Changes of Organic Solutes and Antioxidative Enzyme Activity in Rice Seedling under Salt Stress

  • Seedlings of two rice genotyopes, cvs. Ilpumbyeo and Gancheokbyeo, were exposed to 0, 50 and 100 mM NaCl in nutrient solution for nine days. Plants were collected at the interval of 3 days and organic and inorganic solutes in leaves and roots and antioxidative enzyme activity in leaves were determined. Under salinity, the accumulation of soluble sugars occurred considerably in the older leaves of stressed seedlings compared to younger leaves and roots. The endogenous Na+ contents markedly increased at higher NaCl concentration in leaves and roots of seedlings, though it was higher accumulated in roots. Salinity resulted in an excessive proline accumulation in the stressed plants. A more pronounced increase was observed in Gancheokbyeo leaves. SOD activity in Impumbyeo cannot found any remarkable change, whereas, in Gancheokbyeo, its activity was rapidly decreased. CAT and POD activities increased with an increase in NaCl concentration in both genotypes. In sum­mary, the high capacity of rice seedlings to overcome an unfavorable growth condition such salt stress appears to be related to an adequate partition of organic solutes between shoots and roots and to changes in absorption, transport and re-translocation of salts.

Isolation of Lactic Acid Bacteria Showing Antioxidative and Probiotic Activities from Kimchi and Infant Feces

  • The purpose of this study was to investigate lactic acid bacteria with antioxidative and probiotic activities isolated from Korean healthy infant feces and kimchi. Isolates A1, A2, S1, S2, and S3 were assigned to Lactobacillus sp. and isolates A3, A4, E1, E2, E3, and E4 were assigned to Leuconostoc sp. on the basis of their physiological properties and 16S ribosomal DNA sequence analysis. Most strains were confirmed as safe bioresources through nonhemolytic activities and non-production of harmful enzymes such as β-glucosidase, β-glucuronidase and tryptophanase. The 11 isolates showed different resistance to acid and bile acids. In addition, they exhibited antibacterial activity against foodborne bacteria, especially Bacillus cereus, Listeria monocytogenes, and Escherichia coli. Furthermore, all strains showed significantly high levels of hydrophobicity. The antioxidant effects of culture filtrates of the 11 strains included 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging capacity, 2.2'-azino-bis (2-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical cation scavenging activity, and superoxide dismutase activity. The results revealed that most of the culture filtrates have effective scavenging activity for DPPH and ABTS radicals. All strains appeared to have effective superoxide dismutase activity. In conclusion, the isolated strains A1, A3, S1, and S3 have significant probiotic activities applicable to the development of functional foods and health-related products. These strains might also contribute to preventing and controlling several diseases associated with oxidative stress, when used as probiotics.

상심자와 건조상심자 추출물의 항산화 효과 및 전지방세포 분화억제 효과에 관한 연구 (Study of Mori Fructus and Dried Mori Fructus Extracts on the Antioxidant Effect and the Inhibitory Effect on Adipocyte Differentiation)

  • Objectives This study was to investigate the antioxidative capacity, antiobesity effect and anti-diabetes effects of Mori Fructus and dried Mori Fructus in Raw 264.7 cells and 3T3-L1 cells. Methods 3 different types of Mori Fructus extracts (water 100%, ethanol 30%, ethanol 100%) were used in this study. And 3 different types of dried Mori Fructus extracts (water 100%, ethanol 30%, ethanol 100%) were used in this study. Total polyphenol compund, total favonoid compound, DPPH radical scavenging, ROS activity, NO, cell proliferation were measured in the experiment. Expressions of adipogenic transcription factors including $C/EBP-{\alpha}$, $C/EBP-{\beta}$, $PPAR-{\alpha}$, $PPAR-{\gamma}$, $AMPK-{\alpha}$ were analyzed by Real time PCR. Results Mori Fructus extracts measurements are higher than dried Mori Fructus extracts measurements at Total flavonoid compound and total flavonoid compound. Mori Fructus extracts measurements are lower than dried Mori Fructus extracts measurements at DPPH radical scavenging, ROS activity, NO. In RT-PCR analysis, there is a tendency that dried Mori Fructus extracts inhibit the expression of $C/EBP-{\alpha}$, $C/EBP-{\beta}$ genes. In RT-PCR analysis, there is a tendency that dried Mori Fructus extracts promote the expression of $PPAR-{\alpha}$, $PPAR-{\gamma}$, $AMPK-{\alpha}$ genes. Conclusions Mori Fructus is effective on inhibiting the oxidation and dried Mori Fructus is effective on inhibiting the obesity and diabetes.

