청국장제조시 대두원료의 동결과 쑥추출물의 첨가가 품질 및 이화학적 성분변화에 미치는 영향 (Effect of mugwort extract on the quality and the changes of chemical compositions of the Chung-kookjang prepared with frozen soybean)

  • 청국장제조시 대두원료의 동결과 쑥추출물의 첨가가 청국장의 숙성에 미치는 영향을 검토하고자 청국장 숙성과정중의 아미노태 질소, ${\alpha}-amylase$ activity, protease activity, 향기성분의 변화와 관능특성을 조사하였다. 청국장 숙성과정중 protease activity는 동결처리한 대두를 사용한 청국장에서 약간 높았던 반면 ${\alpha}-amylase$ activity는 차이가 없었다. 아미노태 질소량은 전체적으로 동결처리한 청국장이 다소 높은 값을 보였으며 숙성기간이 길어짐에 따라서 점차적으로 증가하다가 숙성 21일후 감소하였다. 쑥추출물의 첨가에 따른 효소활성의 유의적인 차이는 없었다. 쑥추출물의 첨가에 의해 불쾌취원인의 하나로 알려진 cis-3-hexenol의 생성량이 감소되었고, 동결에 의해 항기물질인 2,6-dimethyl pyrazine의 생성이 촉진되었다. 관능검사 결과 14일 숙성된 청국장이 전체적으로 가장 높은 선호도를 보였으며, 쑥추출물이 첨가된 경우에는 대두의 동결여부에 관계없이 쑥이 첨가되지 않은 경우보다 flavor가 유의적으로 우수하였으나 대두원료의 동결에 따른 차이는 감지되지 않았다.

식물추출물 항산화효능 기전의 일부로서의 활성산소 발생 억제 효과 (Suppression of reactive oxygen species generation as a part of antioxidative effect of plant extracts)

  • 세포 내에서의 활성산소(ROS) 생성을 억제하는 일은 항산화제의 작동방식으로 기존에 알려진 ROS의 화학적 소거보다 조직의 산화적 손상을 줄이는 방법으로서 효과가 뛰어날 것으로 기대되지만, 이러한 전략은 지금까지의 연구들에서 거의 검토되지 않고 있다. 본 연구에서는 항산화 효능이 알려진 식물시료들이 실제로 세포에서 ROS 생성을 제어하여 항산화 효과를 발휘하는지를 쇠뜨기, 가죽나무잎, 달맞이순, 그리고 토마토의 에탄올 추출물을 가지고 조사하였다. 이 네 가지 식물 시료들은 모두 비슷하게 세포내 ROS의 수준을 감소시켰다. 그러나, 가죽나무잎, 달맞이순, 그리고 토마토 시료들만이 미토콘드리아 질을 개선하여 미토콘드리아로부터의 ROS 생성을 감소시켰는데, 이들은 또한 세포와 조직내 lipofuscin과 malondialdehyde의 축적을 줄여서 뚜렷한 산화적 손상을 억제하는 효과를 보였다. 이들은 나아가서 초파리의 수명을 연장시키는 효과도 보였다. 미토콘드리아 질 향상 효과가 거의 없었던 쇠뜨기 시료는 산화적 손상물과 초파리 수명에 거의 영향을 미치지 못하였다. 이러한 결과는 식물 시료들의 항산화 효과가 ROS의 화학적 소거와 미토콘드리아 질의 향상을 통해 발현될 수 있는데, 후자의 효과가 실제로 체내에서 산화적 손상을 억제하는데 중요하게 작용할 가능성을 시사해 준다. 향후, 식물시료들의 항산화 효능에 대해서 이러한 ROS 발생억제 기전을 대상으로 조사하는 일은 그 유용성을 판단하는데 있어서 큰 도움이 될 것으로 기대된다.

사상자 에탄올 추출물의 지방세포 분화 억제 효과 (Inhibitory Effect of the Ethanol Extract of Torilis Japonica Decandolle on Adipocyte Differentiation in 3T3-L1 Cells)

  • 고령화 사회에서 비만을 예방하는 것에 대한 관심이 높아지는 추세에 따라, 남성과 여성 모두 비만 관리에 상당한 비용과 노력을 지불하고 있는 실정이다. 본 연구에서는 항비만 효과를 증명하기 위해서는 3T3-L1 지방전구세포를 이용한 연구가 필수적으로 수행되기 때문에, 3T3-L1 지방전구세포에서 항비만 효과를 지닌 천연물에 조사하였다. 70% 에탄올을 이용한 사상자(Torilis Japonica Decandolle) 추출물이 3T3-L1 지방전구세포에서 성숙한 지방세포로의 분화에 미치는 효과를 Oil red O assay, western blot을 통해 확인하였다. 대조군에 비해 사상자 추출물의 $100{\mu}g/ml$ 농도에서 지방세포 분화와 세포 내 triglyceride (TG) 수준을 효과적으로 억제하였다. TG 함량 감소 메커니즘을 규명하기 위해, peroxisome-proliferatorsactivated-receptor-${\gamma}$ ($PPAR{\gamma}$) 및 CCAAT enhancer-binding-proteins-${\alpha}$ ($C/EBP{\alpha}$), acetyl-CoA carboxylase (ACC)의 인산화 등 항비만 관련 단백질의 표현 수준을 확인하였다. 그 결과, 사상자 추출물은 $PPAR{\gamma}$, $C/EBP{\alpha}$, ACC 인산화 단백질의 발현 정도를 현저하게 감소시켰다. 종합하자면, 사상자 추출물이 비만 예방에 가장 효과적인 후보임을 명백하게 보여준다. 더 나아가서, 천연물인 사상자의 항비만 효과에 핵심적인 역할을 수행하는 활성 화합물을 탐색 및 분리하기 위한 추가 연구가 필요하다고 사료된다.

