Anti-inflammatory Effect of Biotin and Plant extracts

  • Biotin is a water-soluble vitamin used as a skin conditioning agent and promotes the formation of intercellular lipid layers through increased lipid synthesis, which improves the skin's natural barrier function. The anti-inflammatory effects of biotin have been investigated using in vitro assay models, such as MTT assay, measurements of concentrations of nitric oxide(NO), prostaglandin E2(PGE$_2$), and inhibition rate of 5-lipoxygenase(5-LOX). In comparison with biotin, other plant extracts were tested at the same time which were kudzu vine extract, sage extract, paeonia extract, and dipotassium glycyrrhetinate. Nitric oxide is a signal molecule with functions such as neurotransmission, local vascular relaxation, and anti-inflammation in many physiological and pathological processes. NO can cause apoptosis and necrosis of target cells such as keratinocytes and is generated from L-arginine by nitric oxide synthase (NOS). Prostanoids, including prostaglandins and thromboxanes, are generated by the phospholipase $A_2$/cyclooxygenase(COX) pathway, and leukotrienes are generated by the 5-lipoxygenase pathway from arachidonic acid. Prostaglandin E2 recently have been shown to be beneficial in the resolution of tissue injury and inflammation, also has been implicated as an immunosuppressive agent and plasma levels of PGE$_2$ are elevated in patients sustaining thermal injury. Lipoxygenase metabolites from arachidonic acid have been implicated in inflammation, anti-inflammatory activity of the raw materials was evaluated in vitro by the offered inhibition of lipoxygenase.

만분방(慢盆方)이 염증 관련 cytokines의 유전자 발현과 생성량에 미치는 영향 (The Experimental Study on Anti-inflammatory Effect of Manbunbang)

  • Purpose: This study was performed to evaluate the anti-inflammatory effect of Manbunbang extract (MBB). Methods: In order to understand the mechanism of anti-inflammatory effect of MBB, expression of cytokines and its levels in RAW 264.7 cell lines, as well as changes of cytokine gene expressions in serum, spleen, and liver tissues in acute inflammation induced mouse model were investigated. Results: 1. MBB significantly suppressed the expression levels of IL-1${\beta}$, TNF-${\alpha}$ and COX-2 mRNAs at 100 and 50 ${\mu}$g/m${\ell}$ concentrations, and IL-6 and NOS-II genes at 100, 50 and 10 ${\mu}$g/m${\ell}$ concentrations in RAW 264.7 cell lines, compared to those of the control. 2. MBB significantly reduced the production level of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at 100 and 50 ${\mu}$g/m${\ell}$ concentrations in RAW 264.7 cell lines compared the those of the control. 3. MBB significantly reduced the production of IL-1${\beta}$, IL-6 and TNF-${\alpha}$ levels in sera of acute inflammation induced mice. 4. MBB significanlty suppressed the expression level of IL-1${\beta}$, TNF-${\alpha}$ mRNA in spleen tissues as well as IL-6 mRNA in liver tissues in acute inflammation induced mice. Conclusion: From the results above, anti-inflammatory effect of MBB through its immune regulation could be experimentally explained. Wide treatment of inflammatory diseases such as pelvic inflammation using MBB are recommended.

청열해독화어탕(淸熱解毒化瘀湯)의 항혈전(抗血栓) 및 항염(抗炎)효과에 관한 실험적(實驗的) 연구(硏究) (The Experimental Study on Anti-thrombotic and Anti-inflammatory Effect of ChungyeolHaedogHwaeoTang(CHHT))

