Biological Activities in the Leaf Extract of Lythrum salicaria L. (털부처꽃 잎 추출물의 생리활성탐색)

  • In this study, the bioactivities of ethanol (EELS) and water extract (WELS) from the leaf of Lythrum salicaria L. were investigated. In the anti-cancer activity, the growths of both human prostate cancer (DU145) and human colonic carcinoma cell (HT29) were inhibited up 60% by adding 10 mg/$m{\ell}$ of EELS. Anti-inflammatory activity of EELS and WELS have been evaluated on lipopolysaccharide (LPS) induced release of nitric oxide (NO) by the macrophage RAW 264.7 cells. EELS and WELS inhibited inflammatory by 57.3 and 46.9% in 10 mg/$m{\ell}$, respectively. In the anti-oxidative activity, $IC_{50}$ of DPPH radical scavenging activity was respectively 60.71 and $92.90\;{\mu}g/m{\ell}$ by EELS and WELS. In the anti-diabetic activity, $IC_{50}$ of ${\alpha}$-amylase inhibitory activity of EELS and WELS were respectively 5,250 and $5,020\;{\mu}g/m{\ell}$. $IC_{50}$ of ${\alpha}$-glucosidase inhibitory activity was 7.96 and $68.41\;{\mu}g/m{\ell}$ by EELS and WELS. In the anti-obesity, $IC_{50}$ of lipase inhibitory activity was 880 and $9,840\;{\mu}g/m{\ell}$ by EELS and WELS. Finally, EELS and WELS exhibited anti-oxidative, anti-inflammatory, anti-diabetic activity and anti-obesity. It suggests that Lythrum salicaria L. could be potentially used as a resource of bioactive materials for health functional foods.

Anti-obesity Effects of Genistein in Female Ovariectomy-induced Obese Mice (난소절제로 비만이 유도된 암컷 쥐에서 제니스테인의 항비만 효과)

  • To investigate whether genistein regulates menopause-induced obesity, it was studied the effects of genistein on anti-obesity effects in female ovariectomized (OVX) mice, an animal model of postmenopausal women. 7-week-old female mice (C57BL/6J) were randomly divided into three groups. All the animals received a high fat diet or a high fat diet supplemented with genistein for 8 weeks and variables and determinants of obesity were measured. The OVX mice had significantly higher body weight and adipose tissue mass than sham mice. However, genistein supplementation reduced body weight, adipose tissue mass, and adipocyte size of OVX mice. The OVX mice treated with genistein had significantly lower levels of serum triglycerides and total cholesterol than the vehicle-treated OVX mice. Lipid accumulation in liver was also markedly decreased by genistein in OVX mice. The results suggest that genistein can effectively prevent adiposity, adipocyte phertrophy, and llipid disorders caused by ovariectomy. Moreover, this study may contribute to the alleviation of metabolic syndrome, including obesity and hyerlipidemia in postmenopausal women.

Anti-obesity Effect of Jeoreongchajeonja-tang in a High-fat Diet-induced Obesity Mouse model (고지방식이로 유도된 비만 생쥐모델에서 저령차전자탕의 항비만 효과)

  • Objective This study investigated the effects of Jeoreongchajeonja-tang in a high-fat diet-induced obesity mouse model. Methods The study examined 9-week-old male mice (C57bl/6J) divided into four groups: the normal(C57bl/6J-Nr), control (high-fat diet only; HFD-CTL), positive-control (high-fat diet with Garcinia cambogia), and experimental (high-fat diet with Jeoreongchajeonja-tang; HFD-JCT) groups. After 7 weeks, the body weight, food efficiency ratio, organ weight, and visceral fat weight of the mice were measured. Blood serum tests, mRNA, liver histopathology, and epididymis adipocytes were also examined. Results Compared with the Control(HFD-CTL) group, the Experimental(HFD-JCT) group given Jeoreongchajeonja-tang showed significant reductions in absolute body weight and food efficiency ratio. The serum alanine aminotransferase, total-cholesterol, triglyceride, low-density lipoprotein (LDL)-cholesterol, high-density lipoprotein (HDL)-cholesterol, insulin-like growth factor-1, and leptin levels were significantly lower in the experimental group than in the control group. The serum adiponectin levels were significantly higher in the experimental group than in the control group. Compared with the control group, the experimental group showed significant reductions in absolute abdominal subcutaneous fat, epididymal adipose tissue, kidney adipose tissue, intestine adipose tissue, and liver, kidney and spleen adipose tissue weights. The C/EBP-${\beta}$, leptin, and SREBP1c/ADD1 mRNA expression were significantly lower in the experimental group than in the control group, while the UCP-2 and adiponectin mRNA expression were significantly higher. Compared with the control group, the experimental group showed a significant reduction in the absolute adipocyte area in the liver and epididymal adipose tissue. Conclusion Jeoreongchajeonja-tang has an anti-obesity effect. Additional clinical studies are expected.

