Anti-Inflammatory Effect of 3-Bromo-4,5-Dihydroxybenzaldehyde, a Component of Polysiphonia morrowii, In Vivo and In Vitro

  • 3-Bromo-4,5-dihydroxybenzaldehyde (BDB) is a natural bromophenol compound that is most commonly isolated from red algae. The present study was designed to investigate the anti-inflammatory properties of BDB on atopic dermatitis (AD) in mice induced by 2,4-dinitrochlorobenzene (DNCB) and on lipopolysaccharide (LPS)-stimulated murine macrophages. BDB treatment (100 mg/kg) resulted in suppression of the development of AD symptoms compared with the control treatment (induction-only), as demonstrated by reduced immunoglobulin E levels in serum, smaller lymph nodes with reduced thickness and length, a decrease in ear edema, and reduced levels of inflammatory cell infiltration in the ears. In RAW 264.7 murine macrophages, BDB (12.5, 25, 50, and $100{\mu}M$) suppressed the production of interleukin-6, a proinflammatory cytokine, in a dose-dependent manner. BDB also had an inhibitory effect on the phosphorylation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-${\kappa}B$) and signal transducer and activator of transcription 1 (STAT1; Tyr 701), two major signaling molecules involved in cellular inflammation. Taken together, the results show that BDB treatment alleviates inflammatory responses in an atopic dermatitis mouse model and RAW 264.7 macrophages. These results suggest that BDB may be a useful therapeutic strategy for treating conditions involving allergic inflammation such as atopic dermatitis.

Anti-inflammatory Effect of Baekho-tang Extract through Endocannabinoid system (ECS) Control in Atopic Dermatitis (아토피피부염에서 Endocannabinoid system (ECS) 조절을 통한 백호탕 추출물의 염증 완화 효과)

  • Objectives The aim of this study was to identify the effect of Baekho-tang extract on epidermal barrier recovery and inflammation relief in atopic dermatitis-induced mice through Endocanabinoid system (ECS) regulation. Methods In this study, we used 4-week-old NC/Nga mice were divided into 4 group: lipid barrier elimination group (LBEG), palmitoylethanolamide treated group after lipid barrier elimination (PEAT), Baekho-tang extract treatment group after lipid barrier elimination (BHTT) and control group (Ctrl). Each group was assigned 10 animals. We identified that cannabinoid receptor (CB) 1, CB2, CD (Cluster of Differentiation) 68, inducible nitric oxide synthase (iNOS), substance P and Matrix metallopeptidase 9 (MMP-9) through our immunohistochemistry. Results We discovered that when compared to PEAT, 8-hydroxydeoxyguanosine, a marker of oxidative stress in the epidermal barrier, and CB1 and CB2, markers of ECS modulation, were less activated in BHTT. These results led to an anti-inflammatory response in BHTT, with a significant decrease in several inflammatory mediators such as CD 68, iNOS, substance P and MMP-9 compared to PEAT and LBEG. Conclusions These results suggest that the Baekho-tang extract can reduce the inflammation of atopic dermatitis by restoring the structural damage of the skin lipid barrier through ECS modulation.

Combinational Treatment of Oral Hwangtogamibang and External spray on Atopic Dermatitis (아토피 피부염에 대한 황토가미방과 외치방(外治方) 겸용(兼用)에 관한 연구(硏究))

  • The effect of combinational treatment of oral HTGMB and topical CSGMB ("H&C" hereinafter) on the changes of dermal inflammation index and immune system were studied using NC/Nga atopic dermatitis animal model. 1. Through naked eye examination, H&C ameliorated atopic dermatitis compared to the control group. Significant reduction of dermal inflammation index was observed after 12 weeks of treatment. 2. The H&C treated group showed 51% increase in the number of immune cells in DLN, and 59% increase in the number of immune cells is dorsal skin. 3. The H&C treated group showed decrease of 26%, 8%, 59% in CD19+, CD3+/CD69+, B220+/IgE+ cells in DLN respectively. On the other hand, CD3+, CD8+, CD4+ cells were increased by 8%, 31%, 12%, respectively. 4. The H&C treated group showed significant decrease of 38% and 47% in B220+/IgE+, CD11b+/Gr-1+ cells within dorsal skin respectively. Also, a decrease in CCR3+ cells by 21% was observed. 5. Significant decrease of the production of IL-4, IL-5, GM-CSF by 39%, 65%, 60% respectively, in spleen cells activated with CD3 and CD28 were observed in the H&C treated group. The results above strongly suggest significance of anti-atopic dermatitis effect of combinational treatment of oral HTGMB and topical CSGMB through immune modulation. Further applications in clinical use of the treatment are anticipated.

