Tschimganidine reduces lipid accumulation through AMPK activation and alleviates high-fat diet-induced metabolic diseases

  • Obesity increases the risk of mortality and morbidity because it results in hypertension, heart disease, and type 2 diabetes. Therefore, there is an urgent need for pharmacotherapeutic drugs to treat obesity. We performed a screening assay using natural products with anti-adipogenic properties in 3T3-L1 cells and determined that tschimganidine, a terpenoid from the Umbelliferae family, inhibited adipogenesis. To evaluate the anti-obesity effects of tschimganidine in vivo. Mice were fed either a normal chow diet (NFD) or a high-fat chow diet (HFD) with or without tschimganidine for 12 weeks. Treatment with tschimganidine decreased lipid accumulation and adipogenesis, accompanied by reduced expression of adipogenesis and lipid accumulation-related factors. Tschimganidine significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased that of AKT. Depletion of AMPK relieved the reduction in lipid accumulation resulting from tschimganidine treatment. Moreover, tschimganidine administration drastically reduced the weight and size of both gonadal white adipose tissue (WAT) and blood glucose levels in high-fat diet-induced obese mice. We suggest that tschimganidine is a potent anti-obesity agent, which impedes adipogenesis and improves glucose homeostasis. Tschimganidine can then be evaluated for clinical application as a therapeutic agent.

식이유도 비만 동물모델에서 마르멜로추출물의 항비만 효능 비교 연구 (A Comparative Study on Anti-Obesity Efficacy of Cydonia oblonga Miller Fruit Extract in Diet-Induced Obesity Animal Models)

  • Objectives: The objective of this study was to explore the anti-obesity effect of Cydonia oblonga Miller fruit extract (COME) and to compare its anti-obesity efficacy with Garcinia cambogia extract (GCE) in diet-induced obese mice. Methods: Five-week-old male C57BL/6 were allocated into four groups: control diet (CD), high-fat diet (HFD), HFD + 400 mg/kg body weight (BW)/day COME (H+C), or HFD + 400 mg/kg BW/day GCE (H+G) groups. COME or GCE was administered once a day by oral gavage for eight weeks. Body weight, body fat percentage, fat weight, and biochemical parameters in serum were measured. The expressions of transcription factors and their target genes in epididymal adipose tissues were analyzed by reverse transcription polymerase chain reaction. Results: COME reduced body weight, weight gain, body fat percentage, total white adipose tissue weight, adipocyte size, and serum levels of insulin and leptin in high-fat diet-induced obese C57BL/6 mice. COME suppressed the mRNA expressions of CCAAT/enhancer binding proteinα, peroxisome proliferator-activated receptorγ, sterol-regulatory element-binding protein-1c, fatty acid synthase, and adipocyte protein 2 and increased carnitine palmitoyl transferase 1 mRNA expression in epidydimal adipose tissues. The anti-obesity efficacy of COME was found to be similar to that of GCE at the same dose. However, COME more effectively decreased adipose tissue weights, epididymal adipocyte size, serum insulin and leptin compared to GCE. Conclusions: These results demonstrated that COME is not toxic and exhibits anti-obesity efficacy at a level similar to that of GCE, suggesting that COME may be applicable as an anti-obesity agent.

Effects of Hangbisan, an Oriental Medicine, on Body Weight Gain in Diet-Induced Obese (DID) rats

  • This study was designed to determine possible weight loss effects of Hangbisan, an oriental medicine, on Sprague-Dawley (SD) rats. SD rats that were fed a high-fat diet for 6 weeks to induce obesity and subsequently fed with a basic diet containing 10%(w/w) Hangbisan or 10%(w/w) cellulose for 8 weeks. The Hangbisan fed rats demonstrated a significantly (p<0.05) reduced weight gain compared to the cellulose fed rats as well as a reduced level of plasma cholesterol and triglycerides and an increased level of (HDL)-cholesterol. These results suggest that dietary Hangbisan has an anti-obesity effect in the high-fat diet-induced obesity (DIG) rat and therefore a potential use as an anti-obesity agent in the application of oriental medicine compounds.

