식이섭취 빈도수가 흰쥐의 성장 및 각 기관의 조성에 미치는 영향 (The Effect of Feeding Frequency on the Growth and Composition of Individual Organs in Rats)

  • 동물의 성장정도에 따라 식이섭취양식이 성장및 각 기관의 DNA와 RNA함량에 미치는 영향을 경토하기 위하여 , 성장기와 성장완료기의 흰쥐에게 ad-lib. feeding과 1일 3회 급여 그리고 1일 1회 급여등 3가지 섭취 양식으로 정제 식이를 급여하여 비교하였으며, 그 결과는다음과 같다. 1 ) 모든 실험동물에서 3희 식이군은 ad-lib. 군과 비슷한 증체량을 나타냈으나, 1회 식이군은 다른 군보다 낮은 증체량을 나타냈으며, 식이섭취량도 더 적었다. 2) 성장기 쥐의 경우. 1회 식이군은 ad-lib.군에 비해 간, 신장, 비장 등 내장기관의 DNA, RNA, 단백질 함량이 강소되었으나, DNAmg당 각 조성은 다른군과 차이가 없었다. 한편, 성인쥐의 경우는 1회 식이군에서, 간의 크기가 현저히 증가되었으며 DNAmg당 각 조성은 증가하였으나, 간의 DNA함량은 유의적 차이가 없었다. 신장과 비장은 식이섭취양식에 의한 유의적 차이를 보이지 않았다. 3) 성장기 쥐와 성인쥐의 위와 소장은 meal-feeding에 의해 크기가 증대된 경향을 나타냈으며, 혈청콜레스테롤은 유의적 차이가 없었다. 본 결과는 식이섭취양식이 성장기 동물에서는 내장기관의 세포수에 크게 영향을 미치며, 성장완료기 동물에서는 내장기관중 특히 간의 세포크기에 영향을 미침을 시사한다.

표고버섯 열수 추출물이 발암원을 급여한 흰쥐의 간 기능 관련 효소활성에 미치는 영향 (Effects of Hot Water Extracts from Lentinus edodes on Hepatic Functional Enzyme Activities in the Rat Fed Butter Yellow(p-Dimethylaminoazobenzene))

  • 표고버섯의 열수 가용성 다당류(PS)와 간 장해물질이며 발암성을 갖는 butter yellow를 흰쥐의 식이에 6주간 투여하여 혈장과 간의 효소활성 및 과산화지질의 함량을 측정한 결과는 다음과 같다. 1. 혈장 중 GOT 활성은 better yellow 첨가군(BO군)보다 PS의 투여군(BP군 : BO+PS)이 비교적 낮은 수준을 나타내었으며 ${\gamma}$-GTP와 GPT 활성에서도 BO군에 비하여 BP군에서 유의하게 낮은 활성을 나타냈다. 2. 간장 중 glutathion peroxidase, catalase 및 lactate dephdregenase 활성 또한 기초식이군(NO군)에 PS를 토여한 군(NP군)은 NO군과 유의한 차이가 없었으나 BP군은 BO군에 비하여 유의하게 낮은 수준이었다. SOD활성은 각 군간에 유사한 수치로 유의적인 차리를 보이지 않았다. 3. 간장의 microsomal 분획분 및 cytosol 분획분 중의 glutathione S-transferase 활성도는 BP군이 BO군보다 유의하게 낮은 수준을 보였으나 better yellow를 첨가하지 않은 NO군에 비하여 상당히 높은 수준을 나타내었다. 4. 간장 내 glutathione과 malondialdehyde의 함량 또한 PS의 투여로 생성이 저하되는 것으로 나타났다. 이상의 결과를 보아 표고버섯의 열수 추출물은 생체조직의 지질의 과산화를 방지하므로써 better yellow에 의한 간장장애의 치료 또는 독성을 완화하는 효과가 있는 것으로 사료된다.

넙치 치어의 사료 내 myo-inositol 요구량 (Myo-inositol Requirement in Diets for Juvenile Olive Flounder (Paralichthys olivaceus))

