The Screening of Antibiotics Resistance Inhibition of Herb Drugs entered in Korean Official Formulary (I) (공정서 수재생약 정유의 항생제 내성억제작용 검색(I))

  • The essential oils from 73 aromatic herb drugs entered on Korean official formularies were tested for antibioticresistance inhibitory effect. When the oils were combined with ampicillin (Am) or amoxicillin (Amx) they showed significant inhibitory effects on the growth of multi-drug resistant Staphylococcus aureus SA2 in considerably low concentration. The most effective combination were oils from Magnoliae Flos or Piperis longi Fructus ($25{\mu}g/mL$)-Am or Amx ($4{\mu}g/mL$).

Effect of Water Flux on the Determination of Membrane Permeability Using Single-pass Perfusion

  • The single-pass perfusion technique was employed in order to investigate the effect of water flux from the rat jejunum in the normal experimental conditions. Our results suggested that water flux below ${\pm}0.75%/cm$ of jejunal length was considered normal. Water flux was $-0.131\;{\pm}\;0.311%/cm$ of jejunal length in a citrate buffer and should be corrected in order to determine the permeabilities of the compounds. Perfusion rate up to 0.5 ml/min had no effects on the permeability of ampicillin. Neither the effective permeabilities nor the wall permeabilities of aminopenicillins were influenced by water flux during experiments in rats.

  • Biological determination of Pivampicillin hydrochloride was examined and an available method has been established. Unhydrolyzed Pivampicillin hydrochloride has no antibacterial activity, but it exhibits an antibacterial activity hydrolyzed with liberation of ampicillin by nonspecific enzymes present in blood and tissues from various species in vitro and in vivo. Using these properties, Pivampicillin hydrochloride can be determined as ampicillin by use of an agar-diffusion method with Bacillus subtilis as the assay organism.

  • The adsorption of erythrosine in the aliphatic alcohols by antibiotics such as ampicillin anhydrous, ampicillin trihydrate, amoxicillin trihydrate, cephalexin monohydrate was studied. The adsorption isotherms were all described with Freudlich equation. From the various experimerts, it was found that the adsorption of erythrosine by antibiotics in the aliphatic alcohols was affected by the solubility of adsorbents. The adsoption in the mixture of aliphatic alcohols and water is related with the dielectric constant of mixed solvents. According to increase in the pH values of aqueous solution, the adsorption of erythrosine for antibiotics is decreased, but increased in the mixture of aliphatic alcohols and water. The adsorption of erythrosine in the suspension of antibiotics showed a correlation with the temperature. Colloidal silica which is used as stabilizer had a favorable effect on the adsorption of erythrosine in the suspension of antibiotics.

A Trend of Antibiogram of Escherichia Coli Detected by VITEK ESBL Test in Recently Two Years

  • Extended spectrum β-lactamase (ESBL) generated Enterobacteriaceae including Escherichia coli is responsible for resisting antibiotics, which is clinical problem. This study was performed to investigate the isolation rates of 111 strains having positive of VITEK ESBL test 111 ESBL-strains in each month and season and statistically to determine their patterns of antibiotic test. One hundred eleven ESBL-strains were collected among 1,688 strains of E. coli isolated from various clinical specimen of one general hospital in Busan during 2002 to 2003. Month rates of ESBL-strains were 0% to 13.3%, while the seasonal rates were highest at autumns during two years. The resistance to ampicillin, cefazolin, azteronam, ceftriaxone, and cefepime were 100% in 2002. In ampicillin and cefazolin the resistances were 100% during the two years. There were significant differences of cefoxitin and piperacillin/tazobactam between the two years but not significant between specimens. Four groups were divided according to the pattern of resistance and then the highest group had 93∼100% of the resistance to 8 drugs but not resistant to imipenem.

  • Identification and antibiotic susceptibility of Clostridium perfringens isolates from fecal specimens of healthy sheep and dogs were performed from December, 1995 to November, 1996 in Kwang-ju and Chonnam area. C. perfringens was isolated in 3 strains(15.0%) out of 20 healthy sheep and 2 strains(6.7%) out of 30 healthy dogs. In antibiotic susceptibility test of C. perfringens, 80% of the isolates was susceptible to ampicillin, baytril, and penicillin, 60% to cephalothin, and 40% to erythromycin.

  • We monitored antibiotic resistance of Escherichia coli isolates from healthy volunteers of community and chicken butchers from February to July in 2006. From disc diffusion test on 473 E coli isolates from healthy volunteers of the community, the resistance rates of tetracycline, ampicillin, and ticarcillin were 44.8%, 40.8%, and 37.4%, respectively. In the disc diffusion test on 18 E coli isolates from chicken butchers, the resistance rates of tetracycline, nalidixic, streptomycin, and ampicillin were 94.4%, 61.1%, 55.6%, and 50.0%, respectively.

Polymorphism in Intrinsic Antibiotic Resistance of Azospirillum Isolates from Ornamental Plants

  • The polymorphism of Azospirillum isolates from ornamental rhizosphere and two reference strains were examined with respect to intrinsic antibiotic resistance (IAR) profile. All the isolates showed different intrinsic resistances to different antibiotics viz., tetracycline, kanamycin, nalidixic acid, streptomycin, ampicillin, spectinomycin and chloramphenicol. All the strains demonstrated susceptibility to high concentration of all antibiotics used in the present experiment. In addition to these general patterns, we also obseved the multiple antibiotic resistances of Azospirillum strains. The Azospirillum sp. OAD-11 was resistant to tetracycline, streptomycin and ampicillin, and Azospirillum sp. OAD-57 was resistant to tetracycline and streptomycin. Conversely, Azospirillum sp. OAD-9 possessed the dual susceptibility to tetracycline and spectinomycin, whereas Azospirillum sp. OAD-37 was dual susceptible to streptomycin and kanamycin. Such multiple antibiotic resistant/susceptible traits could be useful for the identification of the strains in field experiments or in molecular genetic transfer experiments.

  • Thirty six essential oils from herb drugs entered in Korean official formulary, which are frequently used in oriental region, were tested for antibiotic resistance inhibition. When the oils were combined with ampicillin (Am) or amoxicillin (Amx) they showed significant inhibitory effects on the growth of multi-drug resistant Staphylococcus aureus SA2 in considerably low concentration. The most effective combinations were oils from Acanthopanacis Cortex ($0.49{\mu}g/mL$) with Am and Cnidii Rhizoma and Lonicerae Flos (2.77 and $2.79{\mu}g/mL$, respectively) with Amx as shown in minimum resistance inhibitory concentrations.

Antibiotics susceptibility of Proteus mirabilis isolated from domestic animals in Chonbuk province (동물에서 분리한 Proteus mirabilis의 항생제 감수성에 관한 연구)

  • Isolates of 70 Proteus mirabilis were tested against 10 different antibiotics by a disc diffusion method as recommended by National Committee for Clinical Laboratory Standards (NCCLS). The isolates were resistant in order of tetracycline(100.0%), enrofloxacin(95.7%), ampicillin(74.3%), choramphenicol(62.9%), cephalothin(58.6%), streptomycin(50%), kanamycin(47.2%), neomycin(35.8%), gentamicin(15.7%), and amikacin(2.9%). In the antibiotic resistant patterns, 37 kinds of multiple resistance patterns of P mirabilis isolates were detected. The highest resistant pattern was ampicillin-cephalothin-chloramphenicol-enrofloxacin-tetracycline(11.6%).
