼈 치밀골에서 Crude-BMP의 추출)

  • We tried to extract bone morphogenetic protein (BMP) from the freeze-dried bovine cortical bone (FBCB) for bone graft, which were defatted with chloroform-methanol for 20 days, freeze-dried at $-80^{\circ}C$ for 7 days and sterilized by ethylene oxide gas. Two kg of FBCB were pulverized in a wheel mill to $0.5-2.0mm^3$ cubic in size. The bone particles were demineralized in 0.6N HCI for 10 days at chloroform-methanol$4^{\circ}C$ and defatted with chloroform-methanol for 6 hours at room temperature, which was going to be defatting and demineralized cortical bone (DDM). For extracting BMP, DDM was agitated continuously through 72 hours with magnetic stirrer at $4^{\circ}C$ into 12 times of volume of 6 M guanidine hydrochloride (Gdn-HCl) solution containing proteinase inhibitors to protect BMP such as 2mM N-ethylaleimide, 1mM iodoacetic acid, 1mM phenylmethylsulfonyl fluoride and a sterilizer, 1mM sodium azide. The extraction procedure was repeated for three times. All extracted solution was centrifuged at 10,000 rpm for 30 min and then, the supernatant was dialyzed with 12 times of volume of deionized water at $4^{\circ}C$ for 24-72 hours, which cut off below 6,000-8,000 molecular weight. The dialyzed specimen contained crude-BMP was centrifuged, freeze-dried, and weighted. Through these processing, we could obtained $84.9\%$ as FBCB, $17.8\%$ as DDM and $0.71\%$ as crude-BMP from the wet cortical bone without cancellous bone, marrow and muscles. The crude-BMP were obtained $68.3\%$ from the first extraction, $29.6\%$ from secondary and $2.1\%$ from tertiary, respectively. It was suggested that high yield of crude-BMP migth be explained by three-time repetition of the extraction processing for crude-BMP with Gdn-Hcl sol.

Characteristics of Nitrite Scavenger Derived from Seeds of Cassia tora L. (결명자 추출물의 아질산염 소거인자의 특성)

  • The present study was conducted to elucidate nitrite scavenger derived from seeds of Cassia tora L. Methanolic extract of Cassia tora L was refractionated into ethyl ether, chloroform, ethyl acetate and water farction, and nitrite scavenging abilities of these fractions were investigated. Among these fractions, ethyl acetate fraction had the strongest nitrite scavenging ability(97%/2 mg). Therefore, to further investigate nitrite scavenger, ethyl acetate fraction was fractionated by silica gel column chromatography with a chloroform-methanol($10:0{\sim}0:10$) and then compound A and B were isolated. Compound A had a stronger nitrite-scavenging effect than compound B. Therefore the further separation of compound A was carried out by HPLC(32% acetonitrile. 1 ml/min) using ${\mu}Bondapak$C18 column$(3.9{\times}300 mm)$ and finally compound A-1 was obtained from compound A. Compound A-1 had by far nitrite-scavenging ability as compared with that of ascorbic acid. Compound A-1 was identified as $nor-rubrofusarin-6-{\beta}-mono-D-glucoside$ from the profiles of UV, IR and $^1H-NMR$.

Effect of Complex Extracts of Mushroom and Sharp Toothed Eel on the Skin Conditions with Atopic Dermatitis and Acne Symptoms (갯장어 및 버섯 혼합추출물의 아토피 및 여드름 피부개선 효과)

  • This study was conducted to investigate effect of complex extract of various mushrooms and sharp toothed eel on the skin conditions with atopic dermatitis and acne symptoms. The total phenol concentration was increased in order of hot water> ethanol> ethyl acetate> petroleum ether> chloroform extract. Especially, when the hot water extract was used, it was about 2-3 fold higher than that of ethyl acetate, petroleumether, and chloroform extract. When the complex extract concentration was increased from 1.0 to 50 mg/L, the DPPH scavenging rate increased from 10.1 to 81.4%. The reduction power was sharply increased from 0.05 to 0.27 (700 nm) when the complex extract concentration was increased from 25 to 75 mg/L. However, above 100 mg/L, it was not decreased. In the case of SOD-like activity, it was 45.7% at 100 mg/L. Total numbers of patients with atopic dermatitis were 15 and 5 patients with severe acne symptoms. According to photos taken before and after the treatment and questionnaire results, considerable improvements in skin conditions are observed in the patients with atopic dermatitis and acne. For atopic patients, erythema and edema have been improved but the degree of effect was dependent on the individual's constitution. Concerning acne, the effect of coating of the extracts was prominent for first week and the degree decreased with time till 4 weeks. The complex extracts wereefficient in soothing rash and maturation. Side effects such as a scar were not detected during the application and treatment.

