The aim of this study was to investigate the anti-inflammatory and anti-oxidant activity of Ligularia fischeri leaf extract on adjuvant induced arthritis in experimental mice. The oral administration of the L. fischeri leaf extract (LF), at doses of 100 and 200 mg/kg body weight once a day for 3 weeks, significantly reduced hindpaw swelling and the production of inflammatory cytokines (tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$, and IL-6). Treatment with LF (100 mg/kg) also decreased the serum levels of triglyceride and low density lipoprotein(LDL)-cholesterol, and increased high density lipoprotein(HDL)-cholesterol contents compared with those of a control group. The induction of arthritis significantly increased oxidized proteins such as protein carbonyl, advanced oxidation protein products, and advanced glycation end-products in the lung, heart, and brain. Treatment with LF for 3 weeks reduced the levels of oxidized proteins. These results suggest that L. fischeri extract might be beneficial in the treatment of chronic inflammatory disorders.
Objectives : Corni Fructus (CF) is traditional herbal medicine used on polyuria, low back pain, and tinnitus. This study aimed to evaluate inhibits skin wrinkle formation effect of CF. Methods : To evaluate the produce inhibition effect of CF, SD-rats were distributed into four groups; normal rats (Nor), AGEs (advenced glycation end product)-induced rats (Con), AGEs-induced rats treated with 100mg/kg CF (CF). To induce AGEs, streptozotocin (50mg/kg) was administered intraperitoneally and after 3 days oral administrated 100mM methyl glyoxal for 3 weeks. Results : The oral administration of CF suppressed the reactive oxygen specis (ROS) in serum. The AGEs in skin tissues was significantly reduced through treatment of CF. Furthermore, the expressions of AGEs related proteins such as polyclonal anti-$N^e$-(carboxymethyl)lysine (CML), anti-$N^e$-(carboxyethyl)lysine (CEL), AGE receptors (RAGE) were decreased in CF treated group compared with the control group in skin tissues. Inflammation-related proteins such as Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB), monocyte chemoattractant protein-1 (MCP-1), interleukin-6 (IL-6) reduced in CF treatment group than control group. AGE-induced rats exhibited that the significant decreased collagen however, CF treatment (100mg/kg of body weight) up regulated collagen by improved the expression levels of skin fibril-related genes such as Matrix metalloproteinase (MMP-1). Conclusion : Taken together, our study suggests that CF regulates ROS to prevent accumulation of AGEs and inhibits skin wrinkles. Our finding indicate that CF may be an effective agent for inhibits AGEs formation, and improved skin wrinkle.

Optimization of carnosine and anserine extraction from chicken breast was performed using response surface methodology (RSM) to obtain the maximized physiological activities for anti-glycation and anti-oxidation. The optimum extraction conditions were water extraction for 1.6 h in the case of the 20-wk laying hen muscle and water extraction for 2.12 h in the case of 90-wk laying hen muscle. Higher carnosine and anserine contents were measured in the 20-wk laying hen muscle, along with higher physiological activities, which increased in direct proportion with the dipeptide contents. The extracts prepared from the 20-wk laying hen under optimum conditions showed 57% inhibition of advanced glycated end-product formation, 64% inhibition of lipid peroxidation, and 61% of DPPH radical scavenging effects. On the other hand, 52% inhibition of AGE formation, 62% inhibition of lipid peroxidation, and 53% of DPPH radical scavenging effect were demonstrated within the 90-wk laying hen. In addition, the ratio of carnosine was a key indicator for the physiological activities of the extracts.