Effect of Mixture of Atractylodes macrocephala and Amomum villosum Extracts on Body Weight and Lipid Metabolism in High Fat Diet-Induced Obesity Model (고지방식이 유도 비만 모델에서 백출과 사인 추출 혼합물이 체중 및 지질대사에 미치는 영향)

  • In this study, we investigated the dose-dependent effects of mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts in a ratio of 3:1 on high fat diet (HFD)-induced obesity model. Oral administration of various concentrations with mixtures of AM and AV extracts in a ratio of 3:1 for 6 weeks inhibited HFD-induced increases of body, liver and epididymal fat weights in a dose-dependent fashions. Those effects may be mediated by decreased expressions of lipogenesis-related genes such as acetyl coA carboxylase (ACC) and fatty acid synthase (FAS) in liver. Also, increase of insulin and decrease of adiponectin in serum by HFD supply were inhibited by three different dosages of mixtures of AM and AV extracts in a ratio of 3:1. HFD supply induced increases of serum total cholesterol, triglyceride and LDL cholesterol. However, hyperlipidemia was significantly decreased in dose-dependent manners by treatment with mixtures of AM and AV extracts. Based on the results of the present study, hypolipidemic and anti-obesity effects by mixtures of AM and AV extracts were found in HFD-induced obesity model. Further clinical investigation is needed to develop anti-obesity therapeutic or preventive agents by using mixtures of AM and AV extracts.

The Anti-obesity Effects of Younggyechulgam-tang-ga Hwanggi on Obesity in Mice Induced by High Fat Diet (고지방식이 유도 비만생쥐에 대한 영계출감탕(苓桂朮甘湯) 가(加) 황기(黃芪)의 항비만 효과)

  • Objectives This study was conducted to experimentally evaluate the effects of Younggyechulgam-tang-ga Hwanggi(YGT) on obesity in mice induced by high fat diet. Methods The experiment was conducted with 4-week-old male mice divided into 5 groups. They were a normal diet group(Nor), a high fat diet group(Veh), a positive drug control group-orlistat 40 mg/kg(Oris), a 1.08 g/kg group(YGTL), and a 2.16 g/kg group(YGTH), and were tested for five weeks. Changes in antioxidant activity, body weight, organ weight, ROS, AST, ALT, TC, TG, HDL-C, LDL-C and lipid metabolism protein were checked. Results YGTL and YGTH group significantly reduced body weight compared to Veh group. YGTH group significantly reduced visceral fat weights compared to Veh group. In blood biochemistry analysis, ROS, AST, ALT, TC, TG and LDL-C in YGTL and YGTH group were significantly lower than Veh group. HDL-C increased significance in YGTL and YGTH group. In antioxidation protein analysis, Catalase, GPx and HO-1 have increased significantly in YGTL and YGTH group. YGTH group have increased $PPAR-{\alpha}$, p-AMPK compared to Veh group. but decreased FAS. SREBP-1, p-ACC levels in YGTL and YGTH group were decreased compared to Veh group, however CPT-1, UCP-2 levels in YGTL and YGTH group were increased compared to Veh group. Conclusions YGT has anti-obesity effects by regulating lipolysis and antioxidation in a diet-induced obesity model. Additional clinical studies are needed.

Effects of Combination Therapy with Anti-Obesity Herbal Medicine Including Ephedra Herba and Lorcaserin in Obese Patients: Two Case Reports (마황이 포함된 한방비만 처방과 Lorcaserin 병용 투여 후 호전된 비만 환자 치험 2예)

  • Although there has been many studies about the co-administration of herb formula and western medicine, there is none about obesity. The aim of this study was to report the effects of co-administration of herb formula containing ephedra sinica and lorcaserin on two obese patients for about 2~4 months. During the treatment, we checked body weight and body composition, and collected blood samples to evaluate liver, kidney, thyroid function and lipid profile. This treatment not only decreased body weight but also improved body composition and lipid profile in both patients. There was no hepatic or renal toxicity, nor any serious side effects of treatment. This study demonstrated that co-administration of herb formula containing ephedra sinica and lorcaserin is a safe and effective therapy to lose weight and improve metabolic parameters for obese patients. Further large-scale clinical trials are needed to evaluate the anti-obesity effect of the combination of herbal and western medicines.

