Detection of citrus-infecting viruses and sequence analysis of Satsuma dwarf virus(SDV) and SDV-CiMV in Jeju island

  • To investigate occurrence and variability of satsuma mandarin ( Citrus unshiu)-infecting viruses in Jeju island, several sets of diagnostic RT-PCR primers were designed and applied to samples collected randomly. Each primers set used in this survey was designed to detect Satsuma dwarf virus (SDV, Sadwavirus) and Citrus mosaic virus (CiMV) which is reclassified as an isolate of SDV (SDV-CiMV, Saduavirus). RT-PCR methods could detect SDV-CiMV and CTV from leaf . samples of unshui citrus. CTV was the prevalent and SDV-CiMV was not common in Jeju island. RT-PCR product of SDV-CiMV-JJl2 were cloned and sequenced. Sequence of the isolate revealed that it was 96.9 % identical to SDV-CiMV-Jp isolate at the nucleotide level. SDV-CiMV-JJl2 was propagated on Physalis floridana and sequencing of entire sequences of genome is in progress. Variability of SDV in Jeju island was confirmed by sequence comparisons and restriction mapping analysis.

업사이클링 푸드의 소비가치가 소비자의 구매태도와 구매의도에 미치는 영향 (A Structural Relationship among Consumers' Consumption Values, Purchase Attitudes, and Purchase Intentions toward Upcycled Food Products)

  • This study aims to examine the structural relationship among consumers' consumption values, purchase attitudes, and purchase intentions toward upcycled food products. Survey data were collected from 400 consumers aged 20 and older between June 11 and June 20, 2024. The results revealed that functional value, ethical value, exploratory value, and situational value positively influenced purchase attitude, while social value, ethical value, exploratory value, and purchase attitude significantly influenced purchase intention. Purchase attitude, driven by ethical and exploratory values, was identified as a key factor explaining purchase intention. Based on these findings, the study suggests that developing innovative and original products is essential to attract consumer attention and stimulate the consumption of upcycled food. Additionally, marketing strategies that emphasize ethical responsibility and environmental benefits should be implemented. Furthermore, strengthening ESG management through sustainable business models, such as eco-friendly packaging and transparent information, can enhance positive consumer attitudes and lead to increased product purchases.

Nested PCR 기법을 이용한 인삼 뿌리썩음병원균의 특이적 검출 (Specific Detection of Root Rot Pathogen, Cylindrocarpon destructans, Using Nested PCR from Ginseng Seedlings)

  • Cylindrocarpon destructans는 인삼 및 수목에 뿌리썩음병을 일으키는 토양 전염병 식물병원균이다. 신속 정확한 검출 가능성을 알아보기 위하여 종 특이적인 primer와 nested PCR 기법을 활용하여 인삼 유묘로부터 뿌리썩음 병균 C. destructans로 2차 PCR증폭을 실시한 결과 병원성이 확인된 C.destructans에서만 400bp의 종특이적 증폭산물을 얻을 수 있었다. 종 특이성 primer 와 nested PCR 기법을 이용한 인삼뿌리썩음병균 DNA에 대한 반응 민감도는 최저 약 1fg으로 나타나 단 몇 개의 포자만 존재해도 검출이 가능하였다. 또한, nested PCR 기법은 실제 이병토양에 심었을 경우에도 C.destructans 에 감염된 인삼 유묘로부터만 정확하게 병원균을 검출해 내었다. 종특이적 primer 와 nested PCR 기법을 이용한 본 연구 결과는 실제 재배농가에서 인삼 경작시 뿌리썩음병 진단에 매우 유용하게 활용될 수 있을 것으로 판단된다.

