Purification and Characterization of Fish Oil Obtained from Hydrolysates of Mackerel (Scomber japonicus) By-products (고등어(Scomber japonicus) 부산물 가수 분해물로부터 분리된 유지의 정제 및 특성)

  • This study aimed to investigate an effective method of purifying oil from hydrolysates of mackerel by-products. A hydrolyzed sample was separated into lipids and other material. After degumming of crude lipids with 6% citric acid solution, the phosphorous content (45 mg/kg) decreased to 5.8 mg/kg, and the acid value (18.03 mg /g) decreased to 1.19 mg KOH/g. Following the addition of 8% sodium hydroxide solution, the peroxide value (30.25 meq/kg) dropped to 3.18 meq/kg, and the chromaticity decreased from 0.488 nm to 0.057 nm. Bleaching was performed by adding 5% activated charcoal for 60℃ for 20 min under vacuum conditions. After bleaching, the acid value was 0.17 mg KOH/g, and the peroxide value was 1.21 meq/kg, but the chromaticity was not changed remarkably (0.062 nm). Crystallization was conducted to increase the amount of unsaturated fatty acids. After crystallization, the total amount of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) was 228.83 mg/g. The results suggest that the product can be used as an omega-3 fatty acid resource for functional food.

  • An improvement of methods in fibroin hydrolysates preparat significantly enlarges their applications to practical use. Acidic hydrolysis by hydrohloric acid is one of the methods for silk fibroin depolymeriza6tion. A low yield of final product and long time of the process are the demerits of this method. Possibility of preparation of water-soluble silk hydrolysates with more yield and less expenses is investigated in this study. Such possibility is occurred with the increasing tratment temperature and simultaneously decreasing treatment time, concentration of hydrochoric acid respectively, the concentration of sodium hydroxide used for neutraliza6tion of hydolysates after hydrolysis. Colour is decreased in this case and a small amount of activated, too. Protection of hydrolysates against precipitation after neutraliza6tion, and separation and during concentrating process is the other merit of this method. Creamy-coloured insoluble silk powder is the remainder of hydrolyzed fibroin. This is the only the immobiliza6tion of enzymes and other physiological active substances. Fine particles of this powder can be used as additives for artificial diets and cultural media, as well as raw materials for polymer membranes, etc.

  • Carbohydrate-free caseinomacropeptide (CMP) was isolated from the sweet whey powder by a precipitation method using 12% trichloroacetic acid. The yield of carbohydrate-free CMP was 2.7 g from 100 g sweet whey powder. The electrophoretic pattern and the amino acid analysis of CMP showed that isolated CMP was quite pure, indicating the precipitation with 12% trichloroacetic acid was very effective for isolating carbohydrate-free CMP from the sweet whey powder. Trypsin, covalently immobilized on pore glass beads by carbodiimide (EDC) method, was 20mg per 1g glass beads. CMP was almost completely hydrolyzed by soluble trypsin in 24hr, but not by immobilized trypsin. The tryptic hydrolysates were fractionated on a Bio-Gel P 4 column $(1.5{\times}120\;cm)$and separated peptides were tested for their capacities to inhibit platelet aggregation using a aggregometer. The hydrolysate obtained from CMP after 24hr digestion by immobilized trypsin showed the highest activity.

A Study on the Conversion to Feed Stuff from Cellulosic Biomass (섬유질자원(纖維質資源)의 사료(飼料) 전환(轉換))

