  • The insecticidal protein gene in the pKL-20-1 clone derived from Bacillus thuringiensis serovar. kurstaki plasmid was subcloned in the plant shuttle vector, pGA643. The 7.3 kb fragment was cloned in the BglII and Hpal sites of pGA643 vector and expressed in E. coli S17-1, which produced insecticidal proteins killing Bombyx mori larvae. The clone was named pHL-20. The protoplast formation, calli induction and plantlet regeneration of Nicotiana tabacum was carried out. A tremendous number of mesophyll protoplasts of N. tabacum were formed, up to 7$\times$105 protoplast per ml, for 20 hours in darkness in the enzyme solution of 0.5% cellulase and 0.1% macerosin, pH 5.8. The viabilities of the protoplasts were maintained above 80% for 6 days in the media containing 2mg/1 of NAA and 1mg/1 of kinetin. Calli were induced from the protoplasts and leaves of the N. tabacum on MS medium containing 0.5mg/1 BAP. Under the culture conditions the protoplasts underwent repeated cell division into calli. Plantlets were regenerated from callus cultures derived from protoplast and leaves. Shoots were induced in a medium containing 1mg/1 of BAP.

Variations in Sweetpotato Regenerates from Gamma-ray Irradiated Embryogenic Callus

  • Radiation induced and somaclonal variations were investigated in the regenerates from gamma irradiated and controlled embryogenic callus (EC) of sweetpotato cvs., Yulmi and White Star by morphological, RAPD and AFLP analysis. Most (approx. 90%) of the EC produced somatic embryos developed into plantlets after being transferred to the auxin-free medium. The frequency of morphological variants derived from the irradiated callus ranged from 3 to 7.8% compared to 0.1-1.1% of that derived from the non-irradiated. Morphological variants were selected from the regenerates and analyzed by RAPD and AFLP procedures. RAPD polymorphisms of Yulmi and White Star regenerates from irradiated calli were 8.8% and 6.1%, respectively. However, the polymerphisms among regenerates from the non-irradiation treatment in these two cultivars were non-detectable and 3%, respectively. AFLP polymorphisms of Yulmi and White Star regenerates from irradiated calli were 29.9% and 28.6%, respectively. while the frequencies for those form non-irradiated calli were 8.5% and 5.6%, respectively. Genetic Transformation of Populus nigro x maximowiczii Using Agrobacterium tumefaciens Harboring Antisense OMT Gene

  • An Agrobacterium tumefaciens LBA4404 (harboring antisense OMT gene)-mediated transformation method has been developed for poplar (P.nigra x maximowiczii) using prolonging co-cultivation time. Explants on LT (longterm) were induced transgenic calli one month earlier than those from ST (short-term) co-cultivation and remained healthier on LT than ST. With this approach, LT method reduced time to produce transgenic calli. Shoots were successfully regenerated from transgenic calli on SIM (Shoot Induction Medium) and rooted well on the basal medium spontaneously. The presence of antisense OMT gene was verified both by PCR and Southern analysis. Each transgenic poplar was phenotypically indishtinguishable when compared with controls for their growth pattern, leaf morphologies and xylem coloration.

Somatic Embryogenesis in Withania somnifera (L.) Dunal

  • Somatic embryos were formed from calli obtained from axillary shoots (raised from nodal segments of glasshouse-grown plants under aseptic conditions), internodal segments (from in vitro-raised plants), and root and coty-ledonary leaf segments (from in vitro-raised seedlings) after 8 weeks of initial culture. Embryo formation was the highest (97.33%) from cotyledonary leaf callus on Mura-shige and Skoog's (MS) medium containing kinetin (KN) (3 mg/L). Somatic embryo induction was lesser with different combinations of auxins while it increased to 100% in internodal segment and cotyledonary leaf calli with 6-benzyladenine (BA) (2mg/L) along with 2,3,5-triiodobenzoic acid (TIBA) (2mg/L). The shoots were induced from somatic embryos raised from root, coty-ledonary leaf and internodal segment calli grown on MS medium containing BA in combination with indole-3-acetic acid (IAA). Maximum of 66.67% cultures formed shoots on MS medium containing BA (1mg/L) in combination with IAA (2mg/L). The shoots raised from somatic embryos were rooted on MS medium supplemented with indole-3-butyric acid (IBA) (2mg/L). The plantlets transferred to the field showed 70% survival rate after one year.

인삼 Tumor Callus의 생장 특성 (Characteristics of the Growth of Ginseng Tumor Callus)

  • 인삼 crown gall tumor callus의 특성을 구명하기 위하여 Agrobacterium tumefaciens C58을 감염시켜서 얻어진 crown gall tumor를 phytohormone 무첨가 배지에서 배양하여 생육이 왕성한 tumor callus를 유기하였다. 이들 tumor callus는 정상조직에서 유기한 callus에 비해 외관상으로는 friable한 형태의 덩어리로 자라면서도 매우 단단한 양상을 보였으며, callus증식에 적합한 phytohormone 농도에서는 tumor callus의 생육이 오히려 억제되는 현상을 보였다. Tumor callus의 생육은 고체배양시 암배양이 광배양보다 생체중의 경우에는 2.4배, 건물중은 1.9배 가량이 더 많으며, 2,4-D가 함유된 최적조건에서 배양한 정상 callus의 생체중과는 비슷한 경향을 보였으나, 건물중은 tumor callus가 더 높았다. 그리고 현탁배양시에는 tumor callus가 정상 callus보다 생장속도가 2배 이상 빠른 경향이었다.

