Changes of Chemical Properties during Liquid Fertilizer Processing using Different Organic materials (유기물을 이용한 액비 제조시 발효액의 화학적 특성 변화)

  • This study was carried out to investigate the chemical characteristics of different organic liquid fertilizer during fermentation at plastic house in Chungbuk Agricultural Research and Extension Service. Chicken dung, soybean meal, and rice bran were used for nutrient sources. The obtained results from this study were summarized as follows; Total nitrogen contained the highest in soybean meal as 55 mg·kg-1, and phosphate and potassium contained high in chicken dung and rice bran in organic liquid fertilizer materials. The pH of chicken dung was near 7.0, that of soybean meal and rice bran indicated acidity(pH 3.8~4.4). The electrical conductivity of chicken dung consistently increased during fermentation, and that of soybean meal and rice bran increased and decreased early season and increased at late incubation period. Hydrogen sulfide gas occurrence of chicken dung was 3,200 mg·L-1 at early season and 1,600 mg·L-1 at late incubation period, and that of soybean meal and rice bran treatments were not or very low concentration of hydrogen sulfide gas during manufacturing period. The nitrogen and calcium content of organic liquid fertilizer were the higher in chicken dung and soybean meal than rice bran. The phosphate and magnesium content of rice bran was the high as 5.6 g·kg-1 and 1.5 g·kg-1, respectively. There was no difference in potassium content among the different liquid fertilizers during fermentation.

Studies on the Biological Control of Pine Caterpillar (Dendrolimus spectabilis Butler) by Red Wood Ants (Formica rufa truncicola var. yessoensis Forel) (불개미를 이용한 송총의 생물적방제에 관한 연구)

  • In order to increase utility efficiency of red wood ants, Formica rufa truncicola var. yessonesis Forel as a resource of natural enemy of pine caterpillar, Dendrolimus spectabilis Butler, by finding out ecological and environmental factors in the habitat of red wood ants, the nest distribution and its density in habitat, plant distribution and density, stand-density of red pine, nest building and fixing plants, relative humidity of surface soil, physical and chemical natures of soil, and breeding rate were examined. The obtained results are summarized as follows: 1. The nest of red wood ants was densely distributed, in the lower-and middle top of mountain but no nest was found in the top. 2. The economical distribution of nest of habitat was estimated as $2.85/m^2$ and the lowest density as $1.93/m^2$ and these estimation lead us to confirm that pine caterpillar could be controlled. 3. The ecological characteristics of habitat seemed to be represented as higher stand-density of red pine of 10-20 years of age with large areas of eroded land under trees. The major grasses prevailing in this area were Andropogon brevifolius. Arundinella hirta, Miscanthus purpurasens, Eulia speciosa, Themeda japonica, Cymbopogon goeringii, and Eccoilpus cotulifer 4. Red wood ants seemed to build the nest by using red pine, Arundinella hirta, Miscanthus purpurascens, Themeda japonica or Cymbopogon goeringii as a fixing plant. 5. The limited point of humidity percent in habitat of red wood ants was estimated as $76\%$ during the acting period of May to September and as $72\%$ during pre-period of hibernation of October to November. 6. Soil analysis in habitating region showed higher concentration of organic matters and lower concentration of calcium and magnesium, and habitat was largely composed of silt and fine sand rather than coarse sand. 7. When the separated colony was transplanted to non-habitating red pine forest that seemed to have the similiar conditions as those of habitat, propagation and establishment of nest was possible.

A Study on Cation Extraction and Impurity Separation in Slag (슬래그 내 양이온 추출 및 불순물 분리 연구)

  • The cation extraction and impurity separation were studied in order to investigate the recyclability of a slag produced from the steel refinery industry. Two types of slag (Slag-A, B) were collected and characterized in this study. The initial characterization by X-ray diffraction (XRD) and X-ray fluorescence (XRF) confirmed the existence of various kinds of ions in the slag such as Ca2+ (30 ~ 40%), Fe3+ (20 ~ 30%), Si4+ (15%), Al3+ (10%), Mn2+ (7%), and Mg2+ (3 ~ 5%). Inductively coupled plasma atomic emission spectroscopy (ICP-AES) analysis on the extracted slag using 2 M HCl as a solvent indicated that a higher concentration of Ca2+ was extracted as the S/L ratio was increased. The Ca2+ extraction concentration were found to be 8,940 mg L-1 (Slag-A) and 10,690 (Slag-B) mg L-1 when the S/L ratio for Ca2+ extraction was 0.1. However, the extract was strongly acidic ( < pH 1) at 0.1 S/L. Also the other ions (impurities) were extracted simultaneously in addition to Ca2+. To increase the purity of Ca2+ in order to transform the slag to a high value resource, a pH-swing was conducted. The impurities tended to precipitate at higher rate as the pH was increased. Notably, the Ca2+ rapidly precipitated above a certain pH and at a pH of 10.5, while the selectivity of Ca2+ was over 99%. It is expected that the aqueous solution in which high contents of Ca2+ was selectively dissolved in this study would be suitable for the carbonation process for reducing CO2 and for the production of calcium carbonate.

