Eucommia ulmoides Extract Stimulates Glucose Uptake through PI 3-kinase Mediated Pathway in L6 Rat Skeletal Muscle Cells

  • Eucommia ulmoides (Duchung) is commonly used for treatment of diabetes in Korean traditional medicine. However, the exact mechanism of its anti-diabetic effect has not yet been fully elucidated. In this study, the effect of E. ulmoides extract on glucose uptake was investigated in L6 rat skeletal muscle cells. E. ulmoides extract stimulated the activity of phosphatidylinositol (PI) 3-kinase that is a major regulatory molecule in glucose uptake pathway. Protein kinase B (PKB) and protein kinase C-${\xi}$ (PKC-${\xi}$), downstream mediators of PI 3-kinase, were also activated by E. ulmoides extract. We assessed the activity of AMP-activated protein kinase (AMPK), another regulatory molecule in glucose uptake pathway. Phosphorylation level of AMPK did not change with treatment of E. ulmoides extract. Phosphorylations of p38 mitogen activated protein kinase (p38 MAPK) and acetyl-CoA carboxylase (ACC), downstream mediators of AMPK, were not significantly different. Taken together, our results suggest that E. ulmoides may stimulate glucose uptake through PI 3-kinase but not AMPK in L6 skeletal muscle cells.

홍삼.가시오가피.동충하초 혼합수엑스의 항당뇨작용 (Anti-diabetic Effects of Mixed Water Extract from Ginseng Radix Rubra, Acanthopanacis Cortex, and Cordyceps)

  • The effect of water extract composed of panax ginseng radix rubra, acanthopanacis cortex, and cordyceps (PAC) on diabetic animal models were investigated in two different diabetic animal models. FAC water extract significantly reduced the plasma glucose levels on day 30 as compared with the diabetic control group in $KKA^Y$ obese, hyperglycemic and hyperglycemic and hyperinsulinemic mice, and also reduced the plasma glucose levels as well as total cholesterol in multiple low dose (MLD) strep tozotocin-induced diabetic SD rats. PAC water extract also showed an inhibitory effect on reduction of body weight and on development of MLD STZ-induced diabetic state. Elevated kidney hypertrophy, which is a characteristic feature shown in early stage of diabetic nephropathy and calculated as the ratio of kidney mass (g) relative to the body weight (g), was also markedly improved in PAC water extract- treated group as compared to the diabetic control group. Taken together, these data suggest that PAC water extract may have a potential as a antidiabetic agent in type 2 diabetes mellitus.

도라지 종자 추출물의 처리가 제2형 당뇨 db/db 마우스의 혈당개선에 미치는 효과 (Improvement of blood glucose control in type 2 diabetic db/db mice using Platycodon grandiflorum seed extract)

  • PGSE의 혈당조절 효과를 평가하기 위하여 제2형 당뇨 동물모델을 이용하여 8주간의 동물실험을 진행한 결과, 고농도 PGSE(600 mg/kg)의 투여는 경구 포도당 내성 및 혈당 수준을 유의적으로 감소시켰으며(p<0.05), 당화혈색소도 유의적으로 낮은 수준을 유지시켰다(p<0.05). 또한, 혈청 인슐린과 렙틴 농도 역시 대조군과 비교하여 PGSE 고농도 처리군에서 유의적으로 감소하였다(p<0.05). PGSE 투여는 db/db 마우스의 골격근에서 인슐린 의존적 세포신호전달경로를 유의적으로 활성화시켰으며, AMPK 인산화를 촉진시키고, 골격근내 포도당 흡수를 위한 GLUT4의 세포막으로의 전이를 대조군 대비 약 1.7배 증가시켰다. 이러한 결과를 근거로 할 때 PGSE는 항 당뇨병 치료제로서의 잠재적 가능성을 가진 것으로 판단된다.

