The Analysis of Verbal Interaction in Elementary Science Programs Using Multi-Level Instruction (다수준 포함 교수법을 적용한 초등과학 프로그램에서의 언어적 상호작용 분석)

  • The purpose of this study was to develop elementary science programs that used multi-level instruction and to analyze verbal interaction patterns in science classes that applied these programs. For this study, the 27 children from the fifth grade at B elementary school, located in Gyeonggi-do, were selected and separated into heterogeneous groups of four students. Verbal interactions occuring in two groups during each class were recorded using video. Elementary science programs using multi-level instruction were developed to target a fifth grade second semester 'Lesson 1. Human Body'. This program provided a mission form for each group and evaluation form for each child. A mission form depending on the children's level has different colors and levels of difficulty for questions. The evaluation form is composed of questions suitable for a child's level in reaching the goal with key concepts. The verbal interaction was mostly categorized into the cognitive domain and the affective domain for analysis. The cognitive domain was subdivided into question, response, making solution, receiving opinion, and the affective domain was divided into behavioral participation and students' attitude. Results of study showed that the frequency of the cognitive domain was higher than the frequency of the affective domain. In the cognitive domain, the median-level was of highest frequency in the children. In the affective domain, high-level was of highest frequency in the children. In terms of both the cognitive and affective domains of children, low-level exhibited the lowest frequency. Verbal interaction frequency was no difference between high-level and median level in cognitive and affective aspects, so median-level children were actively participating in activities similarly with high-level children. There were more types of interactions question, response, making solution, students' attitude in the median-low level children's verbal interaction than high-median level children's verbal interaction.

Effects of Probiotic Complex on Performance, Blood Biochemical and Immune Parameters, Digestive Enzyme Activity, Fecal Microbial Population and Noxious Gas Emission in Broiler Chicks (복합생균제가 육계의 생산성, 혈액생화학성분과 면역지표, 소화효소 활성도, 분중 미생물 및 유해가스 발생에 미치는 영향)

  • This study examined the effects of a probiotic complex (PC) containing Lactobacillus plantarum, Bacillus subtilis, and Saccharomyces cerevisiae on growth performance, organ weight, immune parameters, fecal microbial count, and noxious odor in broiler chicks. A total of 216 birds (4-day-old) were fed a basal diet (CON) and a diet supplemented with 0.25% (PC1) and 0.5% (PC2) of PC until 35 days of age. No difference in body weight, feed intake, and FCR was observed among the groups. The intestinal mucosal weight of the PC1 group was greater than that of the CON group without affecting weights of the other organs. Intestinal secretory immunoglobulin A (sIgA) levels in the PC2 group increased significantly (P<0.05) compared with that in the CON group. The PC2 group also had a strong tendency for elevated blood sIgA levels. Dietary PC did not affect the level of interleukin-1β in the blood and mucosal tissues or alter maltase, sucrase, and leucine aminopeptidase activities in the intestinal mucosa. The PC2 group had higher colony-forming units (cfu) for L. plantarum and S. cerevisiae, but lower cfu for E. coli than those in the CON group. Compared to the CON diet, the PC2 diet resulted in a decreased H2S concentration and a tendency toward decreased CH3SH concentration. In conclusion, a 0.5% PC diet showed increased sIgA and desirable microbial population, and decreased noxious odor in the feces, suggesting that PC could be applied as an environmentally friendly feed additive in broiler chicks.

A Survey on Dietary Habits in Gyeongnam and the Development of the Nutrition Education Curriculum with Teacher's Guide for Obese Elementary School Children (경남지역 일부 초등학교 비만아동의 식습관 분석 및 영양교육을 위한 교수학습과정안 개발)

  • The purpose of this study was to develop a nutrition education curriculum with teacher's guide which includes discretionary activities for obese children. A survey was carried out to investigate the recognition of body image and food behaviors according to the obesity index (mild, moderate, severe) in school children (4~6th grade, 158 boys and 60 girls) who were selected based on a physical examination in May, 2006 in the Gyeongnam province. Next, a nutrition education curriculum with teacher's guide was developed on the basis of the findings from the survey and from preceding researches. The results are summarized as follow. The results of this study showed the existence of some nutritional problems such as overeating, prejudice, skipping meals, snacking patterns, etc, which indicate the need for nutritional management for obese children. Most overweight children (80.3%) showed the most interest in the nutrition education program, particularly with regards to dieting for weight control (64.7%). The developed nutrition education curriculum consisted of 8 main subjects and 13 subtitles. The curriculum was prepared for 13 lessons and included songs and singing, making-up lyrics, games about nutrition, discussions of the experience of eating (satiety, thirst, hunger), debates on dietary habits, writing and others to promote the interest for learning. We aimed to develop this program in an attempt to improve the dietary habits of obese school children. This is very important because once a dietary habit is formed in adults, it is difficult to change and the best adjustable stage is during childhood. Therefore, early nutrition education during elementary school can change and build-up the awareness of health in young elementary school children.

