Evaluation of Biological Activities of Invasive Alien Plants for Development of Functional Biomaterials

  • This study aims to confirm the possibility of using the invasive alien plants in Jeju as a functional biomaterial. To achieve this purpose, 70% ethanol extract and solvent fractions were prepared for five invasive alien plants (Hypochaeris radicata, Rumex acetosella, Humulus japonicus Siebold & Zucc., Solanum viarum, Lactuca scariolar) and their antioxidant, antibacterial anti-inflammatory and anti-obesity effects were investigated. The DPPH radical scavenging activity of ethanol extract from invasive alien plants was shown in the order of Rumex acetosella > Hypochaeris radicata > Humulus japonicus. Antimicrobial activity of ethanol extract against food poisoning bacteria (4 species) and oral cavity-induced microorganisms (6 species) was measured. As a result, the extract of Humulus japonicus showed high antibacterial effects against food poisoning bacteria (E. coli, V. parahaemolyticus) and oral microbes (L. casei, S. epidermidis, E. faecalis). In LPS-induced RAW 264.7 cells, the anti-inflammatory effect of ethanol extract from invasive alien plants was investigated. As a result, the NO production inhibition activity was highest in the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract, and the NO production inhibition activity was concentration-dependent. In addition, the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract showed a concentration-dependent inhibitory effect on cytokine (IL-6) production. These extracts also showed inhibitory activity of COX-2, an inflammatory protein. This suggests that NO production inhibition activity by the extract of invasive alien plants is the result of inhibition of iNOS and COX-2 expression. Currently, organic solvent fractions of crude extract are manufactured and the investigation of active ingredients is continuing along with evaluation of biological activity such as anti-inflammatory. These results are expected to be a major data for the study on the separation and utilization of active ingredients with antioxidant, antibacterial and anti-inflammatory effects using foreign plant crude extract and solvent fractions, and are highly likely to be applied to the development of functional food and cosmetics materials.

흑생강(Kaempferia parviflora)의 항응고 및 혈소판 응집저해 활성 (Anti-coagulation and Anti-platelet Aggregation Activities of Black Ginger (Kaempferia parviflora))

  • 흑생강은 생강과 초본과 뿌리식물로 태국 및 라오스에서는 크라차이담으로 불리며, 뿌리는 향신료와 차로 이용되어 왔다. 말린 뿌리는 위장장애, 통풍, 이질, 알러지 치료 및 강장용으로 전통적으로 이용되어 왔으며, 최근에는 항비만, 항산화, 항염증, 혈전 용해 활성 등의 다양한 유용 생리활성이 보고되어 있다. 본 연구에서는 흑생강의 혈액순환 개선 활성을 확인하기 위해, 흑생강 지하부의 ethanol 추출물 및 열수 추출물을 조제하고 각각 이의 순차적 유기용매 분획물인 hexane 분획물, ethylacetate (EA) 분획물, butanol 분획물 및 물 잔류물을 조제하여 각각의 성분을 분석하였고, 혈전 생성과 관련된 항응고 활성, 혈소판 응집저해 활성 및 적혈구 용혈활성을 평가하였다. 그 결과, ethanol 추출물의 EA 분획물 5 mg/ml 농도에서 TT, PT, aPTT를 각각 1.22, 1.49 및 15배 이상 연장시켜 강력한 항응고 활성을 나타냄을 확인하였으며, ethanol 및 열수 추출물의 EA 분획물 모두에서 동량의 아스피린보다 강력한 혈소판 응집저해 활성을 확인하였다. 또한 상기 EA 분획물들은 0.5 mg/ml 농도까지 인간 적혈구에 대한 용혈활성을 나타내지 않았다. 본 연구 결과는 흑생강의 혈전 생성 억제 활성에 대한 최초의 보고이며, 흑생강의 EA 분획물이 인간 적혈구 용혈활성 없이 강력한 항응고 및 혈소판 응집저해 활성을 나타내어 신규의 항혈전제로 이용 가능함을 제시하고 있다.

Hesperetin Stimulates Cholecystokinin Secretion in Enteroendocrine STC-1 Cells

  • Hesperetin (3',5,7-trihydroxy 4'-methoxyflavanone) and its glycoside hesperidin (hesperetin 7-rhamnoglucoside) in oranges have been reported to possess pharmacological effects related to anti-obesity. However, hesperetin and hesperidin have not been studied on suppressive effects on appetite. This study examined that hesperetin and hesperidin can stimulate the release of cholecystokinin (CCK), one of appetite-regulating hormones, from the enteroendocrine STC-1 cells, and then examined the mechanisms involved in the CCK release. Hesperetin significantly and dose-dependently stimulated CCK secretion with an $EC_{50}$ of 0.050 mM and increased the intracellular $Ca^{2+}$ concentrations ($[Ca^{2+}]_i$) compared to the untreated control. The stimulatory effect by hesperetin was mediated via the entry of extracellular $Ca^{2+}$ and the activation of TRP channels including TRPA1. These results suggest that hesperetin can be a candidate biomolecule for the suppression of appetite and eventually for the therapeutics of obesity.

