Herbicidal activity of a new benzenesulfonylurea compound, KSC-13906 (신규 benzenesulfonylurea 계 화합물 KSC-13906의 제초활성)

  • Herbicidal activities of newly synthesized experimental compound, KSC-13906 [Erythro N-{(4,6-dimethoxy pyrimidin-2-yl)aminocarbgnyl}-2-(2-fluoro-1- hydroxy-n-propyl)benzenesulfonamide, US Patent 5,461,025], were investigated in greenhouse and field for development as a paddy herbicide. KSC-13906 exihibited excellent weeding effects and selectivity to rice under submerged paddy condition in geenhouse. At the application rate of $2.5{\sim}5$ g ai/ha treated on surface of water, KSC-13906 effectively controlled three annual weeds such as Echinochloa crus-galli var. oryzicola. Scirpus juncoides, and Monochoria vaginalis as well as five perennial weeds such as Cyperus serotinus, Sagittaria pygmaea, Eleocharis kuroguwai, Potamogeton distinctus, and S. trifolia. However, there was no effect on Aneilema keisak and Lindernia procumbens. Especially, KSC-13906 effectively controlled Echinochloa crus-galli var. oryzicola at growth stages ranging from pre-emergence to 3-leaf stage. Antagonistic interaction was found between pyrazosulfuron-ethyl and mefenacet, but not between KSC-13906 ana mefenacet. KSC-13906 had high selectivity to transplanted rice, but caused phytotoxic effect on direct-seeded rice when treated before 3-leaf stage. Herbicidal activity of KSC-13906 on the Echinochloa crus-galli var. oryzicola was 7-folds greater than that of pyrazosulfuron-ethyl in terms of relative dose ratio of the herbicides. These results suggest that KSC-13906 possesses selective herbicidal potency in rice.

The isolation of Bacillus subtilis KYS-10 with antifungal activity against plant pathogens (식물 병원균에 대한 항진균 활성을 갖는 Bacillus subtilis KYS-10의 분리)

  • This study was investigated for the purpose of the isolation and identification of antagonistic bacteria with antifungal activity against plant pathogens. This bacteria denominated Bacillus subtilis KYS-10 and the optimum growth condition were 4% sucrose, 1% yeast extract, 0.2% $K_2HPO_4$, pH 7, 150 rpm, $30^{\circ}C$, 8 day. The antifungal activities against nine plant pathogens determined inhibition zone size by diffusion methods. The results, G. zeae (scab) 70 mm and P. grisea KACC 40439 (blast), P. capsici KACC 40177 (phytophthora blight) and C. destructans KACC 41077 (root rot of ginseng) 40~43 mm, and C. gloeosporioides KACC 43520 (ripe rot), C. gloesporioides KACC 40003 (anthracnose), S. shiraiana KACC 41065 (stem rot) and S. shiraiana (mulberry sclerotial disease) 35~39 mm and F. Oxysporum KACC 44452 (bulb rot of ginseng) 28 mm. From these experiment results, author suggest that Bacillus subtilis KYS-10 would be developed as a biological control agent thorough the field experimet in the near future.

Physical Characteristics and Antagonistic Effect of Ampelomyces (Ampelomyces의 생리적 특성 및 길항능력)

  • During the period of June, 2005 to May, 2008, 44 host plants infected with powdery mildew were collected in the Jeon-ju and Jang-su districts of Jeonbuk province and in the Jang-sung district of Jeonnam province. The hyperparasites, Ampelomyces were confirmed in 12 plant species. Most of the pycnidium shapes of Ampelomyces were circular or oval shaped, and the sizes were different even within the same host plant, and also the color of pycnidium was ranged from light brown to dark brown. Ampelomyces species were isolated from 4 hosts including Impatiens balsamina L., Cucurbita pepo, Rudbeckia laciniata var. elatier and Youngia sonchifolia, and thus the most appropriate 12 Ampelomyces strains for the current experiment were selected. When analyzing the selected 12 strains' incubational and nutritional characteristics, the malt extract agar was the most appropriate media. When investigating the effect of osmotic pressure on the spore germination, 0.15M NaCl concentration was the optimum germination concentration. When the isolated Ampelomyces sp. was tested in-vitro, it was found to be effective to control in other plant pathogens, isolated Ampelomyces showed no pathogenicity to the plant. strains isolated . studied on rDNA ITS sequence analysis. The rDNA ITS sequence data of Ampelomyces sp. isolate BSLAH16 from Impatiens balsamina L. were analyzed and identified.