Inotodiol Inhabits Proliferation and Induces Apoptosis through Modulating Expression of cyclinE, p27, bcl-2, and bax in Human Cervical Cancer HeLa Cells

  • Inonotus obliquus is a medicinal mushroom that has been used as an effective agent to treat various diseases such as diabetes, tuberculosis and cancer. Inotodiol, an included triterpenoid shows significant anti-tumor effect. However, the mechanisms have not been well documented. In this study, we aimed to explore the effect of inotodiol on proliferation and apoptosis in human cervical cancer HeLa cells and investigated the underlying molecular mechanisms. HeLa cells were treated with different concentrations of inotodiol. The MTT assay was used to evaluate cell proliferating ability, flow cytometry (FCM) was employed for cell cycle analysis and cell apoptosis, while expression of cyclinE, p27, bcl-2 and bax was detected by immunocytochemistry. Proliferation of HeLa cells was inhibited by inotodiolin a dose-dependent manner at 24h (r=0.9999, p<0.01). A sub-$G_1$ peak (apoptotic cells) of HeLa cells was detected after treatment and the apoptosis rate with the concentration and longer incubation time (r=1.0, p<0.01), while the percentage of cells in S phase and $G_2$/M phase decreased significantly. Immunocytochemistry assay showed that the expression of cyclin E and bcl-2 in the treated cells significantly decreased, while the expression of p27 and bax obviously increased, compared with the control group (p<0.05). The results of our research indicate that inotodiol isolated from Inonotus obliquus inhibited the proliferation of HeLa cells and induced apoptosis in vitro. The mechanisms may be related to promoting apoptosis through increasing the expression of bax and cutting bcl-2 and affecting the cell cycle by down-regulation the expression of cyclin E and up-regulation of p27. The results further indicate the potential value of inotodiol for treatment of human cervical cancer.

쑥부쟁이 에탄올 추출물의 급성췌장염 보호 효과 (Protective Effects of Ethanol Extract from Aster Yomena on Acute Pancreatitis)

  • Aster Yomena (AY) has been used in traditional medicine to treat diseases such as obesity, hyperlipidemia, atherosclerosis, diabetes and osteoarthritis. However, protective effect of AY on acute pancreatitis (AP) has not been reported. The present study examined the anti-inflammatory effects of an ethanol extract of AY on cerulein-induced AP. AP was induced in mice by intraperitoneally injecting cerulein ($50{\mu}g/kg$) hourly for 6 times. 70% ethanol extract of AY (0.1, 0.2, and 0.5 g/kg) was orally administered for 1 week before acute pancreatitis induction. The mouse was killed at 6 hours after the final cerulein injection. The pancreas and lung were rapidly removed for histological examination and myeloperoxidase (MPO) assay. Blood samples were taken to determine serum amylase and lipase activity. In addition real-time reverse transcription-polymerase chain reaction (RT-PCR) was also performed to investigate mRNA expression of proinflammatory cytokines such as $TNF-{\alpha}$. $IL-1{\beta}$, and IL-6. Administration of AY significantly ameliorated pancreatic weight to body weight ratio, histological damages and MPO activity during AP. In addition, AY inhibited the serum amylase and lipase activity during AP. Also, mRNA expression of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 were inhibited by AY against AP. Our results revealed that pre-treatment of AY reduces the severity of cerulein-induced AP. Therefore, AY may have a protective effect drug against AP.

Effect of fermented sarco oyster extract on age induced sarcopenia muscle repair by modulating regulatory T cells

  • Sarcopenia is an age-related, progressive skeletal muscle disorder involving the loss of muscle mass and strength. Previous studies have shown that γ-aminobutyric acid (GABA) from fermented oysters aids in regulatory T cells (Tregs) cell expansion and function by enhancing autophagy, and concomitantly mediate muscle regeneration by modulating muscle inflammation and satellite cell function. The fermentation process of oysters not only increases the GABA content but also enhances the content of branched amino acids and free amino acids that aid the level of protein absorption and muscle strength, mass, and repair. In this study, the effect of GABA-enriched fermented sarco oyster extract (FSO) on reduced muscle mass and functions via Treg modulation and enhanced autophagy in aged mice was investigated. Results showed that FSO enhanced the expression of autophagy markers (autophagy-related gene 5 [ATG5] and GABA receptor-associated protein [GABARAP]), forkhead box protein 3 (FoxP3) expression, and levels of anti-inflammatory cytokines (interleukin [IL]-10 and transforming growth factor [TGF]-β) secreted by Tregs while reducing pro-inflammatory cytokine levels (IL-17A and interferon [IFN]-γ). Furthermore, FSO increased the expression of IL-33 and its receptor IL-1 receptor-like 1 (ST2); well-known signaling pathways that increase amphiregulin (Areg) secretion and expression of myogenesis markers (myogenic factor 5, myoblast determination protein 1, and myogenin). Muscle mass and function were also enhanced via FSO. Overall, the current study suggests that FSO increased autophagy, which enhanced Treg accumulation and function, decreased muscle inflammation, and increased satellite cell function for muscle regeneration and therefore could decrease the loss of muscle mass and function with aging.