Repellent and Insecticidal Activity of Sequential Extracting Fractions Obtained from BPH-Resistant Rice Varieties against Brown Planthopper (Nilaparvata lugens) (벼멸구 저항성벼 품종 추출분획물의 기피 및 살충 활성)

  • Rice plant extracts of brown planthopper (BPH) resistant rice varieties, Jangseongbyeo (JSB) and Hwacheongbyeo (HCB) at different growth stages (seedling, tillering, heading and ripening) were sequentially fractioned using hexane, ethyl ether, ethyl acetate, butanol, and distilled water. The extracts were applied to BPH susceptible rice variety, Dongjjnbyeo (DJB), to investigate the insecticidal and repellent effects against BPH. BPH insecticidal effects were not clearly observed with almost all of the extract fractions obtained from both JSB and HCB varieties for 12 h, whereas the ethyl ether and hexane extract fractions showed about 10 to 30% of BPH mortality in 24 to 48 h of application periods. An effective BPH repellent activity was found with the applications of ethyl ether extract fractions obtained from JSB variety. The extract fractions obtained from HCB variety did not show any different repellence among the various fractions. The BPH repellent effects of the extract fractions obtained at different growth stages of either JSB or HCB varieties did not show any correlations. The effect of ethyl ether fraction on BPH repellent was continually increased by 30 h after treatment and thereafter decreased. In addition, the first sub-fraction separated by a flash column chromatography eluted with chloroform:methanol (9:1, v/v) from the BPH effective ethyl ether faction in JSB variety might be meaningful to repel BPH from BPH susceptible target rice plants. The results indicated that the ethyl ether fraction obtained from JSB was higher in repellent activity than in insecticidal activity, and suggesting that there might be specific substance(s) in the first sub-fraction (sF1) of the ethyl ether fraction in JSB that could provide repellent activity against BPH.

Antioxidant and α-Glucosidase Inhibition Activities of Solvent Fractions from Methanolic Extract of Sericea Lespedeza (Lespedeza cuneata G. Don) (비수리(Lespedeza cuneata G. Don) 메탄올 추출물로부터 분획된 용매분획물의 항산화활성과 α-Glucosidase 저해활성)

  • The purpose of this study was to evaluate the antioxidant contents and antioxidant activities of solvent fractions from methanolic extract of sericea lespedeza. To determine the antioxidant compounds in solvent fractions from methanolic extract, total polyphenolic, flavonoid, tannin, and proanthocyanidin contents were measured by spectrophotometric methods. Solvent fractions were evaluated for antioxidative capacity according to DPPH and ABTS radical scavenging activities. Total polyphenolic contents were 12.44, 3.61, 6.39, 27.11, 20.00, and 9.32 mg gallic acid equivalent (GAE)/g extract residue (ER), respectively. Total flavonoid contents were 2.94, 9.92, 7.77, 9.27, 5.11, and 2.66 mg catechin equivalent (CE)/g ER, respectively. Total tannin contents were 8.75, 10.04, 7.42, 17.32, 11.65, and 7.61 mg tannic acid equivalent (TAE)/g ER, respectively. Total proanthocyanidin contents were 346.09, 63.50, 103.76, 288.62, 231.99, and $358.48{\mu}g$ CE/g ER, respectively. DPPH radical scavenging activities of solvent fractions from methanolic extract of sericea lespedeza were 20.62, 5.16, 9.29, 20.80, 20.00 and 20.79 mg Trolox equivalent (TE)/g ER, and ABTS radical scavenging activities were 33.86, 9.24, 17.36, 33.76, 33.49, and 33.86 mg TE/g ER, respectively. SOD-like activities were 4.12, 0.61, 2.01, 9.89, 13.47, and 11.82 units/mL, and ${\alpha}$-glucosidase inhibition activities were 93.85 and 61.64% at concentrations of 50 and $25{\mu}g/mL$ in the water fraction, respectively. The results of this study show that notable antioxidant activities in sericea lespedeza have significant health benefits.