  • 1. pH가 6.4인 상토(床土)에 복비시제품(複肥試製品) 시용(施用)으로 생육기간중(生育期間中)의 상토(床土) pH가 4.0~5.4로 유지(維持)되었다. 2. 복비시제품구(複肥試製品區)의 묘생육상황(苗生育狀況)은 단비구(單肥區)에 비(比)하여 건물중(乾物重)과 엽수(葉數)가 많았으며 건물중(乾物重)/초장비(草長比)도 높았고, 묘립고병(苗立枯病)의 발생률(發生率)도 적었다. 3. 묘(苗)의 무기성함량(無機成分含量)은 생육기간(生育初期)에는 단비구(單肥區)에 비하여 복비시제품구(複肥試製品區)가 다소 낮았으나 생육후기(生育後期)에는 높은 경향(傾向)이었다. 4. 복비시제품(複肥試製品)을 1회(回) 전량기비(全量基肥)로 시용(施用)하여 성력재배(省力化栽培)가 가능(可能)하였고, 건묘(健苗)를 육성(育成)할 수 있었다.

  • Visual features of a mushroom(Lentinus Edodes L) are critical in sorting and grading as most agricultural products are. Because of its complex and various visual features, grading and sorting of mushrooms have been done manually by the human expert. Though actions involved in human grading looks simple, a decision making undereath the simple action comes form the results of the complex neural processing of the visual image. And processing details involved in the visual recognition of the human brain has not been fully investigated yet. Recently, however, an artificial neural network has drawn a great attention because of its functional capability as a partial substitute of the human brain. Since most agricultural products are not uniquely defined in its physical properties and do not have a well defined job structure, a research of the neuro-net based human like information processing toward the agricultural product and processing are widely open and promising. In this pape , neuro-net based grading and sorting system was developed for a mushroom . A computer vision system was utilized for extracting and quantifying the qualitative visual features of sampled mushrooms. The extracted visual features and their corresponding grades were used as input/output pairs for training the neural network and the trained results of the network were presented . The computer vision system used is composed of the IBM PC compatible 386DX, ITEX PFG frame grabber, B/W CCD camera , VGA color graphic monitor , and image output RGB monitor.

저온숙성마늘 농축액을 첨가한 양갱의 품질특성과 항산화 효과 (Effect of Aged Garlic Extract on the Antioxidant Activities and Quality Characteristics of Yanggaeng)

  • This study was performed to investigate the quality characteristics of Yanggaeng, including color, pH, total polyphenol contents, and antioxidant activities, with the addition of aged garlic extract (0, 3, 6, 9 or 12%), aged at low temperatures. Among the color characteristics, the lightness value decreased, and the redness and yellowness values increased, proportional to the amount of garlic extract concentrate added to Yanggaeng. The pH of Yanggaeng significantly (p<0.05) decreased according to the amount of aged garlic extract added. Total phenol contents of control (0% aged garlic extract added Yanggaeng) was the lowest, followed by 3, 6, 9, 12% aged garlic extract added to Yanggaeng (88.15, 118.39, 156.91, 208.79 mg/g, respectively). Antioxidant activities, such as DPPH and ABTS radical scavenging activity, significantly increased with increase in the aged garlic extract concentration. In the sensory evaluation for Yanggaeng, 6% aged garlic extract added to Yanggaeng had the highest score in taste, flavor, and overall acceptance. Based on these results, it is suggest that the addition of 6% aged garlic extract to Yanggaeng can be developed as a product for the elderly.

Gene Cloning, Expression, and Characterization of a Novel ${\beta}$-Mannanase from Bacillus circulans CGMCC 1416

  • A DNA fragment containing 2,079 base pairs from Bacillus circulans CGMCC 1416 was cloned using degenerate PCR and inverse PCR. An open reading frame containing 981 bp was identified that encoding 326 amino acids residues, including a putative signal peptide of 31 residues. The deduced amino acid sequence showed the highest identity (68.1%) with $endo-{\beta}-1,4-D-mannanase$ from Bacillus circulans strain K-1 of the glycoside hydrolase family 5 (GH5). The sequence encoding the mature protein was cloned into the pET-22b(+) vector and expressed in Escherichia coli as a recombinant fusion protein containing an N-terminal hexahistidine sequence. The fusion protein was purified by $Ni^{2+}$ affinity chromatography and its hexahistidine tag cleaved to yield a 31-kDa ${\beta}$-mannanase having a specific activity of 481.55U/mg. The optimal activity of the purified protein, MANB48, was at $58^{\circ}C$ and pH 7.6. The hydrolysis product on substrate locust bean gum included a monosaccharide and mainly oligosaccharides. The recombinant MANB48 may be of potential use in the feed industry.

