포도상구균에서 분비하는 장내독소 B(SEB)에 대한 재조합 단백질을 이용한 면역특이적 난황항체 생산 (Production of Immunospecific Egg Yolk Antibody with Recombinant Staphylococcal Enterotoxin B (SEB) Protein)

  • 본 연구는 식중독은 물론, 아토피성 피부염의 원인물질로 알려져 있는 포도상구균 장내독소 B(Staphylococcal enterotoxin B; SEB)에 대한 특이 난황항체를 개발하고자 실험을 실시하였다. 우선, SEB 유전자를 클로닝한 다음, 대장균 발현시스템을 이용하여 약 30 kDa 정도의 재조합 SEB 단백질을 생산하였다. 재조합 SEB 단백질을 산란계에 2주 간격으로 3회 면역접종을 실시하고, 혈청 및 난황 내 항체가를 측정한 결과, 면역 후 4주경에 항체가가 최고치에 달하였으며, 산란계로부터 획득한 난황항체를 이용한 Western blot 결과, 재조합 SEB 단백질은 물론, 상용화 SEB 단백질과도 특이적으로 반응한다는 것을 규명하였다. 결론적으로, 식중독과 아토피성 피부염 등의 원인물질로 알려진 SEB에 특이적인 난황항체를 생산에 성공하였으며, 이러한 특이적인 난황항체는 식중독 및 아토피성 피부염의 예방 및 치료에 활용 가능할 것으로 사료된다.

Oral Administration of Lactobacillus plantarum HY7714 Protects Hairless Mouse Against Ultraviolet B-Induced Photoaging

  • Ultraviolet (UV) irradiation alters multiple molecular pathways in the skin, thereby inducing skin damage, including photoaging. In recent years, probiotics have gained interest due to their beneficial effects on skin health, such as inhibiting atopic dermatitis and improving skin immunity or inflammation. However, little is known about the effects of probiotics on UVB-induced photoaging. In this study, we evaluated the effect of Lactobacillus plantarum HY7714 against UVB-induced photoaging in human dermal fibroblasts and hairless mice. The results showed that L. plantarum HY7714 treatment effectively rescued UVB-reduced procollagen expression through the inhibition of UVB-induced matrix metalloproteinase (MMP)-1 expression in human dermal fibroblasts. Data from a western blot showed that L. plantarum HY7714 inhibited the phosphorylation of Jun N-terminal kinase, thereby suppressing the UVB-induced phosphorylation and expression of c-Jun. Oral administration of L. plantarum HY7714 clearly inhibited the number, depth, and area of wrinkles in hairless mouse skin. Histological data showed that L. plantarum HY7714 significantly inhibited UVB-induced epidermal thickness in mice. Western blot and zymography data also revealed that L. plantarum HY7714 effectively inhibited MMP-13 expression as well as MMP-2 and -9 activities in dermal tissue. Collectively, these results provide further insight regarding the skin biological actions of L. plantarum HY7714, a potential skin anti-photoaging agent.

The Chloroform Fraction of Carpinus tschonoskii Leaves Inhibits the Production of Inflammatory Mediators in HaCaT Keratinocytes and RAW264.7 Macrophages

  • Inflammation is the immune system's response to infection and injury-related disorders, and is related to pro-inflammatory factors (NO, $PGE_2$, cytokines, etc.) produced by inflammatory cells. Atopic dermatitis (AD) is a representative inflammatory skin disease that is characterized by increasing serum levels of inflammatory chemokines, including macrophage-derived chemokine (MDC). Carpinus tschonoskii is a member of the genus Carpinus. We investigated the anti-inflammatory activity of C. tschonoskii by studying the effects of various solvent fractions prepared from its leaves on inflammatory mediators in HaCaT and RAW264.7 cells. We found that the chloroform fraction of C. tschonoskii inhibited MDC at both the protein and mRNA levels in HaCaT cells, acting via the inhibition of STAT1 in the IFN-${\gamma}$ signaling pathway. In addition, the chloroform fraction significantly suppressed the expression of inflammatory factors induced by lipopolysaccharide stimulation, except COX-2 and TNF-${\alpha}$. These results suggest that the chloroform fraction of C. tschonoskii leaves may include a component with potential anti-inflammatory activity.

  • Ribes fasciculatum var. chinense MAX. (R. fasciculatum) has traditionally been used in Korea to treat inflammatory diseases. However, the exact mechanism that accounts for the anti-inflammatory effect of R. fasciculatum is not completely understood. We aimed to ascertain the pharmacological effects of R. fasciculatum on both compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) in mice. Additionally, to find a possible explanation for the anti-inflammatory effects of R. fasciculatum, we evaluated the effects of R. fasciculatum on the production of inflammatory mediators in LPS-stimulated macrophage cells. Treatment of R. fasciculatum significantly reduced compound 48/80- or histamine-induced the pruritus in mice. R. fasciculatum attenuated the AD symptoms such as eczematous, erythema and dryness and serum IgE levels in AD model. Additionally, R. fasciculatum inhibited the production of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6). The maximal rates of TNF-${\alpha}$ and IL-6 inhibition by R. fasciculatum (1 mg/ml) were approximately 32.12% and 46.24%, respectively. We also showed that R. fasciculatum inhibited the activation of nuclear factor-kappa B in LPS-stimulated macrophages. Collectively, the findings of this study provide us with novel insights into the pharmacological actions of R. fasciculatum as a potential molecule for use in the treatment of allergic inflammatory diseases.

