음곡에 시술한 등심초약침이 LPS로 유도된 흰쥐의 신장염에 미치는 영향 (The Effect of Junci Medulla Herbal-acupuncture at $KI_{10}$ on LPS induced Nephritis in Rats)

  • Objectives : This study aimed to evaluate the effects of Junci Medulla Herbal-acupuncture(JM-HA) at $KI_{10}$(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Rats with nephritis induced by LPS, were treated with JM-HA at $KI_{10}$ 3 times a week. The rats in the NP group and the saline group were treated with a needle prick and a saline injection respectively. To evaluate the effects of JM-HA at $KI_{10}$ on nephritis in rats, WBC, neutrophils in blood, BUN, creatinine, TNF-${\alpha}$ in serum, creatinine, total protein in urine and renal MPO were measured. Results : JM-HA at $KI_{10}$ significantly inhibited the increase of WBC and neutrophils in blood, BUN, creatinine, TNF-${\alpha}$ in serum, and MPO in kidney of LPS-stimulated rats. Conclusion : JM-HA at $KI_{10}$ has therapeutic effects on nephritis in LPS-stimulated rats. Therefore, it is suggested that JM-HA at $KI_{10}$ may be a useful therapy in clinical field after further researches.

  • Objectives : This study aimed to evaluate the effects of Stephaniae tetrandrae Radix Herbal-acupuncture(SR-HA) at KI10(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Male SD rats (n=32) were allocated to four groups; normal, LPS, saline, and SR-HA. The rats in the LPS, saline, and SR-HA groups were injected intra-peritoneally with LPS (2 mg/kg) to induce nephritis. The rats in the saline and SR-HA groups were treated with saline injection and SR-HA respectively at KI10 three times a week. To evaluate the effects of SR-HA at KI10 on nephritis in rats, the authors evaluated the numbers of WBC and neutrophils in blood, and the levels of creatinine, TNF-${\alpha}$, CINC-1 in serum, creatinine, total protein in urine and renal MPO. Results : SR-HA at KI10 significantly inhibited the increase of WBC and neutrophils in blood, TNF-${\alpha}$, CINC-1 in serum, creatine, total protein in urine and MPO in kidney of LPS-stimulated rats. Conclusions : SR-HA at KI10 may have therapeutic effects on nephritis in LPS-stimulated rats.

삼백초약침액(三白草藥鍼液)이 아나필락시스에 미치는 영향 (Effect of Saururus chinensis (Lour.) Baill. Aquacupuncture on Anaphlylaxis in Mice)

  • Objective We investigated the effect of Saururus chinensis (Lour.) Baill. aquacupuncture (SCB) on anaphylaxis in mice. Methods : We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. Results : SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the serum histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability. In addition, SCB showed a significant inhibitory effect on anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. Conclusion : These results indicated that SCB inhibits anaphylatic reaction.

족삼리에 시술한 고빈도 전침자극이 류마토이드 관절염 동물모델에 미치는 영향 (Effects of High Frequency Electro-acupuncture ($ST_{36}$) on the Adjuvant-induced Arthritis in Rats)

  • Objectives: This study was to investigate the effect of high frequency electro-acupuncture (EA) at $ST_{36}$ acupoint on the Freund's Complete Adjuvant (FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Experimental groups were divided into 4 groups; Normal, Control, $ST_{36}$ and Non-Acupoint (NA). $ST_{36}$ group was treated by 120Hz EA at $ST_{36}$ acupoint. Body weight, paw edema volume and ankle joint thickness were measured after treatment. And we investigated the effects of 120Hz EA via WBC count, segmen neurophil, lymphocyte, PGE2 assay and NADPH-d histochemistry. Results and Conclusions : The mean of body weight and ankle joint thickness of $ST_{36}$ group was increased compared with control group. The mean of paw edema volume, WBC count, segment neutrophil and lymphocyte of were not decreased with control group.   • To clarify the distinction between three difference acupoints, the effects of acupuncture on a behavioral performance were evaluated following formalin test. Male Sprague-Dawley rats were used. Each rat received a manual acupuncture at ST36 (zusanli), SP9 (yinlingquan) or BL60 (kunlun) acupoint before formalin injection. The flinching and licking responses were counted by two blinder investigators. The pretreatment of BL60 acupoint was showed significantly inhibition in flinch behavior as compared with control group. These results suggest that acupuncture at BL60 acupuncture may be effective in relieving inflammatory pain.

