  • Physiologic races of Erysiphe graminis f. sp. hordei were isolated from diseased barley plants collected from 93 locations in Korea, 1980 and 1982. Nine races, 3,8,11,19, J13, K1A, K1B, K2, and K3, were identified by using Cherewick's differential plants. Among them, races K1A, K1B, K 2, and K 3 were firstly recorded by authors in Korea. These races were mainly collected from Jeonnam and Gyeongnam province, whereas race 8 was isolated only in Buan, Jeonbug province. On the other hand, race J 13 previously reported in Japan was prevalent as $36.6\%$ of total isolated races, mostly being isolated in southern region of Korea.

  • This experiments were carried out determine planting date for highest Yields and select an covering material for enhancement of environments in Cassia tora L.The highest Yields by regressions estimate produced 352 kg/10a at May 18. of coure, it is possible to plant at June and early-July in spite of slight decreas of Yields, therefore Cassia tora L. is benefit to establish cropping system with barley, wheat or other crops. The covering materials for highest yielding was Black-white nonporous PE(low density) and it out yielded about 53% than conventional cultivations. Cassia tora L. is possible to produce Without agricultural medicines because of little occurence of decrease except occuring of Damping off(Rhizoctonia solanikuhn) at early planting.

First Report of Charcoal Rot Caused by Macrophomina phaseolina on Peanut Plants in Korea (땅콩에서 Macrophomina phaseolina에 의한 균핵마름병 발생 보고)

  • Peanut plants showing mild wilt were found in fields of Iksan, Korea, in August 2021. The diseased peanut plants were collected, and the causal pathogens were isolated using potato dextrose agar (PDA) medium. The isolated IS-1 strain formed white mycelia on PDA, which turned black with age. Sclerotia were produced on the PDA and barley leaves laid on water agar 7 d after incubation at 30℃. The sequences of both the internal transcribed spacer (ITS) region and calmodulin gene of IS-1 showed a 100% similarity with that of Macrophomina phaseolina. A phylogenetic tree constructed using the ITS regions of fungal pathogens causing disease in peanut plants indicated that the IS-1 stain belongs to M. phaseolina. The inoculation of IS-1 sclerotia into peanut seedlings resulted in yellowing and wilt symptoms in aboveground plants and brown to dark rots in roots 35-40 d after inoculation. Overall, the morphological characteristics, molecular identification, and pathogenicity of IS-1 indicate that the causal pathogen is M. phaseolina. This is the first report of charcoal rot caused by M. phaseolina on peanut plants in Korea. Further study is needed to develop the control measures for charcoal rot in peanut plants.

Analysis on Hazard Microorganisms in Raw Materials and Processing Environment for Sunsik Manufacture (선식용 곡류원료 및 제조공정에 따른 유해미생물 오염도 분석)

  • Cereals are the main raw material for sunsik manufacture. As the fundamental processing step, it is very important to confirm the level of the microorganism contamination in the cereals. This study was carried out a micrbiological screening of cereal samples for sunsik from 19 companies located in South Korea. Ten kinds of cereals which were glutinous rice, barley, brownrice, blackbean, blackrice, blacksesame, sorghum, millet, perilla seed, and adlay were investigated. As the results, the contaminations of general bacterial were 3.1~8.6 log(CFU/g). The results of Escherichia coli were 1.0~3.4 log(CFU/g). There was no contamination of Salmonella. spp in any cereal samples except black sesame and mold was detected in barley. The experiment for microbiological contamination during sunsik processing was also investigated in this study. The results of general bacteria were detected as 5.1~8.5, 4.4~7.5, 1.0~2.3, 2.4~4.2, 1.0~4.0, 3.4~4.2, 4.3~5.2, and 3.3~5.5 log(CFU/g) during environment of warehousing, washing, steaming, 1st cooling, drying, 2nd cooling, grinding, and packaging process, respectively. The results of coliform were 1.0~2.0 log(CFU/g) during warehousing respectively. Mold was found in warehousing. In case of the instruments, the contaminations of general bacterial were 4.2~7.5, 0.1~2.0, 0.1~3.2, 3.7~4.0, 2.5~3.0, and 3.8~5.2 log(CFU/g) in cereals tanks, washing machines, grinding machines, packaging machines, and workrooms. The results of coliform were 2.4~4.0, 0.0~4.1 log(CFU/g) in cereals tanks and grinding machines, respectively. Mold were only found in cereals tanks, grinding machines, and workings. Therefore, the risk of hazard microorganisms contmination might be decrased as the exhaustive management is applied to the whole sunsik process.