Sulfuretin Prevents Obesity and Metabolic Diseases in Diet Induced Obese Mice

  • The global obesity epidemic and associated metabolic diseases require alternative biological targets for new therapeutic strategies. In this study, we show that a phytochemical sulfuretin suppressed adipocyte differentiation of preadipocytes and administration of sulfuretin to high fat diet-fed obese mice prevented obesity and increased insulin sensitivity. These effects were associated with a suppressed expression of inflammatory markers, induced expression of adiponectin, and increased levels of phosphorylated ERK and AKT. To elucidate the molecular mechanism of sulfuretin in adipocytes, we performed microarray analysis and identified activating transcription factor 3 (Atf3) as a sulfuretin-responsive gene. Sulfuretin elevated Atf3 mRNA and protein levels in white adipose tissue and adipocytes. Consistently, deficiency of Atf3 promoted lipid accumulation and the expression of adipocyte markers. Sulfuretin's but not resveratrol's anti-adipogenic effects were diminished in Atf3 deficient cells, indicating that Atf3 is an essential factor in the effects of sulfuretin. These results highlight the usefulness of sulfuretin as a new anti-obesity intervention for the prevention of obesity and its associated metabolic diseases.

Anti-Obesitic Effect of Mulberry Root-Bark (Mori radicis Cortex) in Zucker Rats with High Lipid Diet Induced-Obesity (고지방식이에 의해 비만이 유도된 Zucker Rats에서 상백피분말의 비만 개선효과)

  • This study was performed to investigate the anti-obesitic effect of mulberry root-bark on male and female Zucker FA/FA or FA/fa rats. Obesity in the rats was induced by feeding high-lipid diet contained 3% corn oil and 1% cholesterol. Experimental groups in male and female rats were assigned to normal diet group (normal control), high-lipid diet group(positive control) and 3% mulberry root-bark powder in high-lipid diet group (MRC). The mulberry treated-group showed decreases of body weight, FER (food efficiency ratio) value and lipid peroxidation in the liver and increase of high density lipoprotein cholesterol (HDL-C) value, compared with positive control groups. Our findings suggest that mulberry root-bark has a potential role in preventing or improving obesity from the following points of view, body weight, serum lipids and antioxidant enzyme activities.

Histological and Serological anti-Obesity Effect of Mori Folium in Obesity Induced SD Rat by High Fat Diet (상엽(桑葉)의 고지방식이 유도 비만 쥐에 대한 조직학적 및 혈청학적 항비만 효과 연구)

  • Objectives : Mori Folium is used for hyperlipidemia or diabetes. The study was designed to test the anti-obesity effect of Mori Folium on body weight, hepatic fat accumulation and serum lipid level. Methods : The extract from Mori Folium was made by the pharmacy department of Kyung-hee oriental medical hospital. Normal diet, high-fat diet, 30% reduced diet and Mori Folium groups were set. The normal group was administered normal rat food, but the other three groups were administered high fat food. We measured body weight once a week. After 3 weeks experiment, hepatic lipid accumulations were measured. Also we compared serum lipid levels among the 4 groups after 3 weeks. Results : Mori Folium had no effect on body weight, but the 30% reduced diet had an inhibitory effect on body weight gain. Mori Folium had more inhibitory effect on hepatic fat accumulation than a 30% reduced diet. Mori Folium also has more inhibitory effect on serum triglyceride than 30% reduced diet. Conclusions : Mori Folium has anti-obesity effect on hepatic lipid accumulaion and serum triglyceride level in high fat diet induced rats.

Hypolipidemic and Antioxidative Effects of Bangpoongtongsungsankamibang on Diet-Induced Obesity Rats (비만유도 흰쥐에 대한 방풍통성산가미방 (防風通聖散加味方)의 항고지혈 효과 및 항산화 효과)

  • Obesity is associated with a number of pathological disorders such as non-insulin-dependent diabetes, hypertension, hyperlipidemia, and cardiovascular diseases. Bangpoongtongsungsankamibang (BTSK) has been widely used in the oriental medicine for the treatment of several diseases associated with inflammatory abnormalities in cardiovascular and nervous system. The BTSK is the modified prescription of Bangpoongtongsungsankamibang in which sea tangle (Laminaria japonica) were added. This study was carried out to detemine the anti-obestic effects of BTSK. Pretreatment with the BTSK at daily dose of 100 or 200 mg/kg (p.o.) far 4 weeks reduced serum triglyceride, total cholesterol contents in rat induced by Poloxamer-407 or Triton WR-1339, respectively. Furthermore, post-treatment with BTSK far four weeks also inhibited body weight gain, adipose tissue mass and hyperlipidemia induced by the high fat diet for six weeks. The BTSK shifted serum total-, HDL- and LDL-cholesterol levels toward the values of normal group, suggesting that BTSK has hypolipldemlc effects. The rats fed BTSK reduced lipid peroxide and hydroxy radical in the rat blood and increased superoxide dismutase (SOD) activity compared to the control group. Taken together, these results superoxide that BTSK improve hyperlidemia and obesity via the upregulation of anti-oxidative mechanism.
