Impact of Climate Change on Yield Loss Caused by Bacterial Canker on Kiwifruit in Korea (기후변화 시나리오에 따른 미래 참다래 궤양병 피해 예측)

  • We estimated the averaged maximum incidences of bacterial canker at suitable sites for kiwifruit cultivation in 2020s and 2050s using D-PSA-K model with RCP4.5 and RCP8.5 climate change scenarios. Though there was a little difference between the estimation using RCP4.5 and that using RCP8.5, the estimated maximum disease incidences were more than 75% at all the suitable sites in Korea except for some southern coastal areas and Jeju island under the assumption that there are a plenty of infections to cause the symptoms. We also analyzed the intermediate and final outputs of D-PSA-K model to find out the trends on the change in disease incidence affected by climate change. Whereas increase of damage to kiwifruit canes in a non-frozen environment caused by bacterial canker was estimated at almost all the suitable sites in both the climate change scenarios, rate of necrosis increase caused by the bacterial canker pathogen in a frozen environment during the last overwintering season was predicted to be reduced at almost all the suitable sites in both the climate change scenarios. Directions of change in estimated maximum incidence varied with sites and scenarios. Whereas the maximum disease incidence at 3.14% of suitable sites for kiwifruit cultivation in 2020s under RCP4.5 scenario was estimated to increase by 10% or more in 2050s, the maximum disease incidence at 25.41% of the suitable sites under RCP8.5 scenario was estimated so.

  • Dispersal of Xanthomonas axonopodis pv. citri, causing citrus bacterial canker disease on Unshiu orange was investigated at previously infested plots at Seogwipo in Jeju island of Korea. The bacterial pathogen overwintered in lesions started to multiply at tate May, and disease firstly observed one month after detection of phage from lesions. The disease gradually increased, however, it dispersed non-directionally to nearby plants from inoculum sources. Diseased plants were aggregated to form a cluster throughout the experiment. Population dynamics of phage on symtomless leaf surface and the disease severity were compared in the nursery, Increase of phage population on symptomless leaf surface preceded one month to that of the disease severity Population of phage increased constantly from late July to October, however, the disease severity decreased from late August to late October. It was assumed that the decrease of disease severity might be due to disease-induced defoliation.

Detection of Xanthomonas axonopodis pv. citri on Satsuma Mandarin Orange Fruits Using Phage Technique in Korea

  • A phage technique for detection of Xanthomonas axonopodis pv. citri, a causal bacterium of canker on Sastuma mandarin fruits was developed. Phage and ELISA techniques were compared for their sensitivity for detection of Xanthomonas axonopodis pv. citri on orange fruits. Both of techniques revealed a similar efficiency for the bacterial detection; the pathogenic bacteria were observed in pellet from the fruits with over one canker spot with below 2 mm in diameter. In field assays, the increase of phage population(120%) on surface of the fruits related to the disease development one month later indicated that the bacterial pathogens inhabit on the surface. The procedure will be effectively used for detection of only living bacterial pathogen on fruit surfaces of Satsuma mandarin and for the disease forecasting.

  • Latent infections of healthy-appearing oaks of Hypoxylon atropunctatum complicates field studies by interfering with inoculation experiments to follow pathogenesis, fungal development and reproduction of this canker rot fungus. Mutants with unique and easily scorable phenotypes would be useful for inoculation studies. There is a broad range in the capacity of wild-type isolates to utilize nitrate as a sole nitrogen sources. Several types of nitrate-nonutilization mutants (nit1, Nit3, NitM) were selected from nitrate-utilizing wild-type isolates. Also, a few mutants of Hypoxylon atropunctatum were selected that could only grow poorly on basal medium supplemented with various nitrogen sources and even on yeast extract agar. These unknown mutants need to be characterized further. Nit mutants of Hypoxylon atropunctatum were readily selected, grew well and were recovered after inoculation into oak stems. These results suggest that nit mutants could be useful for inoculation studies in trees that contain latent infections.