혈당강하 효과가 있다고 알려진 해당근 추출물을 이용하여 AGEs 생성 저해에 따른 초기 동맥경화증에 대한 효과에 대하여 검토하였다. 먼저 해당근 추출물이 AGEs에 의하여 생성이 촉진된 ROS를 제거하는 효과가 있는지를 확인하기 위하여 항산화 활성을 측정하였다. 총 플라보노이드 함량을 측정한 결과 해당근의 메탄올 추출물은 $128.1{\pm}2.0\;{\mu}g$ quercetin equivalents(QE)/mg DM,총 폴리페놀 함량은 $345.2{\pm}5.7\;{\mu}g$ gallic acid equivalents(GAE)/mg DM으로 나타났다. 환원력 측정을 위한 FRAP assay는 $2.19{\pm}0.1\;mM$$FeSO_4{\cdot}7H_2O$/mg DM으로 $25.5{\pm}0.3$$FeSO_4{\cdot}7H_2O$/mg DM 인 ascorbic acid에 비해 약 9% 정도의 환원력을 가지고 있었으며, 전자공여능의 경우 $DPPHSC_{50}$값이 $34.2{\pm}0.1\;{\mu}g$ DM/mL로 대조군인 ascorbic acid의 $6.8{\pm}0.1\;{\mu}g$ DM/mL과 비교하여 약 20% 정도의 높은 항산화력을 갖고 있었다. 또한, HPLC 분석을 통해 해당근 생물 100 g을 기준으로 240 mg의 EGCG와 50 mg의 kaempferol을 함유하는 것으로 나타났다. AGEs에 의해 유도된 HUVEC의 ROS 생성 저해 효과는 해당근 추출물 100, $500\;{\mu}g/mL$ 처리 시 각각 63, 77%로 나타났다. 또한, monocyte adherent assay에서 해당근 추출물은 $100\;{\mu}g/mL$ 처리시 정상을 0%로 하였을 때 AGEs에 의해 유도된 단핵구 부착을 33%, $500\;{\mu}g/mL$ 처리시 약 75% 정도로 농도 의존적으로 부착을 저해하는 것을 관찰할 수 있었으며, 유도되지 않은 HUVEC의 저항은 $113{\Omega}{\cdot}cm^2$ 에서 glycer-AGEs 처리 시 $88{\Omega}{\cdot}cm^2$로 낮아진 것을 해당근 추출물을 $100\;{\mu}g/mL$ 처리 시 약 $102{\Omega}{\cdot}cm^2$, $500\;{\mu}g/mL$ 처리시 약 $106{\Omega}{\cdot}cm^2$로 저항의 감소를 억제하는 것을 관찰할 수 있었다.
장미과에 속한 낙엽교목인 산사나무의 열매인 산사의 항산화 및 항당뇨 관련 활성을 검토하고, 추출 용매에 따른 차이를 비교하기 위하여 총 페놀 함량, DPPH 라디칼 소거능, ABTS 라디칼 소거능, 최종당화산물 억제능, ${\alpha}$-glucosidase 억제능, aldose reductase 억제능을 측정하였다. 수율은 70% 에탄올 추출물이 33.16%로 가장 높았고, 50% 에탄올 추출물이 27.79%로 그 다음 높았으며, 물, 30% 에탄올, 100% 에탄올은 각각 21.71%, 21.88%, 19.03%를 나타내었다. 총 페놀함량은 50% 에탄올 추출물이 $41.83{\pm}0.07mg$ GAE/g으로 가장 높았고, DPPH와 ABTS 라디칼 소거능도 50% 에탄올 추출물이 각각 $80.79{\pm}0.83%$, $34.92{\pm}0.97%$로 가장 높았다. 최종 당화산물 억제능과 ${\alpha}$-glucosidase 억제능도 50% 에탄올 추출물이 $27.09{\pm}2.27%$와 $58.87{\pm}0.70%$로 가장 높게 나타난 반면에 AR은 물 추출물이 $30.68{\pm}1.41%$로 가장 높게 나타나면서 상이한 경향을 보였다. 이상의 결과를 종합해 보면, 알코올 농도에 따른 산사의 추출물 중 50% 에탄올 추출물이 천연 항산화제로서 가치가 있으며, 당뇨병 예방을 위한 천연물로서의 가능성도 보였으나 당뇨병 합병증 치료에 대한 효과는 미흡한 것으로 나타났다. 향후 좀 더 다양한 fraction에서의 연구와 in vivo 시험 등을 통한 추가적인 연구가 필요할 것으로 사료된다.