Anti-aging and Anti-diabetes Effects of Aconitum pesudo-laeve var. erectum Extracts (진범(Aconitum pesudo-laeve var. erectum) 추출물의 항노화 및 항당뇨 효과)

  • Aconitum pesudo-laeve var erectum has been known to possess anti-inflammatory activity and modulate the intestinal immune system. In addition, it has traditionally been used for the treatment of water retention in the body. In this study, the anti-aging and anti-diabetes effects of water and ethanol extracts from Aconitum pesudo-laeve var. erectum were investigated. The activities of each extract were measured by antioxidant tests such as DPPH and ABTS radical scavenging activity, antioxidant protection factor (PF), TBARs content, and ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibition activity assay. DPPH radical scavenging activity was found in over 50% of water and ethanol extracts at $100{\mu}g/ml$, $50{\mu}g/ml$, respectively. The ABTS radical scavenging activity of ethanol extract was $99.8{\pm}0.1$% at $1,000{\mu}g/ml$ in water, which was highest among the ethanol extract concentrations. PFs measured with ${\beta}$-carotene-linoleate model systems were in the order of ethanol (1.49 PF at $1,000{\mu}g/ml$) > ethanol (1.40 PF at $500{\mu}g/ml$) > water (1.33 PF at $1,000{\mu}g/ml$) > water (1.27 PF at $500{\mu}g/ml$). TBARs content in ethanol extracts ($1,000{\mu}g/ml$) was $0.16{\pm}0.03{\mu}M$, which was lower than that of water extracts and other ethanol extract concentrations. The extracts also showed over 90% of ${\alpha}$-amylase inhibition and over 60% of ${\alpha}$-glucosidase inhibition ratio in water ($1,000{\mu}g/ml$) and ethanol extracts (100~$1,000{\mu}g/ml$). These results suggest that Aconitum pesudo-laeve var. erectum extracts could be used as a cosmetic source and preventive agent for aging and diabetes.

Changes in $A_{1}$, Adenosine Receptor-Adenylyl Cyclase System of Rat Adipocytes Fellowing Induction of Experimental Diabetes by Streptozotocin Treatment (Streptozotocin으로 당뇨병을 유발시킨 쥐의 지방세포에 나타나는 $A_{1}$, Adenosine Receptor-Adenylyl Cyclase System의 변화)

  • Adenosine receptors in rat adipose tissues have been reported to be of $A_{1}$ subclass, and their stimulation leads to inhibition of adenylyl cyclase, resulting in inhibition of lipolysis. In the present study we investigated changes in $A_{1}$ adenosine receptor-adenylyl cyclase system of adipocytes following induction of experimental diabetes in rats. One week following experimental diabetes were induced by intravenous injection of streptozotocin (50 mg/kg body wt.), adipocytes from rats $(170{\sim}230g)$ fed ad libitum were isolated using collagenase. When adipocytes were incubated for 1 h with 1 unit/ml adenosine deaminase and $1\;{\mu}M$ isoproterenol, and assayed for glycerol formation, it was found that the inhibition of lipolysis in diabetic adipocytes by $(-)-N^{6}-(R-phenylisopropyl)adenosine$ (PIA), an $A_{1}$, adenosine receptor agonist, was twice that of control adipocytes. In an effort to delineate the mechanism(s), $[^{3}H]PIA$ binding to adipocytic membranes from diabetic and control rats were determined. Neither the affinities nor numbers of $A_{1}$ adenosine receptor were significantly different from each other (Best fit parameters for the one-site model are: $K_{d}=0.51{\pm}0.09nM$ and $B_{max}=1.60{\pm}0.12\;pmoles/mg$ protein for control membranes; $K_{d}=0.54{\pm}0.21\;nM$ and $B_{max}=1.72{\pm}0.31\;pmoles/mg$ protein for diabetic membranes). However, the inhibiton by PIA of the isoproterenol-stimulated adenylyl cyclase activities was found to be 1.9 times higher in adipocytic membranes from diabetic rats than those from controls. These results suggest that the increased sensitivity of inhibition of lipolysis to PIA in adipocytic membranes from diabetic rats is due to changes in signal transduction pathways, rather than alterations of $A_{1}4 adenosine receptor molecules themselves.

