Inhibitory Effects of Maesaengi (Capsosiphon fulvescens) Extracts on Angiotensin Converting Enzyme and α-Glucosidase (매생이 추출물의 angiotensin converting enzyme 및 α-glucosidase 활성 저해 효과)

  • Physiological activities of hot water (MHW) and 80% ethanol (MEH) extracts from Maesaengi (Capsosiphon fulvescens) were investigated in this study. For the evaluation of antioxidant activities for MHW and MEH, 2,2-diphenyl-1-pic-rylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity were measured. DPPH radical scavenging activity and SOD-like activity of MHW, and MEH were increased weekly in a dose-dependent manner, and were about 10.8, 13.8, 62.4, and 27.1% at 10 mg/ml, respectively. The angiotensin converting enzyme (ACE) inhibitory activities of MHW and MEH were about 5.9% and 49.7% at 1 mg/ml, respectively. The ${\alpha}$-glucosidase inhibitory effect of MHW and MEH were about 1.4% and 67.3% at 1 mg/ml, respectively. To determine the influence of MHW and MEH on alcohol metabolizing activity, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH activity by MHW and MEH were increased weekly in a dose-dependent manner and ALDH activities were not detected. Elastase inhibitory activities of MHW and MEH were 75.9% and 51.2% at 10 mg/ml, respectively.

Analysis of nutritional compositions and biological activity of Agrocybe aegerita (버들송이(Agrocybe aegerita) 영양성분 및 생리활성 분석)

  • The mushroom Agrocybe aegerita was analyzed to evaluate the nutritional value of this potential food and to enhance the cultivation and consumption. Among the nutritional compositions of Agrocybe aegerita, contents of crude proteins, crude fats and ashes were 38.3%, 3.0% and 8.2% respectively. The contents of potassium and phosphorus were higher than that of other minerals. Total phenolics contents were $65.2{\mu}g/m{\ell}$ and $46.0{\mu}g/m{\ell}$ in water extract and ethanol extract, respectively. Total flavonoids were estimated as $12.5{\mu}g/m{\ell}$ contents of water extract and ethanol extract were $7.1{\mu}g/m{\ell}$. Free radical scavenging activity against DPPH of ethanol extract, 79.2% was higher than that of water extract(58.2%). The contents of total phenolics, flavonoids and antioxidant activity by the DPPH scavenging activity of Agrocybe aegerita were higher than those of Pleurotus eryngii and Pleurotus ostreatus. The tyrosinase inhibitory activity showed 63.2% and 65.0% in water extract and ethanol extract, respectively. Angiotensin converting enzyme inhibitory activity of water extract was 78.7% and 75.0% in ethanol extract. In this study, Agrocybe aegerita has abundant essential nutrients and thus is good source of functional health food.

Physicochemical and Functional Properties of Kochujang with Broccoli Leaf Powder (브로콜리잎 분말 첨가 재래식 고추장의 이화학적 및 기능적 특성)

  • Home-made broccoli Kochujang (HMBK) was prepared with the addition of 5% broccoli leaf powder. Its physicochemical and functional properties were measured in extracts (80% methanol, 80% ethanol, and distilled water) and compared with home-made Kochujang (HMK) and factory-produced Kochujang (FPK). Total phenolic content (TPC) was 22% higher in methanol extract from HMBK (524.2 GAE/100 g) compared to HMK (431.0 GAE/100 g). TPC was 8% higher in ethanol extract from HMBK (541.9 GAE/100 g) compared to HMK (499.9 GAE/100 g). DPPH radical scavenging activity was 1.6 times higher in the methanol extracts from HMBK than HMK. In contrast there was no difference in DPPH radical scavenging activity between HMBK and HMK. Oxygen radical absorbance capacities in methanol and ethanol extracts from HMBK were similar to HMK, but both were higher than extracts from FPK (55% and 23% higher, respectively). Inhibition of angiotensin I converting enzyme activity in methanol extracts from HMBK was similar to HMK, but it was 2.6 times higher than inhibition activities from FPK. Interestingly, only ethanol extract from HMBK showed a 10.7% and 18.3% inhibition on cell growth of the human colon adenocarcinoma grade II cell line (HT-29) and human lung carcinoma cell line (NCI-H1229), respectively. These results indicate home-made Kochujang with broccoli leaf powder contains high total phenolics, antioxidant activities, and cancer cell growth inhibition activities.