  • To utilize several species of hard wood as raw materials of feed products, fermentation characteristics of cellulosic substrates to single cell protein was investigated, and results were summarized as follows. Among the microorganisms investigated, Tricoderma viride was selected as one of the most cellulolytic. Mixed culture of fungi did not show a synergistic effect on cellulose degradation. When the fungi were cultured at $28^{\circ}C$ for 7 days in a medium containing wheat bran 25 g, cellulose 0.25 g, proteose peptone 0.025 g and tween 800.025 g, cellulotic activities on carboxy methyl cellulose and filter paper reached maximum at 12 hr. The alkali treatment resulted in increased degradation of substrate from 13 to 18% when treated with enzymes for 12h, and reducing sugar formation increased with decreased size of substrates. Glucose was a very good feedback inhibitor of the enzyme from T.viride than that of xylose. When the substrate was rehydrolyzed, hydrolysis rate was 31% to reducing sugars within 12 hr. Quantative anlysis with HPLC showed the ratio of glucose to xylose in sugar syrups as 1.77 to 1. For the purpose of producing cellulosic-single cell protein from the sawdust of mulberry tree, 15 strains of xylose-assimilating yeast were isolated from 42 samples of rotten woods and compost soils and examined for their ability to utilize xylose. Then three strains were selected by their strong xylose-assimilating activities. The cultivative condition, the growth characteristics, and protein and nucleic acid productivities of three strains were investigated. The results obtained were, 1. Wood hydrolysate of mulberry tree was assimilated by 5 strains of CHS-2, CHS-3, ST-40, CHS-12 and CHS-13. 2. The optimum initial pH and temperature for the growth of strain CHS-13 were 4.4 and $30^{\circ}C$. 3. The specific growth rate of strain CHS-13 was $0.23h^{-1}$ and generation time was 3.01 hrs at the optimum condition. 4. CHS-13 strain assimilated 81 % of sugar in wood hydrolysate. 5. CHS-13 strain was identified as Candida guilliermondii var. guilliermondii 6. When the CHS-13 strain was cultured in the wood hydrolysate containing yeast extract, L-protein content was increased with yeast extract concentration. 7. The L-protein and nucleic acid yields from wood hydrolysate were 0.73 mg/ml and $4.92{\times}10^{-2}\;mg/ml$ respectively. 8. An optimal nucleic acid content of CHS-13 strain was observed in the medium containing 0.2% of yeast extract.

  • This study was carried out to investigate the resistance mechanism by three different kinds of procymidone-resistant and susceptible isolates of Botrytis cinerea which had been collected from green houses. The average resistance level of the resistant strains was 1,000 times higher than that of susceptible ones. Also, it was revealed that the resistance was not originated from components excreted by Botrytis cinerea, based on the result obtained from the treatment with piperonyl butoxide and triphenyl phosphate as an inhibitor of monooxygenase and esterase, respectively. The total lipod content of resistant strains was 1.3 times higher than that of susceptible ones, among fatty acids, palmitic acid, stearic acid, and linoleic and being 3.0, 2.5, and 2.0 times higher, respectinely. Also slight differences in sterol contents and components were observed. The crude chitin content was slightly higher in susceptible strains but contents of N-acetyl glucosamine, a hydrolysate of chitin, were about 2 times higher in resistant ones.

Hypolipidemic Effects of Peptide Fractions of Casein on Serum Lipids in Rats Fed Normal or High Fat Diet (정상 또는 고지방식을 섭취한 흰쥐에서 Casein 펩타이드 분획물이 혈청 지질농도에 미치는 영향)

  • In the recent studies, many researchers are interested in foods as functional components rather than nutrient sources. Cow's milk is considered as an excellent food sources because of its many nutrients. Casein is a major milk protein and has been reported to have hyperlipidemic and hypercholesterolemic effects. But several reporters have suggested that peptide fractions and hydrolysate of casein have hypolipidemic effects differing from intact protein, casein. Therefore, the objective of the study was to investigate how the casein peptide fractions affect lipid metabolism in rats fed normal or high fat diets. The peptide fractions and hydrolysate of casein were obtained by casein hydrolysis with trypsin. The male rats (Sprague-Dawley), weighing approximately 150 g, were fed each experimental diet containing casein (CAS), casein hydrolysate (CH), casein hydrolysate precipitate (Cpt) and two kinds of peptide fractions (CL & CB) for three weeks, respectively. In the exit I, the male rats were fed normal fat diets (7% soybean oil & cholesterol-free; Expt. I), and in the expt II, fed high fat diets (18% beef tallow & 1% cholesterol; Expt. II). Crude protein contents were calculated from nitrogen contents. Amino acid composition of each fraction was also analyzed. The concentration of total lipid, total cholesterol and triglyceride in serum, liver and feces were measured. As the results of study, tole rats fed peptide fractions with normal fat diets (Expt. I) had no effects on total lipid, total cholesterol and triglyceride concentration in serum and liver and fecal excretion. However, in the rats fed hydrophobic casein peptide fractions (CB) with high fat diet, fecal lipids excretion were significantly increased and the lipids concentration of serum and those of liver tended to decrease, numerically.

