Quantitative Analysis of Orcinol and Acute Toxicity of Gyrophora esculenta (석이중 오르시놀 정량 및 급성독성)

  • In previous study, Gyrophora esculenta showed significant inhibitory effect on $\alpha$-glucosidases in vitro and blood glucose elevation in vivo. In the isolating process of active substance, orcinol was separated from Gyrophora esculenta. Orcinol is known to be toxic, therefore, in this study, it was analysed by the TLC densitometry method for quantitative determination from Gyrophora esculenta. The average amount of orcinol of Gyrophora esculenta was 0.2%. For the purpose of removing orcinol, the water extract of Gyrophora esculenta was sequentially fractionated by organic solvents, and the acute toxicity of each fraction was assessed in mice. Among them, the LD50 of butanol fraction was 1.19 g/kg(p.o.) and the weight increase of mice in that group was somewhat retarded.

Comparative Acute Toxicity Studies on Woo Whang Chung Shim Won Suspension and Pill in Mice (우황청심원 환제 및 액제의 급성경구비교독성시험)

  • Acute toxic effects of the Woo Whang Chung Shim Won suspension and pills were studied with the doses of 60, 74, 90, and 110 ml/kg in mice. In this study, we daily examined number of deaths, clinical signs, body weights, and pathological findings for 7 days after administration of Woo Whang Chung Shim Won. All mice given the highest dose (110ml/kg) died at 24 hour after administration of Sam-Sung Woo Whang Chung Shim Won suspension (Byunbang, SS-BS and Wonbang, SS-WS), one male and two female mice given 90 ml/kg dose of SS-BS died, and a few mice given middle dose (74 ml/kg) of SS-WS died. In control group (110 ml/kg) treated with vehicle (SS-BS and SS-WS), three out of 5 males and two out of 5 females mice died during the study. However, in animals treated with Kwang-Dong Woo Whang Chung Shim Won suspension (KD-S) and pill (KD-P), deaths were not observed. In the clinical signs, increase of drooling and decrease of spontaneous motor activities were observed in the highest dose group (110 ml/kg). No significant dose-related change in body weight was observed. The results suggest that the toxic effect of SS-BS and SS-WS may be atttributed to the solution for the Woo Whang Chung Shim Won suspension.

  • This experiment was carried out to study on the safety assessment of Bas tures . Ursus thibetanus extract solution(BU) for Herbal acupuncture. SD rats and ICR mice were used for acute toxicity test the results were summerized as follows; 1. In rats and mice, LD50 value could not be measured. 2. There were no abnormal finding in acute toxicity test treated BU for Herbal-acupuncture.

  • This study was attempted to investigate the acute toxicity of Liocolae vermiculus(Liocola brevitarsis) extract in mice, the effect on GOT, GPT Al.p, LDH activities and level of total cholesterol in serum of $CCl_4-intoxicated$ rats. In acute toxicity test, Liocolae vermiculus extract showed 10% mortality at 2,000 mg/kg, p.o. and at 1,000 mg/kg, i.p.. The Liocolae verculus extract caused a remarkable decrease in serum transaminase as well as Al.p activities in $CCl_4-intoxicated$ rats at $300{\sim}1,000{\;}mg/kg$ dosage ranges. The activities of -LDH and the level of total cholesterol were significantly decreased in all sample-treated group, when compared with the control group. The body weight decreased, and the liver and spleen weight increased in $CCl_4-intoxicated$ rats were significantly recovered by the administration of the extracts.

  • The acute toxicity of the combined vaccine (KGCC-95VI) for the prophylaxis against Japanese encephalitis and Hantaan virus infection. recently developed by Korea Green Cross Corporation, was investigated. KGCC-95VI was administered to the Balb /c mice in two routes, orally and subcutaneously, and into the New Zealand White rabbits subcutaneously. $LD_{50}$ was not accessible as there were no deaths in the group treated even at a dose 800 times the expected clinical dose in both animal species. Between the treated and control groups there were no statistically significant differences in body weight changes and clinical signs during the 14-day observation period, and no pathological gross findings. Accordingly KGCC-95VI is considered not to have the acute toxicity in mice and rabbits.

