Successful Arterial Thromboembolism Therapy in a Cat with Recombinant Tissue Plasminogen Activator Using an Accelerated Dosing Protocol

  • An 8-year old female Korean Short Hair cat with a history of paralysis of both hind limbs less than 1 hour before admission was referred. On physical examination, the left hind limb was cold and there was no pulsation or mobility. On abdominal ultrasound examination, a thrombus 8 mm in length was found at the aortic bifurcation. The patient was diagnosed with hypertrophic cardiomyopathy (HCM) and cardiogenic pulmonary edema through radiologic evaluation and echocardiography. A tissue plasminogen activator (tPA) was applied intravenously using an accelerated dosing protocol (1 mg administered intravenously [IV] bolus, 2.5 mg IV over 30 min, 1.5 mg IV over 1 h) to treat the feline arterial thromboembolism. Within 12 h after administration of tPA, pulsation and mobility of both hind limbs were normal, without any noticeable complications. Clopidogrel was prescribed to prevent additional thrombus formation, and pimobendan, benazepril, and furosemide were prescribed for administration at home. The patient was discharged and survived 377 days.

  • In this study, geopolymer was prepared with reject ash and blast furnace slag using NaOH as an alkali activator and water glass. The aim of this study was to investigate the compressive strength variation according to the contents of NaOH and water glass and replacement ratio of reject ash (RA) and blast furnace slag (BS). The compressive strength measured after 28 days was 38.91 MPa for the geopolymer which consist of 100 % of BS with 1 wt% NaOH and 3 wt% water glass. The major factor for improving compressive strength was the alkali activator proportion and the replacement ratio of RA and BS.

  • This study was undertaker to observe the longitudinal change by orthodontic treatment for early Class III malocclusion in primary and mixed dentition. Cephalometric roentgenograms of 8 children with Class III malocclusion obtained during activator therapy were measured and compared with those obtained before activator therapy. The following results were observed. During treatment with activator; 1. The maxilla became retrueded in A,C,E, and K and protruded in F, G, H, and J. 2. The mandible became retruded in A,C,E, and F and F and protruded in H and J. No difference was observed in G and K. 3. Gonial angle became reduced in A,C,E,F, and K and increased in H. No difference was observed in G and J. 4. The steepness of mandibular plane became reduced in C,E,F,G,H, and J and increased in A. No difference was observed in K. 5. The inclination of upper incisor became more labially in A,C,E,G,H,J, and K. No difference was observed in F. 6. The inclination of lower incisor became more lingually in all cases.

Effect of Sodium Sulfate on Ramie Fabrics Treated with Pectinase (저마직물의 펙티나제 정련 시 황산나트륨의 영향)

  • This study examines the scouring effect of pectinase on ramie fabric and influence of sodium sulfate as an activator for pectinase. The scouring effects were measured by the weight loss and pectin contents. SEM, weight loss, stiffness, moisture regain and dye ability of ramie fabric teated with pectinase/sodium sulfate were also measured. When ramie fabrics were desized with $\alpha$-amylase, the optimum conditions were pH 6.5 at $60^{\circ}C$ for 80 min with 1%(o.w.f) $\alpha$-amylase concentration. When ramie fabrics were scoured with pectinase, the optimum conditions were pH 8.5 at $55^{\circ}C$ for 30 min with 10%(o.w.f) pectinase concentration. Addition of sodium sulfate improved enzyme activity significantly, which increased proportionally with increasing sodium sulfate concentration. When 50 g/l of sodium sulfate was added, the surface became cleaner compared to the enzyme treatment without salt: weight and tensile loss, moisture regain and dyeability of the treated fabrics increased, while pectin contents and stiffness decreased. Therfore, sodium sulfate was effective activator for the pectinase treatment of flax fiber.

A One-Step System for Convenient and Flexible Assembly of Transcription Activator-Like Effector Nucleases (TALENs)

  • Transcription activator-like effector nucleases (TALENs) are powerful tools for targeted genome editing in diverse cell types and organisms. However, the highly identical TALE repeat sequences make it challenging to assemble TALEs using conventional cloning approaches, and multiple repeats in one plasmid are easily catalyzed for homologous recombination in bacteria. Although the methods for TALE assembly are constantly improving, these methods are not convenient because of laborious assembly steps or large module libraries, limiting their broad utility. To overcome the barrier of multiple assembly steps, we report a one-step system for the convenient and flexible assembly of a 180 TALE module library. This study is the first demonstration to ligate 9 mono-/dimer modules and one circular TALEN backbone vector in a one step process, generating 9.5 to 18.5 repeat sequences with an overall assembly rate higher than 50%. This system makes TALEN assembly much simpler than the conventional cloning of two DNA fragments because this strategy combines digestion and ligation into one step using circular vectors and different modules to avoid gel extraction. Therefore, this system provides a convenient tool for the application of TALEN-mediated genome editing in scientific studies and clinical trials.

