Aroma Compounds and Antimicrobial Effect of Garlic from Different Areas in Korea (국내 산지별 마늘의 향기성분 및 항균활성)

  • Physico-chemical characteristics such as width, weight, color and aroma compounds of garlic from 9 different areas were analyzed. Also, antimicrobial activity was tested for their juices. Width of garlic bulb was larger in growing worm-season garlics (Jeju, Namhae, Hapcheon, Hampyeong and Muan) than those of cool-season garlics (Taean, Seosan, Uiseoung) and imported from China. But numbers of nuts were smaller in growing cool-season garlics than others. L color levels of garlics were 80.73~87.40 and a color level have not significantly difference in all samples, b color level was lower in growing Namhae ($20.79{\pm}1.20$) and Muan ($20.91{\pm}1.62$). 25~30 peaks obtained from aroma compounds analysis of various garlics. Among these compounds, 17 kinds of aroma compounds were identified by GC-MS. And, we showed that 15 kinds of compounds were sulfur containing aroma compounds. Diallyl disulfide was contained 34.90~60.54% in various garlic and the highest contents than other compounds in total compounds. It was detected the highest in China garlic but the lowest in growing Muan garlic. All of garlic samples showed antimicrobial activity against Strep. mutans, B. subtilis, E. coli. V. parahaemolyticus, Asp. flavus, and C. albicans. Especially antimicrobial activity was stronger against E. coli, and C. albicans. Antimicrobial activity was enhanced by sample addition volume and the highest in Namhae garlic sample.

Odorant Receptors Containing Conserved Amino Acid Sequences in Transmembrane Domain 7 Display Distinct Expression Patterns in Mammalian Tissues

  • Mammalian genomes are well established, and highly conserved regions within odorant receptors that are unique from other G-protein coupled receptors have been identified. Numerous functional studies have focused on specific conserved amino acids motifs; however, not all conserved motifs have been sufficiently characterized. Here, we identified a highly conserved 18 amino acid sequence motif within transmembrane domain seven (CAS-TM7) which was identified by aligning odorant receptor sequences. Next, we investigated the expression pattern and distribution of this conserved amino acid motif among a broad range of odorant receptors. To examine the localization of odorant receptor proteins, we used a sequence-specific peptide antibody against CAS-TM7 which is specific to odorant receptors across species. The specificity of this peptide antibody in recognizing odorant receptors has been confirmed in a heterologous in vitro system and a rat-based in vivo system. The CAS-TM7 odorant receptors localized with distinct patterns at each region of the olfactory epithelium; septum, endoturbinate and ectoturbinate. To our great interests, we found that the CAS-TM7 odorant receptors are primarily localized to the dorsal region of the olfactory bulb, coinciding with olfactory epithelium-based patterns. Also, these odorant receptors were ectopically expressed in the various non-olfactory tissues in an evolutionary constrained manner between human and rats. This study has characterized the expression patterns of odorant receptors containing particular amino acid motif in transmembrane domain 7, and which led to an intriguing possibility that the conserved motif of odorant receptors can play critical roles in other physiological functions as well as olfaction.

Effects of Biodegradable Mulching Film Application on Cultivation of Garlic (마늘 재배시 생분해성 멀칭 필름 이용효과)

  • The effect of biodegradable mulching film on the growth and development of garlic were investigated in order to develop eco-friendly weed control techniques. The treatments included biodegradable film (Bio-De) and black (Black-PE), green (Green-PE), transparent (Trans-PE) polyethylene mulching films. Non-mulched, bare soil (Non-mulching) was used as a control. Light transmittance value among tested mulching films was the highest in Trans-PE (86.1%) followed by Bio-DE and Green-PE, and the lowest value was observed for the Black-PE (1.1%). All mulching films without exclusion elevated soil temperature, especially Trans-PE and Bio-DE compared to bare soil. Plant height and mean bulb weight were increased due to mulching films with the highest values observed for Trans-PE and Bio-DE treatments. After seven months of field application there were no significant degradation signs on PE plastic films, whereas it was easy to see horizontal cracks on the Bio-DE film surface after five month of usage.