In vitro and Ex vivo Supplementation of Houttuynia cordata Extract and Immunomodulating Effect in Mice (어성초 추출물 첨가가 마우스 면역능 증진에 미치는 영향)

  • Houttuynia cordata THNUB (He; Uh-Sung-Cho) is a medicinal plant which has been widely used as a component of blood-building decoctions. This study was performed to investigate the immunomodulative effect of He in mice. In vitro experiment, the mice splenocytes proliferation and three kinds of cytokines (IL-1$\beta$, IL-6, TNF- $\alpha$) production by mice peritoneal macrophages cultured with six (methanol, hexane, chloroform, ethylacetate, butanol and water) fractions of He were used to indicate the immunomodulative effect. Ex vivo experiment, the different concentrations of He water extract was orally administrated every other day for two weeks. The production of cytokines IL-1$\beta$, IL-6, TNF- $\alpha$) secreted by activated macrophages and the mice splenocytes proliferation were used as an index for the immunocompetence. The supplementation of all six fractions of He enhanced the splenocytes proliferation at the level of 6.58$\pm$1.23∼47.82$\pm$5.48 compared to that of control in the range of 1∼50 $\mu\textrm{g}$/mL. IL-1$\beta$ production was significantly increased with the supplementation of chloroform and water extract of He. Higher level of IL-6 production was detected by the supplementation of ethylacetate, butanol and water extract. TNF - $\alpha$ production was enhanced by the supplementation of all six fractions of He. From the ex vivo study, the highest proliferation of splenocytes was seen from the mice orally administrated with the He water extract at the concentration of 500 mg/kg bw In case of cytokines production, IL-1$\beta$, IL-6, and TNF- $\alpha$ release by activated peritoneal macrophages were augmented by the oral administration of He water extract. These results indicated that He may enhance the immune function by regulating the splenocytes proliferation and cytokines production capacity in mice.

Effect of Extracts produced from Gardenia jasminoides Seed Using Different Types of Solvents on Nitrogen Oxide Scavenging Activities and Lipid Peroxidation Inhibition (치자(Gardenia jasminoides Ellis) 씨 추출물의 질소산화물 소거능 및 지질과산화 저해능에 관한 연구)

  • The object of this study was to measure the bioactivity and antioxidant activity of seed from Gardenia jasminoides Ellis fructus (GJE). GJE seeds were performed the extraction of them by chloroform:methanol (CM, 2:1, v/v), 70% ethanol and n-butanol. Sequentially, total phenol content, nitrogen oxide scavenging activity, antioxidant activity and lipid peroxidation inhibition activity of the extracts were investigated. Solvent extract bioactivity of increasing concentrations (0.2, 0.4, 0.6 mg/mL) were significantly increased (p<0.05). GJE seed extracts showed lower activity than positive control (ascorbic acid, BHA, trolox). The highest concentration of CM extracts was obtained in the same manner as the results of analysis of the total phenol contents of the GJE seed, and 70% ethanol extract showed the highest activity of reducing power. The water soluble carotenoids crocin and flavonoid were effective. As a result of this experiment. the seeds of GJE showed excellent antioxidant, and lipid peroxidation inhibitory properties.

Antioxidant Component and Activity of Dropwort (Oenanthe javanica) Ethanol Extracts (미나리 에탄올 추출물의 항산화성분과 항산화활성)

  • This study was conducted to investigate the antioxidant compounds and antioxidant activity on the dropwort (Oenanthe javanica) and its solvent fraction. The dropwort was extracted with 70% (v/v) ethanol, and then partitioned using the solvents of hexane, chloroform, ethyl acetate, butanol, and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid of 240.61 mg GA eq/g and 105.57 mg catechin eq/g, followed by ethanol extract of 37.50 mg GA eq/g and 26.50 mg catechin eq/g, respectively. The DPPH radical scavenging activity ($IC_{50}$) on the solvent fractions increased in the order of ethyl acetate, butanol, ethanol extract, chloroform, aqueous, and hexane with 0.08, 0.58, 1.07, 2.43, 2.47, and 3.31 mg/mL, respectively. The ABTS radical scavenging activity was the highest value of 382 mg AA eq/g in ethyl acetate fraction. Reducing power and chelating effect on the ethanol extracts and its solvent fraction were in range of 0.23~0.75 and 0~32.01%, respectively. Hydroxyl radical scavenging activity ($IC_{50}$) was the lowest value of $26.71\;{\mu}g$/mL in ethyl acetate fraction.