  • This study was performed to evaluate anti-thrombotic activities and anti-inflammatory effects of ChungyeolHaedogHwaeoTang water extract(CHHT). The results were summarized as follows. In experiment of anti-thrombotic effect; 1. CHHT inhibited human platelet aggregation induced by ADP and epinephrine as compared with the control group, and inhibited pulmonary embolism induced by collagen and epinephrine (inhibitory rate is 37.5%). 2. CHHT increased platelet number significantly, and also CHHT shortened PT and APTT significantly as compared with the control group in thrombus model induced by dextran. In experiment of anti-inflammatory effect; 3. CHHT inhibited $IL-1{\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II mRNA expression as compared with the control group in a concentration-dependent degree, and inhibited NO production significantly at 50, 100 ${\mu}g/ml$, and also inhibited ROS production in a concentration-dependent degree as compared with the control of group in RAW 264.7 cell line. 4. CHHT inhibited $IL-1{\beta}$, IL-6 and TNF-${\alpha}$ production significantly in serum of acute inflammation-induced mice, and decreased $IL-1{\beta}$, IL-6 and TNF-${\alpha}$ production in spleen tissue, and also decreased $IL-1{\beta}$, and IL-6 production in liver tissue, but increased TNF-${\alpha}$ production in liver tissue of acute inflammation-induced Balb/c mice. 5. CHHT increased survival rate from the 3rd day in ICR mice with lethal endotoxemia induced by LPS. These results suggest that CHHT can be useful in treating diverse female diseases caused by thrombosis and inflammation such as menstrual pain, menstrual disorder, leukorrhea, vaginitis, cervicitis, pelvic inflammatory disease and so on.

The effect of Saururus chinensis Baill against oxidative damage and inflammation

  • Objectives : ROS are involved in a wide spectrum of diseases including chronic inflammation and cancer. S.chinensis Baill, a perennial herb commonly called Chinese lizard's tail or Sam-baek-cho in Korea, is used for the treatment of edema and inflammatory diseases in the Oriental folk medicine. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the two extracts, water(WE) and ethyl acetate(EAE) from S.chinensis Baill. Methods : Anti-oxidant activity was evaluated using Fe2+ chelating and hydroxyl radical scavenging assay. DNA cleavage assay, and western blot and immunostaining for phospho-p65 were performed to evaluate anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell. Results : In Fe2+ chelating activity and hydroxyl radical scavenging activity, WE showed more strong scavenging activity for hydroxyl radical than EAE. WE scavenged hydroxyl radical by 12% at 3.2 ${\mu}g/ml$, 21% at 16 ${\mu}g/ml$, 32% at 80 ${\mu}g/ml$, 66% at 400 ${\mu}g/ml$ and 82% at 2000 ${\mu}g/ml$, respectively. In addition, WE showed more strong chelating activity than EAE. WE chelated Fe2+ ion by 1.1% at 3.2 ${\mu}g/ml$, 8.2% at 16 ${\mu}g/ml$, 26.3% at 80 ${\mu}g/ml$, 72% at 400 ${\mu}g/ml$ and 89% at 2000 ${\mu}g/ml$, respectively. Also, WE inhibited oxidative damage via its anti-oxidant activity. In anti-inflammatory effect, EAE inhibited NO production and iNOS expression. In addition EAE suppressed the NF-${\kappa}B$ and MAPK signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, these data indicate that S. chinensis Baill, shows anti-oxidant activity and anti-inflammatory effect.

가감선방활명음(加減仙方活命飮)의 항염작용(抗炎作用)에 대한 실험적(實驗的) 연구(硏究) (The Experimental Study on anti-inflammatory Effects of Kagamsunbanghwalmyungeum(KSBH))

  • Purpose : This study was to investigate the anti-inflammatory effects of Kagamsunbanghwalmyungeum(KSBH) which has been medicated the patient such as breast abscess. Methods : In the study of anti-inflammatory effects, KSBH was investigated using cultured cells and a murine models. As for the parameters of inflammation, level of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were determined in mouse lung fibroblast cell(mLFC) and RAW 264.7 cells. Results : The cytotoxicity on mouse lung fibroblast cells(mLFC) was not served at all concentration of KSBH. KSBH in RAW264.7 cell inhibited IL-l${\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II mRNA genes expression in a concentration-dependent manner, and specially inhibited NOS-II production very significantly at 100${\mu}$g/ ml. KSBH inhibited NO production significantly in a concentration-dependent manner and ROS production in a concentration-dependent manner. KSBH inhibited IL-l${\beta}$, IL-6 and TNF-${\alpha}$ production significantly in serum of acute anti-inflammation-induced mice. KSBH increased the survival rate from the 3rd day on LPS-induced lethal endotoxemia. Conclusion : These results suggest that Kagamsunbanghwalmyungeum(KSBH) can be useful in treating a lot of women mammary diseases caused by inflammation such as breast abscess.