Experimental Studies on the Hair Growth Activity of Trimix Extracts of Mylabris Phalerata Pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata in C57BL/6N mice (반모(斑蝥), 남성(南星), 반하(半夏)(생(生))추출 혼합물의 발모효과에 대한 실험적 연구)

  • To determine whether topical application of trimix extracts of Mylabris phalerata Pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata lead to affects on the hair growth activity in C57BL/6N mice. To examine the hair growth activity of the extracts of Mylabris phalerata Pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata gross, microscopic, and immunohistochemical method were performed. In order to examine the mRNA expression of hair growth related substance, RT-PCR method was performed. Experimental group I on day 14, The most extensive hair growth activity was observed in whole skin area of all the mice whose hair had been clipped. Brdu immunoreactive cells of all the experimental groups were more heavily stained in epidermis, bulge, outer root sheath, inner root sheath, subcutaneous tissue, hair bulb and cutaneous trunci muscles than that of control group on day 12 of hair growing cycle in C57BL/6N mice. VEGF immunoreactive density of all the experimental groups was more heavily stained in epidermis, bulge and cutaneous trunci muscles than that of control group on day 12. FGF and c-kit immunoreactive cells of all the experimental groups were heavily stained in epidermis, outer root sheath, inner root sheath and cutaneous trunci muscles on day 12. PKC-$\alpha$ immunoreactive density of all the experimental groups was mildly stained in epidermis and cutaneous trunci muscles than that of control group on day 12. On day 12, the expression of bFGF (138%, 119%, 120%), VEGF (146%, 144%, 133%), IGF-1 (165%, 141%, 119%) and PLI (121%, 116%, 123&) in each experimental groups was more increased than that of control group. On day 16, The expression of IGF-1 (126%, 149%, 151%) in all the experimental group was more increased than that of control group (100%). The expression of bFGF (92%, 94%) and VEGF (101%, 97%), PL1 (102%, 109%) in all the experimental group was more decreased than that of experimental group I, II on day 12. But the expression of bFGF (109%) and VEGF (127%), and PL1 (105%) in each experimental group III was more increased than that of control group (100%). These experiments suggest that trimix extracts of Mylabris phalerata pall., Arisaematis Rhizoma and Pinelliae Rhizoma Ternata may stimulate the topical hair growth activity and its experimental group I can be useful for treatment of alopecia areata.

Associations between Carotid Intima-media Thickness, Plaque and Cardiovascular Risk Factors (경동맥 내중막 두께 및 죽종과 심 혈관 질환 위험요인과의 관련성)

  • Objectives : This study was conducted to examine the association between the carotid artery intima-media thickness (IMT), plaque and cardiovascular risk factors according to gender and age. Methods : The data used for this study were obtained from 1,507 subjects (691 men, 816 women), aged 20-74 years, who participated in 'Prevalence study of thyroid diseases' in two counties of Jeollanam-do Province during July and August of 2004. The body mass index (BMI) and waist hip ratio (WHR) were calculated by anthropometry The blood pressure, pulse rate, pulse pressure, total cholesterol, triglyceride, HDL cholesterol and fasting blood sugar level were also measured. Ultrasonography was used to measure the carotid artery IMT and plaque. IMT measurements were performed at 6 sites, including both common carotid arteries, and the bulb and internal carotid arteries. The definition of the 'mean IMT' was mean value obtained from these 6 sites. Results : The ${\pm}$ standard deviation IMT values were $0.65{\pm}0.14\;and\;0.60{\pm}0.13mm$ in men and women (p<0.001), respectively. The data were analyzed according to gender and the 50 year age groups. In a multiple linear regression analysis, age and hypertension were positively associated with the mean IMT in both men and women, aged<50 years. Age, total cholesterol and smoking (current) were positively associated with the mean IMT in men $(\geq50\;years)$. Age was positively associated with the mean IMT in women $(\geq50\;years)$, but the HDL cholesterol level was negatively associated. The prevalence of plaques was 44.2%(196/443) in men and 19.4%(89/459) in women, for those greater than 50 years of age. In a multiple logistic regression analysis, age (OR=1.090, 95%CI=1.053-1.129), HDL cholesterol (OR=0.964, 95%CI=0.944-0.984), total cholesterol (OR=1.009, 95%CI=1.002-1.017)and BMI (OH=0.896, 95%CI=0.818-0.983) were independently associated with plaques in men; whereas, age (OR=1.057, 95%CI=1.012-1.103), HDL cholesterol (OR=0.959, 95%CI=0.932-0.986), pulse pressure (OR=1.029, 95%CI=1.007-1.050) and triglycerides (OR=0.531, 95%CI=0.300-0.941) were independently associated with plaques in women. Conclusions: There were significant gender and aging differences in the association between the IMT, plaque and cardiovascular risk factors. Therefore, for the prevention of atherosclerosis, selective approaches should be considered with regard to gender and age factors.