  • 본 연구는 넙치 치어에 있어서 수용성 비타민인 myo-inositol의 사료 내 적정 요구량을 평가하고, 결핍 시 어류에 미치게 되는 생리적 영향을 조사하고자 수행 되었다. 실험에 사용된 어류는 평균무게가 1.22 g인 넙치 초기치어를 사용하였으며, 총 15개의 35 L 원형수조에 각 수조당 48 마리씩(3반복구) 무작위 배치하였다. 반 정제 사료원을 기초로한 총 5개의 실험사료는 52%의 조단백질과 18.3 MJ/kg diet의 에너지함량을 갖도록 조성되었고 myo-inositol 함량을 각각 0, 0, 400, 800, 1600 mg/kg diet으로 기초사료에 첨가하였다(M0, M0+, M400, M800, M1600). 실험사료 중 M0+ 사료는 항생제인 tetracycline hydrochloride 및 (SIGMA, USA)를 실험사료에 0.4%로 첨가함으로써, myo-inositol이 넙치의 장내 미생물들에 의하여 합성되는지를 알아보기 위해 설계되었다. 26주간의 사료공급 실험결과, 사료 내 myo-inositol 첨가에 의한 어류의 성장률 결과들에서 유의적인 차이가 나타났으며, myo-inositol 결핍사료를 섭취한 실험어류에서 성장지연과 같은 결핍증상을 보였다. Hematocrit과 Hemoglobin을 측정한 혈액분석과 간 지방함량 분석에서는 실험구 사이에서 차이가 나타나지 않았으나, 전어체 지방분석 결과에서는 단지 M0와 M400 사이에서 유의적인 차이를 나타내었다. 초기 넙치 치어의 myo-inositol 요구량은 성장률을 기초로 한 broken-line regression model (Robbins, 1986)에서 반 정제사료를 기초로 하였을 때 800 mg/kg로 결정되었다.

  • On the assumption that the supplementation of certain nutrients or foods to the rice diet (low protein, low fat, and low vitamins) may decrease, to some extent, the degree of suffering from abnormal environments, such as vibration, noises, gases, dusts, smog etc. a series of experiments were started. As the first report the nutrition under vibration was studied in this experiment. Sixty (60) young growing male rats weighing about 65 grams were used, grouping to five (5) groups, twelve (12) rats each group. They were fed on the following five (5) experimental diets: rice diet (basal diet), rice diet+casein, rice diet+vitamins, rice $diet+{\alpha}-tocopherol$, and rice diet+ginseng powder (see the tables 1 and 2) for the period of 14 weeks experiment. During the experiment period the half number of the rats of each group were exposed to the three (3) hours vibration every day. The protective effect of each diet against the vibration may be summarized as follows. 1. The growth of rice diet group was impaired significantly under vibration, However, those of other groups (protein-supplemented, vitamin-supplemented, ${\alpha}-tocopherol-supplemented$ and ginseng-supplemented groups) were impaired much less compared with rice diet group. 2. The feed efficiency of the rice diet group was decreased significantly under vibration. It is estimated that the biological availability of nutrients was impaired under this environment. On the other hand, the feed efficiencies of protein supplemented, of vitamin supplemented, and of ginseng supplemented groups were not decreased under vibration, statistically. 3. There is tendency that the food spillages of vibration groups are higher than those of non-vibration groups. Especially it seems true in the case of rice diet group. The food spillage may be, to some extent, related with mental nervousness of animals. From the point that the food spillage of ginseng supplemented group is significantly lower than those of other groups it is thought ginseng acts some good role in protecting nervous system from suffering from vibration. 4. In all groups except protein supplemented group, liver fat of vibration group tends to be higher than that of non-vibration group. 5. It shows that, in general, the serum alkaline phosphatase activity of the vibration group is significantly higher than that of the non-vibration group. It seems that there may be, to some extent, corelation between the amount of liver fat and serum alkaline phosphatase activity. 6. There is tendency that, in rice diet group, the organs of vibration group are smaller than those of non-vibration group, especially lung is so. It is thought that this may be due to the poor growth of whole body size in vibration group.

높은률의 단백질 함유 식이와 지방 세포의 지방축적과 상호 관계(II) -동 열량의 저, 중, 고 단백식이가 흰쥐의 간 세포활성에 미치는 영향- (Relationship between Higher Protein Contents in the Diet and Adipose Tissue Fat Accumulation (II) -Effect of isocaloric low, medium and high protein diets on the cellular activities of rat liver-)