Visible injury and growth inhibition of black pine in relation to oxidative stress in industrial areas

  • The objective of our study was to investigate the major reasons for the different growth and visible injury on the needles of black pine growing in Ulsan and Yeocheon industrial complex areas, South Korea. After 12 years of growth, we collected climatic and air pollutant data, and analyzed soil properties and the physiological characteristics of black pine needles. Annual and minimum temperatures in Ulsan were higher than those in Yeocheon from 1996 to 2008. Ozone ($O_3$) was the pollutant in greatest concentration in Yeocheon, and whereas the $SO_2$ concentration in most areas decreased gradually during the whole period of growth, $SO_2$ concentration in Yeocheon has increased continuously since 1999, where it was the highest out of four areas since 2005. Total nitrogen and cation exchange capacity in Yeocheon soil were significantly lower than those of Ulsan. The average growth of black pine in Yeocheon was significantly smaller than that in Ulsan, and the growth of damaged trees represented a significant difference between the two sites. Photosynthetic pigment and malondialdehyde content and antioxidative enzyme activity in the current needles of black pine in Yeocheon were not significantly different between damaged and healthy trees, but in 1-year-old needles, there were significant differences between damaged and healthy trees. In conclusion, needle damage in Yeocheon black pine can be considered the result of long-term exposure to oxidative stress by such as $O_3$ or $SO_2$, rather than a difference in climatic condition or soil properties, and the additional expense of photosynthate needed to overcome damage or alleviate oxidative stress may cause growth retardation.

Effects of Grapefruit Seed Extract on Oxidative Stability and Quality Properties of Cured Chicken Breast

  • This study investigated the antioxidative and functional effects of a curing agent containing grapefruit seed extract (GSE) on the quality and storage characteristics of chicken breast. The total polyphenol and total flavonoid contents of GSE were 45.06 mg/g and 36.06 mg/g, respectively. The $IC_{50}$ value of 2,2-diphenyl-1-picrylhydrazyl hydroxyl scavenging of GSE was $333.33{\mu}g/mL$. The chicken breast comprised six groups: no-treatment (N), 0.2% ascorbic acid + 70 ppm sodium nitrite (C), 0.05% GSE (G0.05), 0.1% GSE (G0.1), 0.3% GSE (G0.3), and 0.5% GSE (G0.5). The pH and cooking loss of cured chicken breast decreased with increasing GSE levels, and the water holding capacity increased with increasing GSE levels. The hardness and chewiness of GSE-treated chicken breast were higher than those of N and C. Hunter's L and a color values increased significantly after GSE addition. Moreover, 0.1% GSE (G0.1) increased the flavor and total acceptability scores. The 2-thiobarbituric acid and volatile basic nitrogen values of the 0.5% GSE group decreased significantly compared with those of C group. Total microbial counts of GSE-treated chicken breast were higher than those of C, but that lower than those of N. Adding GSE to chicken breast delayed lipid peroxidation and had antimicrobial effects during cold storage. GSE improved shelf life and palatability; therefore, it could be used as a natural antioxidant and functional curing agent ingredient in meat products.

Effect of Dietary Supplementation of the Combination of Gallic and Linoleic Acid in Thigh Meat of Broilers

  • This study was performed to investigate the combined effect of dietary supplementation of gallic and linoleic acid (GL) on the antioxidative effect and quality of thigh meat from broilers. Broilers received 3 dietary treatments: i) commercial finisher diet (control), ii) 0.5% GL (gallic:linoleic acid = 1 M:1 M), and iii) 1.0% GL during the 22 to 36 d. The pH value of broiler thigh meat was increased by GL supplementation. Water holding capacity of the thigh meat was enhanced by the 1.0% dietary GL supplementation. Antioxidative effect (total phenolic content, DPPH radical scavenging activity, $ABTS^+$ reducing activity, reducing power, and TBARS value) in the thigh from the broilers improved significantly with 1.0% GL. Linoleic acid and docosahexaenoic acids were higher in the broilers fed both levels of dietary GL. However, volatile basic nitrogen content and microbiological quality was not shown to be different between control and treated group. Results indicate that 1.0% dietary supplementation of GL can improve the antioxidant activity of broiler thigh meat and may enhance the meat quality.

