Safety and Risk Assessment of 3-Monochloro-1,2-propanediol (3-MCPD) (3-Monochloro-1,2-propanediol(3-MCPD)의 안전성 및 위해성 평가)

  • 3-Monochloro-1,2-propanediol(3-MCPD) is currently being a matter of concern because of its toxicity. 3-MCPD produced during the acid hydrolysis of soybean products has been reported to be mutagenic, neurotoxic, nephrotoxic and spermatotoxic. Howerer, the carcinogenicity of 3-MCPD is a controversial issue over the past several decades. 3-MCPD characteristically showed a variety of toxicities in reproductive system such as, decrease in sperm number and sperm motility, infertility, loss of sperm function, and weight decrease in ovary. Due to the toxicity of 3-MCPD, exposure to 3-MCPD has been proposed to be reduced to as low a level as technologically feasible. 3-MCPD can be detected in soy sauce or non-soy sauce products. The legal limit for 3-MCPD this year has been suggested to be 20 ppb($\mu\textrm{g}$/kg)in the European Community. In Korea, the permissible level of 3-MCPD is expected to be 0.3 ppm. In this study, 3-MCPD was toxicologically evaluated in terms of risk assessment in humans.

Spontaneous Multicentric Malignant Schwannoma in a Male Fischer 344 Rat

  • We describe here a multicentric spontaneous malignant schwannoma obtained from one male F344 rat, and this animal was the subject of a carcinogenicity study for which it was treated with diisodecyl phthalate. The animal of the control group not treated with diisodecyl phthalate showed dyspnea and severe lordosis. On the necropsy, two tan, firm, encapsulated masses were observed in the subcutis of the lumbosacral region and the left inguinal region of the abdominal cavity, respectively; the masses were $25{\times}17{\times}8$ mm and $16{\times}14{\times}8$ mm in size, respectively. Histologically, the tumor consisted of spindle and pleomorphic cells that grew in various patterns, that was, sweeping fascicles and herringbone and local organoid patterns. The pleomorphic neoplastic cells had more than two nuclei. Additionally, the diagnosis of malignant schwannoma was confirmed by the immune reactivity of the tumor cells for S-100 protein.

Effect of Genotoxicity or Carcinogenecity Chemicals on the ROS Production (유전독성, 발암성 화학물질이 ROS 생성에 미치는 영향)

  • In the present study, ROS detection of L5178Y cells that were treated with twenty test compounds in order to find out hydrogen peroxide ($H_2O_2$) induction for genotoxicity and carcinogenic toxicity. Twenty test compounds were consist of four classes, such as genotoxic carcinogens, genotoxic noncarcinogens, nongenotoxic carcinogens, and nongenotoxic noncarcinogens. Genotoxic carcinogens are 1,2-dibromoethane, glycidol, melphalan, diethylstilbestrol and urethane. Genotoxic noncarcinogens are 8-hydroxyquinoline, emodin, acetonitrile and diallylphthalate, L-ascorbic acid. Nongenotoxic carcinogens are methyl carbamate, O-nitrotoluene, 1,4-dioxane, tetrachloroethylene and 2,3,7,8-tetrachlorodibenzo-p-dioxin. And nongenotoxic noncarcinogens are D-mannitol, 1,2-dichlorobenzene, caprolactam, bisphenol A and chlorpheniramine maleate.

Importance of Peer Review in Toxicological Pathology and Its Practical Approach (독성병리 Peer Review의 중요성과 실제적 접근방법)

  • Evaluation of toxicological pathology is to some extend subjective judgments by pathologist and the accuracy of pathologist's works is based on the individual training and experiences. It has been required to establish a peer review system for toxicologic pathology and these review system has been employed by various practice of toxicological pathology. It would be pointed out that the possible causes of drifts in pathology are due to 1) lack of knowledge or experience of individual pathologists, 2) poorly maintained consistence of grading among animals in study, 3) different interpretation of findings between pathologists, or 4) pathology data processing. Example cases of diagnostic errors and current practice of peer review including tissue selection criteria, documentation and problem resolution for short-term and carcinogenicity studies were introduced. For sound regulatory system and high integrity of practice in toxicological pathology, current approaches of peer review system were reviewed.

A Comparative Study of Radiation and Gas Sterilization. (방사선항균법과 개스항균법의 비교연구)

  • Ethylene oxide gas has been used as a cold sterilant for heat-sensitive medical equipments and as a fumigant for food for more than 30 years, and it is used more widely than radiation although radiation sterilization has made significant inroads in recent years. But according to recent studies of toxicities such as mutagenicity, haemolytic effect and possible carcinogenicity of Ethylene oxide (ETO) and its two main reaction products, Ethylene chlorohydrin (ETCH) and Ethylene glycol (ETG), Environmental Protection Agency in U.S.A. has suggested some regulations on residual gas in drug products and medical devices for human use. The mutagenic activity of ETO compared with that of X-ray has an equivalency of 1 ppm/hr for ETO as compared to 20 mrad for X-ray, and one could suggest the present maximum allowable concentration for ETO (50 ppm) should be 400 times lower than the radiation standard (2.5 mrad/hr). Although radiation sterilization has advantages of simplicity of operation and complete reliability, changes of physico-chemical properties with possible formation of toxic substances may occur. It is therefore necessary to make some regulations of our own for residual toxicities orginated from each sterilization method.

