산마늘 기능성 활성물질 탐색 및 자원화 (Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

고지혈증 흰쥐에서 생약제제 KH-204의 발기부전 치료연구 (The Effect of Herbal Formula KH-204 on Erectile Dysfunction in Hyperlipidemic Rat)

  • This study was designed to investigate the effects KH-204 on erectile dysfunction in hyperlipidemic rat. KH-204 has been evaluated antihyperlipidemic and antierectile dysfunction effects on experimental hyperlipidemic rats induced by high fat diet. After oral administration of the water extract KH-204 (50, 100, 200, 300 mg/kg) to hyperlipidemic rats for 8 weeks, the variables including body weight, total cholesterol, HDL and LDL levels in serum, the expression of eNOS and nNOS in penis were measured. And erectile function was determined by measurement of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve. Oral administration of KH-204 significantly inhibited the increases of serum total cholesterol and LDL-cholesterol levels and the decreased of serum HDL-cholesterol levels in hyperlipidemic rats induced by high fat diet. The penile expression level of the two enzyme (eNOS, nNOS) were increased significantly after oral administration of the KH-204 50 mg/kg. Erectile function after 10 volts stimulation was significantly decreased in the hyperlipidemic rat compared with the normal rat, but increased in KH-204 group compared with hyperlipidemic group. These results suggest that KH-204 is effective for erectile dysfunction in hyperlipidemia.

Antidiabetic activity of Argyreia speciosa (sweet) (Burm.f.)Boj. in normoglycemic and Streptozotocin-induced diabetic rats

  • Effect of ethanol (ASE) and water (ASW) extracts of Argyreia speciosa on blood glucose and lipid profile was investigated in normoglycemic and Streptozotocin (STZ)-induced diabetic animals. In oral glucose and sucrose tolerance test, treatment with ASE and ASW (100 and 200 mg/kg) and Glidenclamide (10 mg/kg) significantly improved the glucose and sucrose tolerance in normal animals. In addition, respective treatment for fifteen-day resulted in significant percentage reduction in serum glucose (SG) ie., 30.39% (lower dose of ASE) and 33.21% (higher dose of ASW). In standardized STZ (50 mg/kg, iv)-induced diabetic rats, a single dose of ASE and ASW treatment exhibited reduction in SG levels at different time intervals compared to basal levels. Administration of both the doses of ASE and ASW for fifteen-day days exhibited greater percentage reduction in glycemia (24.6%, 24.7%, 23.9% and 21.9% respectively) and also ameliorated restored to near normal value of all tested lipid parameters. Further, treatment also exhibited significantly improved glucose tolerance over the period of 120 min compared to diabetic control group. Eventhough treatment failed to increase serum insulin levels significantly but peripheral utilization of insulin was increased as evident by insulin tolerance test. Taken together, present study supports the traditional usage of title plant in the treatment of diabetes mellitus.

Antihyperlipidemic Effect of Dietary Lentinus edodes on Plasma, Feces and Hepatic Tissues in Hypercholesterolemic Rats

  • We investigated diet supplementation with shiitake mushroom fruiting bodies on biochemical and histological changes in hypercholesterolemic rats. Six-wk old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. A diet containing 5% Lentinus edodes fruiting bodies given to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and the LDL/high-density lipoprotein ratio by 34.33, 53.21, 75.00, 34.66, 25.73, and 71.43%, respectively. Feeding mushroom also significantly reduced body weight in hypercholesterolemic rats. However, it had no detrimental effects on plasma albumin, total bilirubin, direct bilirubin, creatinine, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that L. edodes significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red-O staining showed normal findings for mushroom-fed hypercholesterolemic rats. These results suggest that shiitake mushrooms could be recommended as a natural cholesterol lowering substance in the diet.