Genistein from Vigna angularis Extends Lifespan in Caenorhabditis elegans

  • The seed of Vigna angularis has long been cultivated as a food or a folk medicine in East Asia. Genistein (4',5,7-trihydroxyisoflavone), a dietary phytoestrogen present in this plant, has been known to possess various biological properties. In this study, we investigated the possible lifespan-extending effects of genistein using Caenorhabditis elegans model system. We found that the lifespan of nematode was significantly prolonged in the presence of genistein under normal culture condition. In addition, genistein elevated the survival rate of nematode against stressful environment including heat and oxidative conditions. Further studies demonstrated that genistein-mediated increased stress tolerance of nematode could be attributed to enhanced expressions of stress resistance proteins such as superoxide dismutase (SOD-3) and heat shock protein (HSP-16.2). Moreover, we failed to find genistein-induced significant change in aging-related factors including reproduction, food intake, and growth, indicating genistein exerts longevity activity independent of affecting these factors. Genistein treatment also led to an up-regulation of locomotory ability of aged nematode, suggesting genistein affects healthspan as well as lifespan of nematode. Our results represent that genistein has beneficial effects on the lifespan of C. elegans under both of normal and stress condition via elevating expressions of stress resistance proteins.

비접촉 생체신호 측정 기반 헬스케어 시스템 설계 및 구현 (Design and Implementation of Healthcare System Based on Non-Contact Biosignal Measurement)

  • 급격한 노령화가 의료 시설의 부족과 이로 인한 국민 건강의 질적 하락을 가져올 가능성이 점차 높아지고 있으며 의료비 상승의 부담을 해소하기 위해 선진국에서는 서비스 단가를 낮추기 위한 의료 기관들의 원격의료가 확대되고 있는 추세이다. 유헬스케어는 인체에서 발생하는 물리적, 화학적 현상의 변화를 감지하여 처리 가능한 전기적 신호로 변환하고 측정된 신호 중에서 원하는 정보만을 선택하기 위한 분석과정, 시각화 과정을 통해 결과를 피드백하여 관련정보, 경고, 알람 등을 사용자에게 제공하는 과정을 통해 서비스되고 있다. 하지만 센서를 신체에 직접 부착하는 전통적인 생체계측 방법은 일상생활에 불편을 주고 거부감이 발생할 수 있다. 따라서 일상생활에 불편함을 주지 않고 지속적으로 생체 정보를 측정할 수 있는 방법이 필요하다. 본 논문에서는 일상생활에 불편을 주지 않고 지속적으로 생체정보를 모니터링 할 수 있는 IR-UWB 기반의 비접촉. 무구속적인 호흡측정 시스템을 제안한다.

화학적 저산소증이 유도하는 뇌신경세포 손상에 있어서 미성숙 진귤 과피 발효 추출물의 보호 효과 (Anti-apoptotic effect of fermented Citrus sunki peel extract on chemical hypoxia-induced neuronal injury)

  • Purpose: Neuronal apoptotic events induced by aging and hypoxic/ischemic conditions is an important risk factor in neurodegenerative diseases such as ischemia stroke and Alzheimer's disease. The peel of Citrus sunki Hort. ex Tanaka has long been used as a traditional medicine, based on multiple biological activities including anti-oxidant, anti-inflammation, and anti-obesity. In the current study, we examined the actions of fermented C. sunki peel extract against cobalt chloride ($CoCl_2$)-mediated hypoxic death in human neuroblastoma SH-SY5Y cells. Methods: Cell viability was measured by trypan blue exclusion. Expression of apoptosis related proteins and release of cytochrome c were detected by western blot. Production of intracellular reactive oxygen species (ROS) and apoptotic morphology were examined using 2',7'-dichlorofluorescin diacetate (DCF-DA) and 4',6-diamidino-2-phenylindole (DAPI) staining. Results: Exposure to $CoCl_2$, a well-known mimetic agent of hypoxic/ischemic condition, resulted in neuronal cell death via caspase-3 dependent pathway. Extract of fermented C. sunki peel significantly rescued the $CoCl_2$-induced neuronal toxicity with the cell viability and appearance of apoptotic morphology. Cytoprotection with fermented C. sunki peel extract was associated with a decrease in activities of caspase-3 and cleavage of poly (ADP ribose) polymerase (PARP). In addition, increase in the intracellular ROS and release of cytochrome c from mitochondria to the cytosol were inhibited by treatment with extract of fermented C. sunki peel. Conclusion: Based on these data, fermented C. sunki peel extract might have a protective effect against $CoCl_2$-induced neuronal injury partly through generation of ROS and effectors involved in mitochondrial mediated apoptosis.