Anthocyanin의 Delphinidin이 MDA-MB-231 유방암세포에 미치는 영향 (Effects of Delphinidin in Anthocyanin on MDA-MB-231 Breast Cancer Cells)

  • 본 연구에서는 블루베리에 포함된 anthocyanin 중 delphinidin이 인간 유래의 MDA-MB-231 유방암세포의 성장을 억제시키고 apoptosis를 유발하는지 살펴보고 in vivo 실험에서도 항암효과가 나타나는지 확인하였다. 그 결과 cell viability를 보기 위한 MTT assay에서는 농도 의존적으로 암세포의 성장을 억제시켰으며, apoptosis의 확인을 위한 DAPI stain에서 농도 의존적으로 chromatin condensation이 유의적으로 증가하는 것을 확인하였다. 또한 western blot에서 암 억제 유전자인 p53 단백질이 증가하였고, anti-apoptotic 분자인 Bcl-2 단백질과 p-$GSK3{\beta}$ 단백질은 감소하였다. In vivo 실험에서는 대조군과 비교해 10 mg/kg delphinidin을 투여한 군에서 종양의 크기가 감소하였으며, TUNEL assay를 통해 apoptosis 세포 또한 통계학적으로 유의적인 증가가 관찰되어 종양 억제 효과를 확인하였다. 이상의 결과들로 볼 때, MDA-MB-231 유방암세포에서 delphinidin은 암세포의 증식을 억제시키고, apoptosis를 유발시키는 효과를 보이므로 암 예방제나 치료제로 개발될 수 있을 것으로 사료된다.

마(Dioscorea batatas DECENE) 점질물이 Alloxan 유발 당뇨 마우스의 혈당 및 지질 성분에 미치는 영향 (Effects of Mucilage from Yam (Dioscorea batatas DECENE) on Blood Glucose and Lipid Composition in Alloxan-induced Diabetic Mice)

  • 한국산 마(Dioscorea batatas DECENE, Yam)에서 점질물(mucilage)의 fraction 1, 2를 분획하여 6주령의 ICR 마우스에게 당 부하 검사(glucose tolerance test)를 실시한 결과 마 점질물 fraction 1이 마 점질물 fraction 2에 비해서 더 나은 효과를 나타내었기 때문에 마 점질물 fraction 1을 시료로 채택하여 6주령의 ICR 마우스에게 Alloxan으로 당뇨를 유발시킨 후 10일간 경구 투여를 실시하고 식이 섭취량 및 체중의 변화를 관찰하고 혈액중의 포도당 농도와 혈청과 간장 중의 지질 성분과 각종 장기의 무게 변화를 측정한 결과는 다음과 같았다. 식이 섭취량은 당뇨대조군과 마점질물투여군이 정상대조군에 비해 거의 2배에 가까운 식이 섭취량을 보였다. 체중의 변화는 정상대조군과 당뇨대조군은 증가한데 반해 마점질물 투여은 유의적으로 체중이 감소하는 경향을 보였다 공복시 혈중 포도당 농토는 시료 투여 직전에 정상 대조군은 $68.00{\pm}6.04\;mg/dL$였으며, 당뇨 대조군과 시료 투여군은 $400.50{\sim}403.50\;mg/dL$였다. 당뇨 대조군의 경우 시료 투여 첫째날부터 종료시까지 고혈당을 유지하였으며, 마점질물 투여군에서는 실험기간 5일째까지는 당뇨 대조군에 비해 혈당이 감소하지 않았지만, 시료투여 10일째에 마점질물투여군은 당뇨대조군에 비해 낮은 혈당치를 나타내었다. 혈액중 중성지방농도는 당뇨 대조군이 정상 대조군에 비해 약 1.7배 정도 유의적으로 높았다. 마점질물투여군은 당뇨 대조군에 비해 중성지방의 농도가 유의적으로 감소하였다. 비장의 무게는 y500F1군이 당뇨 대조군에 비해 유의적으로 감소하였다. 이상에서 마점질물 fraction 1을 Alloxan 유발 당뇨 마우스에 투여하였을 때 체중의 감소 및 혈중 포도당과 중성지방농도를 감소시키는 경향을 나타내어 당뇨병에 대한 개선 효과를 위한 기능성 식품으로서의 개발 가능성이 높다고 사료된다.