아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 혈청 지질 성분에 미치는 영향 (Effects of Feeding the Mixture of Linseed and Sunflower Seed Oil on the Lipid Components of Serum in Dietary Hyperlipidemic Rats)

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0 % and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil and antihyperlipidemic drugs for 2 weeks. Analysis of the lipid component of the serum showed following results. Concentration of total cholesterol in serum was significantly lower in the other groups except group 2 (lard 3.0 % + olive oil 12.0%) than in the control group , especially in groups 3 (cholestyramine 2.0%) and 5 (lard 3.0% + linseed oil 12.0%). HDL-C and the ratio of HDL-C to total cholesterol concentration were higher in groups 1, 3, 5 and 9 (lard 3.0% + sunflower seed oil 12.0%) , while the atherosclerotic index was low in groups3, 5. Concentrations of free cholesterol and choesteryl ester in serum were significantly lower in the other groups except group 2 than in the control group, especially lower in grop 5. Concentration of LDL was significantly higher in group 2 while it was remarkably lower in groups 3 and 5, while those of triglyceride in groups in 3 and 4 (lipraroid) were significantly higher than in the control groups. From the data on concentration of total choelsterol , HDL-cholesterol, LDL, phospholipid and triglyceride in serum, the results suggested that the feeding mixed with 3.0% lard and 12.0% linseed oil or 2.0% cholestyramine were most effective for the improvement of the serum lipids.

  • This study was carried out to investigate the effect of the feeding mixture of linesed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hypprlidemic rats. Concentration s of the total cholesterol and phospholipid in liver were significantly higher in group 2 (olive oil 12.0%) and lower in the other groups than in the control group, especially lower in groups 3 (cholestyramine 2.0%) and 9 (sunflower seed oil 12.0%) . Concentration of triglyceride was lower in the other groups except group 4 (liparoid), especially lowe rin group 9 than in the control group. In the fatty acid composition of liver lipids, C18:2 was the major fatty acid. Contents of n-6 PUFA increased , while those of n-3 PUFA decreased in groups composition of the test lipids. From the data on concentration s of total cholesterol. Phospholipid and triglyceride in liver, we concluded that the feeding mixed with 3.0% lard and 12.0 % sunflower seed oil were most effective for the improvement of the live lipids. The fatty acid composition in liver lipids were affected by the fatty acid composition of the test lipids.

Antihyperlipidemic and Antioxidant Effects of the Mixture of Ginseng Radix and Crataegi Fructus: Experimental Study and Preliminary Clinical Results

  • The mixture of Ginseng Radix and Crataegi Fructus (Gen-CF) was developed to increase the pharmacological effect of ginseng in the treatment of hypercholesterolemia and prevention of cardiovascular disease. This study evaluated the effects of Gen-CF on serum lipids of hypercholesterolemic rats in vivo, as well as its antioxidant activities in vitro, and explored its clinical effects on patients with hypercholesterolemia. In vitro, Gen-CF displayed 1,1-diphenyl-2-picrylhydrasyl and superoxide radical scavenging activities, and inhibited hemolysis induced by 2,2'-azobis-2-amidinopropane dihydrochloride in a dose-dependent manner. In vivo, Gen-CF significantly inhibited the increases of total cholesterol, low-density lipoprotein cholesterol and triglyceride in high cholesterol-diet and Triton WR-1339 models. It also significantly inhibited the decrease of high-density lipoprotein cholesterol in these models. In the clinical trial, Gen-CF significantly lowered total cholesterol, low-density lipoprotein cholesterol, triglyceride, total lipid and phospholipid, with no adverse events, including hepatic or renal toxicity. The data suggest that Gen-CF has the potential to treat hypercholesterolemia and prevent cardiovascular disease.

약용식물 혼합물의 고지혈증 개선 효과 (Antihyperlipidemic Effect of Complex of Medicinal Plant Products on Hyperlipidemic Rats Induced by High Cholesterol Diet)

  • The HWND_G02 (Salvia miltiorrhiza Bunge, Crataegus pinnatifida, Polygonum multiflorum Thunberg, Cnidium officinale Makino) and HWND_G03 (Cinnamonum cassia Blume, Salvia miltiorrhiza Bunge, Crataegus pinnatifida, Polygonum multiflorum Thunberg, Cnidium officinale Makino, Allium macrostemon Bunge) are new natural mixture composed with several oriental herbs. The aim of the present study was to investigate the effects of HWND extracts on high cholesterol diet (HCD)-induced hyperlipidemic rats. Male Sprague-Dawley rats were divided into five groups: control, HCD, atorvastatin (5 mg/kg, po), ethanolic extracts of HWND_G02 (1,000 mg/kg, po) and HWND_G03 (1,000 mg/kg, po) were administered to the HCD-induced hyperlipidemic rats for 4 weeks to evaluate their anti-hyperlipidemic activities. HWND extracts markedly decreased body and liver wight gain, and recovered serum lipid levels, such as total cholesterol (TC), triglycerides (TG), low density lipoprotein-cholesterol (LDL-C) and high density lipoprotein-cholesterol (HDL-C) in the HCD-induced hyperlipidemic rats. Furthermore, the lipid levels (TC and TG) and the lipid accumulation were significantly lowered in the liver tissue of HWND-administrated rats. After a HCD, each group had a lower atherogenic index (AI) compared to the HCD group. In conclusion, these data suggest that HWND extracts could be the candidate for the material to prevent hyperlipidemia.