  • 카제인 8%, 13% , 18% 수준의 동열량 함유 식이를 8%를 기준으로 하여 1일동일 열량 섭취하도록 급식시킨 이유쥐의 성장, 지방축적, 에너지 소비를 관찰하였다. 30일 급식시켰을 때 낮은 수준의 단백식이 (8%)군의 몸무게는 높은 수준(18%)의 단백섭취군 보다 43.01g더 가벼운 것으로 나타났다(최종 몸무게증가 ; ${85.57}{\pm}{7.5g}$${128.58}{\pm}{11.64g}$, p<0.001). 도살체당 지방축적량은 8%단백식이군의 작은 몸의 크기에도 불구하고, 13%,18% 단백식이군과 비교할 때 차이를 보이지 않았다. 이와 반대로, 체질소 함량은 13%와 18% 단백식 군들이 8% 보다 훨씬 높았다. 총 실험기간(32일)동안 8%, 13%, 18%군의 에너지 소비는 각기 4,576.61kJ, 5,440.80kJ, 5,6067.67kJ로 추정되었다. 낮은 수준의 단백 식이군이 섭취한 과다에너지는 손실된 것으로 보인다. 간세포의 malic enzyme활성은 식이변화에 따라 변동이 없는 것으로 나타났다. 이러한 실험결과들로 부터, 낮은수준의 단백식이의 성장지연은 성장에 필요한 열량부족에서 온 것이 아니라, 질소공급의 부족에서 초래된 것이며, 단백질 급여 제한은, 에너지 섭취를 같게한 고단백군들과 비교할 때, 지방이외 무게는 감소시키는 반면, 체지방 합성과 축적능력에는 영향을 미치지 않는다는 결론을 얻었다.

Modulated Gene Expression of Toxoplasma gondii Infected Retinal Pigment Epithelial Cell Line (ARPE-19) via PI3K/Akt or mTOR Signal Pathway

  • Due to the critical location and physiological activities of the retinal pigment epithelial (RPE) cell, it is constantly subjected to contact with various infectious agents and inflammatory mediators. However, little is known about the signaling events in RPE involved in Toxoplasma gondii infection and development. The aim of the study is to screen the host mRNA transcriptional change of 3 inflammation-related gene categories, PI3K/Akt pathway regulatory components, blood vessel development factors and ROS regulators, to prove that PI3K/Akt or mTOR signaling pathway play an essential role in regulating the selected inflammation-related genes. The selected genes include PH domain and leucine- rich-repeat protein phosphatases (PHLPP), casein kinase2 (CK2), vascular endothelial growth factor (VEGF), pigment epithelium-derived factor (PEDF), glutamate-cysteine ligase (GCL), glutathione S-transferase (GST), and NAD(P)H: quinone oxidoreductase (NQO1). Using reverse transcription polymerase chain reaction (RT-PCR) and quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR), we found that T. gondii up-regulates PHLPP2, $CK2{\beta}$, VEGF, GCL, GST and NQO1 gene expression levels, but down-regulates PHLPP1 and PEDF mRNA transcription levels. PI3K inhibition and mTOR inhibition by specific inhibitors showed that most of these host gene expression patterns were due to activation of PI3K/Akt or mTOR pathways with some exceptional cases. Taken together, our results reveal a new molecular mechanism of these gene expression change dependent on PI3K/Akt or mTOR pathways and highlight more systematical insight of how an intracellular T. gondii can manipulate host genes to avoid host defense.

  • 최근 성인병 중에서 가장 높은 빈도로 발병하는 것이 고혈압인데, 이 고혈압의 예방하기 위해서 가장 손쉽게 일상적으로 섭취할 수 있는 것이 바로 우유의 성분인 카제인염이다. 일반적으로 우유 펩타이드는 혈압조절자인 ACE를 방해함으로써 혈압을 줄여주는 항고혈압 효과와 관련이 있으며, 또한 혈액의 혈소판 응집을 저해해 심근경색이나 뇌경색의 원인인 혈전을 방지한다라고 알려져 있다. 따라서 이러한 기능을 가지고 있는 우유의 카제인염을 다양한 단백질가수분해 효소로 처리하여 가수분해 시간에 따른 특성 그리고 ACE 저해효과가 높은 가수분해물을 제조하기 위해서 대표적인 막여과 방법인 한외여과법이 이용되고 있는 것을 살펴보았다. 향후 가수분해물들의 (1) 항산화 효과, 세포독성 및 용해성, 기포성 등이 더 향상될 수 있도록 효소선발과 효소처리에 대한 폭 넓은 조사와 연구, (2) 고품질 대량생산법 확립, 그리고 (3) 건강기능성식품으로의 산업화 이용이 가능하도록 지속적인 기초 연구가 신속히 진행되어야 할 것이다.