  • Obesity is a physical condition that results from excessive storage of fat in the body. The present study examined the anti-obesity effects of the selected natural medicine, Galla rhois extract (GRE) and solvent fractions on 3T3-L1 preadipocytes and in vivo studies. Here, we show that EtOAc fraction of Galla rhois inhibits the differentiation of the 3T3-L1 preadipocytes induced by differentiated medium in a dose-dependent manner. To investigate the effect of the GRE-EtOAc fraction on obesity in high fat diet-fed C57BL/6 mice, which included a normal diet (ND), high-fat diet (HFD) and HFD+GRE concentration-dependent, were fed to the mice for 6 weeks. The GRE-EtOAc fraction was inhibited the highest adipocyte differentiation in vitro, the GRE supplement significantly decreased body weight and visceral fat mass compared to the HFD group. The total cholesterol and triglyceride levels in the plasma were significantly decreased by GRE supplementation compared with those of the HFD group. Also, we aimed to determine the differentiation inhibition and the modulation of differentiation genes brought about by the Galla rhois in adipocyte. A cDNA microarray-based method was introduced for the high contents screening (HCS) of gene expressions. This technology has revolutionized gene expression studies by providing the means to measure mRNA levels in thousands of genes simultaneously in simple and complex biological samples. 13 genes were founded to be affected in their expression levels by more than 5-fold up-regulation after 4 days treatment with the EtOAc fraction from Galla rhois. Otherwise, 21 genes were founded to be affected in their expression levels by more than 5-fold down-regulation treated with the EtOAc fraction. Therefore, Galla rhois extract may be considered for use in a therapeutic agent to control obesity.

고지방식이(高脂肪食餌)로 유도된 동물모델에서 백수오(白首烏)의 항비만(抗肥滿) 효과 (Anti-obesity effect of Cynanchi Wilfordii Radix on High fat diet-induced obese mice)

  • Objectives : Obesity is a public health concern associated with chronic diseases including hyperlipidemia, diabetes, fatty liver, atherosclerosis and cancer. As several anti-obesity drugs have been limited owing to their side effects, the development of new anti-obesity drugs through herbal medicines has been increasing. Cynanchum Wilfordii Radix (CW) traditionally is consumed for various health benefits including immune enhancing, anti-inflammation and anti-tumor activities. The aim of the present study is to evaluate the effects of CW on High fat diet (HFD)-induced obese mice. Methods : The mice were randomly divided into four groups (n=7). The mice were respectively fed a normal diet (ND), a high-fat diet (HFD), HFD plus CW (50 mg/kg/day), HFD plus CW (100 mg/kg/day). All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : HFD-fed mice showed an increase in the body weight and serum biochemistry parameters levels (total cholesterol and triglycerides) as well as organic tissue weights. However, the administration of CW to obese mice induced a reduction in their body weight, food efficiency ratio, blood biochemistry parameters and weight of liver and fat compared with the HFD fed mice. Additionally, we observed that CW inhibited the lipid accumulation in liver and serum lipid parameter induced by HFD. Conclusions : Taken together, the findings of this study suggest that CW may be a potential agent for use in the treatment of obesity and obesity-related metabolic diseases.

죽엽(竹葉)과 황금(黃芩) 복합물의 항비만 효과 (Synergistic combination effect of anti-obesity in the extracts of Phyllostachys pubescence Mael and Scutellaria baicalensis Georgi)

  • Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and considerable adverse effects affecting tolerability and safety. Therefore, most anti-obesity durgs have been withdrawn. We tried to develop anti-obesity agent by combinations from herbs that are used in food ingredients as well as in traditional medicines. Methods : The 80% (v/v) ethanol extracts from Bamboo (Phyllostachys pubescence) leaf (BL) and Scutellaria baicalensis (SB) and their 1:1 combination (BLSB) was evaluated on high fat diet induced obese mice compared to Omega-3 as a positive control. The mice were divided into six groups (n=5), one group fed a normal diet (ND), and the others fed a high fat diets for eight weeks. Two weeks after starting feeding the diets, the high fat diet groups were orally administered vehicle and Omega-3, BL, SB, and BLSB at dosage of 200 mg/kg/day for six weeks. All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : BLSB group showed significant reductions in body weight gain and fat weights of liver and epididymal adipose tissue compared to BL or SB alone as well as control. Total-cholesterol and LDL-cholesterol levels significantly decreased, and HDL-cholesterol level increased. In liver tissue, macrovesicular steaotisis was remarkably improved and its fat cell size was also significantly decreased. Conclusions : These results suggested that a combination preparation of bamboo leaf and S. baicalensis has anti-obesity effect and have synergistic effect compared to bamboo leaf or S. baicalesis.