Extraction of Antioxidants from Lonicera japonica and Sophora japonica L.: Optimization Using Central Composite Design Model (금은화와 회화나무꽃으로부터 항산화성분의 추출 : 중심합성계획모델을 이용한 최적화)

  • In this study, an antioxidant was extracted from Lonicera japonica and Sophora japonica L, which was optimized by using the central composite design (CDD) model of response surface methodology (RSM). The response value of CDC model establishes the extraction yield and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. The extraction time, volume ratio of ethanol/ultrapure water, and extraction temperature were selected as quantitative factors. According to the result of CDC, optimal extraction conditions of Lonicera japonica were as follows; the extraction time of 2.08 h, volume ratio of ethanol/ultrapure water of 41.53 vol.%, and extraction temperature of $55.08^{\circ}C$. At these conditions the expected results indicated that the yield and DPPH radical scavenging activity were estimated as 37.88 wt% and 40.37%, respectively. On the other hand, optimal extraction conditions of Sophora japonica L. could be found as the extraction time of 2.13 h, volume ratio of ethanol/ultrapure water of 62.89 vol.%, and temperature of $50.42^{\circ}C$. Under the conditions, the (possible) maximum values of yield and DPPH radical scavenging activity were found as 35.43 wt% and 55.7%, respectively.

Optimization of Extraction Process for Total Polyphenols from Angelica Using Response Surface Methodology (반응표면분석법을 이용한 안젤리카로부터 폴리페놀 성분의 추출공정 최적화)

  • In this study, polyphenols were extracted from Angelica, which are known to have a high antioxidant content and the extraction process was optimized using the response surface methodology. The extraction yield and the total polyphenols were set as response values for the methodology. Quantitative factors in the extraction process were the extraction time, volume ratio of alcohol/ultrapure water, and extraction temperature. When considering both the main and interaction effects, the greatest influence factor on the extraction yield and total polyphenols was the extraction time. The optimum extraction time and temperature and alcohol/ultrapure water volume ratio for angelica were 2.8 h, $56.6^{\circ}C$ and 64.0 vol% respectively. The extraction yield and total polyphenols when using the conditions were calculated to be 24.6% and 8.76 mg GAE/g. respectively. Determination coefficients of regression equations for the extraction yield and total polyphenols were 81.4 and 75.4%, respectively. Also the overall satisfaction level was found to be 0.80 and the significance was confirmed within 5%.

  • To develop the functional pear fruitlet product, we prepared fermented pear fruitlet product (FPFP) from mixed fermentation of Saccharomyces cerevisiae, Kluyveromyces fragilis and Lactobacillus plantarum. Then, we investigated their several physiological functionalities. Among several physiological functionalities, antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of the FPFP was the highest of 87.4% and its antioxidant activity was also showed 69.6%. FPFP from mixed fermentation by yeasts and Lactobacillus plantarum after thawing of frozen pear at $20^{\circ}C$ showed higher physiological functionalities than those of single fermentation by Saccharomyces cerevisiae or Bacillus subtilis after $40^{\circ}C$ of thawing.

Improvement on Yield and Functional Properties of Autoclave-Treated Salmon Frame Extracts using Commercial Enzymes (효소 처리에 의한 고온가압 연어 frame 추출물의 수율 및 건강 기능성 개선)

  • This study was conducted to improve yield and functional properties of autoclave-treated salmon frame extracts (SFETA) using commercial enzymes (Alcalase 2.4 L FG, Flavourzyme 500 MG, Neutrase 0.8 L and Protamex 1.5 MG). Yield and angiotensin I converting enzyme (ACE) inhibitory activity of all enzymatic hydrolysates improved compared to those of control (undigested extracts), which were the highest in hydrolysates incubated with Protamex 1.5 MG for 4 hrs (P4-treated hydrolysates) and 2 hrs (P2-treated hydrolysates), respectively. However, antioxidant activities of all enzymatic hydrolysates showed less than 29%. According to the trichloroacetic acid soluble-N, volatile component intensity and sensory evaluation, when compared to control, taste of P4-treated hydrolysates improved, while its fish odor strongly smelt. Therefore, for efficient use of P4-hydrolysates, the fish odor should be improved by Maillard reaction of extracts or pre-treatment of salmon frame.