Suppression Effect of Gray Mold and Late Blight on Tomato Plants by Rhamnolipid B (Rhamnolipid B에 의한 토마토 잿빛곰팡이병과 역병의 억제효과)

  • A Pseudomonas strain SG3 producing biosurfactant and showing antifungal and insecticidal activities was isolated from agricultural soil severely contaminated with machine oils. The antagonistic bacterium inhibited mycelial growth of all of the tested fungal pathogens. The fermentation broth of SG3 also effectively suppressed the development of various plant diseases including rice blast, tomato gray mold, tomato late blight, wheat leaf rust, barley powdery mildew and red pepper anthracnose. An antifungal substance was isolated from the fermentation broth of SG3 by ethyl acetate partitioning, silica gel column chromatography and preparative HPLC under the guide of bioassay. The chemical structure of the antifungal substance was determined to be rhamnolipid B by mass and NMR spectral analyses. The antifungal biosurfactant showed a potent in vivo antifungal activity against gray mold and late blight on tomato plants. In addition, rhamnolipid B inhibited mycelial growth of B. cinerea causing tomato gray mold and zoospore germination and mycelial growth of P. infestans causing tomato late blight. Pseudomonas sp. SG3 producing rhamnolipid B could be used as a new biocontrol agent for the control of plant diseases occurring on tomato plants.

Bacillus oryzicola sp. nov., an Endophytic Bacterium Isolated from the Roots of Rice with Antimicrobial, Plant Growth Promoting, and Systemic Resistance Inducing Activities in Rice

  • Biological control of major rice diseases has been attempted in several rice-growing countries in Asia during the last few decades and its application using antagonistic bacteria has proved to be somewhat successful for controlling various fungal diseases in field trials. Two novel endophytic Bacillus species, designated strains YC7007 and $YC7010^T$, with antimicrobial, plant growth-promoting, and systemic resistance-inducing activities were isolated from the roots of rice in paddy fields at Jinju, Korea, and their multifunctional activities were analyzed. Strain YC7007 inhibited mycelial growth of major rice fungal pathogens strongly in vitro. Bacterial blight and panicle blight caused by Xanthomonas oryzae pv. oryzae (KACC 10208) and Burkholderia glumae (KACC 44022), respectively, were also suppressed effectively by drenching a bacterial suspension ($10^7cfu/ml$) of strain YC7007 on the rhizosphere of rice. Additionally, strain YC7007 promoted the growth of rice seedlings with higher germination rates and more tillers than the untreated control. The taxonomic position of the strains was also investigated. Phylogenetic analyses based on 16S rRNA gene sequences indicated that both strains belong to the genus Bacillus, with high similarity to the closely related strains, Bacillus siamensis KACC $15859^T$ (99.67%), Bacillus methylotrophicus KACC $13105^T$ (99.65%), Bacillus amyloliquefaciens subsp. plantarum KACC $17177^T$ (99.60%), and Bacillus tequilensis KACC $15944^T$ (99.45%). The DNA-DNA relatedness value between strain $YC7010^T$ and the most closely related strain, B. siamensis KACC $15859^T$ was $50.4{\pm}3.5%$, but it was $91.5{\pm}11.0%$ between two strains YC7007 and $YC7010^T$, indicating the same species. The major fatty acids of two strains were anteiso-$C_{15:0}$ and iso $C_{15:0}$. Both strains contained MK-7 as a major respiratory quinone system. The G+C contents of the genomic DNA of two strains were 50.5 mol% and 51.2 mol%, respectively. Based on these polyphasic studies, the two strains YC7007 and $YC7010^T$ represent novel species of the genus Bacillus, for which the name Bacillus oryzicola sp. nov. is proposed. The type strain is $YC7010^T$ (= KACC $18228^T$). Taken together, our findings suggest that novel endophytic Bacillus strains can be used for the biological control of rice diseases.

  • For weeding on the after-barley cropping soybean, the tank-mix use of postemergence herbicides acifluorfen and haloxyfop-methyl was tested. Acifluorfen has shown the transient leaf-burn on the newly formed soybean leaves, but no growth inhibitions. Under the treatment of acifluorfen only, grassy weeds occurred successively and explosively. However, under the combinated spraying of both herbicides, the grassy weeds could be controlled in a slight antagonistic, the broadleaved weeds in additive, and the total weed species including sedges in synergic tendencies, respectively. Also, to achieve the full-season weeding efficacies for the prominent crop yields, both herbicides are suggested to be mixed with more dosages than 0.245 kg ai/ha of acifluorfen and 0.15 kg ae/ha of haloxyfop-methyl, respectively.