Acute Toxicity Study on the Extract of Mori Fructus (상심자의 급성독성에 관한 연구)

  • Acute toxicity on the water extract of Mori Fructus was examined in male and female mice. The water extract of Mori Fructus was orally administered at a dose of 5 mg/kg, 50 mg/kg, 300 mg/kg and 2,000 mg/kg and had been observed for two weeks. No mortality and abnormal clinical signs were shown for the observation period. At the terminal sacrifice, there were no difference in net body weight gain, organ weight and gross pathological findings among the groups treated with different doses of the water extract of Mori Fructus. The results suggested that under the condition employed in this study $LD_{50}$ would be more than 2,000 mg/kg. All the data obtained the experiments lead to the water extract of Mori Fructus should have very low acute toxicity.

Acute Toxicity of a Combined Preparation of the Standardized Extracts of Scutellaria baicalensis GEORGI and Salvia miltiorrhiza BUNGE (황금 및 단삼 표준화시료의 급성독성에 관한 연구)

  • Acute toxicity of a combined preparation of the standardized extracts Scutellaria baicalensis GEORGI and Salvia miltiorrhiza BUNGE in a ratio of 3:1 was examined in male and female ICR mice. Mice were treated with the test substance intragastrically at a dose of 0 mg/kg, 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. No death or abnormal clinical sign was shown during the observation period. Also there were no difference in net body weight gain, organ weight, and gross pathological findings at the terminal sacrifice. The results suggested that acute oral toxicity of a combined preparation of the standardized extracts is very low at the conditions employed in this study.

A Study on the Acute Toxicity of Leejung-tang(Lizhong-tang) and Fermented Leejung-tang(Lizhong-tang) Extract in ICR Mice (ICR 마우스를 이용한 이중탕 및 발효이중탕의 급성독성 연구)

  • Objectives : This study was carried out to investigate the acute toxicity and safety of $Leejung$-$tang$($Lizhong$-$tang$) and fermented $Leejung$-$tang$($Lizhong$-$tang$) extract. Methods : To evaluate their acute toxicity and safety, 0(control group), 1250, 2500 and 5000 mg/kg of $Leejung$-$tang$($Lizhong$-$tang$) and fermented $Leejung$-$tang$($Lizhong$-$tang$) extracts were orally administered to 35 male and 35 female ICR mice. After a single administration, we observed survival rates, general toxicity, changes of body weight, and autopsy. Results : Compared with the control group, we could not find any toxic alteration in any of treated groups (1250, 2500 and 5000 mg/kg). Conclusions : $LD_{50}$ of $Leejung$-$tang$($Lizhong$-$tang$) and fermented $Leejung$-$tang$($Lizhong$-$tang$) were over 5000 mg/kg and it is very safe to ICR mice.

  • Objectives : Socheongryong-Tang (小靑龍湯, SCRT) has been widely used to treat respiratory disease. In this study, we investigate the acute toxicity and safety of fermented Socheongryong-Tang extract. Methods : To evaluate the acute toxicity and safety, 0 (control group), 1250, 2500 and 5000 mg/kg of Socheongryong-Tang and fermented Socheongryong-Tang extracts were orally administered to male and female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight for the 14 days and autopsy. Results : Compared with the control group, we could not find any toxic alteration in all treated group (1250, 2500 and 5000 mg/kg). Conclusions : $LD_{50}$ of Socheongryong-Tang and fermented Socheongryong-Tang extracts might be over 5000 mg/kg and it is safe to ICR mice.

Acute Toxicity of Kami-Okchun-San in ICR-Mice (가미옥천산의 ICR 마우스 경구 투여에 의한 급성 독성시험 연구)

  • Kami-Okchun-San(OCS) is known as an effective herbal medicine on Type 2 diabetes. We peformed to investigate acute toxicity of OCS on ICR mice. ICR mice in acute toxicity experiment were administered orally with dosages of 3,200mg/kg (low dosage group), 4,000mg/kg (middle dosage group), 5,000mg/kg (high dosage group) per single time, respectively. Body weights, clinical signs, motalities and histopathological finding were observed daily for 14 days according to the Regulation of Korean Food and Drug Administration(1999. 12. 22). Single oral administration of OCS with different dosages, no animals died of the test drug. Autopsy of animal revealed no abnormal gross findings. Therefore, LD50 value of OCS for ICR mice was more than 5,000mg/kg on oral route. Normally increasing changes were observed in body weight, drinking water and food intake in every dosage group. Hematological parameters were also observed normally in all animals. No histopathological lesions were observed in both control and treated animals. Above data suggest that no toxic dose level of OCS in ICR mice is considered to be more than 5,000mg/kg. Therefore, it was concluded that OCS have no effect on acute toxicity and side effect in ICR mice.