쿠퍼 세포에서 Nrf2 활성화 매개 죽력의 염증 및 인플라마좀 억제 효능 (Anti-inflammation and Anti-inflammasome Effects of Bambusae Caulis in Liquamen mediated by Nrf2 Activation in Kupffer cells)

  • Objectives : Bambusae Caulis in Liquamen (BCL), a traditional herbal medicine, is a distilled product of condensation from the burning of fresh bamboo stems. We previously identified the anti-oxidant capacity of BCL in hepatocytes and suggested that BCL is a promising therapeutic candidate for treating oxidative stress-induced hepatocellular damage. Despite the importance of the role played by Kupffer cells in liver disease, the efficacy of BCL on Kupffer cells is unclear. Therefore, this study aimed to determine whether BCL could suppress LPS-induced inflammation and LPS+ATP-induced inflammasomes in Kupffer cells. Methods : We used ImKCs, a murine immortalized Kupffer cell line to examined whether BCL inhibited LPS-induced inflammation response and oxidave stress. And, we prepared a total of 18 L of BCL, purchased from Bamboo Forest Foods Co., Ltd. (648 Samdari, Damyang-eup, Damyang-gun, Jeollanam-do, Republic of Korea), was concentrated using a decompression concentrator. Result : The LPS-induced release of inflammatory cytokines was abolished by BCL treatment. Also, BCL treatment suppressed the LPS+ATP-induced expression of inflammasome proteins (NLRP3, IL-1, and IL-18), and inhib β ited the release of IL-1 . BCL decreased LPS-or LPS+ATP-induc β ed reactive oxygen species production. In addition, BCL increased nuclear translocation of Nrf2 and the expression of HO-1 in a time-dependent manner. Conclusion : These results suggest the efficacy of BCL with respect to its anti-inflammatory and anti-inflammasome effects mediated by Nrf2 in Kupffer cells.

益氣化瘀湯加味方이 고지방 식이로 유발된 생쥐의 항고지혈 및 항산화에 미치는 효과 (Anti-hyperlipidemic and Anti-oxidant effects of Kgihwaetang-gamibang in High Cholesterol Diet-induced Mice Model)

  • This study was conducted to investigate the anti-hyperlipidemic and anti-oxidative activities of Ikgihwaetang-gamibang (IK). We observed the effect of IK on lipid factor, inflammation factor, body weight and organ weight, and liver after administering IK to hyperlipidemia mice triggered by high fat diet. Additionally, we used Raw 264.7 cells to investigate the changes in anti-oxidative activity and the effects of anti-inflammation. Administration of IK resulted in reduction of total cholesterol, LDL cholesterol, TG, and glucose in blood serum while a clear increase in HDL cholesterol. The production of IL-1β and IL-6, and ROS and NO also decreased. The results showed that IK has anti-hyperlipidemic and anti-oxidative effects, and suggested its potential use in prevention and treatment of hyperlipidemia and its associated diseases.