Effect of De-hulling on Ileal Amino Acids Digestibility of Soybean Meals Fed to Growing Pigs

  • A study was carried out to determine the effect of de-hulling on apparent and true ileal amino acids digestibility of soybean meals for growing pigs. Twenty barrows (Duroc${\times}$Large white${\times}$Longer white) were fitted with a simple T-cannula at the distal ilium. Digestibility of 20 experimental diets was determined, nine of them were de-hulled soybean meal diets, and nine of them were regular soybean meal diets and two low protein casein diets for determination of endogenous amino acid correction for true digestibility determination. A TEX>$5<{\times}5<$ Latin Squares Design was adopted in this trail. The results showed that de-hulling increased apparent ileal digestibility of isoleucine, threonine, aspartic, tyrosine and indispensable and dispensable amino acid (p<0.05) in soybean meals. Furthermore, dehulling is also increased apparent digestibility of arginine, leucine, lysine, phenylalanine, alanine, glutamic acid, serine and gross amino acids (p<0.01). However, there were no significant differences found for histidine, methionine, tryptophan, cystine and glycine (p>0.05). Similar responses were found for true ileal digestibility. In three dehulled and non-dehulled pairs soybean meals from the same respective sources, de-hulling increased apparent digestibility of lysine, methionine, threonine and cystine 1.42%, 2.06%, 2.18% and 1.40% respectively. True digestibility of lysine, methionine, threonine and cystine was increased 1.65%, 1.94%, 2.30% and 1.82% respectively. A prediction equation for true ileal amino acid digestibility (including lysine and arginine) was established by multivariate linear regression. The independent variables included relevant amino acid, organic matter, crude protein, ether extract and nitrogen free extract. The coefficient R2 values of lysine and agrinine were 0.596 and 0.531 respectively. According to the crude protein content, a prediction equation for lysine and arginine content in soybean meal was also established by single linear regression. The coefficient $R^2$ values of lysine and agrinine were 0.636 and 0.636 respectively.

Re-evaluation of the Optimum Dietary Vitamin C Requirement in Juvenile Eel, Anguilla japonica by Using L-ascorbyl-2-monophosphate

  • This study was conducted to re-evaluate the dietary vitamin C requirement in juvenile eel, Anguilla japonica by using L-ascorbyl-2-monophosphate (AMP) as the vitamin C source. Five semi-purified experimental diets were formulated to contain 0 ($AMP_0$), 30 ($AMP_{24}$), 60 ($AMP_{52}$), 120 ($AMP_{108}$) and 1,200 ($AMP_{1137}$) mg AMP $kg^{-1}$ diet on a dry matter basis. Casein and defatted fish meal were used as the main protein sources in the semi-purified experimental diets. After a 4-week conditioning period, fish initially averaging $15{\pm}0.3$ g (mean${\pm}$SD) were randomly distributed to each aquarium as triplicate groups of 20 fish each. One of five experimental diets was fed on a DM basis to fish in three randomly selected aquaria, at a rate of 3% of total body weight, twice a day. At the end of the feeding trial, weight gain (WG) and specific growth rate (SGR) for fish fed $AMP_{52}$ and $AMP_{108}$ were significantly higher than those recorded for fish fed the control diet (p<0.05). Similarly, feed efficiency (FE) and protein efficiency ratio (PER) for fish fed $AMP_{52}$ were significantly higher than those for fish fed the control diet (p<0.05). Broken-line regression analysis on the basis of WG, SGR, FE and PER showed dietary vitamin C requirements of juvenile eel to be 41.1, 41.2, 43.9 and 43.1 (mg $kg^{-1}$ diet), respectively. These results indicated that the dietary vitamin C requirement could range from 41.1 to 43.9 mg $kg^{-1}$ diet in juvenile eel when L-ascorbyl-2-monophosphate was used as the dietary source of vitamin C.

Betulin suppressed interleukin-1β-induced gene expression, secretion and proteolytic activity of matrix metalloproteinase in cultured articular chondrocytes and production of matrix metalloproteinase in the knee joint of rat

  • We investigated whether betulin affects the gene expression, secretion and proteolytic activity of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well as in vivo production of MMP-3 in the rat knee joint to evaluate the potential chondroprotective effect of betulin. Rabbit articular chondrocytes were cultured and reverse transcription-polymerase chain reaction (RT-PCR) was used to measure interleukin-$1{\beta}$ ($IL-1{\beta}$)-induced gene expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 and type II collagen. Effect of betulin on IL-$1{\beta}$-induced secretion and proteolytic activity of MMP-3 was investigated using western blot analysis and casein zymography, respectively. Effect of betulin on MMP-3 protein production was also examined in vivo. The results were as follows: (1) betulin inhibited the gene expression of MMP-3, MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5, but increased the gene expression of type II collagen; (2) betulin inhibited the secretion and proteolytic activity of MMP-3; (3) betulin suppressed the production of MMP-3 protein in vivo. These results suggest that betulin can regulate the gene expression, secretion, and proteolytic activity of MMP-3, by directly acting on articular chondrocytes.