Screening of Protein Tyrosine Phosphatase 1B Inhibitory Activity from Some Vietnamese Medicinal Plants

  • Protein tyrosine phosphatase 1B (PTP1B), a negative regulator of insulin signaling, has served as a potential drug target for the treatment of type 2 diabetes. The MeOH extracts of twenty-nine medicinal plants, traditionally used in Vietnam as anti-diabetes agents, were investigated for PTP1B inhibitory activity in vitro. The results indicated that, most materials showed moderate to strong inhibitory activity with $IC_{50}$ values ranging from $3.4\;{\mu}g/mL$ to $35.1\;{\mu}g/mL$; meanwhile, eleven extracts (37.9%) could demonstrate PTP1B activity with $IC_{50}$ values less than $15.5\;{\mu}g/mL$; sixteen extracts (55.2%) could demonstrate PTP1B activity with $IC_{50}$ values ranging from $15.5\;{\mu}g/mL$ to $35.1\;{\mu}g/mL$. The study may provide a proof, at least in a part, for the ethno-medical use in diabetes disease of these plants.

Inhibition of Adipocyte Differentiation by MeOH Extract from Carduus crispus through ERK and p38 MAPK Pathways

  • In this study, the effects of a methanol (MeOH) extract of Carduus crispus L. (Asteraceae) on adipogenesis was investigated in 3T3-L1 cells. To differentiate preadipocytes to adipocytes, confluent 3T3-L1 preadipocytes were treated with a hormone mixture, which included isobutylmethylxanthine, dexamethasone, and insulin (MDI). The methanol extract of C. crispus significantly decreased fat accumulation by inhibiting adipogenic signal transcriptional factors in MDI-induced 3T3-L1 cells in a dose-dependent manner. In MTT assays and on PI-staining, methanol extract of C. crispus inhibited the proliferation of 3T3-L1 cells during mitotic clonal expansion (MCE). The anti-adipogenic effect of the Carduus extract seemed to be associated with the upregulation of extracellular signal-regulated kinase (ERK) and p38 mitogen-activated protein kinase (MAPK) pathways within the first 2 days after MDI treatment. These results suggest that methanol extract of C. crispus might be beneficial for the treatment of obesity.

노인의 저근육형 비만에 따른 만성염증 억제를 위한 후생유전학적 영양에 관한 고찰 (A review of epigenetic nutrients on chronic inflammation associated with sarcopenic obesity in the elderly)

  • 노인에게서 두드러지게 나타나고 있는 저근육형 비만은 근육감소를 동반한 체지방의 증가로 신체상의 뚜렷한 체성분의 변화를 야기 시킨다. 이때 골감소증을 동반하여 신체기능의 감소 및 골절장애 그리고 대사성 관련 질환의 위험도가 올라가는 것으로 보고되고 있다. 노화로 인한 체성분의 변화는 단순한 저근육형일 경우와 비만일 때 보다 급격히 증가된 복부내장 지방조직에서 분비되는 염증성 사이토카인, C-반응성 단백질(CRP), 인터루킨(IL)-6, IL-8 및 종양 괴사 인자(TNF-${\alpha}$)들이 단백질 대사를 저해하여 근육량의 감소를 더욱 촉진시키며, 염증관련 대사질환의 유병률에 중요한 요인이다. 본 연구에서는 DNA 메틸화가 당뇨병, 심혈관질환, 암과 같은 만성염증성 질환에 관계하고 있다는 최근 연구 결과를 기초로 하여 항염증 영양소와 생리활성을 갖는 식품인자들의 충분한 섭취가 염증조절에 중요하게 기여할 것으로 생각되며, 또한 염증성 질환의 주요 표식자인 DNA 메틸화와 히스톤 변형을 유발하는 효소의 활성 또는 비 암호화된 RNA의 발현을 조절함으로써 근육량 증가와 체지방 감소에 중요한 역할을 하는 것을 살펴보았다. 따라서 최근 새롭게 인식되는 후생유전학적 연구의 중심에 있는 항염증 영양소의 효과와 체성분 변화와의 긍정적 관계를 중심으로 저근육형 비만의 예방 및 인구고령화에 건강한 노화를 위한 효과적인 방법을 제시하였다.

