  • This study examined the antioxidative and lipid accumulation inhibitory effects of 14 plants from Mongolia and Myanmar on 3T3-L1 and HepG2 cells. The total phenolic and flavonoid contents (TPC and TFC) of 14 plant extracts were measured, and the antioxidative activities were analyzed using DPPH, ABTS, FRAP, and ORAC. After measuring the pancreatic lipase levels and performing the thiobarbituric acid assay, the degree of lipid accumulation was determined by lipid (Oil Red O) staining and triglyceride assay in 3T3-L1 and HepG2 cells. M. paniculate (259.43 mgGAE/g) and C. benghalensis (130.78 mgNAE/g) had the highest TPC and TFC, respectively, among the 14 plants. R. acicularis Lindl. had the highest antioxidant activity in DPPH. The ABTS, FRAP, and ORAC results showed that the antioxidant activity of 11 species was higher than that of the positive control. The pancreatic lipase inhibitory effect of C. angustifolium Scop. was reduced to 23.65% at 0.1 mg/mL, and the level of lipid peroxidation of C. abrorescens Lam. was 0.63 nmol/mg. Five selected plants inhibited the lipid accumulation and triglyceride content, respectively, in 3T3-L1 and HepG2 cells. These results provide scientific evidence for developing functional foods using 14 plants from Mongolia and Myanmar, which have antioxidant activities and lipid accumulation reduction effects.

  • Hemp is known as one of the most productive and useful plants, which grows quickly in a moderate climate with only moderate water and fertilizer. Traditionally in Korea, hemp bast is used to natural fibres, and remaining such as stem and root is treated as waste. Those of hemp by-products can be transformed to bio fuel such as bio-oil and activated carbon. To understand pyrolysis characteristics, thermogravimetric analysis were carried out in TGA, in which hemp by-products were mostly decomposed at the temperature range of $270{\sim}370^{\circ}C$. The corresponding kinetic parameters including activation energy and pre-exponential factor were determined by differential method over the degree of conversions. The values of activation energies for pyrolysis were increased as the conversion increased from 10 to 90%.

  • The coffee grounds generated during the coffee extraction process contain several resources, but the technology for their recycling has not been commercialized yet, causing various environmental problems. Due to the recent increase in coffee consumption worldwide, the amount of coffee grounds produced has been continuously increasing, reaching more than 750 million tons. In Korea, about 120,000 tons of coffee waste are annually generated; however, most of them are landfilled or incinerated. Although there is still a shortage of coffee waste recycling technologies compared to the amount of coffee grounds produced, various recycling approaches are being actuated in many countries including Korea. In this study, the generation of coffee grounds at home and abroad, the status of coffee grounds recycling, and the associated technology development trends were investigated. The coffee grounds recycling has been studied in the fields of energy, adsorbent, construction, agriculture, and bio-foods. Research is most active in the energy and biotechnology areas; in particular, since the oil in the coffee grounds is valuable as a feedstock for biomass energy, the technology related to energy recovery is currently under development worldwide. • BACKGROUND/OBJECTIVES: The fruit of Cydonia oblonga Miller (COM) is used traditionally in Mediterranean region medicine to prevent or treat obesity, but its mechanism of action is still unclear. Beyond a demonstrated anti-obesity effect, the fruit was tested for the mechanism of adipogenesis in 3T3-L1 preadipocytes. MATERIALS/METHODS: 3T3-L1 preadipocytes were cultured for 8 days with COM fruit extract (COME) at different concentrations (0-600 ㎍/mL) with adipocyte differentiation medium. The cell viability was measured using an MTT assay; triglyceride (TG) was stained with Oil Red O. The expression levels of the adipogenesis-related genes and protein expression were analyzed by reverse transcription polymerase chain reaction and Western blotting, respectively. RESULTS: COME inhibited intracellular TG accumulation during adipogenesis. A COME treatment in 3T3-L1 cells induced upregulation of the adenosine monophosphate-activated protein kinase (AMPK)α phosphorylation and downregulation of the adipogenic transcription factors, such as sterol regulatory element-binding protein 1c, peroxisome proliferator-activated receptor γ, and CCAAT/enhancer binding protein α. The COME treatment reduced the mRNA expression of fatty acyl synthetase, adenosine triphosphate-citrate lyase, adipocyte protein 2, and lipoprotein lipase. It increased the mRNA expression of hormone-sensitive lipase and carnitine palmitoyltransferase I in 3T3-L1 cells. CONCLUSIONS: COME inhibits adipogenesis via the AMPK signaling pathways. COME may be used to prevent and treat obesity.