Anti-glycemic effect of L-carnosine in streptozotocin-induced diabetic mice (Streptozotocin으로 유도된 당뇨 마우스에서 L-carnosine의 혈당강하 효과)

  • Carnosine is a dipeptide $(\beta-alanyl-L-histidine)$ found in mammalian brain, eye, olfactory bulb and skeletal muscle at high concentrations. Its biological functions include antioxidant and anti-glycation activities. The objectives of this study were to investigate anti-diabetic effects of carnosine as determined by blood glucose levels, glucose tolerance test (GTT), glycosylated hemoglobin, and serum biochemical and lipid levels in streptozotocin-induced diabetic mice. There were five experimental groups including normal (ICR mice), control (saline), and three groups of carnosine at doses of 6, 30, and 150 mg/kg b.w.. Carnosine was orally administered to the diabetic mice everyday for 12 weeks. There was no significant difference in body weight changes in carnosine-treated groups compared to the control. The treatments of carnosine at the dose of 6 mg/kg significantly decreased the blood glucose level compared with the control at 2 and 4 weeks. The treatments of carnosine at the doses of 6 and 30 mg/kg significantly decreased the blood glucose levels in GTT and glycosylated hemoglobin compared with the control. Carnosine significantly increased total proteins compared with the control. Carnosine at the dose of 6 mg/kg significantly decreased total cholesterol and triglyceride in the serum compared to the control. These results suggest that carnosine at a low level has a hypoglycermic effect resulting from reduction of blood glucose and that a carnosine-containing diet or drug may give a benefit for controlling diabetes mellitus in humans.

Phototaxis of Fish 7. Response of Sea-Bass to the Colored Lights (어류의 주광성에 관한 연구 7. 색광에 대한 농성어의 반응)

  • The author carried out an experiment to find out the response of sea-bass, Epinephelus septemfasciatus (Thunberg) to the color lights. The experimental tank (360L$\times$50W$\times$55H cm) was set up in a dark room. Six longitudinal sections with 60 cm intervals are marked in the tank to observe the location of the fish. Water depth in the tank was kept 50 cm level. Light bulbs of 20 W at the both ends of the tank projected the light horizontally into the tank. Two different colored filters were selected from four colors of red, blue, yellow, and white, and they were placed in front of the light bulbs to make different colors of light. Light intensity were controlled by use of auxiliary filters intercepted between the bulb and the filter. The fishes were acclimatized in the dark for 50 minutes before they were employed in the experiment. Upon turning on the light, the number of fish in each section was counted 40 times in 30 second intervals, and the mean of the number of fish in each section was given as the gathering rate of the fish. The colors favourited by the fish was found in the order of blue, red, white and yellow in day time, and red, blue, yellow and white at night time. The gathering rate of fish on illumination period was not constant and fluctuated with irregularity. The difference of the gathering rate on two different colors of light was small and the difference was larger in night time then in day time.

Molecular characterization and expression pattern of a novel Keratin-associated protein 11.1 gene in the Liaoning cashmere goat (Capra hircus)

