Basic Physiological Activities of Bifidobacterium infantis Maeil-K9 and Lactobacillus plantarum KCTC3099 Selected by Anticarcinogenic Activities. (항암 활성능이 우수한 Bifidobacterium infantis Mneil-K9과 Lactobacillus plantarum KCTC3099의 기초 생리활성)

  • This study was conducted to confirm the availability of lactic acid bacteria as probiotics haying inhibitory effects to cancer cell line. Five lactic acid bacteria showing anti-cancer activity were compared by acid tolerance, bile tolerance, antibiotics resistance, milk fermentation, stability, and cell adherence activity to colon epithelial cell. The results obtained are as follows : In acid tolerance, all strains did not have a resistance below pH 3.0 and 3.5 except Lactobacillus plantarum KCTC3099. In antibiotics resistance, Lactococcus lactis and L. plantarum KCTC3099 were resistant to cotrimoxazol (128 mg/1), and Bifidobacterium adolescentis Maeil-K8 and B. infantis Maeil-K9 were resistant to doxycylin and gentamycin (4 mg/1). In case of cell adherence ability to Caco-2 cell, B. infantis Maeil-K9 was found to be superior to others as 3.1%, while the others were less than 0.5%. When the strains were cultured to milk base, viable counts of the strains tested increased more 1 log cycle than inoculation, but acid production was very low except L. plantarum KCTC3099. Also, L. plantarum KCTC3099, B. adolescentis Maeil-K8, and B. infantis Maeil-K9 were stable in fermented milk base during storage. In conclusion, L. plantarum KCTC3099 and B. infantis Maeil-K9 were confirmed to be superior for the availability as probiotics.

Fermentation Characteristics of Wine Yeast Strains (포도주 양조용 효모의 발효특성)

  • In fermentation characteristics, R2 was suitable for fast start and fermentation, while UCD530 was tool suitable far complete fermentation at 25$^{\circ}C$. T73 and AC- strains produced much more total acid compared to other strains and it was concerned with producing large amount of succinic acid and acetic acid. OC-2, UCD530, Beaujolais, and BC strains revealed low fermentation efficiency (below 62.0%), but EC1118, RC212, T73 and BM45 strains showed opposit result (above 70.0%). D254 and Wadenswil 27 seemed to have excellent cohesive ability because these two yeast strains made somewhat hard precipitation at the bottom after complete fermentation. T73 and CEG gave higher amounts of acetic acid (above 630 mg/L), while UCD530, W-3 and Beaujolais recorded low concentration (below 200 mg/L). In sulfur dioxide tolerance, preferable culturing temperature and times were 25$^{\circ}C$ and 72 hr respectively. The strains R2, BM45 and L2056 revealed high sulfur dioxide tolerance (above 30mg/L), white 71B, Wadenswil 27 and Beaujolais showed the opposite result (below 5mg/L)

Acid Tolerance of the Acid-Resistant Mutant of Leuconostoc paramesenteroides Improved for Kimchi Starter. (김치 Starter용으로 개량된 Leuconostoc paramesenteroides의 내산성 변이주가 갖는 내산성 특성)

  • To investigate the increased acid tolerance of a acid-resistant mutant Leuconostoc paramesenteroides P-100 improved as a kimchi starter, proton permeability, ATPase acitivity, glycolysis activity, $Mg^2$sup +/ releasement, and membrane fatty acid composition were studied and comprised with its wild type Leuconostoc paramesenteroides Pw. In the proton permeability experiment, the maximum values of the average half time (t$\_$1/2/) of pH equilibration through the cell membrane of the Pw and the P-100 were about 6.4 min and 7.8 min in 150 mM KCI solution, respectively. In the 3% NaCl solution, the t$\_$1/2/ values of the Pw and the P-100 were 5.5 min and 6.9 min, respectively. The values and pHs of maximal specific activities of ATPase originated from the Pw and the P-100 were 0.5 unit/mg protein and 0.78 unit/mg protein at pH 6.0, respectively. The result of pH dependence of glycolysis showed that the P-100 had higher activities than that of Pw except at pH 7.0. The releases of magnesium from the Pw and the P-100 were observed about 54.5% and 23.2% at pH 4.0 after 2 hours, respectively. The results of comparison of membrane fatty acid composition of the Pw with the P-100 showed that C$\_$8:0/, C$\_$9:0/, C$\_$10:0/, C$\_$11:0/, C$\_$18:0/, and C$\_$19:0,cyclo/ were major different fatty acids between two strains and the content of C$\_$18:1/, and C$\_$19:0,cyclo/ were 2.8%, N.D (not detected) in the Pw and 0.4%, 2.3% in the P-100. These results indicated that acid tolerance of the P-100 was significantly improved in comparison with its wild type Pw.