감마선 조사로부터 당뇨합병증에 효과적인 소재를 개발하기 위하여 본 연구를 수행하였으며, mangiferin의 감마선 조사산물이 최종당화산물 생성 저해에 우수한 활성이 나타나 효능성분의 동정을 위하여 ODS gel을 활용한 column chromatography를 수행하여 3종의 mangiferin 유도체 화합물을 분리하였고, 각 화합물의 화학구조는 NMR 스펙트럼 데이터 해석을 통하여 mangiferdiol(1), mangferinol(2) 및 isomangiferinol(3)로 동정하였다. 이들 화합물에 대해 최종당화산물 생성 저해 활성을 평가한 결과 mangferinol(2)이 가장 강한 $5.6{\pm}0.8{\mu}M$의 $IC_{50}$ 값을 나타내었고, isomangiferinol(3)가 $7.6{\pm}0.9{\mu}M$의 $IC_{50}$ 값을 나타내었다. 또한, mangiferdiol(1)이 $12.1{\pm}1.3{\mu}M$의 $IC_{50}$ 값을 나타냈으며, 이들 활성은 mangiferin의 ketone기가 소실되고 새롭게 생성된 methyl기와 hydroxymethyl기의 결합 양상에 따른 화합물의 구조에 따라 다름이 시사되었다. 향후 이들 활성물질의 활성 기작에 대한 추가적인 연구가 필요하며 기존 유효 성분보다 우수한 최종당화산물 생성 저해 활성을 가지는 새로운 선도화합물 발굴을 위한 기초자료로 활용할 수 있으리라 판단된다.
The endocrinology of type 2 diabetes (T2D) and its predisposing factors have been studied extensively while its skeletal effects have received negligible research despite this being a global disease. The cellular and molecular association between proximal humeral fractures and T2D has not been fully elucidated. We aimed to study bone cell quantities and immunolabel osteogenic and antiosteogenic cytokines. The study used 12-week-old rats (23 males) consisting of 8 Sprague Dawley (SD) and 15 Zucker Diabetic Sprague Dawley (ZDSD). Weekly mass measurements were taken while fasting blood glucose levels were recorded every 2 weeks with oral glucose tolerance tests conducted once every 4 weeks. Upon termination at the age of 28 weeks, humeri were fixed in 10% buffered formalin, prior to decalcification in ethylenediaminetetraacetic acid. The bone samples were then processed in ascending grades of alcohol using an automatic processor before embedding in paraffin wax. Sections were cut at 5 ㎛ thickness in a series for Haematoxylin and Eosin stain, and immunohistochemistry was performed with the anti-tartrate-resistant acid phosphatase (TRAP), anti-alkaline phosphatase (ALP), anti-bone morphogenetic protein 3 (BMP3), anti-transforming growth factor beta 1 (TGFβ1), anti-aged glycation end product (AGE) antibodies in the sequence. ZDSD rats had more adipocytes, BMP3 and AGEs expression with higher numbers of TRAP positive osteocytes and fewer ALP cells although no differences were found in TGFβ1 immunopositivity. We also found that T2D increases the number of AGEs immuno-positive cells, as well as its extracellular expression, thus providing a conducive environment for the interaction of the osteogenic cytokine and its antagonist to suppress osteoblastogenesis. ZDSD groups had higher adipocyte numbers therefore increased marrow adiposity in T2D.