  • Interactions of herbicide mixtures were assessed for effective control of annual and perennial paddy weeds by isobole method(90% control) using Echinochloa crusgalli Beauv. var. oryzicola Ohwi, Scirpus hotarui Ohwi, Sagittaria pygrnaea Miquel, and Cyperus serotinus Rottb which are dominated in the paddy field of Korea. Mixture of butachlor and pyrazolate showed additive effect for control of E. crusgalli Beauv. var. oryzicola Ohwi, S. hotarui Ohwi and C. serotinus Rottb, but synergistic effect for control of S. pygmaea Miquel. Interaction of bifenox and bromobutamide showed synergistic effect to E. crusgalli Beauv. var oryzicola Ohwi and C. Serotinus Rottb, but slightly antagonistic effect to S. pygmaeo Miquel.

Comparison of Herbicidal Action between Pyrazosulfuron - ethyl and Imazaquin (Pyrazosulfuron - ethyl과 Imazaquin의 살초작용 비교)

  • To know whether pyrazosulfuron-ethyl(PYR) and imazaquin(IMA), known as a acetolactate synthase(ALS) inhibitors, have a same herbicidal action pattern in rice(Oryza sativa) or barnyardgrass (Echinochloa crus-galli), an inhibition pattern and a response characteristics in combination with dymron or butachlor were investigated. In contrast to the phytotoxicity of rice treated with IMA, the one treated with PYR was completely tended to be recovered after 25 days after treatment. Safening effect of dymron against PYR was effectively developed to transplanted-rice, while such an effect was not shown in combination with IMA. In combination with PYR and butachlor, antagonistic effect was observed in both simultaneous or sequential treatment on bamyardgrass, however, additive effect was rather shown in combination with IMA and its activity was dominantly dependent on the first applied compound. $I_{50}$ of PYR and IMA on the ALS extracted from barnyardgrass was $4{\times}10^{-7}$M and $2.8{\times}10^{-6})$M, respectively. Butachlor did not affect their activities on ALS in vitro. These results suggest that PYR and IMA might have a different action each other in the pathway to a final herbicidal activity even though their primary action site is ALS.

Development of Biofungicide Using Bacillus sp. KBC1004 for the Control of Anthracnose of Red Pepper (길항세균 Bacillus sp. KBC1004를 이용한 고추탄저병의 생물학적 방제제 개발)

  • To develop an effective biopesticide to control pepper anthracnose disease, an isolate which showed strong inhibitory effect on the mycelial growth and conidial germination of Colletotrichum acutatum was selected among the antagonistic bacterial isolates collected from pepper grown soil. The bacterial isolate was identified as Bacillus sp. KBC1004 using 16S rRNA sequence analysis. The liquid culture of KBC1004 was freeze-dried and formulated as a wettable powder(WP). The wettable powder form of KBC1004 required at least 24 hours to activate and to inhibit the conidial germination of C. acutatum. In vitro bioassay using the detached green pepper fruits, biocontrol activity of the WP was not recognizable in simultaneous inoculation, but significant disease suppression was observed pre-treatment (24 hr) of the WP before pathogen inoculation. In field experiment, 4 times foliar applications of the 1/500 diluted wettable powder from the end of June showed great control efficacy similar to that of the chemical fungicide application. These results suggest that the formulated WP product could be an alternative mean to control of pepper anthracnose disease in environmentally friendly farming practices.

The Effect of female Hormone on Knee Joint Laxity (여성 호르몬이 무릎의 느슨함(laxity)에 미치는 영향)

  • The purpose of this study was to determine whether hormone levels change knee laxity in healthy females. Twenty three healthy females were recruited for the study. Serum estradiol and progesterone levels were recorded three times during the subjects' menstrual cycles. The first measurements were taken between day 3 and 7 of the follicular phase and the second data collection coincided with ovulation, 24 to 48 hours after the estrogen surge detected by an ovulation predictor kits. Based on a 28 day cycle, the third data collection occurred approximately 7 days later during the luteal phase. Knee joint laxity was recorded at the same intervals with a KT 2000 arthometer. Hormone levels and phases were compared to passive knee joint laxity with multiple regression analysis. Estradiol and progesterone levels differed significantly across the three tests. Knee joint laxity increased during ovulation. Based on a multiple regression analysis, estradiol and progesterone levels predicts 77.9% to 80.9% of the laxity at 20lb and 30lb loads. An antagonistic relationship between estradiol and progesterone was found when testing for knee laxity. Serum hormone levels have moderate power in predicting knee joint laxity. Individual hormonal profiling in female athletes would allow researchers to access the structural properties of the ACL, such as the laxity which may provide beneficial information to understand female ACL injury mechanism in sports activity.
