Antihyperlipidemic Activity of the Ethyl-acetate Fraction of Stereospermum Suaveolens in Streptozotocin-induced Diabetic Rats

  • Objectives: Dyslipidemia in diabetes mellitus is a significant risk factor for the development of cardiovascular complications. The aim of this study was to evaluate the effect of the ethyl-acetate fraction of an ethanolic extract from Streospermum suaveolens on lipid metabolism in streptozotocin (STZ)-induced diabetic rats. Methods: Diabetes was induced by intraperitonial injection of STZ (50 mg/kg). Diabetic rats were treated with an ethyl-acetate fraction orally at doses of 200 and 400 mg/kg daily for 14 days. On the $15^{th}$ day, serum lipid profiles, such as total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), and high-density lipoprotein (HDL), were estimated in experimental rats. The atherogenic (AI) and the coronary risk (CRI) indices were also evaluated. Results: The ethyl-acetate fraction at doses of 200 and 400 mg/kg significantly (P < 0.001) and dose-dependently reduced serum cholesterol, triglycerides and LDL, but increased HDL towards near normal levels as compared to diabetic control rats. The fraction also significantly (P < 0.001) lowered the atherogenic index (AI) and coronary risk index (CAI) in a dose-dependent manner. Conclusion: The present study demonstrated that the ethyl-acetate fraction of Stereospermum suaveolens exhibits a potent antihyperlipidemic activity in hyperglycemic rats and suggests that the plant may have therapeutic value in treating the diabetic complication of hyperlipidemia.

Cherry Tomato Supplementation Reduces Cardiovascular Risk

  • Tomatoes are rich in nutrients and have many beneficial advantages on human health. Four cherry tomato supplementation diets (CTSDs) were prepared from the juice and cake of fresh and processed (heat-treated) cherry tomatoes. Rats were fed CTSDs for 28 days and the changes in health indices in the serum were analyzed. CTSDs significantly decreased (P < 0.05) food efficiency ratio compared with the control. CTSD feeding significantly increased (P < 0.05) the high-density lipoprotein cholesterol level compared with the control, which resulted in a significant (P < 0.05) decrease in coronary artery risk index and atherogenic index. Furthermore, CTSD feeding increased serum serotonin level. These results indicate that CTSD shows antihyperlipidemic effect.

트랜스형 불포화 지방산이 mice 혈중 지질농도에 미치는 영향 (Effect of Trans-unsaturated Fatty Acid on Serum Lipid Levels in Mice)

  • 본 연구에서는 각기 다른 형태의 지방산이 ICR mice의 혈중 지질 농도에 미치는 영향을 평가하고자 하였다. 이를 위하여 8주령 수컷 ICR mice를 일반 식이섭취군(C), 10%의 트랜스 불포화 지방산 섭취군(TFA-1), 30% 트랜스 불포화 지방산 섭취군(TFA-2), 50% 트랜스 불포화 지방산 섭취군(TFA-3), 50% 포화 지방산 섭취군(SFA), 불포화 지방산 섭취군(USFA) 으로 나누어 식이 하였다. 혈중 총 콜레스테롤의 수치는 TFA-3 군과 SFA 군의 지질함량이 다른 군에 비하여 높았으며, 중성지방지수 역시 높게 나타났다. 총 콜레스테롤과 중성지방수치를 이용하여 나타낸 LDL 콜레스테롤의 경우에도 50%의 트랜스 지방산을 섭취한 군에서 포화 지방산을 섭취한 군과 유사한 결과가 나타났다. 이에 반해, 10%, 30%의 트랜스 지방산을 섭취한 군에서는 일반식이 섭취군과 비슷한 결과가 나타났다. 이는 많은 양의 트랜스 불포화지방산을 짧은 기간 동안 섭취하더라도 포화지방산을 섭취한 것과 유사한 효과를 나타낸다는 것을 의미한다.