표고버섯과 보리에서 추출한 ${\beta}-glucan$이 Alloxan 유발 당뇨 마우스의 혈당 및 지질 성분에 미치는 영향 (Effects of ${\beta}-glucan$ from Lentinus edodes and Hordeum vulgare on Blood Glucose and Lipid Composition in Alloxan-induced Diabetic Mice)

  • 표고버섯과 보리에서 추출한 ${\beta}-glucan(M500,\;B500)$을 Alloxan으로 당뇨를 유발시킨 6주령의 ICR 마우스에 10 일간 경구 투여를 실시하고 식이 섭취량 및 체중의 변화를 관찰하고 혈액중의 포도당 농도, 혈액과 간장 중의 지질 성분 및 각종 장기의 무게 변화를 측정하였으며 결과는 다음과 같다. Alloxan 유발 당뇨 마우스에서 B500군과 M500군은 정상대조군에 비해 거의 2 배에 가까운 식이 섭취량이 관찰되었으나 이에 따른 체중 변화는 시료 투여군에서 당뇨 대조군과 정상 대조군보다 유의적으로 체중이 감소하는 경향을 보였다(p<0.05). 혈중 포도당 농도는 당뇨 대조군의 경우 실험기간 시작부터 종료시까지 고혈당을 유지하였으나, B500군과 M500군은 당뇨 대조군에 비해 유의적으로 낮은 혈당치를 나타내었다(p<0.05). 혈중 중성지방 함량은 당뇨 대조군이 정상 대조군에 비해 약 1.7배 정도 증가하였다. B500군은 당뇨 대조군에 비해 중성지방의 농도가 유의적으로 감소하였으나, M500군에서는 유의적인 차이가 없었다(0.05). 혈액중 총 콜레스테롤 농도와 LDL 콜레스테롤 농도는 각 군에서 유의적인 차이를 보이지 않았다. 이상에서 표고버섯과 보리의 ${\beta}-glucan$을 Alloxan 유발 당뇨 마우스에 투여하였을 때 체중 및 혈중 포도당 농도가 감소되었고, 특히 보리 ${\beta}-glucan$은 혈중 중성지방 농도를 감소시키는 경향을 보였으므로 표고버섯과 보리의 ${\beta}-glucan$이 당뇨병에 대한 개선 효과가 있다고 인정되며 이를 이용한 기능성 식품으로의 개발 가능성이 높다고 사료된다.

회화나무 열매 추출물에 의한 지방세포 분화 및 지방생성 억제 (Inhibition of Adipocyte Differentiation and Adipogenesis by the Extract from Sophora japonica Fruit)

  • The world-wide rate of obesity is increasing continuously, representing a serious medical threat since it is associated with a variety of diseases including type 2 diabetes, cardiovascular disease, and numerous cancers. Sophora japonicais used as a traditional herb for medicinal purposes in eastern Asia. However, the anti-obesity effects of S. japonicafruit have not been explored. The aim of this study is to investigate the inhibition of adipocyte differentiation and adipogenesis by an ethanol extract of S. japonicafruit (EESF) in 3T3-L1 pre-adipocytes. Our results demonstrate that EESF suppressed the terminal differentiation of 3T3-L1 pre-adipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet number and lipid content through Oil Red O staining. EESF significantly reduced the accumulation of cellular triglyceride, which was associated with a significant inhibition of the levels of pro-adipogenic transcription factors, including PPARγ, C/EBPα and C/EBPβ. In addition, EESF potentially down regulated the expression levels of adipocyte-specific proteins, including aP2 and leptin. In particular, EESF treatment effectively enhanced the activation of the AMPK signaling pathway; however, the co-treatment with compound C, an inhibitor of AMPK, significantly restored the EESF-induced inhibition of pro-adipogenic transcription factors and adipocyte-specific genes. These results indicate that EESF may exert an anti-obesity effect by controlling the AMPK signaling pathway, suggesting that the fruit extract of S. japonica may be a potential anti-obesity agent.