Assessing the Activity Concentration of Agricultural Products and the Public Ingestion Dose as Result of a Nuclear Accident

  • Background: A model to assess the activity concentration of agricultural products and the public ingestion dose as result of a nuclear accident is necessarily required to manage the contaminated agricultural systems by the accident, or to estimate the effects of chronic exposure due to food ingestion at a Level 3 PSA. Materials and Methods: A dynamic compartment model, which is composed of three sub-modules, namely, an agricultural plant contamination assessment model, an animal product contamination assessment model, and an ingestion dose assessment model has been developed based on Korean farming characteristics such as the growth characteristics of rice and stockbreeding. Results and Discussion: The application study showed that the present model can predict well the characteristics of the activity concentration for agricultural products and ingestion dose depending on the deposition date. Conclusion: The present model is very useful to predict the radioactivity concentration of agricultural foodstuffs and public ingestion dose as consequence of a nuclear accident. Consequently, it is expected to be used effectively as a module for the ingestion dose calculation of the Korean agricultural contamination management system as well as the Level 3 PSA code, which is currently being developed.

  • 중요한 먹을거리 중의 하나인 농산물, 하지만 근래에 들어 소비자들은 농산물을 믿고 구매하기가 쉽지 않다. 생산지도 확실치 않은 외국의 값싸고 품질을 믿을 수 없는 농산물의 수입, 국내의 일부 과다한 농약사용, 그리고 환경오염에 따른 재배지 토양의 오염 등 단지 판매자와 생산자의 양심에만 맡길 수 있는 문제들이 아니다. 정부는 이를 해결하고자 원산지 표시제를 시행하고 있지만 이조차도 지켜지지 않는다면 무용지물에 불과할 것이다. 건강과 먹을거리에 관심이 더 높아진 요즘, 소비자들은 자신이 구매하려는 농산물에 대해 좀 더 정확하고 믿을 수 있는 정보를 얻고 싶어 한다. 이에 본 논문에서는 RFID(Radio-Frequency IDentification) 기술을 기반으로 농산물에 RFID 태그를 부착하여 소비자가 이를 구매 시 RFID 리더기를 통해 RFID 태그를 읽어 해당 농산물의 믿을 수 있는 생산정보와 안전성정보를 소비자에게 정확하고 손쉽게 제공하는 농산물 생산이력정보 제공 시스템을 제안하였다.

Identification of Papaya Ringspot Potyvirus type W infecting squash in Korea

  • A flexuous rod-shaped virus was isolated from Cucurbita pepo leaves showing green mosaic and puckering symptoms at Anseong, Korea. Based on the biological tests, electron microscopy, and reverse transcription-polymerase chain reaction (RT-PCR), the isolate was identified as Papaya ringspot virus type Watermelon (PRSV-W). In the biological test, host range of PRSV-W was limited in the families Cucurbitaceae and Chenopodiaceae. Most susceptible cucurbit species, such as Cucurmis lanatus, Cucurmis sativus, Cucurbita pepo, and Citrullus lanatus, responded to mechanical inoculation by PRSV-W that induce green mosaic, malformation, puckering, and narrow laminae. The local lesion symptoms were produced on the inoculated leaves of Chenopodium maranticolor and C. quinoa PRSV specific primers which amplifies the part of the coat protein (CP) genes, generated a 648 bp product from 6 isolates of PRSV-W, but no amplification had been detected in other viruses including CMV, CGMMV, KGMMV, ZYMV and WMV. In electron microscopy, PRSV particles were flexuous, approximately 780 nm in length and 12 nm in width. PRSV-W is one of the worldwide viruses which has the great economic importance in cucumber, melon, squash, watermelon, and other cultivated cucurbits with ZYMV and WMV. This is the first report of PRSV-W on cucurbits in Korea.