  • Purpose: This study was performed to evaluate anti-inflammatory effects of Cheongyeoljohyeoltangkamibang water extract (CYJHT). Methods: In the study of anti-inflammatory effects. CYJHT was investigated using cultured cells and murine models. As for the parameters of inflammation. levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were determined in mouse lung fibroblast cells(mLFCs). RAW 264.7 cells and acute inflammation-induced mice. Results: 1. CYJHT showed a safety in cytotoxicity and toxicity of liver. 2. CYJHT effected scavenging activity on 2.2-diphenyl-1-picrylhydrazyl(DPPH) free radical, superoxide dismutase(SOD) and superoxide anion radical(SAR). 3. CYJHT in RAW 264.7 cell decreased IL-l$\beta$ mRNA expression at 100, 50 ${\mu}g$/ml and also decreased TNF-$\alpha$ mRNA expression at 100 ${\mu}g/ml$ and decreased COX-2. NOS-II mRNA expression and decreased IL-6 mRNA expression in a concentration-dependent manner. 4. CYJHT in RAW 264.7 cell decreased IL-l$\beta$ significantly at 100, 50 ${\mu}g$/ml and decreased IL-6. TNF-$\alpha$ significantly at 100 ${\mu}g$/ml. 5. CYJHT inhibited IL-l1$\beta$, IL-6 and TNF-$\alpha$ production significantly in serum of acute inflammation-induced mice. 6. CYJHT decreased IL-1$\beta$, IL-6 and TNF-$\alpha$ mRNA production significantly in spleen tissue. and also decreased IL-l$\beta$. TNF-$\alpha$ mRNA production significantly in liver tissue of acute inflammation-induced mice. Conclusion: These results suggest that CYJHT can be useful in treating diverse female diseases caused by inflammation such as menstrual pain. menstrual disorder. leukorrhea. pelvic inflammatory disease and so on.

지부자(地膚子)의 신생혈관 및 염증매개 단백질 발현에 미치는 영향 (Effects of Kochiae Fructus Extracts on the Expression of Angiogeneis and Inflammation Related Proteins)

  • Hypoxia induced angiogenesis and inflammation are essential processes for metastasis and progress of solid tumors. We examined the anti-angiogenic and inflammation related activity of Kochiae Fructus (KF) extract. To investigate the roles of the KF extract, we performed MTS assay, western blots using HaCaT cells and $HepG_2$ cells. The results are as follows. The protein level of $HIF-1{\alpha}$ was reduced when induced by $CoCl_2$ in $HepG_2$ cells treated with KF extract and induced by IGF-11 in HaCaT cells treated with KF extract. KF extract reduced the mRNA level of VEGF in HaCaT cells and KF extract reduced the protein level of iNOS in HaCaT cells. These results suggest that KF extract contributes to the anti-angiogenic and anti-inflammatory activities and also we could assume that KF extract act as antioxidant or anti-inflammatory agents via reduction of $HIF-1{\alpha}$.

육계 유래 Cinnamaldehyde의 투여와 항염증 및 진통효과의 평가 (Antinociceptive and Anti-inflammatory Properties of Cinnamomum cassia Derived-cinnamaldehyde in Rodents)

  • The aim of present study was to investigate the possible influence and the related mechanism of Cinnamomum cassia derived-cinnamaldehyde on the inflammation or nociception. Cinnamomum cassia was referred to be treated for common cold or dyspepsia in the traditional medicine. However, there are no reports on the antinociceptive or anti-inflammatory properties of cinnamaldehyde, the primary ingredient of Cinnamomum cassia. We hypothesized that cinnamaldehyde would play a role in the modulation of inflammation or nociception evoked by carrageenan, acetic acid or heat. Male Institute of Cancer Research mice were used and the size of edema, frequency of writhing and latency of abnormal behaviors such as licking, flicking, shaking or jumping were measured and recorded. The present study was carried out to evaluate the antiinflammatory and antinociceptive effects of cinnamaldehyde. The administration of cinnamaldehyde (30 and 100 mg/kg) inhibited carrageenan-induced paw edema only at the final phase, suggesting the blockade of synthesis or release of prostaglandins. It also reduced the frequency of the acetic acid-induced writhing reflex in mice. In addition, the administration of cinnamaldehyde prolonged the latency for extraordinary reaction at the hot plate in mice. In conclusion, cinnamaldehyde has anti-inflammatory and analgesic properties and is a potential therapeutic for inflammation and nociception.