Acupuncture treatment of torticollis in a foal

  • A 6-month-old thoroughbred filly foal had torticollis and circled towards the right side. A local veterinarian treated her twice using dexamethasone for 1 week but there was little clinical improvement. Needles were inserted into Ting points of both hind limb and one ocular acupoint (shang jiao area from GB01). Injection acupuncture (dexamethasone, twice/week) was used at GV16, GB20, BL10 and LU07, for the neck and head. Acupoint GB34 was added to those points at session 4. In addition, a trigger point in the left neck was injected with 0.2 ml ($200{\mu}g$ of apitoxin) of bee-venom diluted with 1 ml of 2% lidocaine from session 1 to session 3. At session 2, the symptoms had ameliorated a little. At session 3, they were much improved; the right ear was completely normal and the neck could be moved about 60% of normal range. At session 4, nodding was possible and stiffness of the neck was much improved, having returned to about 80% of normal range. Three days after session 4, the symptom of torticollis had disappeared completely and the foal could walk in a straight line. In conclusion, the present patient was a case with equine torticollis which showed favourable therapeutic response by combination of needle-acupuncture plus injection-acupuncture with dexamethasone and apitoxin.

임신오조(姙娠惡阻)의 외치법(外治法)에 대한 고찰(考察) (A study on the external treatment of hyperemesis gravidarum)

  • Hyperemesis gravidarum is one of the most common symptom in the early pregnancy and if it cause severe malnutrition by means of heavy vomiting as a basic sign of disease of pregnant syndrome, appropriate treatments are necessary. In the clinics the methods of treatment of hyperemesis gravidarum include herb-medication usually. But herb-medication therapy is rather difficult because during pregnancy it may cause vomiting by the smell and taste of herbs. So the author investigated the literatures referred to the external treatments of hyperemesis gravidarum excluding herb-medication and the results obtained here were as follows. 1. The external methods of treatment of hyperemesis gravidarum are various and include moxibustion, the method of applying drug at the umbilicus, ear-acupuncture method, pressure massage therapy, naso-spray method, acupoint injection, intervenous injection 2. Among the external methods of treatment of hyperemesis gravidarum , the method of applying drug at the umbilicus is most variable and almost all prescriptions use Zingiber officinalis Rose, characteristically. 3. Ear-pressure massage method to treat the hyperemesis gravidarum uses the car-acupoints of Gan, We, Shin-mun, Gyo-gam. 4. Among the external methods of treatment of hyperemesis gravidarum , moxibustion, pressure massage, and acupoint injection select frequently Nae-gwan(PC6), Chok-samni(ST36) and Chung-wan(CV12). 5. In the both the internal and external methods of treatment of hyperemesis gravidarum one have to differentiate syndromes in the viewpoint of oriental medicine and treat with the principls of treatment and prescriptions which are fit to each differentiation model under the principle of regulating the stomach and relieving vomiting.

족삼리(足三里) 전침(電鍼)이 백서(白鼠)의 슬관절염(膝關節炎) 통증(痛症)에 미치는 영향(影響) (Effects of Acupuncture applied to Food Samli on the Rat Model of Knee Arthritic Pain)

  • Objectives : The usage of acupuncture has gained popularity as an alternative method of treatment for certain chronic pain conditions. However, the efficacy of acupuncture in various diseases has not been fully established and the underlying mechanism is not clearly understood. In the present study, the effect of electroacupuncture (EA) applied to foot samli$(ST_{36})$ on the carrageenan-induced knee arthritic pain was examined. Methods : A common source of persistent pain in humans is the knee arthritis. Knee arthritis was induced by injection of 2 % carrageenan $50\;{\mu}l$ into the knee joint cavity. When rats developed pain behaviors, EA was applied for 30 min. under enflurane anesthesia with repeated train stimuli at the intensity of 10X of muscle twitch threshold. The weight bearing force of the hind limb was measured for an indicator of pain level after each manipulation. Results : The average weight borne by the hind limb during normal gait was 55% of total body weight, which was reduced to less than 10% after knee arthritis. EA improved the weight bearing of the arthritic hind limb significantly for the duration of 4 hr. EA applied to $ST_{36}$ point produced a significant improvement of stepping force of the arthritic foot lasting for at least 4 h. However, $GB_{31}$ point did not produce any significant increase of weight bearing force. The analgesic effect was specific to the acupuncture point since the analgesic effect on the knee arthritis model could not be mimicked by EA applied to a nearby point, $GB_{31}$. The relations between EA-induced analgesia and endogenous nitric oxide(NO) and inducible NO synthase(iNOS)/neuronal NOS was also examined. Results were turned out that both NO production and nNOS/iNOS protein expression which is increased by arthritis were suppressed by EA stimulation applied to $ST_{36}$ point. Conclusions : The data suggest 1) that EA produces a potent analgesic effect on the rat model of chronic knee arthritis pain in a point specific manner and 2) that EA-induced analgesia modulate endogenous NO through the suppression of nNOS/iNOS protein expression.

