Antianaphylactic Effect of Sinpo-tang by Anal Therapy (肛腸療法에 의한 辛蒲湯의 抗아나필락시 效果)

  • Objective : Anal Therapy is another way of taking medicine and applies to each field of clinical treatment extensively. Sinpo-tang(SPT) has been used for the treatment of a allergic rhinitis. In this study, the auther investigated the anti anaphylactic action of Sinpo-tang by anal therapy was investigated on cutaneous allergic reaction models. Methods : Results : 1. Sinpo-tang (0,001-0.1 g/kg) dose-dependently inhibited the compound 48/80-induced ear swelling response in mice. Inhibitory effect of Sinpo-tang was significant (P < 0.05) at the doses of 0.01, 0.1 g/kg. 2. Sinpo-tang (0.001-0.1 g/kg) inhibited the cutaneous allergic reaction activated by anti-dinitrophenyl IgE in rats. Of special note, Sinpo-tang (0.1 g/kg) inhibited the cutaneous allergic reaction by $68\%$. 3. This inhibitory effect of Sinpo-tang was confirmed by observation of alcian blue/nuclear fast red stained-mast cells in the cutaneous tissue. 4. Sinpo-tang (0.01-1 g/L) dose-dependently inhibited the compound 48/80-induced histamine release from the peritoneal mast cells. Conclusions :These results indicate that anal therapy of Sinpo-tang may be beneficial in the treatment of mast cell-mediated anaphylaxis by inhibition of histamine release from mast cells in vivo and in vitro.

Rubus croceacanthus Leveille inhibits mast cell-mediated immediate-type allergic reactions and tumor necrosis factor-$\alpha$ secretion

  • We studied the effect of Rubus croceacanthus Leveille (RCL) on mast cell-mediated anaphylactic reactions. RCL inhibited compound 48/80-induced systemic anaphylactic shock. When RCL was given at concentrations ranging from 0.01 to 1 mg/$m\ell$, the histamine release from rat peritoneal mast cells induced by compound 48/80 was reduced in a dose-dependent manner. RCL also inhibited passive cutaneous anaphylaxis activated by anti-dinitrophenyl IgE. In addition, RCL inhibited phorbol 12-myristate 13-acetate and A23187-induced tumor necrosis factor-${\alpha}$ secretion from human mast cell line HMC-1 cells. These results indicate that RCL may possess a strong anti-anaphylactic activity.

  • Effects of Panax ginseng on allergic reactions were studied uslng various in vivo and in vitro experimental models such as 48-hr passive cutaneous anaphylaxis, mediators-induced skin reactions, histamine release from rat peritoneal mast cells, hexosaminidase release from RBL-2H3 cells, and lipoxygenase assay . In all of anti-allergic experiments we conducted, ginseng components (50% ethanol extract or ginseng total saponin or ginsenosides) extracted from Korean red ginseng, did not show significant anti-allergic actions. In 48-hr passive cutaneous anaphylaxis and mediators-induced skin reactions, 50% ethanol extract did not suppress hypersensitivity reactions. Total saponin, 50% ethanol extract, and 8 major ginsenosides did not show inhibitory effects on lipoxygeanse activity. Ginseng total saponin did not inhibit histamine release from rat peritoneal mast cells. All of the ginseng components mentioned above were also tested on RBL-2H3 cells, but none of them inhibited hexosaminidase release from this cell line. These results suggest that Panax ginseng does not have effects on allergic reactions at the level of 50% ethanol extract or total saponin used. All of 8 major saponin components tested ($Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_2$), did not inhibit lipoxygenase activity and degranulation events.

The Effect of Caffeic Acid on Wound Healing in Skin-incised Mice

  • This study was carried out to investigate the wound healing effect of caffeic acid in skin-incised mice. Caffeic acid showed significant effects on anti-inflammatory activity and wound healing, such as myeloperoxidase activity, lipid peroxidation, phospholipase $A_2$ activity and collagen-like polymer synthesis, in incised-wound tissue. On the other hand, it significantly stimulated collagen-like polymer synthesis in NIH 3T3 fibroblast cells, while inhibited both silica-induced reactive oxygen species generation and melittin-induced arachidonic acid release and $PGE_2$ production in Raw 264.7 cells, and histamine release in RBL 2H3 cells stimulated by melittin or arachidonic acid. Therefore, caffeic acid appears to have a potent antioxidant and anti-inflammatory effect in cell culture system, which may be related to wound healing in skin-incised mice.