Chemical Components and Physiological Functionalities of Brassica campestris ssp rapa Sprouts (순무 싹의 화학성분과 생리기능성)

  • This study was carried out to investigate physicochemical and functional properties of dried Brassica campestris ssp rapa (BR) sprouts. The proximate compositions of BR sprouts as dry matter basis were 2.35% of moisture content, 22.51% of crude protein, 21.60% of crude lipid, 4.35% of crude ash, and 49.19% of carbohydrate, respectively. The free sugars were identified as glucose and fructose. Analyzing total amino acids, 18 kinds of components were isolated from BR sprouts. The essential amino acid contained in BR sprouts accounted for 47.00% of total amino acid, while the non-essential amino acid accounted for 53.00%. The contents of vitamin A and vitamin E were 0.09 mg% and 3.06 mg%, respectively. Tartaric acid was the major organic acid. Among the minerals in dried BR sprouts, the content of potassium was the highest (882.50 mg%) and those of magnesium and calcium were comparatively high (342.85 mg%, 274.30 mg%). BR sprouts ethanol extract significantly inhibited the HMG-CoA reductase activity in a concentration-dependent manner in vitro. Furthermore, nitrite scavenging ability and DPPH radical scavenging activity of the ethanol extract of BR sprouts were 64.25% and 69.29% at a concentration of 1,000 ${\mu}g$/mL, respectively. These results suggest that BR sprouts possess potential antioxidative capacity and HMG-CoA reductase inhibitory activity.

Analysis of Sap Production and Composition of Acer okamotoanum from Artificial Stands in Different Tapping Time in Jin-ju Region in Korea (진주지역 인공식재 우산고로쇠의 수액 채취 시기별 출수량 및 성분 분석)

  • This study was conducted to analyze the sap flow and composition by tapping date in Acer akamotoanum from an artificial forest in the City of Jinju. The sap was collected from five saplings (diameter at breast height10-20 cm) in two phases with an interval of a month. During phase 1 (January 10, 2019 - February 9, 2019) with daily mean temperature of 1.16±2.15℃, 60.59 liters of sap was collected over 32 days. Phase 2 (February 12, 2019 - February 22, 2019) with daily mean temperature of 2.55±1.30℃ yielded 13.38 liters of sap over 11 days, which was a reduction to 22% of the total sap flow in phase 1. The analysis of sugar composition in the collected sap from phase 1 and 2 showed that sucrose concentration was 2.5% at its highest during the earlier days of collection and decreased to 0.8% at the end. Glucose and fructose concentrations were measured near 0, and were lower than sucrose concentration. The most prominent inorganic elements in the collected sap were potassium (41.69 - 89.75 mg/kg), calcium (24.66 - 48.14 mg/kg) and magnesium (10.18 - 27.25 mg/kg). The contents of sucrose and inorganic components between the samples collected from phase 1 and 2 did not show significant differences. The sap flow amounts of A. okamotoanum from different tapping periods showed a notable variation, but there were no significant differences in the contents of sugar or inorganic elements.

Effect of Fly Ash Fertilizer on Paddy Soil Quality and Rice Growth (비산재로 제조한 비료가 논토양 질과 벼 생육에 미치는 영향)

  • Coal ash can be added to agricultural soils to increase the chemical properties of soil such as pH, cation exchange capacity and nutrient availability of - B, Ca, Mo etc-. Therefore, the main purpose of this study was to evaluate the feasibility of fly ash as a soil amendment in paddy soils. Selected fly ash was mixed with bentonite and calcium hydroxide at the ratio of 80:15:5 (w/w) and manufactured as a pellet type at the size of 10 mm. Field experiments were conducted to evaluate the effects of fly ash fertilizer on the soil quality and crop growth compare to the control (no fertilizer) and, - traditional fertilizer. Results showed that soil pH and organic matter in paddy soils after applying the manufactured fly ash fertilizer were not increased compared to the other two treatments. However, the concentration of available phosphate and silicate in paddy soils were higher than those of the control and traditional fertilization. With regard to crop growth, no significant difference was observed between three different treatments. However, the content of protein in the rice grain cultivated with the fly ash fertilizer was higher than in the rice cultivated by other two treatments. Overall, fly ash fertilizer could increase the concentration of available silicate and phosphate in the paddy soil and improve the rice quality. In conclusion, fly ash can be utilized in agricultural soils as soil amendment, especially in the rice paddy soil.