조릿대의 잎과 줄기 추출물 분획의 염증 및 비만 억제 효과 비교 (Comparative Study for Anti-inflammatory and Anti-obesity Effect of Fractions from Leaf and Stem of Sasa Borealis)

  • Sasa borealis has long been used in folk remedies and traditional Korean medicine for analgesic, sedative and anticarcinogenic purposes. To excavate novel natural compounds for treating obesity, we separated leaf and stem and fractionated. We screened phenolic contents, anti-oxidative and anti-inflammatory properties with the fractions. Dichloromethane (DCM) and ethyl acetate (EA) fraction from both leaf and stem exhibited high anti-oxidative properties demonstrated by DPPH, ABTS assay in accordance with their phenolic contents. In addition, DCM and EA fraction from leaf inhibited nitric oxide (NO) production as well as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in Raw 264.7 cell. Finally, DCM and EA fraction from both leaf and stem significantly inhibited 3T3-L1 preadipocyte differentiation and decreased intracellular lipid level. These results suggest that DCM and EA fraction from Sasa borealis may contain therapeutic agent for obesity by attenuating inflammation and oxidative stress.

Bioconversion Products of Whey by Lactic Acid Bacteria Exert Anti-Adipogenic Effect

  • Microbial bioconversion using lactic acid bacteria (LAB) provides several human health benefits. Although whey and whey-derived bioactive compounds can contribute to an improvement in human health, the potential anti-obesity effect of whey bioconversion by LAB has not been well studied. This study aimed to investigate whether bioconversion of whey by Pediococcus pentosaceus KI31 and Lactobacillus sakei KI36 (KI31-W and KI36-W, respectively) inhibits 3T3-L1 preadipocyte differentiation. Both KI31-W and KI36-W reduced intracellular lipid accumulation significantly, without decreasing 3T3-L1 preadipocyte proliferation. In addition, obesity-related transcription factor (peroxisome proliferator-activated receptor γ) and genes (adipocyte fatty acid-binding protein and lipoprotein lipase) were down-regulated significantly in 3T3-L1 cells in the presence of KI31-W and KI36-W. Collectively, these results suggest that bioconversion of whey by LAB exhibits anti-adipogenic activity and may be applied as a therapeutic agent for obesity.

  • 본 연구에서 PTS는 지방전구세포에서 PPARγ, C/EBPα 및 FABP4의 발현 억제를 통해 지방전구세포의 adipogenesis를 억제하고, AMPK 및 HSL의 활성화, ATGL의 발현증가, 그리고 perilipin-1의 발현감소를 통해 지방세포의 지질을 분해하여 지방세포 내 과도한 지질형성을 억제하는 것으로 판단된다. 이러한 결과들은 PTS는 비만 예방 및 치료를 위핸잠재적 식의약 소재로의 개발 및 활용이 가능할 것으로 사료된다.

마황을 포함하지 않은 한약처방 단일중재에 의한 단순비만 치료 : 증례보고 (Simple Obesity Treatment by Single Intervention of Herbal Medicines without Ephedra Herba: A Case Report)

  • Objective: Glucagon-like peptide 1 (GLP-1), one of the gut peptide hormones, has an action to induce satiety, and its effect as an anti-obesity agent is known. Recently, it has been reported that many herbal medicines have an anti-diabetic effect through inhibition of DPP-4 enzyme and inducing of GLP-1 secretion. It is therefore suggested that GLP-1 may be effective for the treatment of obesity. In this study, we report a case of male obese patients treated with herbal medicine as a GLP-1 secretagogue. Methods: In this study, the patient took a fixed prescription of herbal medicine for 10 weeks and recorded his weight at each visit. Results: This prescription produced significant weight loss (BMI loss>5%). In the follow-up period after two weeks, the trend of weight loss was observed continuously. Conclusion: This prescription can be an alternative to ephedra herba-based obesity treatment.