  • Most countries including Korea and Indonesia have strong policy for implementing biofuels like biodiesel. Shortage of the oil feedstock is the main barrier for increasing the supply of biodiesel fuel. In this study, in order to improve the stability of feedstock supply and lower the biodiesel production cost, the feasibility of biodiesel production using two types of Indonesian tropical crop oils, pressed at different harvesting times, were investigated. R. Trisperma oils, a high productive non-edible feedstocks, were investigated to produce biodiesel by esterification and transesterification because of it's high impurity and free fatty acid contents. the kindly provided oils from Indonesia were required to perform the filtering and water removal process to increase the efficiency of the esterificaton and transesterification reactions. The esterification used heterogeneous acid catalyst, Amberlyst-15. Before the reaction, the acid value of two types oil were 41, 17 mg KOH/g respectively. After the pre-esterification reaction, the acid value of oils were 3.7, 1.8 mg KOH/g respectively, the conversions were about 90%. Free fatty acid content was reduced to below 2%. Afterwards, the transesterification was performed using KOH as the base catalyst for transesterification. The prepared biodiesel showed about 93% of FAME content, and the total glycerol content was 0.43%. It did not meet the quality specification(FAME 96.5% and Total glycerol 0.24%) since the tested oils were identified to have a uncommon fatty acid, generally not found in vegetable oils, ${\alpha}$-eleostearic acid with much contents of 10.7~33.4%. So, it is required to perform the further research on reaction optimization and product purification to meet the fuel quality standards. So if the biodiesel production technology using un-utilized non-edible feedstock oils is successfully developed, stable supply of the feedstock for biodiesel production may be possible in the future.

  • In this study, to reduce the production cost of poly(3-hydroxyalkanoates) (PHA), optimal cell growth and PHA biosynthesis conditions of the isolated strain Pseudomonas sp. EML8 were established using waste frying oil (WFO) as the cheap carbon source. Gas chromatography (GC) and GC mass spectrometry analysis of the medium-chain-length PHA (mcl-PHAWFO) obtained by Pseudomonas sp. EML8 of WFO indicated that it was composed of 7.28 mol% 3-hydrxoyhexanoate, 39.04 mol% 3-hydroxyoctanoate, 37.11 mol% 3-hydroxydecanoate, and 16.58 mol% 3-hydroxvdodecanoate monomers. When Pseudomonas sp. EML8 were culture in flask, the maximum dry cell weight (DCW) and the mcl-PHAWFO yield (g/l) were showed under WFO (20 g/l), (NH4)2SO4 (0.5 g/l), pH 7, and 25℃ culture conditions. Based on this, the highest DCW, mcl-PHAWFO content, and mcl-PHAWFO yield from 3-l-jar fermentation was obtained after 48 hr. Similar results were obtained using 20 g/l of fresh frying oil (FFO) as a control carbon source. In this case, the DCW, the mcl-PHAFFO content, and the mcl-PHAFFO yields were 2.7 g/l, 62 wt%, and 1.6 g/l, respectively. Gel permeation chromatography analysis confirmed the average molecular weight of the mcl-PHAWFO and mcl-PHAFFO to be between 165-175 kDa. Thermogravimetric analysis showed decomposition temperature values of 260℃ and 274.7℃ for mcl-PHAWFO and mcl-PHAFFO, respectively. In conclusion, Pseudomonas sp. EML8 and WFO could be suggested as a new candidate and substrate for the industrial production of PHA.