Aristolochia ringens extract ameliorates oxidative stress and dyslipidaemia associated with streptozotocin-induced hyperglycaemia in rats

  • The study was designed to assess antioxidant and antidyslipidaemic effects of terpenoid-rich extract from the root of Aristolochia ringens V. Hyperglycemia-induced oxidative stress and dyslipidemia were established in rats by single intraperitoneal administration of 65 mg/kg bw streptozotocin. Based on therapeutic dose determined in previous study, streptozotocin-induced rats were orally administered with 75 and 150 mg/Kg bw of A. ringens extract for 14 days. Total protein, serum lipid profiles and biomarkers of oxidative stress in liver and kidney of the experimental rats were determined. Atherogenic and cardiovascular disease risk indices were computed. Streptozotocin-induced hyperglycaemia significantly (p < 0.05) decreased activities of superoxide dismutase, catalase and glutathione transferase as well as the amount of reduced glutathione in both tissues indicating oxidative stress induced kidney and liver injury due to glucotoxicity. In comparison to non-treated hyperglycaemic rats, activities of the antioxidant enzymes and concentration of glutathione-H were significantly (p < 0.0001) increased, whereas malondialdehyde was reduced in the tissues of rats treated with both 75 and 150 mg/Kg bw of the extract. The extract also caused significant (p < 0.001) reduction in elevated levels of total cholesterol, triglycerides and low density lipoprotein-cholesterol levels, whereas concentration of the attenuated high density lipoprotein-cholesterol was increased in serum of the treated rats. Reduced atherogenic and cardiac risk indices were projected for the A. ringens extract-treated groups. Results from this study showed that extract from A. ringens root was rich in terpenoids and may reduce risks of complications associated with hyperglycemia-induced oxidative stress and dyslipidemia.

Investigation of Blood Betatrophin Levels in Obese Children with Non-Alcoholic Fatty Liver Disease

  • Purpose: The prevalence of obesity has significantly increased among children and adolescents worldwide and is becoming an important health care problem in parallel with the increased prevalence of obesity pediatric non-alcoholic fatty liver disease. Betatrophin is a newly define hormone that is commonly secreted by liver and plays role in glucose tolerance. This study aimed to investigate the relationship between serum betatrophin levels and non-alcoholic fatty liver disease in obese children. Methods: The study included 40 obese children with a body mass index (BMI) above 95th centile, and 35 non-obese subjects with a BMI 3-85th centile, whose age and gender were similar to those of the patient group. For the evaluation of metabolic parameters fasting serum glucose, insulin, alanine aminotransferase, aspartate aminotransferase, lipid profile and serum betatrophin levels were measured. Total cholesterol: high-density lipoprotein cholesterol and low-density lipoprotein cholesterol: high-density lipoprotein cholesterol ratios were calculated as "atherogenic indices." Results: Serum betatrophin levels of the obese subjects were similar to that of non-obese subjects (p=0.90). Betatrophin levels were not correlated with the metabolic parameters. Conclusion: In the present study, levels of betatrophin are not different between obese and insulin resistant children and non-obese subjects, and they are not correlated with atherogenic indices. To elucidate the exact role of betatrophin in obesity, further studies are required to identify the betatrophin receptor and/or other possible cofactors.

Anti-hyperlipidemic Effect of Vitex doniana Ethanol Extract in Poloxamer Induced Hyperlipidemia

