韓國産 抗腫瘍性 資源의 Screening에 대하여(II) (Screening for Antitumor Efficacy fro the wild plants in Korea(II))

  • Many plants, which collected from Korea, were applied to antitumor and cytotoxic screeing tests against sarcom 180 a ascitec in mice, V-79 KB and P388 cultured cells. The results are summarixed as follows:1) The total packed cell volum method has been used for the antineoplastic screening for from natural higher plants in Korea. By this method, we have found out that the root, leaf and stem of Tripterygiu, regelii Spragne & Taketa having strong antineoplastic activity and also Rumex Japonicus Houtt. Eragrositis ferru-ginea Beauv. and Patrinia scabio-saefolia Fischer showed significant activity to anticancer tumor while cynanchum wilfordii Hemsley, and Rosa polyantha Sieb. et Zacc. showed slight activity to antitumor. 2) Among the 13 tested plants, the root and stem of Tripterygium regelii Spragne & Taketa and Amethystanthus excisus Nakai showed strong antitumor activity by the V79 cytotoxic cell screening test. 3) Twelve plants, which are glowing in mountainous area of Korea tested to anticancer activity. From the results, Eragrositis ferru-ginea Beauv., Angelica gigas Nakai, Geranium sibiricum L., Patrinia scabio-saefolia Fisher, Cynanchum wilfordii Hemsley, and Rubia akane Nakai have been proved to be anti-cancer plants by using P388 cell cultured method. 4) Tripterygiu, resgelii Spragne & Taketa, Eragrositis ferru-ginea Beauv., Patrinia scabio-saefoli Fisher, Cynanchum wilfordii Hemsley and Rasa polyantha Sieb. et Zacc., var. genuina Thunb. showed strong anti-tumor activity both total packed cell volume method and Cytotoxicity method.

  • Some Mannich bases of Antineoplaston A10 which is antitumor agent under chlinical investigation were synthesized and tested fro cytotoxicity. The tested compounds (2a, 2b, 2d) showed good activity comparable to that of carboplatin.

  • Pharmacological studies have been carried out with the methanol extract of the whole plant of Pseudostellaria palibiniana Ohwi (Caryophyllaceae). The results showed that it had stimulation effect on heart, hypotensive actions depending on extract contents, stimulation effect on respiration, contraction on excised intestines, enhancement of tension on excised uterus, antineoplastic activity on Ehrlich carcinoma, liver protective activity against $CCl_4$ intoxication, writhing and diuretic activities.

  • Antineoplastic activity against human gastric and colon carcinoma cell lines was measured in 49 extracts from 46 plants using MTT (3-[4, 5-dimethyl thiazol-2-yl]-2, 5-diphenyl tetrazolium bromide) method. Six extracts from five plants have been reported to have antineoplastic effect. Extracts from remaining 41 plants failed to show significant cytotoxic effect at the concentration of less than $230\;{\mu}g/ml$.

저분자량 키토산 올리고당의 항종양성 (Antineoplastic Effect of Low Molecular Weight Chitooligosaccharide on Various Tumor Cell Lines)

  • 저분자량 키토산 올리고당의 세포 독성을 실험하였다. 저분자량 키토산 올리고당은 정상세포주인 Vero E6(Africa green monkey kidney cell)에 대한 세포 독성을 거의 나타내지 않았다. 정상세포주에 대한 저분자량 키토산 올리고당의 $IC_{50}$값은 $1,060.28{\mu}g/m{\ell}$이었다. 저분자량 키토산 올리고당은 폐암 세포주인 A549, 방광암 세포주인 J82, 대장암 세포주인 SNU-C4, 위암 세포주인 SNU-1, 유방암 세포주인 ZR75-1 등과 같은 사람의 종양세포주에 대한 in vitro 항종양성을 나타내었다. 종양세포주에 대한 저분자량 키토산 올리고당의 $IC_{50}$값은 A549, J82, SNU-C4, SNU-1, ZR75-1 세포주의 경우에 각각 $477.42{\mu}/m{\ell}$, $480.40{\mu}g/m{\ell}$, $436.84{\mu}g/m{\ell}$, $373.55{\mu}g/m{\ell}$, and $539.95{\mu}/m{\ell}$이었다.

  • To find out the novel anticancer agent for the treatment of solid tumors, 1-arylsulfonyl-2-alkoxy-4-arylimidazolines were designed and prepared from substituted styrenes through three steps. Compared to 5-fluorouracil, these analogues exhibit moderate cytotoxicity against human solid tumor cell lines, A-549(lung carcinoma) and SK-Mel-2(malignant melanoma). The structure-activity relationship indicates the high potential of this series for the development of novel antineoplastic agent.

  • DNA topoisomerases I (topo I) and II are essential enzymes that relax DNA supercoiling and relieve torsional strain during DNA processing. including replication. transcription. and repair. Topo I relaxes DNA by cleaving one strand of DNA by attacking a backbone phosphale with a catalytic lyrosine (Tyr723. human topo I). This enzyme has recently been investigated as a new target for antineoplastic drugs. Inhibitors to the enzyme intercalate between the DNA base pairs. interfering religation of cleaved DNA, therefore inhibit the activity of topo I. (omitted)

항종양활성천연약물연구의 전망 (DISCOVERY OF NEW ANTITUMOR AGENTS FROM MEDICINAL PLANTS)

  • Many types of compounds have been isolated from higher plants till now, that is, alkaloids, terpenes, lignans, steroids and so on. One of them, named as RA series Cyclic hexapeptides isolated from Rubia akane and R. cordijofia also have strong antineoplastic activity against various types of tumors. Till now 10 kinds ofRA series compounds were isolated and named as RA - I, II, III, IV, V, VI, VII, VIII, IX and X. Moreover,monogl-ucoside of RA - V newly Isolated from same plant. Many kinds of derivatives including natural RAcompounds were tested for QSAR, and one of them, RA - VII was screened up as a most suitable substance asan antitumor agent. RA - VII(=RA-700) has strong cytotoxic activity against KB cells, P388 Iymphocyticleukemia and MM2 mammary carcinoma cells. RA - VII has been under investigation for Phase I clinical trials.

  • 21plants, which collected from Korea and Japan, were applied to antitumor and cytotoxic screening tests against sarcoma 180 a ascitec in mice. The results are summariged as follows : 1) The total packed cell volume method has been used for the antimeoplastic screening for from natural higher plants in Korea. By this method, we have found out that Selaginella involves, Patrinia hispida, Archyranthes japonica and Solanum nigrum having significant activity and also Cydonia sinensis and Rubia akane showed slight activity to antitumor 2) The total packed cell volune method has been used for the antineoplastic screening for from natural higher plants in Japan. Among the 21 tested plants, lsodon japonicus having strong antitumor activity and also Torilis japonica, Aralia elata, Leonurus sibiricus and Rubia cordifolia showed significant activity to anticancer tumor while Forsythia spp and Solanum nigrum showed slight activity to antitumor. 3) Among the 21 tested Korea plants, lsodon excisus and Forsythia Koreana showed strong antitumor activity by the V79 cytotoxic cell screening test.