Herbicidal Activities of Trichosanthes kirilowii Maxim Extracts (하늘타리 추출물의 제초활성)

  • Currently, methods for controlling weeds in organically produced crops have not been as effective as conventional methods. This research was carried out to determine the herbicidal effects of leaf, stem, fruit, root extracts of Trichosanthes kirilowii. The extraction methods used were water, boiling water and ethanol. The characteristics of potential herbicidal components among extraction methods were investigated by using the following solvent fractions: hexane, chloroform, ethyl acetate, butanol, and water. Generally, water extracts provided the best on inhibition of germination rate, plant height, and root length in cucumber and barley. Specifically, extractions made from fruit parts of T. kirilowii provided the greatest inhibition effect on plant growth in cucumber and barely. Inhibition of germination rate, plant height, and root length in cucumber and barley in solvent fractions was the best in water fractions, but there were no significant differences among the other fractions. Digitaria siliaris and Solanum nigrum were controlled 80-100% by 5% extractions of water fraction. However, there were no herbicidal effects from foliar treatment in cucumber, barley, black nightshade, and henry crabgrass by 5% extractions of the water fraction. These results show that extractions of T. kirilowii can be used for controlling some weeds in organically produced crops.

Cultural Condition for the Mycelial Growth of Phellinus igniarius on Chemically Defined Medium and Grains (화학합성배지 및 곡물을 이용한 Phellinus igniarius의 균사체 배양조건)

  • The chemical media composition and culture conditions were optimized for mycelial growth of Phellinus igniarius 26005. The method of solid-state fermentation, cultivation of basidiomycetal strains in various grains, was developed. Media composition for optimal growth of Phellinus igniarius 26005 was made of 7.0% malt extract, 0.3% bacto soytone, and 0.2% yeast extract. The optimum condition for mycelial growth was $28^{\circ}C$ and pH 7.0, respectively. For the mass cultivation of mycelia, the hydrated grains with cold water, were put into the plastic bottle. The mycelial growth rate in the bottled grains was high in the early stage with inoculation of homogenized mycelium. The activity of mycelium was maintained by adding sterilized water in the middle of cultivation. The glucosamine content which determins the mycelial growth rate in solid material was in the order of job's tears>barley>black soybean>wheat>malt soybean>brown rice>sorghum>glutinous rice.

Analysis of Selenium in Grain with ORC Collision-Removal of Br Interference using Mathematical Calibration (ORC ICPMS에서의 곡류중의 셀레늄 분석-수학적 보정을 이용한 Br의 간섭제거)

  • The concentration of selenium in grain samples was determined using isotope dilution method in ORC-ICPMS. The experimental conditions were optimized to $H_2$ mode and the flow rate was $4.0\;mL\;min^{-1}$. ORC in $H_2$ mode proved to eliminate most of polyatomic interferences except $BrH^+$ when Br is present in sample matrix. Chemical removal of Br was very difficult and the mathematical correction was successfully employed. The fraction of $BrH^+$ generated from Br at the current experimental condition was 14.1%. The signal on m/z 82 was corrected and calculated for isotope dilution. The analytical reliability of the propose method was successfully evaluated by analyzing the certified standard reference material NIST SRM 1566 and 1567. The method was applied to real samples and the results are $0.034{\pm}0.001\;{\mu}g\;g^{-1}$ for white rice, $0.059{\pm}0.002_5\;{\mu}g\;g^{-1}$ for brown rice, $0.029{\pm}0.001_4\;{\mu}g\;g^{-1}$ for black rice, and $0.034{\pm}0.002\;{\mu}g \;g^{-1}$ for barley. The detection limits ($3\sigma$) for Se was $0.012\;ng\;g^{-1}$.