Improving Effect on Aatopic Dermatitis with Treatment of Selected Herbs ; Polygoni Multiflori Radix, Diospyros Kaki, ilite and its Mixture in NCNga Mice (백하수오(白何首烏), 시엽(枾葉), 일라이트 조성물의 Atopy 병태모델 치료효과)

  • We observed the efficacy of natural herbs and mixture in treating atopic dermatitis using anti-human IgE treated Human HMC-I cell and NCNga mice model. First, we selected three herbs, Cynonchum witfordii, Diospyros kaki, Ilite which were used to treat skin disease in Traditional Korea Medicine. Using Human HMC-I cell treated with anti-human IgE, we investigate in vitro whether each herb effects on IL-4, IL-13, $TNF-{\alpha}$ expression and $TNF-{\alpha}$, Histamine secretion value. Finally, we conducted study whether the mixture of the selected herbs is more effective than each herb which consist the mixture and control group. The results show that the mixture is better in improving atopic dermatitis condition.

The Effects of Forsythiae Fructus n-BuOH Fraction on Atopic Dermatitis (연교(連翹) n-BuOH 분획물의 아토피 피부염 억제 효과)

  • Objectives Previous studies have found out that Forsythiae Fructus (FF) extracts have anti-atopic activities by in vitro experiment. In order to understand more about FF extracts' benefit, we subdivided FF extracts depending on systematic fractionation method by using Methylene chloride (MC), Ethyl acetate (EtOAc), n-BuOH and n-hexane (n-Hx). This study is designed to examine the effect of FF fractions on the PMA- ionomycin-induced activation of RBL-2H3 mast cell lines in vitro and on the DNCB-induced activation of NC/Nga mice in vivo. Methods For this study, we examined IL-4, IL-13 production by ELISA analysis, IL-4, IL-13, IL-31, IL-31RA and TNF-${\alpha}$ mRNA expression by real-time PCR and manifestations of AP-1 and MAPKs transcription factors by western blotting in vitro. Through in vitro experiment, we selected FF n-BuOH fraction that seems the best effective in atopic dermatitis then induced it on NC/Nga mice by DNCB. We measured mice's WBC, eosinophil and neutrophil in heart blood, IL-4, IL-5, IFN-${\gamma}$ in the spleenocyte culture supernatant, the absolute cell numbers of CD4+, CD8+, B220+CD23+, CD3+CD69+ and Gr-1+CD11b+ in the PBMCs, ALN and dorsal skin, IL-5, IL-13, IL-31, IL-31RA in the dorsal skin by real-time PCR and the distribution of immune cells by H&E on dorsal skin and ANL and toluidine blue staining on dorsal skin. Results FF n-BuOH fraction suppressed IL-4, IL-13 production and mRNA expression of IL-4, IL-13, IL-31, IL-31RA and TNF-${\alpha}$. Results from the western blot analysis showed that FF n-BuOH fraction reduced the activation of the mast cell specific transduction factors involved in AP-1 by suppressing JNK and ERK phosphorylation. In the gross, atopic dermatitis induced by DNCB in NC/Nga mice were improved by oral administration of FF n-BuOH fraction. Oral FF n-BuOH fraction also decreased the level of IgE in mice's serum and the level of IL-4 and IL-5 in the spleenocyte culture supernatant, cell numbers of CD8+, B220+CD23+ in the PBMCs, CD4+ in the ALN and CD4+, Gr-1+CD11b+ in the dorsal skin and suppressed mRNA expression of IL-5, IL-13, IL-31, IL-31RA in the dorsal skin. Histological examination showed that infiltration levels of immune cells in atopic dermatitis induced NC/Nga mice were improved by FF n-BuOH fraction. Conclusions FF n-BuOH fraction can reduce pruritus by suppressing IL-31, IL-31RA secretion and modulate molecular mediators and immune cells associated with atopic dermatitis induced in NC/Nga mice which may have played a significant role in recovering atopic dermatitis symptoms.

Inhibitory Studies of Hwangryunhaedok-tang on Development of Atopic Dermatitis in NC/Nga Mice (황련해독탕이 NC/Nga Mice에서 유발된 아토피 피부염에 미치는 영향)

  • Objectives : HRHDT has been known as a useful prescription with antibiotic, anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-atopic dermatitis effect of HRHDT, we treated HRHDT in NC/Nga mice model skin induced contact hypersensitivity. Methods : Contact hypersensitivity, a local inflammatory response of skin, was induced by spreading the back skin of NC/Nga mice with 0.4${\sim}$1% DNCB. HRHDT was prepared by dissolving 3% HRHDT in solution and treated 3 weeks on the back skin. Results : HRHDT significantly reduced TEWL and erythema by 0.4${\sim}$1% of DNCB treatment compared to control group. HRHDT also reduced IgE on serum obtained from blood of DNCB-treated mice. Conclusions : These results showed that HRHDT could be used as a pharmaceutical material with anti-atopic dermatitis effect by reducing IgE in contact, hypersensitivity atopic dermatitis NC/Nga mice model by DNCB.