Modulation of adipogenesis and lipolysis by green tea in 3T3- Ll adipocytes

  • Green tea have been widely reported as functional foods because of their various bioactivities. In the present study, we used 3T3-Ll cells model of white adipocytes to clarify whether green tea and its main pharmaceutically effective compounds (EGCG, caffeine and theanine) prevent obesity. Cellular viability, glycerol-3-phosphate dehydrogenase activity, glycerol release and HSL mRNA levels were checked. Glycerol release into the medium was significantly increased by the cells treated with green tea extract. Glycerol release into the medium was significantly increased by the cells treated with green tea extract. Caffeine and theanine from green tea showed some level of lipolytic activity, and glycerol-3-phosphate dehydrogenase activity was remarkably decreased by EGCG. These results suggest that green tea has anti-obesity effect through inhibition of adipogenesis and stimulation of lipolysis. Catechins and theanine of green tea might be the factors responsible for the modulation of lipid metabolism and adipocyte differentiation.

Inhibitory Effect of the Ethyl Acetate Fraction from Tulip Tree Leaf (Liriodendron tulipifera L.) on Adipogenesis in 3T3-L1 Cells

  • The inhibitory effects of adipogenesis on ethyl acetate (EtOAc) fraction from leaves of the Tulip tree (TT) were evaluated. Exposure to TT EtOAc fraction (25~200 ${\mu}g/mL$) for a 72 hr incubation period did not significantly change cell viability. TT EtOAc fraction, with concentrations of 100 and 200 ${\mu}g/mL$, inhibited lipid accumulation in 3T3-L1 adipocytes in a dose dependent manner in adipogenesis. The expression of $PPAR{\gamma}$ and $C/EBP{\alpha}$, essential adipogenic markers, was significantly decreased when TT EtOAc fraction was added to cells for 8 days as compared with the untreated control group. These results suggest that TT EtOAc fraction might be a potential therapeutic agent as an effective, natural alternative material for obesity treatment.

Generation of Transgenic Mice with Overexpression of Mouse Resistin

  • The hormone resistin is associated with type II diabetes mellitus in rodent model. Resistin impairs glucose tolerance and insulin action. A new class of anti-diabetic drugs were called thiazolidinediones (TZDs) downreguates a resistin. Resistin gene expression is induced during adipocyte differentiation and resistin polypeptide is secreted by adipocytes. But, the correlation between increased adiposity and resistin remains unknown. The objectives of this study was to clone a mouse resistin CDNA and to generate transgenic mice overexpressing mouse resistin gene. The pCMV-mus/resistin gene was prepared from previous recombinant pTargeT$^{TM}$-mus/resistin by digestion of Bgl II, and has used for microin- jection into pronuclei of one cell embryos. Mouse resistin expression was detected in transgenic F$_1$mice by RT-PCR. The transgenic mouse with resistin gene expression has heavier body weight which was measured higher level of plasma glucose than that of normal mouse. And in diet-induced experiments, in fasting group, resistin expression was higher than that of re-feeding group. This result demonstrates that the resistin gene overexpressing mice may be became to obesity and be useful as an animal disease model to be diabetes caused by insulin resistance of resistin.n.

항고혈압제제를 복용 중인 성인 고혈압 환자의 한열 특성에 관한 연구 (Study on Cold-Heat Pattern Identification in Hypertensive Patients with Antihypertensive Agents)

  • Objectives : The aim of this study was to investigate the correlation between Cold-Heat pattern tendency and demographic traits, physical character, and vital signs in hypertensive patients taking anti-hypertensive agents. Methods : 28 hypertensive patients 30 to 69 years old were recruited. We assessed their general characteristics, physical characteristics (height, weight, waist circumference, etc.), vital signs without respiratory rate (blood pressure, pulse rate, temperature) and administered a Cold-Heat pattern questionnaire. After that, we analyzed statistical data on separate groups according to Cold-Heat characteristics or other criteria. Results : 1. Heat group patients had statistically higher scores in waist, body mass index (BMI), waist circumference and pulse rate. 2. Over weighted group patients had statistically lower scores in cold Pattern Identification. 3. Higher BMI group (above 23/25) patients had statistically significantly higher Heat scores and lower Cold scores, higher waist circumference group patients had lower Cold scores. Conclusions : From the above result in hypertensive patients taking hypertensive agents, Heat group had a obesity tendency and the inverse is also valid. Later, progressed study based on more samples and varied data will contribute to diagnosis the Cold-heat Pattern identification in hypertensive patients.