Objectives : Collagen decrease of Skin appears through various path ways. One of causes may be the Advanced glycation endproducts (AGEs) that combine formation of glucose and protein. The aim of this study was to explore the prevent wrinkle formation of Paeoniae radix (PR) and heated Paeoniae radix (HPR) via AGEs path way. Methods : AGEs formation inhibitory activities of PR and HPR measured using bovine serum albumin, glucose, and fructose. To evaluate the protective effects of PR and HPR in diabetic rats induced with streptozotocin (STZ) and methyl glyoxal (MGO), SD rat were distributed into four groups. Normal rats (Nor), AGEs-induced rats (Con), AGEs-induced rats treated with 100 mg/kg PR(PR), AGEs-induced rats treated with 100 mg/kg HPR (HPR). To induce AGEs, streptozotocin (50 mg/kg) was administered intraperitoneally and after 3 days administrated 100mM methyl glyoxal for 3 weeks. Results : The oral administration of HPR inhibited AGEs in skin tissues compared with PR. The increased reactive oxygen species (ROS) levels in the serum were diminished by HPR treatment. The analyses of kidney and skin tissues proteins indicated that HPR treatment effectively reduced AGEs related protein levels as compared to that by PR treatment. Also, HPR decreased anti-oxidant related protein levels in skin tissues such as catalase, glutathione peroxidase. Moreover, it inhibited the reduction of COL1A2 by decreasing MMP-1. Conclusion : Based on these results, it was suggested that PR and HPR could have Improving effects on wrinkle formation. These evidences provide useful information for the development wrinkle formation treated agent.

Apolipoprotein A-I and A-IV are protein constituents of high-density lipoproteins although their functional difference in lipoprotein metabolism is still unclear. To compare anti-atherogenic properties between apoA-I and apoA-4, we characterized both proteins in lipid-free and lipidbound state. In lipid-free state, apoA4 showed two distinct bands, around 78 and $67{\AA}$ on native gel electrophoresis, while apoA-I showed scattered band pattern less than $71{\AA}$. In reconstituted HDL (rHDL) state, apoA-4 showed three major bands around $101{\AA}$ and $113{\AA}$, while apoA-I-rHDL showed almost single band around $98{\AA}$ size. Lipid-free apoA-I showed 2.9-fold higher phospholipid binding ability than apoA-4. In lipid-free state, $BS_3$-crosslinking revealed that apoA-4 showed less multimerization tendency upto dimer, while apoA-I showed pentamerization. In rHDL state (95:1), apoA-4 was existed as dimer as like as apoA-I. With higher phospholipid content (255:1), five apoA-I and three apoA-4 were required to the bigger rHDL formation. Regardless of particle size, apoA-I-rHDL showed superior LCAT activation ability than apoA-4-rHDL. Uptake of acetylated LDL was inhibited by apoA-I in both lipid-free and lipid-bound state, while apoA-4 inhibited it only lipid-free state. ApoA-4 showed less anti-atherogenic activity with more sensitivity to glycation. In conclusion, apoA-4 showed inferior physiological functions in lipid-bound state, compared with those of apoA-I, to induce more pro-atherosclerotic properties.
본 연구는 BRECs에서 AGEs로 유도된 세포의 이동 및 침윤에 있어서 AuNP의 역할에 관한 연구이다. 소 망막으로부터 내피세포를 분리하고, 세포 생존율은 MTT assay로 확인하였다. Wound migration assay는 BRECs의 이동력을 확인하기 위해 수행하였다. Tube formation은 on-gel system을 통해 확인하였다. AuNP의 apoptosis 유도는 caspase-3 assay를 통해 확인하였다. AGE-BSA은 세포증식 및 이동에 있어서 증가함을 보여주었다. 또한 AuNP는 AGE-BSA 존재 유무에 상관없이 세포의 증식, 이동, 신생혈관 형성을 억제하였고, caspase-3을 통해 apoptosis를 유도하였다. 이러한 결과, AuNP는 AGE로 유도된 신생혈관 형성 및 세포의 이동성을 억제하는 약재제로서, 당뇨성 합병증에 있어서 잠재적으로 중요한 분자가 될 것이다.