  • Objective: An experiment was conducted to determine the relationship between the KAP11.1 and the regulation wool fineness. Methods: In previous work, we constructed a skin cDNA library and isolated a full-length cDNA clone termed KAP11.1. On this basis, we conducted a series of bioinformatics analysis. Tissue distribution of KAP11.1 mRNA was performed using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) analysis. The expression of KAP11.1 mRNA in primary and secondary hair follicles was performed using real-time PCR (real-time polymerase chain reaction) analysis. The expression location of KAP11.1 mRNA in primary and secondary hair follicles was performed using in situ hybridization. Results: Bioinformatics analysis showed that KAP11.1 gene encodes a putative 158 amino acid protein that exhibited a high content of cysteine, serine, threonine, and valine and has a pubertal mammary gland) structural domain. Secondary structure prediction revealed a high proportion of random coils (76.73%). Semi-quantitative RT-PCR showed that KAP11.1 gene was expressed in heart, skin, and liver, but not expressed in spleen, lung and kidney. Real time PCR results showed that the expression of KAP11.1 has a higher expression in catagen than in anagen in the primary hair follicles. However, in the secondary hair follicles, KAP11.1 has a significantly higher expression in anagen than in catagen. Moreover, KAP11.1 gene has a strong expression in inner root sheath, hair matrix, and a lower expression in hair bulb. Conclusion: We conclude that KAP11.1 gene may play an important role in regulating the fiber diameter.

Effect of Housing on Physiological Responses and Energy Expenditure of Sheep in a Semi-arid Region of India

  • An investigation was carried out to study the effect of two housing systems on physiological responses and energy expenditure of sheep in a semi-arid region of India. Two types of housing management were adopted. First was a shed- $6{\times}3\;m^2$ structure with all the four sides of 1.8 m chain link fencing with a central height of 3 m. The roof was covered with asbestos sheets and with mud floorings. Second was an open corral- $6{\times}3\;m^2$ open space with all the four sides covered with 1.8 m chain link fencing. Thirty-four (32 ewes and 2 rams) sheep of native Malpura breed aged about 18 months (body weight 28 kg ewes; 35 kg rams) were grazed together on a 35 ha plot of native range. All the sheep were grazed as a flock from 08.00 to 17.00 h during a yearlong study. The flock was divided into two groups (16 ewes+1 ram) in the evening and housed as per the systems (Shed and Open Corral). Dry and wet temperatures were recorded at 06.00 h and 21.00 h using a wet and dry bulb-thermometer both inside the shed and in the open corral and temperature humidity index (THI) was calculated. There was significant (p<0.05) difference in the THI between shed and open corral in all the seasons, indicating that shed was always warmer compared to open corral. Rectal temperature (RT) of both the groups of sheep was similar during morning as well as evening throughout the seasons. There were significant (p<0.05) differences in the skin temperature (ST) and respiration rate (RR) between the two groups at both the measurements in all the seasons. Highest energy expenditure (EE) was recorded inside the shed at 21.00 h (224 kJ/h) during monsoon and lowest at 6.00 h during winter (119 kJ/h). There was a significant (p<0.05) difference between the EE inside the shed and that in the open corral. It was concluded that housing had significant effects on the physiological responses and EE of sheep. Provision of housing at night was stressful during monsoon (with less rainfall) and summer, whereas it was protecting the sheep from acute cold during winter in a semi-arid region of India.

The isolation of Bacillus subtilis KYS-10 with antifungal activity against plant pathogens (식물 병원균에 대한 항진균 활성을 갖는 Bacillus subtilis KYS-10의 분리)

  • This study was investigated for the purpose of the isolation and identification of antagonistic bacteria with antifungal activity against plant pathogens. This bacteria denominated Bacillus subtilis KYS-10 and the optimum growth condition were 4% sucrose, 1% yeast extract, 0.2% $K_2HPO_4$, pH 7, 150 rpm, $30^{\circ}C$, 8 day. The antifungal activities against nine plant pathogens determined inhibition zone size by diffusion methods. The results, G. zeae (scab) 70 mm and P. grisea KACC 40439 (blast), P. capsici KACC 40177 (phytophthora blight) and C. destructans KACC 41077 (root rot of ginseng) 40~43 mm, and C. gloeosporioides KACC 43520 (ripe rot), C. gloesporioides KACC 40003 (anthracnose), S. shiraiana KACC 41065 (stem rot) and S. shiraiana (mulberry sclerotial disease) 35~39 mm and F. Oxysporum KACC 44452 (bulb rot of ginseng) 28 mm. From these experiment results, author suggest that Bacillus subtilis KYS-10 would be developed as a biological control agent thorough the field experimet in the near future.