  • To determine the effect of Vitex doniana (leaves stem and root bark) ethanolic extracts on lipid profiles of Poloxamer 407 (P407) induced hyperlipidemic and normal rats. Fifty four mixed sex rats weighing 100-200g were divided into nine groups comprising six animals per group. At the end of the 21 day, the animals were sacrificed and blood sample were collected for determination of serum levels of: Total cholesterol (TC), Triacylglycerides (TAG), High-density lipoprotein cholesterol (HDL-c) and Low-density lipoprotein cholesterol (LDL-c). The studies showed that all induced treated groups significantly (P<0.05) lower serum levels of TC, TAG, LDL-c and significantly (P<0.05) increased HDL-c when compared to the P407 induced hyperlipidemic control. The normal treated groups showed no significant (P>0.05) difference in the serum levels of TC, TAG, LDL and HDL when compared to the normal control group. Calculation of atherogenic risk predictor indices of the induced treated groups showed that all the extracts significantly (P<0.05) lowered the LDL-c/HDL-c, log (TAG/HDL-c) and significantly (P<0.05) increased HDL-c/TC ratio when compared to the P407 induced hyperlipidemic control group. The atherogenic risk predictor indices of normal treated groups showed no significant difference (P>0.05) in LDL-c/HDL-c, Log (TAG/HDL-c) and HDL-c/TC ratio when compared to the normal control group. The study demonstrates the phytotherapeutic effect of Vitex doniana (leaves, stem and root bark) ethanolic extract in poloxamer 407 induced hyperlipidemia.

Relationships of Potential Atherogenic Indices to Anthropometric Measurements, Dietary Intake and Dietary Behavior in Korean Obese Children

  • Epidemiological studies have indicated that a relationship between overweight and cardiovascular disease exists. To assess the relationship of unfavorable changes in serum lipid concentrations, apolipoprotein B, lipoprotein(a), blood sugar and blood pressure to anthropometric variables, nutrient intakes and dietary behavior,64 obese children were chosen as subjects and 24-hour dietary intake, dietary behavior as well as blood pressure, apolipoprotein B, lipoprotein(a), three kinds of cholesterol concentrations were measured in November 1976. More than half(57.8%) of the children had serum cholesterol concentrations over 175mg/d1, and 26.6% of them had over 200mg/d1 and this group could be classified as a high risk group. Abdominal obesity as measured with waist circumference and waist hip ratio was negatively co..elated to high density lipop개tein(HDL) cholesterol. Dietary behavior rather than nutrient intakes appeared to have associations with unfavorable lipid profiles. Total dietary behavior scores and fish and bean product consumption had strong correlations with potentially atherogenic lipoprotein concentrations. This study suggests that fir the early prevention of cardiovascular disease, waist hip ratio together with obese rate should be included in selective programs to identify risk group of children in Korea. furthermore, interventions in cardiovascular disease risk groups in obese children should emphasize maintenance of ideal body weight through reducing body fat and adopting desirable dietary behaviors such as increasing intake of fish or bean products. 만성퇴행성 질환에 미치는 다시마(Laminaria japonica)와 후코이단 성분의 영향 (Effects of Sea Tangle (Laminaria japonica) and Fucoidan Components on Chronic Degenerative Diseases)

  • This study was designed to investigate the effects of sea tangle (Laminaria japonica) extract and fucoidan components on chronic degenerative diseases. Sprague-Dawley(SD) male rats (210$\pm$5g) were fed experimental diets: Dasi-Ex group: dasima extract powder of 4.0% added to control diet; Fuco-I, II and III groups: fucoidan powder of 1, 2 and 3% added to Dasi-Ex group for 45 days. Triglyceride (TG) levels in serum were significantly lower (10~15%) in Fuco-I, II and III groups compared with control group. Total cholesterol levels were significantly decreased (7~10% and 15~ 35%) in brain mitochondria and microsomes of Fuco-II and III group compared with control group. LDL-cholesterol levels were remarkably decreased (20~30%) in Dasi-Ex and Fuco-I, II, III groups, but HDL-cholesterol levels were significantly increased (10~12%) in Fuco-II and III groups only compared with control group. The ratios of HDL/total cholesterol resulted in a marked increase (3 5~55%) in Dasi-Ex and Fuco-I, II, III groups, but atherogenic indices were remakably decreased (40~50%) in Dasi-Ex and Fuco-I, II, III groups compared with control group. Membrane fluidities were remarkably increased (45~70% and 38~42%, respectively) in brain mitochondria and microsomes of Fuco-II and III groups compared with control group. Administrations of fucoidan added to dasima effectively decreased TG, total and LDL-cholesterol, and atherogenic index, while also effectively increased HDL-cholesterol, HDL/total cholesterol ratio, and membrane fluidity, suggesting chronic degenerative diseases were very effectively prevented by the administration of fucoidan component.

