This study was performed to evaluate the antioxidant and antimicrobial activity of methanol extract from Rosmarinus officinalis L. and its fractions. The ethyl acetate fraction of rosemary had a higher antioxidant activity in both DPPH ($3.22\;{\mu}g/ml$) and ABTS ($5.05\;{\mu}g/ml$) compared to other extracts and fractions. Based on the results of the FRAP assay, the ethyl acetate fraction of rosemary showed a value of $5.9{\pm}0.3\;{\mu}M/{\mu}g$, and buthanol fraction and rosmarinic acid exhibited values of $4.8{\pm}0.2\;{\mu}M/{\mu}g$ and $5.1{\pm}0.1\;{\mu}M/{\mu}M$, respectively. Measurements of the antimicrobial activities of the extracts, fraction against gram positive, negative bacteria revealed that the methanol extract, hexane, ethyl acetate, and chloroform fraction of rosemary caused Staphylococcus aureus and Escherichia coli to form clear zones greater than 12 mm. Furthermore, the methanol extract and chloroform fraction showed high antibacterial activity, with inhibition zone exceeding 13 mm. The methanol extract and chloroform fraction of rosemary had broad antimicrobial spectrums and low MIC values. Therefore, methanol extracts of rosemary could serve as potential antibacterial agents to inhibit pathogen growth in food and hand sanitizers.

Seungmagalgeuntang (SG) is broadly used in traditional Oriental medicine especially in Korea, China, and Japan, for its many pharmacological effects. This study was carried out to investigate the antioxidative, antimicrobial, and anticytotoxic activities of SG and fermented seungmagalgeuntang (FSG). DPPH radical scavenging activities of SG and FSG were 70% and 74%, respectively, which increased slightly by fermentation. Nitrite scavenging activities were strongly altered at pH 1.2, (36.4% in SG and 38.3% in FSG) by addition of $200{\mu}g/g$. Superoxide dismutase-like activities were from 21.5% to 23.3% at a concentration of 0.4 mg/mL, and the highest value were observed in FSG. Total flavonoid contents of SG and FSG were 47.1 and $52.1{\mu}g/L$, respectively which shows an increase upon fermentation. In the antimicrobial activity test, $MIC_{50}$ values of SG and FSG were $800{\mu}g/mL$ for Candida albicans and 3,200 and $1,600{\mu}g/mL$ for Staphylococcus epidermidis and Staphylococcus aureus, respectively. Antibacterial effects were higher in FSG compared to SG. Anticytotoxic cadmium toxicities ranged from 63.5% to 76.1% at $10{\mu}g/mL$ of SG and FSG, and the highest value was observed in FSG. In the sensory evaluation, color, flavor, and overall preference values were higher in FSG.

Objectives : This study was conducted to measure the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica against various species of anaerobic bacteria. Methods : In order to evaluate the antioxidative and antimicrobial activities of water- and ethanol-extracts, DPPH radical scavenging and superoxide dismutase (SOD)-like activities were measured in various species such as Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Propionibacterium acnes that induce skin inflammation. Also the total amount of phenol in each water- and ethanol-extract was measured to identify its role in the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica. The antibacterial activity of the extracts was measured by confirming the lowest concentrations in disk diffusion and minimum inhibitory concentration (MIC) tests. Results : It was observed that RJE (Rhus javanica-EtOH extracts) show the highest content of total phenol. In addition, for RJE the total phenol content was higher from samples taken from domestic sources than from samples taken from foreign sources. DPPH radical scavenging activity was increased by treatment with PGE (Punica granatum-EtOH extracts), RJE and RJW (Rhus javanica-water extracts). It was observed that SOD-like activity was highest in the treatment with PGE. All of the extracts showed antimicrobial activity on S. epidermidis, S. aureus, P. acnes and E. coli, including those from Punica granatum, and it was noted that the activity was higher with RJE than with RJW. Conclusions : These results provide evidence that ethanol extracts of Punica granatum and Rhus javanica may have a beneficial role as antioxidants and antibiotics. Extracts from domestic samples of Betula platyphylla var japonica appeared to have a greater efficacy than extracts from foreign samples.
The natural products are used to be development of new antibacterial substances against human pathogenic bacteria. Adherence to the tooth surface by S. mutans is an important step in initiation of dental caries. This study was to examine antibacterial activity and anti-adhesive effect of Scutellaria baicalensis extract against S. mutans. Extracts of S. baicalensis were tested for antimicrobial activities by paper disc methods and radial diffusion assay methods, and bacterial adherence assay using 3 type of hydroxyapatite. The antibacterial level of ethyl acetate extract, IPK-3 on the growth of S. mutans was 125 mg/ml of minimum inhibitory concentration (MIC). The maximum growth of S. mutans in medium added with IPK-3 extract (50 mg/ml) was delayed to 30 hr, while the highest at 24 hr in control medium. The pH values of the control medium was 5.63 at 18 hr, but the media supplemented with IPK-3 extract was pH 6.50 at 12 hr. In adhesive inhibition assay, S. mutans was labelled with the fluorescent indicator DAPI and measured with fluorescence microscope. Adhesion of S. mutans on hydroxyapatite beads was inhibited by IPK-3 extracts. These results suggest that S. baicalensis extract can be used as an effective material for antibacterial activity and adhesive inhibition against S. mutans.