An Anti-Cancer Drug Candidate CYC116 Suppresses Type I Hypersensitive Immune Responses through the Inhibition of Fyn Kinase in Mast Cells

  • Mast cells are the most prominent effector cells of Type 1 hypersensitivity immune responses. CYC116 [4-(2-amino-4-methyl-1,3-thiazol-5-yl)-N-[4-(morpholin-4-yl)phenyl] pyrimidin-2-amine] is under development to be used as an anti-cancer drug, but the inhibitory effects of CYC116 on the activation of mast cells and related allergy diseases have not reported as of yet. In this study, we demonstrated, for the first time, that CYC116 inhibited the degranulation of mast cells by antigen stimulation ($IC_{50}$, ${\sim}1.42{\mu}M$). CYC116 also inhibited the secretion of pro-inflammatory cytokines including TNF-${\alpha}$ ($IC_{50}$, ${\sim}1.10{\mu}M$), and IL-6 ($IC_{50}$, ${\sim}1.24{\mu}M$). CYC116 inhibited the mast cell-mediated allergic responses, passive cutaneous anaphylaxis (ED50, ~22.5 mg/kg), and passive systemic anaphylaxis in a dose-dependent manner in laboratory experiments performed on mice. Specifically, CYC116 inhibited the activity of Fyn in mast cells and inhibited the activation of Syk and Syk-dependent signaling proteins including LAT, $PLC{\gamma}$, Akt, and MAP kinases. Our results suggest that CYC116 could be used as an alternative therapeutic medication for mast cell-mediated allergic disorders, such as atopic dermatitis and allergic rhinitis.

율피추출물의 사람 비만세포주 사이토카인 발현 억제 효과 (Inhibitory Effects of Chestnut Inner Shell Cytokine Production from Human Mast Cell Line)

  • Objectives : Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by eczematous inflammtion of the skin. The chestnut inner shell extracts (CI) has been used as a cosmetic material for a long time in Korea. However, the precise anti-allergy effects of CI have yet to be clearly elucidated. In this study, we attempted to evaluate the effect of CI on mast cell-mediated allergy inflammation. Methods : To find the anti-allergy and inflammatory effect of CI, we investigated the inhibitory effect of CI on the production of inflammatory mediators using by enzyme-linked immunosorbent assay in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore (A23187) stimulated-human mast cell (HMC-1). Results : In this study, we found that CI did not show cytotoxic effect at up to 10 ug/ml on HMC-1. CI inhibited the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6 and IL-8 in stimulated HMC-1. Maximal rate of TNF-${\alpha}$, IL-6 and IL-8 inhibition by CI (10 ug/ml) were about 47.6%, 44.1% and 22.5% respectively. In addition, we showed that Fr.3 isolated from n-Butyl alcohol layer of CI attenuated the production of TNF-${\alpha}$, IL-6 and IL-8 in HMC-1. Conclusion : Taken together, the findings of this study provide us with a novel insight action of CI as a potential molecule for use in the treatment of allergic inflammation diseases.

길경(桔梗)발효 추출물의 알레르기성 소양행동 억제효과연구 (Anti-pruritic Effect of Ethanol Extracts from Platycodon grandiflorum and its fermented production in Scratching Behavior Mouse Models)

  • Objectives : Platycodon Root is frequently used in traditional medicine to treat inflammatory diseases of the throat. The purpose of this study was to characterize the effect of the EtOH extract of fermented Platycodon grandiflorum on the ameliorative effects on the Antipruritic Effect of atopic dermatitis mouse model induced by compound 48/80 and ovalbumin (OVA)-induced allergic responses in mice. Methods : In the present study, we examined the anti allergic effect of Platycodon grandiflorum (PR) and its fermented production (FPR) in several mouse model. We measured acute ear edema in a mouse model caused by TPA and consecutively histological change of Ear tissue was observed by hematoxylin and eosin (H&E) staining. and also Scratching behaviors by compound 48/80 was investigated. The levels of allergic mediators such as immunoglobulin (Ig) E, and anti-oxidant markers such as SOD and MDA in the sera of OVA induced allergic mice were measured by enzyme-linked immunosorbent assay. Results : FPR inhibited compoud 48/80-induced scratching behavior in mice, as well as acetic acid-induced writhing in mice. The anti-scratching behavioral effect of FPR was more potent than PR. FPR extract significantly decreased the serum levels of IgE and MDA compared with those of OVA control group. Conclusions : These results indicate that Anti allergic effect of Platycodon grandiflorum is enhanced by fermentation with Saccharomyces cerevisae and FPR may be useful for protection from the itching reactions, which are IgE-mediated representative skin allergic diseases.