Interrelationship between Dopaminergic Receptors and Catecholamine Secretion from the Rat Adrenal Gland (흰쥐 부신에서 카테콜아민 분비작용과 도파민 수용체간의 상관성)

  • It has been known for some time that dopamine-containing cells are existed in sympathetic ganglia, i.e., small, intensely fluorescent cells. However, its role and mechanism of action as a peripheral neurotransmitter are poorly understood so far. In the present study, an attempt was made to examine the effect of apomorphine, which is known to be a selective agonist of dopaminergic $D_2$. receptor on secretion of catecholamines (CA) from the isolated perfused rat adrenal gland. The perfusion of a low concentration of 10uM apomorphine into an adrenal vein for 20 min produced significant reduction in CA secretion induced by 5.32 mM ACh, 56 mM KCl, 100 uM DMPP and 100 uM McN-A-343. Increasing apomorphine concentration to 30 uM led to more markedly decreased CA secretion as compared to the case of 10 uM apomorphine and also did inhibit clearly CA release by $10^{-5}M$ Bay-K-8644. Furthermore, in adrenal glands preloaded with a higher dose of 100 uM apomorphine, CA releases evoked by ACh, excess $K^+$, DMPP and McN-A-343 were almost abolished by the drug. The perfusion of $3.3{\pm}10^{-5}M$ metoclopramide, which is well-known as a selective dopaminergic $D_2$ antagonist, produced significantly inhibitory effect of CA release by ACh, DMPP and McN-A-343 but did not affect that by excess $K^+$. However, preloading of 30uM apomorphine in the presence of metoclopramide did not modify the CA secretory effect of excess $K+$ and DMPP. These experimental results demonstrate that apomorphine causes dose-dependent inhibition of CA secretion by cholinergic receptor stimulation and also by membrane depolarization from the isolated perfused rat adrenal gland, suggesting that these effects appear to be exerted by inhibiting influx of extracellular calcium into the rat adrenal medullary chromaffin cells through activation of inhibitory dopaminergic receptors.

Glutamate-and NMDA-induced calcium influx at synaptosomes and the difference of their actions (Glutamate와 NMDA에 의한 Synaptosome에서의 칼슘 유입과 이들의 작용의 차이)

  • Glutamate and aspartate may evoke an increase in membrane permeability to monovalent cations and $Ca^{++}$. However, it is uncertain whether $Ca^{++}$ influx is mediated by voltage dependent $Ca^{++}$ channels or by excitatory amino acid activated channels. In addition, the influences of excitatory amino acids on $Ca^{++}$ uptake by neuronal tissues as well as the responses of their actions to extracellular $Mg^{++}$ concentration are different. $K^{+}$ induced $Ca^{++}$ uptake by synaptosomes was dependent on extracellular $Mg^{++}$ up to 5 mM and at concentration of 10 mM, $Ca^{++}$ influx was rather reduced. In $Na^{+}$ rich media, glutamate-and aspartate-induced $Ca^{++}$ uptake was increased by $Mg^{++}$ in a dose independent manner. However, the response for NMDA was inhibited by $Mg^{++}$ at concentrations above 2 mM. $K^+$-and glutamate-induced $Ca^{++}$ influx s were inhibited by 2,4-dinitrophenol, chlorprom-azine and verapamil but not by tetraethylammonium chloride. Tetrodotoxin effectively inhibited the action of glutamate but did not affect that of $K^+$. The response for MNDA was inhibited by 2, 4-dinitrophenol and tetrodotoxin, slightly inhibited by verapamil, and not affected by tetraethylammonium chloride. In $Na^{++}$ rich medium, depolarizing action of glutamate, aspartate and MNDA on synaptosomes was not demonstrated, whereas these agents stimulated $Ca^{++}$ uptake and caused $Ca^{++}$ influx induced depolarization at mitochondria. On the other hand, the activities of synaptosomal ATPases were not affected by excitatory amino acids at 5 mM. The results suggest that glutamate or NMDA induced $Ca^{++}$ influx at synaptosomes exhibits different responses for extracellular $Mg^{++}$ Ex citatory amino acids induced $Ca^{++}$ influx at synaptosomes may be associated with increased permeability of membrane for $Na^{++}$ and $Ca^{++}$ except $K^{++}$ and membrane depolarization due to increased ionic permeability.