  • A lipase producing bacterium was isolated from button mushroom bed, which showing high clear zone on agar media containing Tributyrin as the substrate. The strain was identified as Burkholderia cepacia by analysis of 16S rDNA gene sequence. Crude lipase (CL) was partially purified from 70% ammonium sulfate precipitation using the culture filtrate of B. cepacia. Immobilized lipases were prepared by cross-linking method with CL from B. cepacia and Novozyme lipase (NL) onto silanized Silica-gel as support. Residual activitiy of the immobilized CL (ICL) and immobilized NL (INL) was maintained upto 61% and 72%, respectively. Biodiesel (Fatty acid methyl ester, FAME) was recovered by transesterification and methanolysis of Canola oil using NaOH, CL and ICL as the catalysts to compare the composition of fatty acids and the yield of FAME. Total FAME content was NaOH $781mg\;L^{-1}$, CL $681mg\;L^{-1}$ and ICL $596mg\;L^{-1}$, in which the highest levels of FAME was observed to 50% oleic acid (C18:1) and 22% stearic acid (C18:0). In addition, the unsaturated FAME (C18:1, C18:2) decreased, while saturated FAME (C16:0, C18:0) increased according to increasing the reaction times with both CL and ICL, supporting CL possess both transesterification and interesterification activity. When reusability of ICL and INL was estimated by using the continuous reaction of 4 cycles, the activity of ICL and INL was respectively maintained 66% and 79% until the fourth reaction.

  • In an effort to reduce the onset of global warming, the International Maritime Organization Marine Environment Protection Committee (IMO MEPC) proposed the reduction in ship speeds as a way of lowering the proportion of carbon dioxide ($CO_2$) in the Green House Gas emissions from ships. To minimize fuel costs, shipping companies have already been performing slow steaming for their own fleets. Specifically, the slow steaming approach has been adopted for most ocean-going container lines. In addition, because of the increased marine fuel cost that is required to enable increased capacity, there is an urgent need for more advanced fuel-saving technologies. Therefore, in this present study, we propose a fuel-cost reduction method that can improve the performance of diesel engines. We introduce a predetermined amount (0.025% of the amount of fuel used) of fuel additive (oil-soluble calcium-based organometallic compound). For improved experimental accuracy, as the test subjects, we utilize a large two-stroke diesel engine installed in land plants. The loads of the test engine were classified as low, medium, and high (50, 75, and 100%, respectively). We compare the engine performance parameters (power output, fuel consumption rate, p-max, and exhaust temperature) before and after the addition of fuel additives. Our experimental results, confirmed that we can realize fuel-cost savings of at least 2% by adding the fuel additive in low load conditions (50%). Likewise, the maximum combustion pressure was found to have increased. On the other hand, we observed that there was a reduction in the exhaust temperature.

  • The purpose of this study was to evaluate the influence and safety of spraying Farm Clean-S (FC-S) containing 70% Chamaecyparis obtusa essential oil on the elimination of red mite in laying hens. In total, 300 laying hens (ISA brown) aged 72 weeks and infected with red mite were assigned to individual cages for five treatments with five replicates for 18 days. Treatments consisted of distilled water (negative control; NC), commercial spray product (positive control; PC), FC-S 0 (original concentration), FC-S 50 (50 times diluted) and FC-S 100 (100 times diluted). The number of red mites, egg production and weight of each group were evaluated daily throughout the whole experimental period. Egg quality and blood biochemical parameters were measured at the end of the experiment. Safety tests were confirmed by feed intake, respiration, lethargy, diarrhea and mortality symptoms after the feeding trial. As the result, more than 80% of red mites were eliminated by the FC-S 0 spray treatment, and FC-S 50 also showed a higher elimination percentage than did PC (P<0.01). However, the elimination effect of the FC-S 100 spray was lower than that of the PC, but higher than that of the NC (P<0.01). These results confirmed that the optimum FC-S dilution rate ranged from 0~50 times with distilled water to eliminate the red mite effectively. The performance, egg quality, blood parameter and safety tests were not significantly different among treatments. Thus, FC-S 0 and FC-S 50 spray treatments can effectively and sustainably control red mite infestations without detrimental effects on the performance, egg quality and plasma biochemical properties of laying hens.