긴볼레기말의 항고지혈증 효과에 관한 연구 - Triton WR-1339 주사에 의한 고지혈증 유발 생쥐의 간세포내 지방 축적 감소를 중심으로 (The Antihyperlipidemic Effect of Colpomenia bullosa Extract -Based on the Decreasing Effect of Lipid Accumulation in Hepatocyte of Murine with Hyperlipidemia induced by Triton WR-1339)

  • 본 연구는 긴볼레기말 추출물의 항고지혈증 효과를 조사하기 위해 ICR 생쥐에 Triton WR-1339(TX) 복강주사로 인위적인 고지혈증을 유발시킨 후 긴볼레기말 추출물(30mg/kg)를 복강주사하여 시간의 경과에 따른 간세포내에서의 지방 축적 변화를 조직화학적으로 관찰하였다. TX 주사후 그물구조의 세포질을 가진 간세포가 간엽 전체에서 관찰되었고, 일부 간소엽에서는 간세포 손상으로 인한 간세포판 소실이 나타났다. 또한 간세포내 지방축적도 증가하여 전체 간소엽의 간세포에서 지방의 과출현을 확인 할 수 있었고, 지방의 크기도 대조군에 비해 증가된 것으로 관찰되었다. 그러나 긴볼레기말 추출물 주사군에서는 그물구조의 세포질을 가진 간세포의 수가 TX 주사군에 비해 감소되었고, 대부분의 간소엽에서 정상적인 간세포판의 배열을 확인할 수 있었다. 간세포내의 지방 축적과 크기도 감소된 경향으로 관찰되었다. 이상의 결과로 볼 때 해조류 긴볼레기말 추출물은 고지혈증이 유발된 생쥐 간세포 내에서의 과도한 지방축적을 감소시키는 항고지혈증 효과을 하는 것으로 사료된다.

고지방식이로 고지혈증이 유도된 흰쥐에서 황화된 수용성 β-glucan의 항고지혈증 효과 (Antihyperlipidemic Activities of a Chemically Engineered Sulfated Mushroom β-glucan on High Fat Dietary-induced Hyperlipidemia in Sprague-Dawley Rats)

  • 본 연구는 영지버섯 균사체로부터 얻은 수용성 베타글루칸(SBG)이 고지방식이로 유도한 흰쥐의 항고지혈 효과 및 혈중 지질수준에 미치는 영향을 조사하였다. 5주령의 수컷 흰쥐에 고지방식이를 2주간 급여하여 비만을 유도하고, 그 후 2주 동안 다음과 같은 5가지 group의 식이를 급여하였다: 1) 일반사료 식이군(NC), 2) 고지방사료 식이군(HC), 3) 고지방사료 및 고농도(200 mg/kg) SBG 식이군(HC-HSBG) 4) 고지방사료 및 저농도(20 mg/kg) SBG 식이군(HC-LSBG), 5) 고지방사료 및 로바스타틴(lovastatin) 식이군(HC-Lov). 실험 결과, 식이 효율(Food efficiency ratio)은 고지방식이를 급여한 군들에서 일반사료 식이군에 비하여 증가하였으나 통계적 유의성은 없었다. 혈액학적 결과에서 고농도 및 저농도의 SBG와 로바스타틴 투여군은 고지방식이만 급여한 군(HC)보다 호중구와 중성구 수치가 유의성 있게 증가하였다. 또한 혈청 내 총콜레스테롤, 중성지질 및 LDL-C는 고농도 및 저농도 SBG 투여군 모두에서 유의성 있게 감소되었다. 이 같은 결과는 화학적으로 황화된 수용성 베타글루칸(SBG)이 흰쥐에서 혈중 콜레스테롤 수치를 낮추어 항고지혈증 효과를 나타내는 것을 의미한다.