구미지역 보육시설 유아들의 신체계측과 혈액생화학적 지표에 따른 건강상태 (Health Status by Anthropometrie Values and Serum Biochemical Indices of Preschool Children in Day-Care Centers in Gumi)

  • This study was conducted to investigate health status of preschool children utilizing anthropometric values and serum biochemical indices. The subjects were 1,159 boys and 1,019 girls in Gumi day-care centers aged 4 to 6 years. The mean height and weight of 4, 5 and 6 year old children were 103.0 cm and 17.1 kg, 107.2 cm and 18.3 kg and 113.4 cm and 20.6 kg, respectively. By the obesity index criteria, $5\%$ of boys and $4\%$ of girls were obese whereas $10\%$ of boys and $9\%$ of girls were underweight. By the Kaup index criteria, $7\%$ of boys and $4\%$ of girls were obese. By the percentage of body fat criteria, $5\%$ of boys and girls were obese. The mean concentrations of serum total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, Atherogenic Index and serum glucose were 154.1-156.5 mg/dL, 80.1-81.9 mg/dL, 46.8-47.4 mg/dL, 90.9-92.8 mg/dL, 2.34-2.39 and 76.4-77.8 mg/dL, respectively. Serum triglyceride level, AI and body fat percentage of obese children were significantly higher than other children and were significantly lower in HDL-cholesterol level (p < 0.05). In addition there were significant correlation between anthropometric values and serum biochemical indices : Kaup index and obesity index were positively correlated to the systolic blood pressure and diastolic blood pressure in boys and girls (p < 0.01) and to the triglyceride concentration in boys (p < 0.05). These results indicate the need of developing and implementation of nutrition education program for preschool children to prevent further development of childhood obesity and to maintain normal serum lipid profiles.

여대생의 HDL-콜레스테롤 수준에 따른 임상건강지표와 영양섭취 실태 (Food Intake Patterns and Blood Clinical Indices in Female College Students by HDL-cholesterol Levels)

  • Recently, a lot of epidemiological studies revealed that low HDL-cholesterol level was a better predictor of risk for coronary heart disease than total cholesterol. This study investigated the anthropometric parameters, clinical blood indices, and dietary factors influencing serum HDL-cholesterol level by using a cross-sectional study for Korean female college students. The subjects were 94 female college students. They were divided into three groups according to their serum HDL-cholesterol levels, low HDL-cholesterol (<50 mg/dL, n=20), medium HDL-cholesterol (50 $\leq$, < 60 mg/dL, n=39) high serum HDL-cholesterol groups (60 $\leq$ mg/dL, n = 35). This study examined their demographic data and dietary intake throughout a questionnaire. Clinical blood indices were measured using an automatic blood chemistry analyzer (Selectra E), after 12 hours of fasting. BMI, body weight, fat mass, and waist circumferences were significantly increased according to low serum HDL-cholesterol levels. Serum lipid analysis showed a significantly higher level of TG, LDL-/HDL-Ratio, atherogenic index in the low HDL-cholesterol group. Serum levels of GPT, uric acid and alkaline phosphatase in the low HDL-cholesterol group were significantly higher than in the other group. The average consumption of energy was 1627 kcal and 77.76% of estimated energy requirement (EER). The mean ratio of calories from carbohydrate: protein: fat was 57:15:28. The low HDL-cholesterol group was significantly higher than the other groups in eggs, fat and oils consumption. Interestingly, milk and diary products consumption of low HDL-cholesterol group was half (p < 0.05) of those of the other groups. In conclusion, serum HDL-cholesterol levels appeared to be decreased by increasing BMI, fat mass, waist circumference, and serum TG level. In addition, some dietary factors seemed to be related to serum HDL-cholesterol levels. However, further research is needed to elucidate the exact relationship between serum HDL-cholesterol level and dietary factors.