이중배양법에 따른 Lactococcus lactis의 아토피 유발인자 억제 효과 증대 (Double-culture Method Enhances the in Vitro Inhibition of Atopy-inducing Factors by Lactococcus lactis)

  • 유산균이 활성화된 비만세포에서 발현하는 IL-4와 IL-13을 조절할 수 있는지를 분석하였고, GATA-1, GATA-2, NF-AT1, NF-AT2, NF-κB p65 전사인자의 활성을 억제하는지를 실험적으로 규명하였다. 이전 연구에서 T cell에서 CD4+/CD25+/foxp3+ 증가를 실험하여 항아토피 기능성이 있는 유산균을 탐색하였고, Staphylococcus aureus에 대한 항균력을 증가시키는 유산균의 이중배양법을 확인하였다. 여기서는 RBL-2H3 비만세포를 이용하여 이 배양법으로 배양한 유산균이 아토피 피부염의 원인이 되는 allergy 염증반응에서 얼마나 억제능을 갖는지 알아보았다. 그 결과 Lc. lactis culture with medium containing Lb. plantarum supernatants > Lc. lactis > Lc. lactis culture broth with medium containing Lb. plantarum culture broth > Lb. plantarum의 순으로 나타났다. 이 cell 수준(level of mast cells)에서의 순서는 이전 연구의 level of microorganisms (anti-S. aureus)에서의 아토피 유발인자 억제능 순서와 같다. 따라서 세포수준에서도 Lb. plantarum 배양상층액을 첨가한 배지에 Lc. lactis 배양한 경우가 활성화된 비만세포의 allergy 반응으로의 분화 및 활성을 가장 잘 억제하고 관련 유전자 발현을 선택적으로 조절하는 anti-allergy 효과를 나타낸다고 사료된다.

개에서 0.1% FK-506 연고의 피부 비만세포와 호산구에 미치는 영향 (Effects of 0.1% Tacrolimus on Canine Skin Mast Cells and Eosinophils)

  • Goat-anti-canine IgE polyclonal antibody로 유발한 개 피부염에 대한 0.1% FK-506 연고의 항염효과를 평가하였다. 임상적으로 건강한 성견 5두를 사용하여 실험군은 대조군, 위약적용군, FK-506 적용군으로 구분하였다. 항염효과는 팽진의 크기, 피부생검 검사를 통한 비만세포와 호산구수를 측정하여 평가하였으며 비만세포는 Toluidine blue, 호산구는 Luna 염색법을 사용하였다. 염증유발 15분 후의 팽진의 크기는 FK-506 적용군에서 유의성 있게 감소하였다 (p < 0.01). 6, 12, 24 시간 후 염증유발부위에 대한 피부생검을 각각 실시하였다. 정상피부의 비만세포는 $12.32{\pm}3.27cells/mm^2$ 이었으며 12시간 후에 FK-506 적용군은 $5.79{\pm}0.86cells/mm^2$으로 대조군의 $1.89{\pm}0.49cells/mm^2$, 위약군의 $1.74{\pm}0.50$ 에 비해 유의하게 증가하였다(p < 0.01). 정상피부의 호산구는 $2.73{\pm}2.24cells/mm^2$ 이었다. 대조군, 위약 적용군, FK-506 적용군의 호산구 수는 12 및 24 시간 후 FK-506 적용군에서 대조군과 위약 적용군에 비해 유의하게 증가하였다(p < 0.01). 따라서 FK-506 연고는 급성염증과 아토피증상과 유사한 후기염증에서 항염효과 있는 것으로 사료된다.

참당귀 초임계 이산화탄소 선택 추출물의 항염 효능 (The Anti-inflammatory Effect of Angelica gigas Nakai Supercritical Carbon Dioxide Selective Extracts)

  • 본 연구에서는 참당귀(Angelica gigas Nakai)의 핵심 유효 성분인 decursin과 decursinol angelate를 높은 수율로 추출한 초임계 이산화탄소 선택 추출물의 항염증 효능을 에탄올 추출물과 비교하여 측정하였다. 항염증 효능을 측정하기 위해 염증 매개 인자인 nitric oxide(NO), 염증성 사이토카인인 interleukin(IL)-6, IL-8 생성량을 측정하였다. NO 생성량은 lipopolysaccharide(LPS)로 염증 반응을 유도한 Raw 264.7 세포를 대상으로 Griess assay를 통해 측정하였고, IL-6 및 IL-8 생성량은 tumor necrosis factor(TNF)-α로 염증 반응을 유도한 human keratinocyte cell line(HaCaT) 세포를 대상으로 enzyme-linked immunoadsorbent assay(ELISA)를 통해 측정하였다. NO 생성량은 초임계 이산화탄소 추출물이 에탄올 추출물에 비해 뛰어나게 억제하였다. IL-6 및 IL-8 생성량 또한 에탄올 추출물은 오히려 증가시키는 반면, 초임계 이산화탄소 추출물은 6.25 ㎍/mL의 농도에서 통계적으로 유의하게 억제하였다(P<0.01). 우리는 이러한 결과를 통해 참당귀 초임계 이산화탄소 선택 추출물이 아토피 피부염을 완화시키는 항염증 코스메슈티컬 소재로써 활용 가능함을 확인하였다.