Changes of Haematological Constituents in the Nile tilapia, Oreochromis niloticus Exposed to HCB and PCBs (HCB와 PCBs에 노출된 나일틸라피아, Oreochromis niloticus의 혈액성분의 변화)

  • The study was carried out to investigate the changes of haematological constituents induced by dietary exposure of Hexachlorobenzene (HCB) and Polychlorinated biphenyls (PCBs) in nile tilapia, Oreochhromis niloticus. The nile tilapia were fed pellet with 0.05, 0.25 and 0.50 mg/kg body weight/day of HCB and PCBs for 30 days, respectively. HCB ($\ge$ 0.25 mg/kg) induced a significantly decreased the red blood cell count at 30 days, and the hemoglobin concentration and hematocrit value were significantly decreased after 20 days to 0.50 mg/kg. The diminution of red blood cell count, hemoglobin concentration and hamatocrit value by PCBs was found after 20 days at $\ge$0.25 mg/kg. The concentrations of calcium decreased significantly at 30 days at $\ge$0,25 mg/kg of PCBs-diet. However, magnesium and inorganic phosphorus did not show any noticeable changes in the range of 0.05$\~$o.50 mg/kg of HCB and PCBs compared to control group during the experimental period, The low concentration of total protein was observed at the last day of experiment to 0.50 mg/kg of HCB-diet, while total protein initiated to increase from 20 days at the 0.25 mg/kg of PCBs-diet. The glucose was significantly increased after 20 days at $\ge$0,25 mg/kg of HCB and PCBs, A significant increment GOT and GPT activities was noticed after 20 days at 0.50 mg/kg of HCB and PCBs, These results indicate that hematological and serum biochemical changes in the nile tilapia by dietary exposure to HCB and PCBs are affected at more than 0,25 mg/kg after 20 days.

Effect of a Herb Mixture (HIM-I) on the Protection of the Hematopoietic-Immune System and Self-renewal Tissues against Radiation Damage (면역조혈계 및 재생조직의 방사선 손상에 대한 생약복합물(HIM-I)의 방호 효과)

  • In previous studies, we evaluated the effect of the 6 energy-tonic or blood-building prescriptions of traditional oriental medicine, and observed that Si-Wu-Tang and Bu-Zhong-Yi-Qi-Tang showed high activity in the protection of the gastrointestinal and hematopoietic organs in irradiated mice. But any of these prescriptions did not show a high activity in the activation of the immune cells. We performed this study to design an herb mixture which protects the self-renewal tissues and also promotes recovery of the immune system against radiation damage. In order to meet all the requirements, we designed a new mixture of 3 edible herbs listed in Korean Food Code. The mixture of Angelim gigas radix, Cnidium officinale rhizoma and Paeonia japonica radix was decocted with hot water, and the activities of the water extract (HIM-I) were evaluated. HIM-1 stimulated the immune cells in a much higher extent than the traditional prescriptions, and promoted dramatically the growth of bone marrow stem cells in vitro. Also, HIM-1 protected digestive and hematopoietic organs against radiation as effectively as the 2 prescriptions, Si-Wu-Tang and Bu-Zhong-Yi-Qi-Tang. On the other hand, it showed high in vitro antioxidative activity that might be considered as a mechanism of the protective effects against radiation. Although the detailed mechanisms of those effects remain to be elucidated, these results indicated that HIM-I might be a useful agent for protection and recovery of body from various risk factor as well as radiation, especially since it is a relatively nontoxic natural product.