Antioxidant and Anti-inflammatory Effects of Atoberry in Atopic Dermatitis-like NC/Nga Mouse Model (아토피 피부염 유사 NC/Nga 마우스 모델에서 아토베리의 항산화 및 항염증효과)

  • Oxidative stress has been implicated in cutaneous damage in various inflammatory skin diseases, including atopic dermatitis (AD). Atoberry is the herb medicine extract which is composed with Spirodelae Herba, Xanthii Fructus, Houttuyniae Herba, Taraxaci Herba, Retinervus Luffae Fructus, Platycodi Radix, and Scutellariae Radix. In this study, we investigated the antioxidant and anti-inflammatory effects of Atoberry in AD-like skin lesion NC/Nga mice. Murine AD-like skin lesions were made by painting Dermatophagoides farinse (Df) extract. Atoberry significantly increased electron donating ability (DPPH), nitrite scavenging (NO) and superoxide dismutase (SOD) activities in dose dependant. Topically applied Atoberry significantly reduced clinical severity score, ear thickness and histological grade in AD-like skin lesion NC/Nga mice. In addition, the serum levels of IgE, NO and prostaglandin E2 were significantly reduced by Atoberry. Futhermore, skin tissue levels of SOD, catalase and glutathione peroxidase (GPx) were significantly reduced by Atoberry. These results demonstrate that topical application of Atoberry may be improve the AD-like skin lesion by antioxidant and anti-inflammatory effects.

Anti-atopic Effect of Hot Water and Supercritical Carbon Dioxide Fluid Extract of Persimmon (Diospyros kaki) Peels (감껍질 열수 및 초임계 유체 추출물의 항아토피 효과)

  • This study aimed to investigate the anti-atopic effect of hot water (PPWE) and supercritical-carbon dioxide fluid extract of persimmon peels (SPPE) on atopic dermatitis (AD)-like skin lesions in hairless mice. Histological analyses demonstrated that SPPE treatment more strongly inhibited the dermal infiltration of inflammatory cells in AD-like skin lesions than that by PPWE. Compared to PPWE, SPPE significantly decreased the dermatitis clinical score and the epidermal thickness and potently suppressed serum IgE and interleukin (IL)-4 production in hairless mice with AD. Furthermore, compared to PPWE, SPPE potently inhibited the production of nitric oxide, prostaglandin $E_2$, and proinflammatory cytokines such as IL-6 and IL-$1{\beta}$ in lipopolysaccharide-stimulated RAW264.7 macrophages. These results suggested that SPPE exhibited anti-atopic dermatitis activity via the regulation of inflammatory responses.

Anti-inflammatory and Antioxidant Effects of Zizyphus jujuba Miller var. inermis extracts (대추 추출물의 항산화 및 아토피 피부염 관련 항염증 효과)

  • In the present study, the effect of Z. jujuba Miller var. inermis Rehder extracts on the secretion of atopic dermatitis (AD)-related cytokines and hyaluronidase activity was investigated. We prepared four fractions, butylene glycol (JB), ethanol (JE), and water (JW), with Z. jujuba Miller var. inermis extracts. JW significantly reduced the secretion of interleukin-8 and JE reduced the secretion of tumor necrosis factor-alpha in human keratinocyte HaCaT cells. Also, hyaluronidase activity was measured by enzyme assay and the fractions inhibited the activity in a dose-dependent manner. In addition, the human dermal fibroblast, HDF-n cells were treated with the extracts and antioxidant activities were measured. The results showed that the extracts increased the free radical scavenging activity and the superoxide dismutase activity. Taken together, Z. jujuba Miller var. inermis extracts reduced the secretion of AD-related cytokines and inhibited the hyaluronidase. In addition, the extracts showed antioxidant activity.

The Effect of Anti-atopic Cosmetic in Hairless Mice (항 아토피 화장품이 아토피 동물모델 Hairless Mice에 미치는 영향)

  • The efficacies of anti-atopic preparations were investigated in hairless mice suffering from 2,4-dinitro-chlorobenzene (DNCB)-induced atopic dermatitis-like lesion. Solid lipid nanoparticle (SLN) containing ceramide and astaxanthin was prepared by a melt-homogenization method using Aminsoft-CT 12 (Cocoyl glutamate) as an emulsifier. And then, the SLNs were coated with silk fibroin (SF) by taking advantage of an electrostatic interaction between the surface of SLNs and SF. SLNs were included in lotion (FL) and cream (FC) types of preparations. Anti-atopic efficacies of the preparations were investigated in terms of appearance of skin surface, spleen index, serum IgE level, and serum cytokine level. SLN-containing preparations suppressed IgE production and IL-4 expression, but promoted $IFN-{\gamma}$ expression.