Effects of Rice Added with Mulberry Leaves and Fruit on Blood Glucose, Body Fat and Serum Lipid Levels in Rats (뽕잎오디쌀이 흰쥐의 혈당농도와 체지방 및 혈청지질 분획에 미치는 영향)

  • This study was performed to investigate the effects of rice added with mulberry leaves and fruit on blood glucose, body fat and serum lipid levels in Sprague-Dawley male rats for 4 weeks. Mulberry rice diet was prepared with mixture of 50% of purified AIN-diet and 50% of formulated rice consisting of soybean, barley, polished rice, black rice, uncleaned rice bud, mulberry leaves and fruit. The experimental animals were fed the mulberry rice diet (group A) and the purified AIN-diet (group B, diabetic control) for 4 weeks in rats injected with streptozotocin. In terms of observing hypolipidemic effect of mulberry rice, rats were fed either high-fat diet (13% fat) with additional lard, corn oil, cholesterol to AIN-diet (group C, control) or mulberry rice diet mixed with high fat diet (group D) for 4 weeks. Blood glucose level at the 4th week of group A had increased 5 mg/dL compared with that of day 0, while group B increased 51 mg/dL. However, the glucose levels of the groups A and B at the final day were 156 mg/dL (12.4% decrease compared with B) and 178 mg/dL, respectively. As for hypolipidemic effect, weight gain and body fat were 8% lower in the group D and serum triglyceride level also 19% lower in the group D compared with those of group C (p<0.05).

Quality and Physicochemical Characteristics of Rice Cooked along with Various Mixed Grains and by Following Different Cooking Methods (취반방법에 따른 혼합잡곡밥의 품질 및 이화학 특성)

  • This study evaluated the quality characteristics, the presence of polyphenolic compounds, and radical scavenging activity of rice cooked along with various mixed grains (barley, black soybean, adzuki beans, foxtail millet, proso millet, sorghum, glutinous rice) by following two different cooking methods (normal and pressure cooker). The amylogram and water characteristics of mixed grains showed significant differences based on the presence of different types of mixed grains. The chromaticity, palatability characteristics, presence of phenol compounds, and radical scavenging activity of rice cooked along with different mixed grains showed significant differences according to the nature of mixed grains. Total polyphenol contents of before cooking, cooked-rice added to mixed grains cooked in the normal cooker and a pressure cooker were 4.46~5.16, 0.58~0.93 and 0.65~0.96 mg GAE/g, and total flavonoid contents were 250.74~548.89, 129.26~207.04 and $127.41{\sim}218.15{\mu}g\;CE/g$, respectively. The DPPH radical scavenging activity of before cooking, cooked-rice added to mixed grains cooked in the normal cooker and a pressure cooker was 79.25~181.61, 22.07~53.64 and 7.51~39.97 mg TE/100 g, and ABTS radical scavenging activity was 203.25~328.24, 47.28~84.94 and 58.27~99.51 mg TE/100 g, respectively. Accordingly, it is necessary to different combinations of mixed grains according to the cooking method at home and grain industry.

  • In 2005, average temperature was lower, and average rainfall was less than those of previous year. The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Bacterial blight, bacterial grain rot, and panicle disease of rice, black rot of pear, and white rot and bitter rot of apple were severe. Especially, brown rot of rice occurred four times higher than those of previous year. Panicle blight of rice increased about 3 times, compared with the previous year, presumed that the higher rainy days, rainfall and RH promoted spread of the fungal pathogens to panicles of rice. The diseases of rice leaf blast, sudden wilt syndrome, downy mildew and powdery mildew of chinese melon in plastic greenhouse, and virus diseases of hot pepper occurred distinctly less than those of the previous year. Another diseases surveyed occurred similar or less.