Effects of Vegetable Sprout Power Mixture on Lipid Metabolism in Rats Fed High Fat Diet (새싹채소 혼합분말이 고지방 식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of vegetable sprout powder on serum and adipose tissue lipid metabolism in rats fed high-fat diet for 4 weeks for induction hyperlipidemic model rat. Weight-matched male Sprague-Dawley rats were assigned to five groups according to dietary fat level (10% or 20% of diet wt.) and mixture of vegetable sprout powder levels (5% or 10% 10% or 20% of diet wt.). Vegetable sprout powder was the mixture of same amounts of dried barley, broccoli, rapeseed, alfalfa, radish, mustard, buckwheat and brussels sprouts. Experimental groups were normal fat diet with 5% cellulose (NF-C), high fat diet without fiber (HF-N), high fat diet with 5% cellulose (HF-C), HF-C diet with 5% vegetable sprout powder (HF-CSL), and HF-C diet with 10% vegetable sprout powder (HF-CSH). The body weight of HF-N group increased 16% compared with the NF-C group, while it was decreased by 15% and 22% for HF-CSL group and HF-CSH group, respectively. Fat mass and fat cell size of adipose tissue were lower in HF-CSL group and HF-CSH group compared with HF-C group, and lower in HF-CSH group compared with HF-CSL group. Serum triglyceride, total cholesterol and LDL-cholesterol contents were markedly decreased by vegetable sprout powder containing diet, while the serum HDL-cholesterol and phospholipid contents were higher in vegetable sprout powder containing diet in a dose-dependent manner. Leptin and insulin levels in serum showed a decrease in HF-CSH group. Significantly increased contents of triglyceride, total cholesterol, LDL-cholesterol, leptin and insulin in the serum of HF-N group were returned to normal or even below normal levels by feeding 10% vegetable sprout powder diet. The increased activities of NADP-malate dehydrogenase (ME), glucose-6-phosphate dehydrogenase (G6PDH) and 6-phosphogluconate dehydrogenase (6PGDH) and lipoprotein lipase (LPL) in adiposetissue by HF-N group were decreased to the activity of normal fat group by feeding vegetable sprout powder in a dose-dependant manner. These results indicate that lipid metabolism in rats fed high-fat diet was suppressed by feeding vegetable sprout powder.

Antioxidant Effect of Chungkukjang Supplementation against Memory Impairment induced by Scopolamine in Mice (Scopolamine으로 유도된 기억 손상 마우스에서 청국장 식이의 항산화 효과)

  • In this study, the antioxidant effect of Chungkukjang supplementation against memory impairment and oxidative stress in scopolamine (2 mg/kg i.p)-injected mice was investigated. The experimental animals were divided into five groups and fed experimental diets for 12 weeks; normal diet group (C), scopolamine + normal diet group (S), scopolamine + 63.0% soybean Chungkukjang supplementation group (SS), scopolamine + 45.0% Yakkong Chungkukjang supplementation group (SY), and scopolamine + 50.0% black foods such as black rice, black sesame seeds, and sea tangle added Yakkong Chungkukjang group (SYB). For the results of food intake, body weight gain, and brain weights, levels of scopolamine-injected groups were lower than the levels of the control group. The reduced brain weight of the scopolamine-injected group (S) was regulated to control level by supplementation of three types Chungkukjang. In the oxidative stress indicator, nitric oxide and malondialdehyde levels in serum of scopolamine-injected mice were higher than those of other groups. However, supplementation of soybeans, Yakkong and black foods added Yakkong Chungkukjang was proven to regulate them. Antioxidant enzyme activities such as superoxide dismutase (SOD) and glutathione-S-transferase (GST) in serum showed no significant differences among the groups. The reduced levels of vitamin A and vitamin E in serum and brain tissue of scopolamine-injected mice were controlled by supplementation of three types of Chungkukjang. Total antioxidant capacity (TAC) of scopolamine-injected group was lower than those of other groups. However, TAC was significantly elevated by Chunggukjang supplementation. Therefore, antioxidative effects of soybeans, Yakkong, and black foods added Yakkong Chungkukjang supplementations against oxidative stress in scopolamine-injected in mice could expected.

Study on Anti-obesity and Hypoglycemic Effects of Lycium chinense Mill Extracts (구기자 추출물의 항비만 및 혈당강하 효과)

  • This study was designed to evaluate the anti-obesity and hypoglycemic effects of Gugija (Lycium chinense Mill) extracts in 3T3-L1 adipocytes. We investigated the $\alpha$-amylase and $\alpha$-glucosidase inhibitory activities of extracts from Gugija. Gugija was extracted by 70% EtOH and 80% MeOH and aqueous, respectively. A single oral dose of Gugija extract inhibited the increase of blood glucose levels significantly at 0, 30, 60, 90 and 120 min and decreased incremental response areas under the glycemic response curve. These results suggest that Gugija 70% EtOH extracts may delay carbohydrate digestion and reduce postprandial hyperglycemia. In addition, triglyceride content in 3T3-L1 adipocytes decreased at higher concentrations of Gugija 70% EtOH extract. Free fatty acid content in 3T3-L1 adipocytes was increased at higher concentrations of Gugija 70% EtOH extract. Also, glucose transporter 4 (GLUT4), the key insulin signaling pathway transcription factor, was remarkably increased by the Gugija 70% EtOH extract when compared to those of control cells in protein expression levels. Therefore, Gugija can be developed as an effective anti-obesity and hypoglycemic agent.