오디 추출물의 알코올 분해능 및 항산화 효과 (Study on the Alleviation of an Alcohol Induced Hangover and the Antioxidant Activity by Mulberry Fruit)

  • 본 연구에서는 식용 가능한 오디를 용매별로 추출하여 항산화 효과 및 숙취 해소 활성능을 알아보고자 하였다. 오디Fig. 1. Contents of ADH ability of each solvent fraction of mulberry fruit extract. 추출물에 들어있는 총 페놀 함량을 정량하였으며, 오디 추출물의 전자공여능, SOD 유사 활성, TBARS 아질산염 소거능 등을 통하여 오디의 항산화 활성을 측정하였고, 알코올 분해능은 alcohol dehydrogenase activity assay 방법을 이용하여 조사하였다. 오디 추출물의 총 페놀 화합물은 60% methanol 추출물에서 가장 높게 나타났다. 전자공여능(EDA)은 오디의 40% ethanol 추출물에서 BHT와 유사한 활성을 나타났고, SOD 유사 활성은 BHT보다 오디 추출물이 낮은 활성을 나타내었다. 아질산염 소거능은 오디의 60% ethanol과 40% methanol 추출물이 각각 98%, 97%로 ascorbic acid와 상응하는 결과를 보였다. 알코올 분해능에서는 오디의 60% ethanol 추출물이 가장 높은 활성을 나타내었다.

토사자(Cuscutae Semen) 에탄올 추출물의 항산화 효과 및 항돌연변이 활성 (Antioxidative and Antimutagenic Activity of Ethanol Extracts from Cuscutae Semen)

  • 본 연구에서는 토사자 에탄올 추출물의 항산화 효과와 항돌연변이 활성을 분석하였다. 토사자 에탄올 추출물은 100 ppm, 200 ppm의 농도에서 BHT보다 높은 항산화 활성을 나타내었다. 토사자 추출물은 본 연구에서 사용한 농도에서는 돌연변이를 유발하지 않았으며, 직접돌연변이원인 4-NQO에 의해 유발된 돌연변이를 90% 이상 억제시키는 효과를 보여주어 우수한 항돌연변이능을 가지고 있는 것으로 나타났다. 또한 토사자 에탄올 추출물은 폴리페놀 화합물을 다량 함유하고 있어 이들 항산화 물질이 토사자의 항산화 효과와 항돌연변이 활성에 영향을 주었을 것으로 사료되었다. 본 연구 결과 토사자 에탄올 추출물은 항산화 효과와 함께 돌연변이 유발을 억제하는 항돌연변이 활성을 가지는 것으로 나타나 천연항산화제 및 고부가가치의 기능성식품 신소재로서 이용될 가능성을 보여주었다.

Anti-Oxidative and Anti-Inflammation Activities of Pork Extracts

  • This study was conducted to evaluate the antioxidative and anti-inflammatory effects of boiled pork powder (BPP) and hot water extract powder (HWEP) from 4 cuts of meat from Landrace × Yorkshire × Duroc (LYD). The highest DPPH radical scavenging activities determined were from BPP of Boston butt (13.65 M TE) and HWEP of loin (19.40 M TE) and ham (21.45 M TE). The 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activities of BPP from shoulder ham (39.28 M TE) and ham (39.43 M TE) were higher than those of other meat cuts, while HWEP of ham exhibited the highest ABTS radical scavenging activity. A higher oxygen radical absorbance capacity was determined for BPP from ham (198.35 M TE) and in HWEP from loin (204.07 M TE), Boston butt (192.85 M TE), and ham (201.36 M TE). Carnosine content of BPP and HWEP from loin and were determined to be 106.68 and 117.77 mg/g on a dry basis, respectively. The anserine content of BPP (5.26 mg/g, dry basis) and HWEP (6.79 mg/g, dry basis) of shoulder ham exhibited the highest value as compared to the extracts from the other meat cuts. The viability of RAW 264.7 cells was increased with increasing HWEP from loin and ham treatment. In addition, the expression of IL-6 and TNF-α was significantly reduced by HWEP from loin and ham, in a dose dependent manner. These results suggested that boiled pork and hot water extract of pork have antioxidative and cytokine inhibitory effects.

  • Objectives: This study was to investigate the antioxiative capacity, antiobesity effects of Atractylodes Rhizoma Alba, Houttuyniae Herba, Lonicerae Flos, Scutellariae Radix, and Coptidis Rhizoma on Raw 264.7 and 3T3-L1 cell lines. Methods: Three different types of herb extracts (A. Rhizoma Alba, H. Herba, L. Flos, S. Radix, and C. Rhizoma; water 100%, ethanol 30%, ethanol 100%) were used in this study. Total polyphenol compound, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, reactive oxygen species (ROS) activity, NO production and cell proliferation were measured. Results: Total polyphenol compound measurement of L. Flos, A. Rhizogenes, and C. Rhizoma extracts were higher than A. Rhizoma Alba, H. Herba. DPPH radical scavenging activity, ROS activity and NO production of S. Radix, C. Rhizoma extracts were lower than L. Flos, A. Rhizoma, and H. Herba. Conclusions: Metformin and S. Radix, C. Rhizoma, A. Rhizoma Alba, and L. Flos extracts combination groups showed synergistic effect on adipocyte differentiation inhibition and antioxidative activity.