Effects of Crormakalim on the Release of Mediators in Hypersensitivity of Guinea pig (Cromakalim이 해명의 과민반응 매개체 유리에 미치는 영향)

  • Potassium $(K^+)$ channels are present in airway smooth muscle cells, and their activation results in hyperpolarization and relaxation. Because these effects may have therapeutic relevance to hypersensitivity and asthma, we examined the effect of a potassium channel activator, cromakalim (BRL 34915, CK) on the release of mediators from superfused tracheal and parenchymal strips after passive sensitization with $IgG_1$ antibody. Both tissues were superfused with CK $(2{\times}10^{-6}\;M)$ for 30 min and challenged with CK and antigen (Ox-HSA). Using monodispersed, partially purified, highly purified guinea pig lung mast cells, we also examined the effect of CK on mediator release from these cells after passive sensitization with $IgG_{1}$ antibody $({\alpha}-OA)$. Guinea pig lung mast cells were purified using enzyme digestion method, count current elutriation, and discontinuous Percoll density gradient. After CK pretreatment, passively sensitized mast cells were challenged with varying concentration of antigen (OA, immunological stimuli) or with varying concentration of calcium ionophore (CaI, non-immunological stimuli). Histamine (Hist) release was determined by spectrophotofluorometry, and leukotrienes (LT) by radioimmunoassy. CK pretreatment decreased Hist by 35% and LT release by 40% in the antigen-induced tracheal tissue after $IgG_1$ sensitization but did not decrease the contractile response. In the antigen-induced parenchymal tissue CK decreased Hist release by 25% but poorly decreased LT. Both immunologic and non-immunologic stimuli caused a dose-dependent release of Hist and LT from monodispersed, partially purified and highly purified lung mast cells. Verification of LT release was obtained by the use of 5-lipoxygenase inhibitor, A64077 (Zileuton). CK decreased Hist and LT release by 20% respectively in the OA-induced guinea pig lung mast cells after $IgG_1$ sensitization. The inhibitory effects of CK on the Hist and LT release in the Ox-HSA-induced airway smooth muscle tissues or in the OA-induced and CaI-induced mast cells after $IgG_1$ sensitization were completely blocked by TEA and GBC. These studies show that guinea pig lung mast cells seem to be an important contributor to LT release, and that CK (which has been known as an airway smooth muscle relaxant) can in part act to inhibit mediator release in the antigen-induced airway smooth muscle, and that CK may also act to inhibit mediator release in the OA-induced and CaI-induced highly purified mast cells. These results suggest that Hist and LT release evoked by mast cell activation might in part be associated with $K{^+}4 channel activity.

Comparison of Ingredients and Antioxidant Activity of the Domestic Regional Wolfiporia extensa (국내 지역별 매립 복령의 성분 및 항산화 활성 비교)

  • This study was conducted for comparison of ingredients, phytochemical compounds and antioxidant activity of Wofiporia extensa cultured in Gangwon-do, Gyeongsang-do, and Jeolla-do. Three contents of Wofiporia extensa were analyzed as oxygen (46~48%), carbon (38~39%), hydrogen (6.05~6.1%) and nitrogen (0.17~0.21%). The mineral contents of 50% ethanol Wofiporia extensa extracts were measured as sulfur (S) 145~149 ppm, Magnesium (Mg) 69~72 ppm, phosphorus (P) 122~154 ppm and calcium (Ca) 210.61~509.98 ppm. Wofiporia extensa from Gyeongsang-do (509.98 ppm) contained a significantly higher quantity of Ca than that from Gangwon-do (210.62 ppm) and Jeolla-do (223.88 ppm). In the gas chromatograph-mass spectrometry (GC-MS) analysis, oleic acid was identified in three 50% ethanol Wofiporia extensa extracts. In the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay for antioxidant activity, the $IC_{50}$ values of Wofiporia extensa cultured in Gangwon-do, Gyeongsang-do and Jeolla-do were calculated as 2.966 mg/mL, 23.03 mg/mL, and 4.16 mg/mL and 3.521 mg/mL, 12.17 mg/mL, and 7.40 mg/mL. In the ferric reducing antioxidant power (FRAP) assay, the $IC_{50}$ values of Wofiporia extensa cultured in Gangwon-do, Gyeongsang-do, Jeolla-do were 6.585 mg/mL, 19.06 mg/mL, and 18.97 mg/mL, respectively. In summary, Wofiporia extensa cultured in Gangwon-do had stronger antioxidant activity and higher concentration of oleic acid than that of Geyongsang-do and Jeolla-do. However, Wofiporia extensa cultured in Geyongsang-do contained a much higher concentration of Ca than that of Gangwon-do and Jeolla-do.