Characteristics of Bacterial Canker of Kiwifruit Occurring in Korea and Its Control by Trunk Injection (우리나라 참다래 궤양병 발생 특성 및 수간주입에 의한 방제)

  • 암수 딴그루인 참다래 품종 중에서 우리나라에서 재배되고 있는 대표적인 결실수(암나무)인 Hayward와 수분수(숫나무)인 Matua에서 궤양병의 발생 차이를 조사한 결과 Hayward의 이병엽율은 18.0%인 반면에 Matua의 이병엽율은 2.4%였다. 또한 Hayward와 Matua의 잎에 형성된 병반에서 분리된 궤양병균의 균총수는 생체중 1 g당 각각 1.8$\times$10\ulcorner개와 2.1$\times$10\ulcorner개로 Hayward에서 Matua보다 약 10배 가량 많은 궤양병균이 검출되었다. Hayward 신초에 존재하는 궤양병균의 밀도를 조사한 결과 줄기 조직보다는 잎 조직에서 많은 궤양병균이 검출되었으며 새로 나온 잎일수록 궤양병균의 밀도는 낮게 검출되었다. 궤양병 방제를 위하여 스트렙토마이신 수간주입에 의한 궤양병 치료효과는 50.7~53.7%였다. 수간주입시 약량을 주당 2 L에서 3 L와 4 L로 증가시킴에 따라 치료효과도 32.3%에서 58.1%와 71.0%로 각각 증대되었는데, 수간주입 약제와 수간주입 방법의 개선을 통하여 궤양병 치료효과를 더욱 증대시킬 수 있을 것으로 기대된다.

Isolation and Identification of Antagonistic Microorganisms for Biological Control to Apple Tree Diseases, Canker(Valsa ceratosperma) (사과 부란변 방제를 위한 길항미생물 분리 및 동정)

  • For the purpose of acquiring microbial agents that can be utilized to billogically control the major airborne disease to apple trees, such as canker(Valsa ceratosperma), the effective microorgaisms were isolated, tested for antagonistic activity to the pathogen causing major disease to apple trees and identifed. Screening of more than 3, 000 species of microorganisms collected in nature for them antagonistic action to the pathogen, Valsa ceratosperma causing disease to apple tree resulted in selection of effective species. Out of the 11 species, one species designated as CAP141 demonstrated outstanding activity. The bacterial strain, CAP141 exerted antagonistic efficiency of 65% on Valsa ceratosperma. The CAP141 was identified as a bacterial strain to Bacillus subtilis based on morphology, culture conditions, and physio-biochemical characteristics.

가칭 Yucca의 운문병

  • The strains of fungus which belong to Coniothyrium genus were isolated from leaf spoot of Yucca. The isolated strain can grow easily and forms characteristic colonies on Saido's soy agar. There are definite differences, according to other authors, (By Miyake et Hara 1911) Pycnidio-spore are usually formed ellipsoidal, dark brown, 4$\times$2. 5-4. $O\mu$ in size, whereas isolated strains are globose, yellowish brown, 2.8-4.2$\mu$ in diameter. The positive inoculation test can be attained by burned petridish method, preserving it in incubator at 28$^{\circ}C$, and the pycnidio-spore can be reisolated from the inoculated portion. (Photo.7) Up to the present day there is no report for leaf spot of Yucca caused by Coniothyrium sp.. Comparing the writer's isolate with those fungi described(leaf spot of alloe, rose canker and elm canker etc.) hitherto, there are no accordance in the symptom and especially in morphological charactgers and Pathogenicitys.

First Report of Botryosphaeria dothidea Responsible for Branch Dieback and Canker on Hovenia dulcis in South Korea

  • In 2021, severely infected trees showing distinct symptoms including branch dieback, necrotic lesions on branches, cankers on the shoots, and dark brown discoloration on the stem on Hovenia dulcis were found in the provincial forest of Jeollabuk-do located in Jangsu, South Korea. The causal agent of the pathogen was identified as Botryosphaeria dothidea based on morphology and sequence comparisons. This is the first confirmed report of B. dothidea causing branch dieback and canker on H. dulcis.

  • Eleven fungi were isolated from the cankered stems, branchs and twigs of chestmut trees collected from Chuncheon, Gapyung, Wonseong, Inje and Kanghwa during 1985-1986. Among them, Botryosphaeria dothide (Moug. ex Fries) Ces. et. de Notaris, Cryptodiaporthe castanea (Tulasne) Wehmeyer, Endothia singularis Sheaet Stevens, E. parasitica (Murrill) P. J. et H. W. Anderson and Pseudovasella modonia (Tulasne) Kobayas were identified as perithecial stage. Dathiorella castaneae Camara et Vasconcelos, Fusicoccum castaneun Saccardo, Catinula japanica st. nov., Endothiella singularis (H. et. P. Syd.) Shear et Stev. nom. seminud, E. parasitica And., nom. seminud., and Coryneum castaneae (Sacc.) comb. novo were identified as conidial stage Botryosphaeria dothidea causing canker or diebark but and Endothia parasitica causing Endothia canker were already reported in Korea. The other fungi are first reported in Korea.