  • Objective We investigated the effect of Saururus chinensis (Lour.) Baill. aquacupuncture (SCB) on anaphylaxis in mice. Methods : We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. Results : SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the serum histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability. In addition, SCB showed a significant inhibitory effect on anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. Conclusion : These results indicated that SCB inhibits anaphylatic reaction.

  • Experimental studies were done to research effects of Leejungtang and Leejungtanggamibang on the anti-allergic effects. We studied the vacular permeability response induced by histamine and serotonin, homologous PCA provoked by IgE-like antibody against EWA, contact dermatitis induced by picryl chloride and delayed type hypersensitivity response to SRBC. The results were obtained as follows: 1. In effects of the Leejungtang and Leejungtanggamibang on the vascular permeability responses to intradermal serotonin, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 2, In effects of the Leejungtang and Leejungtanggamibang on the vascular permeability responses to intradermal histamine, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 3. In the homologous PCA provoked by IgE-like antibody against white albumin, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 4. In the delayed type hypersensitivity responses to picryl chloride, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 5. In the delayed type hypersensitivity responses to sheep red blood cell the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect.

Studies on the Inhibitory effect of anaphylactic reaction by Shinisan (辛夷散에 의한 아나필락시 反應의 抑制에 관한 硏究)

  • Shinisan has been used for treatment of allergic disease in Oriental Medicine. However, its effect in experimental models remains unknown. The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. The present report describes an inhibitory effect of Shinisan on mast cell-mediated immediate type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal ($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Shinisan inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80. Shinisan inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Shinisan also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Shinisan had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Shinisan inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

Effects of Mix-1 on Anti-CD40 Antibody and Recombinant IL4- Induced Cytokine Production and Immunoglobulin E in Highly Purified Mouse B Cells

  • In the oriental medicine, a mixture of herbs has been commonly used as important components to control allergic and inflammatory diseases. In the present study, we prepared a mixture of Dictamni Radicis Cortex(Baiksunpee), Houttuyniae Herba(Uhsungcho), and Aurantii Immaturus Fructus(Jisil) to examine its anti-allergic effects in activated mouse splenic cells and found that Mix-1 is involved in regulating levels of B cell activating factors (CD23 and CD11a), IL-1β, IL-6, IL-10, TNF-α, and 1gE as well as HRF expression. It was observed that Mix-1 did not have cytotoxic effects on mLFC. Mix-1 showed inhibition of CD23 and CD11 alpaha expression in mouse B cells, and also decreased the production of IL-6, TNF-α, and 1gE. Both RT-PCR and ELISA analyses indicated that IL-6 and TNF alpha production were regulated at the gene expression level. In contrast, IL-10 mRNA and protein levels were increased in activated B cells by Mix-1 treatment. We also found that Mix-1 inhibited B cell proliferation and inhibited histamine releasing factor(HRF) expression, suggesting its inhibitory effect on histamine secretion. These data indicated that Mix-1 has an anti-allergic effect in activated macrophages and further suggest the possible application of Mix-1 as a therapeutic agent for the treatment of allergy-related diseases.

  • Objective : This study was carried out for the purpose of proving the effect of anti-allergic efficacy on B cells and the mast cells of the BALB/C mouse by the abstraction from a Moutan cortex. Methods & Results : In order to know what the effect of an abstraction from Moutan cortex and about the expression of CD23 and IgE, IC-2 cell (mouse mast precursor cells that was dependent on IL-3), it was necessary to be activated. We then analyzed it from the flow of cytometry on the increase and the divorce of the B cells activated by anti-CD40. In order to know what the effect of it was on the organization of cytokine gene expression from the increase and divorce of the B cells and allergic acting by Moutan cortex, we found it necessary to examine the IC-2 cells and B cells. At the same time, as we examined the histamine release of IC-2 cells by ELISA method, we also examined the effect of Moutan cortex on the increase and divorce of the B cells by 3H-thymidine uptake method. We then analyzed the release of IL-4, IgE and histamine. Conclusions : As a results, Moutan Cortex promoted blood supply by extending the blood vessel of nasal mucous, which was contracted by the hypertrophied nasal mucous.