Compressive Strength and Optimal Mixing Ratio of Alkali Activated Cement Concrete Containing Fly Ash (플라이 애쉬를 활용한 알칼리 활성시멘트 콘크리트의 압축강도와 최적혼합비)

  • This is a fundamental research to utilize alkali activated cement(AAC) in concrete. The compressive strength of AAC concrete were measured for the various mixing ratios of activator/fly ash, and the mixing ratios of water glass, NaOH, and water among the activators. The mixing ratio of fine and coarse aggregates was maintained constantly. The relationships between the compressive strength and mixing ratios were analyzed to find the optimal mixing ratio of AAC concrete. As the results, the optimal mixing ratio of activator/fly ash in AAC concrete was 0.7, and that of water glass, NaOH, water among the activator was 4.0:1.0:2.5 for the maximum compressive strength.

  • FLOWERING LOCUS T (FT) and TERMINAL FLOWER 1 (TFL1) in Arabidopsis are homologous proteins that perform opposite functions: FT is an activator of flowering, and TFL1 is a repressor. It was shown before that change of a single amino acid (His88) of TFL1 to the corresponding amino acid (Tyr) of FT is enough to convert the floral repressor to an activator. However, structural basis of the functional conversion has not been understood. In our molecular dynamics simulations on modified TFL1 proteins, a hydrogen bond present in native TFL1 between the His88 residue and a residue (Asp144) in a neighboring external loop became broken by change of His88 to Tyr. This breakage induced conformational change of the external loop whose structure was previously reported to be another key functional determinant. These findings reveal that the two important factors determining the functional specificities of the floral regulators, the key amino acid (His88) and the external loop, are correlated, and the key amino acid determines the functional specificity indirectly by affecting the conformation of the external loop.

Cell Type-Specific and Inducible PTEN Gene Silencing by a Tetracycline Transcriptional Activator-Regulated Short Hairpin RNA

  • Inducible and reversible gene silencing in desired types of cells is instrumental for deciphering gene functions using cultured cells or in vivo models. However, efficient conditional gene knockdown systems remain to be established. Here, we report the generation of an inducible expression system for short hairpin RNA (shRNA) targeted to PTEN, a well-documented dual-specificity phosphatase involved in tumor suppression and ontogenesis. Upon induction by doxycycline (DOX), the reverse tetracycline transcriptional activator (rtTA) switched on the concomitant expression of GFP and a miR-30 precursor, the subsequent processing of which released the embedded PTEN-targeted shRNA. The efficacy and reversibility of PTEN knockdown by this construct was validated in normal and neoplastic cells, in which PTEN deficiency resulted in accelerated cell proliferation, suppressed apoptosis, and increased invasiveness. Transgenic mice harboring the conditional shRNA-expression cassette were obtained; GFP expression and concurrent PTEN silencing were observed upon ectopic expression of rtTA and induction with Dox. Therefore, this study provides novel tools for the precise dissection of PTEN functions and the generation of PTEN loss of function models in specific subsets of cells during carcinogenesis and ontogenesis.

Hormonal Regulation of Leptin, Resistin, and Plasminogen Activator Inhibitor-1 Gene Expression in 3T3-L1 Adipocytes

  • Leptin, resisitn and PAI-1 (plasminogen activator inhibitor-1) are synthesized and secreted by rodent fat cells and recently postulated to be an important link to obesity. This study was conducted to characterize the hormonal regulation of leptin, resistin, and PAI-1 gene expression in the 3T3-L1 adipocytes. The cells were treated with 0.5 $\mu$M insulin, 1 $\mu$M dexamethasone (Dex), or 0.05 $\mu$M triiodothyronine (T3) for 72 hours. The mRNA levels of each peptide were measured by semi-quantitative RT-PCR. The mRNA level of the leptin-producing ob gene was significantly increased by insulin, Dex, and T3 by 3.2-, 3.1- and 2.7-fold, respectively, compared to the control (p < 0.05). The level of resistin mRNA was increased by insulin, Dex, and T3 by 2.7-, 2.5- and 2-fold, respectively, compared to the control (p < 0.05). Likewise, the level of PAI-1 mRNA was significantly increased by insulin, Dex, and T3 compared to the control (p < 0.05). Taken together, our results suggest that insulin, Dex, and T3 may regulate the gene expression of leptin, resistin, and PAI-1 in 3T3-L1 adipocytes.

Luminescence Characteristics of ZnGa2O4:Mn2+,Cr3+ Phosphor and Thick Film

  • In this study, $ZnGa_2O_4$ phosphors in its application to field emission displays and electroluminescence were synthesized through the precipitation method and $Mn^{2+}$ ions. A green luminescence activator, $Cr^{3+}$ ions, and a red luminescence activator were separately doped into $ZnGa_2O_4$, which was then screen printed to an indium tin oxide substrate. The thick films of the $ZnGa_2O_4$ were deposited with the various thicknesses using nano-sized powder. The best luminescence characteristics were shown at a thickness of 60 ${\mu}m$. Additionally, green-emission $ZnGa_2O_4:Mn^{2+}$ and red-emission $ZnGa_2O_4:Cr^{3+}$ phosphor thick films, which have superior characteristics, were manufactured through the screen-printing method. These results indicate that $ZnGa_2O_4$ phosphors prepared through the precipitation method have wide application as phosphor of the full color emission.