Manufacture and Physiological Functionalities of Some Natural Plant Fermentation Broths and Liquor (몇 가지 천연식물 발효액의 제조 및 생리기능성)

  • Three kinds of plant fermentation broths and one liquor were prepared using persimmon, Ulmus davidiana var. japonica and barley, and investigated its physiological functionality. Anti-aging antioxidants activity was the highest of 99% in Ulmus dacidiana bar. japonica liquor and also was high in the persimmon vinegar(68.3 %). SOD-like activity show also 80.6% in the Ulmus davidiana var japonica liquor however the other fermentation broths were less 25%. Antihypertensive angiotensin I-converting enzyme inhibitory activity and fibrinolytic activity were weak or not detected in all the fermentation broths. Anti-dementia acetylcholinesterase inhibitory activity were showed 54.7% in the Ulmus davidiana var. japonica liquor. Our results showed that the Ulmus davidana var. japonica liquor has the potential to become a new functional liquor because of high antioxidant activity and anti-dementia activity.

Extraction of Flavonoid Components from Persimmon Leaf, Thistle and New Green (감잎, 엉겅퀴, 뉴그린으로부터 플라보노이드성분의 추출)

  • In this study, we extracted active components from thistle, persimmon leaf, and new green which are known to have a high content of antioxidants and also analyzed the 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavening activity and flavonoid content. Both ultrapure water and alcohol were used as extraction solvents and the ratio of both solvents, sample, amunts extraction time, and extraction temperature were varied. The optimal extraction condition of each natural compounds were 2.5~3.5 h of the extraction time and 50 g/L of the sample amount. The optimal ratio of ultrapure water and alcohol and extraction temperature were as follows; persimmon leaf (55~65 vol%, $50{\sim}60^{\circ}C$), thistle (40~50 vol%, $55{\sim}65^{\circ}C$) and new green (55~65 vol%, $50{\sim}60^{\circ}C$). In addition, the antioxidant capacity and flavonoid content of the extract increased in the order of persimmon leaf, thistle, and new green.

Network Pharmacology: Prediction of Astragalus Membranaceus' and Cornus Officinalis' Active Ingredients and Potential Targets to Diabetic Nephropathy (네트워크 약리학을 통한 당뇨병성 신병증에서의 황기와 산수유의 활성 성분 및 잠재 타겟 예측)

  • The purpose of this study is to predict the effects of macroscopic and integrative therapies by finding active ingredients, potential targets of Astragalus membranaceus (Am) and Cornus officinalis (Co) for diabetic nephropathy. We have constructed network pharmacology-based systematic and network methodology by system biology, chemical structure, chemogenomics. We found several active ingredients of Astragalus membranaceus (Am) and Cornus officinalis (Co) that were speculated to bind to specific receptors which had been known to have a role in the progression of diabetic nephropathy. Four components of Am and eleven components of Co could bind to iNOS; two ingredients of Am and six ingredients of Co could docking to cGB-PDE; one component of Am and nine components of Co could bind to ACE; three ingredients of Co with neprilysin; three components of Co with ET-1 receptor; four ingredients of Am and fourteen ingredients of Co with mineralocorticoid receptor; one component of Am and seven components of Co with interstitial collagenase; one ingredient of Am and ten ingredients of Co with membrane primary amine oxidase; one component of Am and four components of Co with JAK2; two ingredients of Am and one ingredient of Co with MAPK 12; one component of Am and five components of Co could docking to TGF-beta receptor type-1. From this work we could speculate that the possible mechanisms of Am and Co for diabetic nephropathy are anti-inflammatory, antioxidant and antihypertensive effects.
