Survey on Fungal Diseases of Ornamental Plants (I) (관상식물(觀賞植物)의 진균병(眞菌病) 발생조사(發生調査) (I))

  • The 34 species of the ornamental plants belong to 19 families which seemed to be infected by fungal pathogens were collected at 6 localities in Korea. The diseases were diagnosed with their symptoms, the characteristics of the pathogens and the pathogenicity on host plants. The results were summarized as follows : The 37 diseases by 16 genera of fungal pathogens were investigated on 32 species of 32 genera of the ornamental plants. Among them 22 diseases by 11 genera of fungal pathogens were turned out to be new diseases which have not been described on the host plants of 19 species in Korea.

  • The 32 species of the shade trees and ornamental plants belong to 23 families which seemed to be infected by fungal pathogens were collected at 5 localities in Korea. The diseases were diagnosed with their symptoms, the characteristics of the pathogens and the pathogenicity on host plants. The results were summarized as follows ; The 45 diseases by 21 genera of fungal pathogens were investigated on 30 species of the shade trees and ornamental plants. Among them 22 diseases by 13 genera of fungal pathogens were turned out to be new diseases which have not hitherto been described on the host plants of 16 species in Korea.

Effect of Light Source on Organic Acid, Sugar, and Flavonoid Concentrations in Buckwheat

  • The major free sugars of buckwheat plants were fructose, glucose, and maltose but their contents and compositions were influenced by the different wavelength of light. Free sugar contents of Clfa 39 (Fagopyrum tataricum) were higher than those of Yangjul-maemil (Fagopyrum esculentum) regardless of the light sources. As treated with red and blue light, the free sugar contents in the leaves of buckwheat plants were slightly increased, but their contents in the stems and flowers were lower than those of natural light condition. Under the natural light condition, maltose was detected in every tissues of buckwheat plants, but as treated with blue and red light, it was not detected in the flowers of buckwheat plants. Citric, malic and acetic acid were detected as major organic acids in buckwheat plants. Red and blue lights decreased the total organic acid contents in buckwheat plants as compared with natural light condition. It was considered that blue light are less active than red light for the accumulation of organic acids. Tataric acid was detected only in the leaves of buckwheat plants, however, as treated with red and blue light, it was not detected in the leaves of Clfa 39. Flowers of Yangjul-maemil contained a considerable amount of rutin and quercitrin. Only small amount of quercitrin was detected in leaves, but it was not detected in stems. On the other hand, Clfa 39 leaves contained a considerable amount of rutin, quercetin and small amount of quercitrin, but quercitrin and quercetin were detected only in the stems of Clfa 39. Red and blue lights significantly decreased the contents of rutin, quercitrin, and quercetin in buckwheat plants as comparing with natural light condition. Rutin content in the flowers of Clfa 39 was increased under the red and blue light conditions.

New Disese of Wheat and Barley Caused by Fusarium (Calonectria) nivale in Korea (Fusarium(Calonectria) nivale에 의한 맥류의 신병해)

  • Early in the growing season of 1976, rain and cool weather favored the blighting of leaves of young plants of barley by Fusarium nivale. The fungus was recovered in culture from infected foliage, and the perithecia of Calonectria nivalis were demonstrated to be present as well as the Fusarium state. On 22 April 1976, in Suweon, plants pulled at random revealed stem lesions from which F. nivale was cultured. On S May 1976 near Kwangju. Perithecia were found embedded within leaf sheaths and blades of mature wheat and barley plants. It was evident in the 1976, 1977 season that Fusarium(Calonectria) nivale was common but unrecognized as an early season pathogen of barley in Korea. The probable source of primary inoculum was the infected refuse from the previous season's barlry and wheat crops. Stem lesions caused by this fungus were considered to he detrimental to the maximum yield of barley or wheat.

  • The monitoring system for integrated management of on-site wastewater treatment plants(biofilter) was designed and its operation program was developed. In design process, the research on monitoring parameters which will be able to represent condition and operation of the pilot plants was accomplished, and these parameters came to reveal with ORP(Oxidation-Reduction Potential), water level, pump and power on/off. Proposed monitoring system is composed with measurement, control, communication and display device, and PCB(Prototype Circuit Boards) and microcontroller (PIC16F877) technique are applied to its design of control device for performing specific function. also, The operation program of PC setup is developed in order to provide a convenience to the manager.

A Simple and Reliable Method for Preparation of Cross-Contamination-Free Plant Genomic DNA for PCR-Based Detection of Transgenes

  • A simplified but reliable method was developed for the polymerase chain reaction (PCR)-based detection of genetically modified (GM) plants. The modified CTAB (mCTAB) method enabled us to prepare a high quality of genomic DNA from several hundred plant leaf samples in one day. Using DNA samples prepared from seven dicots and two monocots, approximately 1.75-kb regions spanning 17 S to 25 S ribosomal RNA genes were successfully amplified in a 2X PCR pre-mix containing BLOTTO. Further fidelity assessment of the mCTAB method by PCR analysis with Roundup Ready soybean (RRS) and non-RRS plants showed that the DNA samples prepared alternately from each of two lines were evidently free of cross-contamination. These results demonstrate that the mCTAB method is highly recommended for the rapid detection of transgenes in large numbers of leaf samples from diverse transgenic plants.