지치(Lithospermum erythrorhizon S.)의 캘러스배양에서 Shikonin 유도체 생산에 미치는 저선량γ선의 효과 (Effects of Low Dose Gamma Radiation on the Formation of Shikonin Derivatives on Callus Cultures of Lithospermum erythrorhizon)

  • The effects of low dosage ${\gamma}$-radiation on the cell growth and the formation of shikonin derivatives were investigated in callus cultures of Lithospermum erythrorhizon under different medium and light conditions. Gamma radiation significantly affected the cell growed and formation of shikonin derivatives, depending on the culture conditions. In the cell cultures grown on M-9 medium, 2Gy and 16Gy of ${\gamma}$-radiation increased the calli growth and the formation of shikonin derivatives, respectively under 16hr day light condition. When calli were cultured for 60 days in the dark after irradiation of ${\gamma}$-radiation, cell growth was increased at low dosage of 1Gy and 2Gy in LS medium containing BA 2mg/L and IAA 0.2mg/L. Interestingly, calli grown in M-9 medium by 2Gy irradiation for 60 days significantly stimulated the formation of shikonin derivatives(13.21mg/g cell fresh wt), which was approximately 6 times higher than untreated cells.

  • Pelargonium aridum과 P. zonale의 자엽과 유식물의 절편체를 2 mg/L NAA와 0.2 mg/L BA를 첨가한 MS배지에서 배양하였을 때 캘러스를 형성하였다. 이들 캘러스는 동일배지에서 계대배양양하였다. 계대배양된 캘러스는 P. aridum의 경우 0.1∼l mg/L NAA와 0.25∼2 mg/L BA에서, P. zonale의 경우 0.1∼0.5 mg/L NAA와 1∼2 mg/L BA를 첨가한 MS 배지에 옮겨주었을 때 가장 많은 shoot를 형성하였다(P. aridum은 explant당 0.78개, P.zonale은 0.65개). 대부분의 shoot은 1/2 MS기본배지에서 배양하였을 때 발근하였다. 이들 재분화 개체들은 화분으로 옮겨진 추 온실에서 발육하여 개화하였다

오차드그래스 성숙종자로부터 캘러스 유도 및 고효율 식물체 재분화 (High-frequency Plant Regeneration from Mature Seed-derived Callus Cultures of Orchardgrass)

  • In an effort to optimize tissue culture conditions for genetic transformation of orchardgrass (Dactylis glomerata L.), an efficient and high-frequency plant regeneration system from seed-derived calli was established. Embryogenic calli induced on MS medium containing 3mg/L 2,4-D and 0.1mg/L BA had significantly improved regeneration ability. Plant regeneration rate was 92% when embryogenic calli were cultured on N6 medium supplemented with 1mg/L 2,4-D and 3mg/L BA. Among three kinds of medium, MS and N6 medium were optimal for embryogenic callus induction and plant regeneration, respectively. Ho difference in callus induction frequency was observed among four cultivars of orchardgrass, however, "Roughrider" cultivar showed higher regenerability with the frequency of 61%. Addition of maltose to the regeneration medium as a carbon source dramatically increased regeneration frequency up to 69%. A short tissue culture period and high-frequency regeneration system would be beneficial for molecular breeding of orchardgrass through genetic transformation.

  • Friable callus와 compact callus를 배양 단계별로 단백질을 정량한 결과 friable callus가 가장 낮았고 배양 0주째부터 단백질 함량이 서서히 증가하여 배양 3주째 최대함량이 되었으며, 4주째부터는 감소하기 시작했다. 전기영동법으로 단백질의 패턴을 조사한 결과 compact callus와 friable callus간에 특징적인 차이를 보이는 단백질 밴드가 관찰되었다. Friable callus는 약 28 KD, 31 KD과 35 KD에서 특이적으로 나타났으며. compact callus에서는 30 KD부근의 밴드에서 많은 변화가 일어났다. 전체 단백질의 구성 아미노산은 배앙 2∼3주에 가장 높은 함량을 보였고, friable callus에서 그 함량이 가장 낮았다.

  • 발아된 벼(Oryza sativa L. cv. Nakdong)의 하배 축에서 callus를 유도하고, 현탁배양한 세포에서 원 형질체를 분리하여 MS 배지에서 재분화를 수행하였다. Callus는 하배축으로부터 $2.5mg/{\ell}2$,4-0를 첨가한 LS 배지에서 최고 65%의 치상효율로 유도되었으며, 형태척으로 선별 가능한 embryogenic callus 는 $2mg/{\ell}2$,4-D, O.2mgj P kinetin과 $0.1mg/{\ell} GA_3$ 가 첨가된 AA, 배지에서 현탁배양하였다. 4개월 이상 현탁배양한 세포로부터 분리한 원형질체는 voltage를 400V jern와 Imsec 통안 electropora­tion했을 때 $2.5mg/{\ell}2$,4-D, $0.1mg/{\ell}$kinetin과 lOmM proline이 첨가된 PCM 배지에서 1.1%의 치 상효율을 보였다. 형성된 microcalli는 LS2.5 배지 에 치상된 feeder cell에 $0.2\mu\textrm{m}$membrane filter를 얹은 위에 옮겨 암소에서 2주 동안 배양한 후, $30{\pm}/3{\mu}E$.m^{-2}S^{-1}의 형광하에서 2주 통안 배양했을 때 2mm 직경의 노란색 callus를 형성하였다. 형성된 callus를 재분화 배지에 치상하였을 때, 녹점 green spot)에서 shoot 형성과정을 거쳐 완전한 식물체로 분화되었으며 재분화율은 1l~33%였다.