Stability Test and Analysis of Rubia cordifolia for Carcinogenicity Study (천초근의 발암성 연구를 위한 분석 및 안정성 시험)

  • The marker constituent was isolated from Rubia cordifolia as a major compound. Quantitative method for the marker of the natural medicine was developed using HPLC-DAD and using established method the extract of Rubia cordifolia was evaluated. In addition, long term and accelerated stability test in the extract was examined for six months. No significant change in content of the marker constituent of the extract observed during the period of long term test.

  • Reciprocal exchanges of DNA in sister chromatids (SCEs) are induced by various carcinogens and mutagens, although the quantitative relationship between the number of mutations and SCEs induced varies among chemicals. Nevertheless, the analysis of SCEs production by various agents often proposed as a sensitive and quantitative assay for mutagenicity and cytotoxicity. Mercury, even if which has no evidences for mutagenicity and carcinogenicity, is reported to exert some cytotoxic effects, such as chromosomal aberrations or bad influences to ovulation and reproduction in experimental animals, etc.. In this study, tests for sister chromatid exchanges have been carried out on normal human lymphocytes in whole blood culture to add mercury chloride ($HgCl_2$) or methylmercury chloride ($CH_3\;HgCl$) for 72 hr. The results indicate the dose-dependent relationship between the frequencies of SCEs and the concentrations of $HgCl_2,\;CH_{3}HgCl$ and 5-bromo-2'-deoxyuridine (BrdU). Lymphocyte proliferation has depressed in the higher concentration of mercury.

Spontaneous lingual papillomas in fischer 344 rats (Fischer 344 랫드의 혀 유두종(Lingual papilloma) 자연발생 예)

  • Two cases of lingual masses were observed among 500 Fischer 344 (F344) rats which were used as control and treated animals in a 2 year carcinogenicity study in Toxicology Research Center, Korea Research Institute of Chemical Technology. The masses grossly appeared as tan, pedunculated, fungiform on the dorsal aspect of the base of the tongues. They were approximately $1.5{\times}1.2{\times}0.3cm$ in size. The microscopic features consisted of acanthosis, hyperkeratosis, papillary projection with connective tissue cores and multifocal chronic active inflammation with hair shafts. The results observed support the epigenetic mechanism of tumorigenesis which is caused by Physical stimuli of foreign bodies. Both of the masses were diagnosed as papillomas with the incidence rate of 0.4%(1/250) in each sex on the basis of the gross and microscopic features.

A study for the possibility of international harmonization on alcohol labelling under WTO/TBT (WTO/TBT협정하에서 알코올 라벨링에 대한 국제적 조화 가능성에 관한 연구)

  • This paper determines if mandated alcohol health information labelling could be an obstacle to international trade by discussing international trade rules affecting member nations. WTO members could use alcohol labelling rules to protect human health, reduce the consumption of alcohol and provide product information. When a member uses alcohol labelling as a technical regulation or standard, it should comply with TBT Articles 2.2 and 2.4. If a member uses some specific warning messages for their objectives, it should be considered the contribution of the measure, trade restrictiveness, and alternatives. If WTO members want to employ their own alcohol labelling schemes for addressing carcinogenicity, intoxicating effects, dependence potential, effects on children, adolescents and pregnancy, it could be a technical barrier and also create adverse effects on international trade. Therefore, the harms of alcohol are supported by scientific evidence, and for this reason, the international harmonization for alcohol labelling should be discussed openly to prevent negative effects on trade.

Effects of Pueraia mirifica on the Experimental Hepatocarcinogenesis in Rats and Gap Junctional Intercellular Communication (랫드의 실험적 간암 발생과정과 Gap Junction을 통한 세포간 정보전달에서 Pueraia mirfica의 효과)

  • To investigate the modifying effect of Kwao Kreu, Pueraria mirifica (PM), we performed two kind of studies which are the non-surgical medium-term carcinogenicity study and the modulation of gap junctional intercellular communication study. The first study, a non-surgical medium-term carcinogenicity bioassay was done to investigate the modifying effect of Kwao Keru, Pueyaria mirifca (PH), a rejuvenating folk medicine from Thailand, on the male F344 rat liver. Specific pathogen free, male 6-week-old F3444 rats were divided into ten groups. To induce hepatocarcinogenesis, those in all groups were given a single i.p. injection of DEN (200 mg/kg) and were received two i.p. injection of DGA (300 mg/kg) at the ends of weeks 2 and 5. Rats of group 3-6 were given sodium phenobarbital (PB 0.05% in drink). A diet containing 10 mg/kg PM was given to group 2 during the post-initiation phase and to groups 4 and 5 during promotion and initiation phase, respectively. Group 6 was given the experimental diet alone throughout the experiment (8 weeks). Rats of group 7, 8, 9 and 10 were fed 1000 mg/kg PH in the same manner as group 2, 4, 5 and 6. All animals were sacrificed at 8 weeks after DEN administration. Result of the immunohistochemical staining of the glutathione S-transferase placental form (GST-p) indicated that the numbers and areas of the preneoplastic leisions were not significantly changed in all PM treatment group comparing to control group. Also the numbers and areas of GST-p positive foci among group 7, 8, 9 and 10 were not significantly changed in comparing to control group. To study the effect of PM on the modulation of gap junctional intercellular communication, the present study was performed scrape-loading dye transfer (SL/DT) assay in human keratinocytes. The results showed that PM could not modulate GJIC. These results indicate that Pueraria mirifica may have no carcinogenic effects on experimental hepatocarcinogenesis in rats and gap junctional intercellular communication in human keratinocyte.