Suppression of Cocaine Intake by Acupuncture at HT7

  • Objectives: Cocaine has been well known as a representative drug of abuse for its strong reinforcing effect linked to the mesolimbic reward system including dopamine, and such reinforcement leads to the addiction. Acupuncture has been used widely in eastern Asia for the treatment of various diseases including mental disorders and psychiatric problems, and is gathering more interest as one of the complementary and alternative medicines in western countries. In a previous study, we demonstrated that acupuncture at HT7 could affect dopamine release in the mesolimbic system of rats sensitized to alcohol and morphine. This study was designed to investigate whether acupuncture at HT7 could attenuate cocaine intake or not. Material & Methods: Male Sprague-Dawley rats weighing 270-300 g at the start of experiment were trained to self-administer food pellets under a fixed ratio 1 schedule. After the success of acquisition of 100 pellets within 3 h for 3 consecutive days, animals were subjected to surgery whereby Silastic tubing was implanted into right jugular vein and secured with mesh under the anesthetization using pentobarbital injection (50 mg/kg, i.p.). Following recovery, rats were trained to self-administer cocaine (0.25 mg/kg) in daily 2 h sessions under fixed ratio 1 schedule over 10 days. Each treatment was performed on the next day of each establishment of baseline. Results: Results show that acupuncture at HT7, but not at control points, reduced cocaine intake significantly. Acupuncture at HT7 decreased selectively active lever response from $63.15{\pm}3.35$ to $51.46{\pm}3.99$ corresponding $82.12{\pm}5.31%$ compared to basal level. Also, it was demonstrated that the effect of acupuncture was mainly occurred at the half period. Nevertheless, acupuncture at HT7 did not influence the food taking behavior. Conclusions: From the results of this study, it may be suggested that acupuncture at HT7, at least in part, could contribute to the treatment of cocaine abuse.

형용자침이 발열 흰쥐의 체온하강과 중추성 면역에 미치는 영향 (A Study on Acupuncture of Spring-point(형혈); Effect on Body Temperature and Central Immune System)

  • Objective : There are many reports that acupuncture has thermoregulatory effects on human and animals. To investigate the effect and mechanism of antipyretic action of acupuncture, we observed the body temperature and cytokine expressions in the hypothalamus of rats. Methods : Lipopolysaccharide (LPS, i.p., 2.5mg/kg) was injected to conscious rats (Sprague-Dawley, male, n=4l) to cause hyperthermia and simple needling (stainless steel, 0.25 mm o.d., 5 mm insertion for 10 sec with no manipulation) was performed bilaterally with the measurement of rectal temperature. Next, we sacrificed rats to remove brain and determined the level of mRNA for interleukin-6 (IL-6), $interleukin-1{\beta}{\;}(IL-1{\beta})$, interleukin-2 (IL-2) and $interferon-{\gamma}{\;}(IFN-{\gamma})$ in the hypothalamus by using reverse transcriptase-polymerase chain reaction (RT-PCR). Resul1s : Needling on forepaw (acupoint HT8) and needling on hindpaw (acupoint BL66 and acupoint LR2) significantly inhibited LPS-induced fever of rats (P<0.01, 10 min after treatment respectively), but same treatment on proximal tail (non-acupoint) did not cause any change on fever. The levels of IL-6 and $IL-1{\beta}$ mRNA in the hypothalamus was significantly enhanced by LPS-injection, while the level of IL-6 and $IL-1{\beta}$ mRNA was markedly reduced after treatment on BL66 (P<0.01). Treatment on forepaw reduced it slightly, but not significantly. Equivalent stimulation on proximal tail did not cause any changes. Conclusions : Our results indicate that acupuncture stimulation on various body parts has differential thermoregulatory effects on LPS-induced fever of rats with site-specificity. And, we suggest that its antipyretic action might be accompanied with the suppression of hypothalamic production of pro-inflammatory cytokine of immune system, IL-6 and $IL-1{\beta}$.