Antioxidant Activities of Various Extracts from Korean Yam (Dioscorea batatas DECNE.)

  • The objective of this study was to investigate the antioxidant activity of Korean yam (Dioscorea batatas $D_{ECNE.}$) by different extraction solvents including 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v). Raw yam was analyzed for its color property, total phenol content and antioxidant activity. Yam possessed high $L^*$ value and $H^{\circ}$ value, which were $81.64{\pm}2.59$ and $83.36{\pm}0.15$, respectively. Raw yam was found to have great antioxidant activity evaluated through ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power. Total phenol contents of various extracts from raw yam increased in the following order: 70% methanol extract ($37.62{\pm}0.88mg\;CAE/g$), 70% ethanol extract ($43.38{\pm}0.66mg\;CAE/g$) and CM extract ($67.17{\pm}0.12mg\;CAE/g$), respectively. The same trend was also could be found in the ABTS radical scavenging activity, DPPH radical scavenging activity and ferric reducing antioxidant power assays. These results implied that Korean yam might play an important role in antioxidation and serve as the bio-health functional food to take a good part in prevention of human diseases and aging.

한국 한방화장품 발달 배경 및 연구 현황 (The Background and Current Research Applied to Development of Korean Cosmetics Based on Traditional Asian Medicine)

  • Traditional Asian medicine has an extensive evidence base built upon thousands of years of experience within Asia, of curing various diseases. Only recently, within the past two centuries, have modern medical scientists developed interest in traditional Asian medicine. Asian Medicine seems to be regarded only as an adjunctive medicine and viewed as alargely un-proven alternative medicine to complement western medicine, used in some cases to establish a new paradigm of "integrative medicine". This article reviews how Korean herbal cosmetics emerged by applying traditional Asian medicine to the science of cosmetics. The characteristics of Korean herbal cosmetics are examined through examples of history, concepts and traditions. With the advancements in biotechnology, studies are now being conducted on the dermatological effects and processing methods of herbal ingredients, including ginseng. The authors explain the current research on the identification on the active ingredients of herbs, extraction methods, and bio-processing of ingredients to improve the biological efficacies of herbs on the skin. A summary of studies focused on modern reinterpretations of ageing theories, such as 'Seven year aging cycle', are provided. In conclusion, the development of Korean cosmetics products are based on the accumulated knowledge of thousands of years of experience including; 1) practical heritage of traditional Asian medicines such as Donguibogam; 2) excellent medicinal plants, such as ginseng, which are native to Korea; and 3) innovative attempts to modernize materials, processes, and principles.

골절 방지 시스템의 개발을 위한 낙상 동적 특성 분석 (Dynamic Characterization of Fall for Development of Fracture Prevention System)

  • The social activities of the elderly have been increasing as our society progresses toward an aging society. As their activities are increased, the occurrence of falls that could lead to fractures are increased. Falls are serious health hazards to the elderly and we need more thorough understanding of falls including the progress of falls and the impact area in various fall directions. Many of the traditional methods of falls research dealt with voluntary falls by younger subject since older subject can easily get fracture from voluntary falls. So, it has been difficult to get exact data about falls of the elderly. Here, we tried to capture the characteristics of the movements of major joints using three dimensional motion capture system during falls experiments using a moving mattress that can safely induce unexpected falls. Healthy younger subjects participated in the actual falls experiment and the moving mattress was actuated by a pneumatic system. The kinematic parameters such as velocities and accelerations of major segments were imported to a computer simulation environment and falls to hard surfaces were simulated in a computational environment using a realistic human model of aged persons. The simulation was able to give approximations to contact forces which can occur during actual falls.

비결정질철산화물 원위치 형성을 통한 비소오염토양 안정화 및 X선 분광분석법의 활용에 대한 연구 (Study on Stabilization of Arsenic in Soil through in situ Formation of Amorphous Fe Oxides and use of X-ray Absorption Spectroscopy)

  • This study was conducted to investigate the in situ formation of amorphous Fe oxides as a stabilization technology in As-contaminated soil. After addition of ferric nitrate and the neutralizing agent, most of extractable fractions of As in soil (i.e., SO42- and PO43--extractable As) was converted into As bound to amorphous Fe oxides. In addition, results of solubility bioavailability research consortium (SBRC) test indicated that a significant amount of As in untreated soil changed to a non-bioaccessible form after stabilization. The reason was attributed to the newly formed amorphous Fe oxides in the stabilized soil, which was confirmed by linear combination of fitting (LCF) using X-ray absorption spectroscopy (XAS) analysis. Interestingly, after five months of aging of the stabilized soil, ferrihydrite and schwertmannite newly formed in the soil were transformed to crystalline Fe oxides such as goethite, and further decrease in SBRC extractable fraction of As was observed. The results suggest that co-precipitated As with amorphous Fe oxides can be further immobilized with time, due to the crystallization of amorphous Fe oxides.