This research was performed to investigate the possibilities of industrial usage of camellia (Camellia japonica L.) by examining the antioxidant and antimicrobial effects of methanol extract with different sections. Content of total phenolics, DPPH radical scavenging activities and antibacterial activity of young leaf, mature leaf, flower bud, flower, bark, and seed of camellia were compared in vitro experimental models. Total phenolics was contained the higher in young leaf (74.62 mg), flower bud (65.02 mg) and flower (62.42 mg) but less than 20.95 mg per 100 g of dry weight in other parts of Camellia japonica L. And effects of antioxidant measured by DPPH radical scavenger activity ($RC_{50}$, reduce concentration 50%), was shown higher $7.16{\sim}18.14\;{\mu}g/m{\ell}$ in methanol extract of young leaf, flower bud and flower than $61.23\;{\mu}g/m{\ell}$ of BHT as a chemical oxidant. Also, the antimicrobial activity of Camellia japonica L. extracts determined using a paper disc method against food-borne pathogen and food spoilage bacteria, the young leaves extracts showed the most active antimicrobial activity against 7 kinds of harmful microorganisms. Flower bud extracts showed the highest antibacterial activity against P. aeruginosa and Enterobacter spp. C1036. In addition, the minimum inhibitory concentration (MIC) of young leaf extract against B. subtillis,S. fradiae,S. aureus,E. coli,P. aeruginosa, Enterobacter spp. C1036, and S. typhimurium were revealed 1 to 15 ${\mu}g/m{\ell}$. As a result, antimicrobial activity of camellia extracts was shown higher gram positive bacteria than gram negative bacteria.
The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.
Yam, Prunella was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Anti-microbial activity of each extract was investigated. Hexane extract was tested for anti-microbial effect on Streptocaccus mutans, one of causative factor of dental caries. Methanol extracts of 7 plants were investigated to anti-microbial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. Methanol extract of Yam and Prunella revealed anti-microbial activity against S. mutans, P. gingivalis, and S. aureus. Also, hexane fraction of Yam revealed anti-microbial activity against S. mutans. In sequence of hexane, chloroform, ethylacetate, butanol fraction by Prunelia acted as potent anti-microbial agent on P. gingivalis. The measured MIC of hexane fraction of Yam and Prunella on S. mutans KCTC 5316 strain was 0.25 mg/ml and 0.5 mg/ml and the MIC of hexane fraction of Prunella on S. aureus was 0.5 mg/ml. The hexane fraction of Yam and Prunella suppressed viable ceil counts(VCC) of S. mutans, especially after 24 hrs. The Prunella hexane fraction suppressed VCC of S. aureus, after 12 and 24 hrs. Tested concentrations were 0.1, 0.25 and 0.5 mg/ml. the results were compared with control (0 mg/ml). The pH of S. mutans media and GTase activity were determined to evaluate the anticariogenic activity of Yam, Prunella hexane fraction. The pH were increased from 5.6 to 7.0-7.2 in concentration of 2.0 mg/ml. Yam hexane extraction revealed 35% inhibition to GTase activity and Punella inhibited 25% of GTase. These results suggest that the hexane extracts of Yam and prunella have Antibacterial activities against S. mutans, P. gingivalis, S. aureus and have preventive effect on dental caries.
The methanolic extracts from the elm (Ulmus davidiana var japonica) root barks which have been known as an oriental medicine Eubackpie for the treatment of edema, ulcer and inflammation were examined for their bacteriocidal effects on mastitis pathogens of milk cows. The growth of the pathogenic bacteria such as Staphylococcus aureus and Streptococcus sp. No. 12 was markedly inhibited by the chloroform fraction among the extracts, and these antibacterial activities were also supported by MIC test. Water-soluble mucilage was obtained from the root barks with a yield of 36.8~40.2% by dry weight basis. This substance appeared to be a kind of polysaccharide with $61.0{\pm}1.0%$ carbohydrate content, and to have hygroscopic and astringent properties due to its high viscosity and water absorption.
This study was performed to examine the cytotoxicity and antimicrobial activity of synthetic or natural preservative. Fibroblast cells L929 were used for cytotoxicity test and Pseudomonas aeruginosa ATCC27853, Staphylococcus aureus ATCC25923, Escherichia coli ATCC25922, Trichoderma reesei ATCC6967 were used for antibacterial and antifungal activities. Benzalkonium chloride(BAC) as a synthetic preservative and chitosan as a natural preservative were used for this study. Minimum inhibitory concentration(MIC) of BAC was 0.1~0.01% for P. aeruginosa and 0.001~0.0001% for S. aureus and 0.1~0.01% for T. reesei, MIC of chitosan was 2% for P. aeruginosa and 1I % for S. aureus. This study suggest that chitosan might be useful as an eyedrop.
This study aimed to evaluate the antimicrobial activity of 2-phenylethynyl-butyltellurium (PEBT) in Escherichia coli and the relation to its pro-oxidant effect. For this, we carried out the disk diffusion test, minimum inhibitory concentration (MIC) assay, and survival curve analysis. We also measured the level of extracellular reactive oxygen species (ROS), activity of the antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT), and level of non-protein thiols (NPSH). PEBT at 1.28 and 0.128 mg/disk exhibited antimicrobial capability in the disk diffusion test, with an MIC value of 1.92 mg/ml, whereas PEBT at 0.96, 1.92, and 3.84 mg/ml inhibited bacterial growth after a 9-h exposure. PEBT at 3.84, 1.92, and 0.96 mg/ml increased extracellular ROS production, decreased the intracellular NPSH level, and reduced the SOD and CAT activities. Glutathione or ascorbic acid in the medium protected the bacterial cells from the antimicrobial effect of PEBT. In conclusion, PEBT exhibited antimicrobial activity against E. coli, involving the generation of ROS, oxidation of NPSH, and reduction of the antioxidant defenses in the bacterial cells.