Cutaneous hydration effect of collagen hydrolysate containing collagen tripeptides (콜라겐 트리펩타이드를 고함량으로 함유하는 콜라겐 가수분해물의 피부 보습 효과)

  • Skin ageing is associated with compromised performance of its fundamental barrier functions, with undesirable changes in appearance. Since this may introduce a detrimental impact on the quality of life, significant effort to discover effective ingredients against ageing is being invested. Recently, collagen hydrolysates containing tripeptides such as GlyPro-Hyp (GPH) have been developed with anticipation of improved effects compared to that of existing collagen hydrolysate-products. To evaluate the cutaneous hydration effect of collagen tripeptides (CTP), meaningful biomarkers in human dermal fibroblasts (HDF) and NC/Nga Tnd mice were analyzed in this study. Increased levels of ceramide kinase, hyaluronic acid, collagen 1A, and hyaluronan synthase-2 (HAS2), and decreased levels of hyaluronidase-1 (HYAL1) and CD44 in HDF cells were demonstrated. Furthermore, significant reduction of transepidermal water loss (TEWL), scratching behavior, HYAL1, $TNF-{\alpha}$ and IL-6 and increased water content and HAS2 were verified by in vivo tests. These results strongly suggest the potential of CTP as a skin hydration agent.

Separation of Wood Components by Acetone (아세톤에 의한 목재 조성분의 분리)

  • The purpose of this study was to seek the optimum condition for effective separation of the chemical constituents of wood biomass by means of hydrolysis of acetone solution in presence of acid salt as a catalyst. Out of diverse acid salts the catalytic effect of aluminum sulfate ($Al_2(SO_4)_3$) was the most excellent during the hydrolysis of wood biomass in the acetone solution and the optimum concentration was 0.01 M (6.3 wt%). In the condition of mixture ratio of acetone and water to 9 : 1 as well as optimum concentration of aluminum sulfate two wood biomass species, oak wood (Quercus mongolica Fischer) and Pine wood (Pinus densiflora Sieb. et Zucc.), was hydrolyzed for 45 minutes at $200^{\circ}C$ and the degree of hydrolysis was determined to 92.7% and 92.4%, respectively. Extending the reaction time to 60 minutes in the mixture ratio of acetone and water to 8 : 2 the degree of hydrolysis of oak wood was also ca. 92.7%. In the case of Pinus, however, the similar hydrolysis ratio was obtained at $210^{\circ}C$. As the temperature and hydrolysis time increased, the quantitative amount of lignin recovered from the hydrolysate clearly increased, whereas the total amount of carbohydrates in the hydrolysate decreased rapidly. Considering the recoverable amount of lignin and carbohydrate in the hydrolysate, the best condition for the hydrolysis of wood biomasses were confirmed to the mixture ratio of acetone and water to 8 : 2, the concentration of aluminum sulfate of 6.3 wt%, hydrolysis temperature of $190^{\circ}C$ for 60 minutes. In this condition the total amounts of carbohydrate in the hydrolysates of oak wood and pine wood were estimated to 47.6% and 51.4%, respectively. The amount of lignin recovered from the hydrolysates were ca. 18.2% for oak wood and 13.7% for pine wood.

Effects of ABA, reduced nitrogen source and osmoticum for somatic embryogenesis in Liriodendron tulipifera (백합나무의 체세포배 유도에 미치는 ABA, 환원질소원 및 삼투압제 효과)

  • This study was conducted to evaluate effects of various kinds or concentrations in abscisic acid (ABA), reduced nitrogen sources (casein hydrolysate, casamino acid and L-glutamine) and osmoticum for production of somatic embryos (SEs) from pro-embryogenic mass (PEM) in yellow poplar (Liriodendron tulipifera). In comparison of various concentrations of ABA, the highest number (640/10 mg PEM) of SEs was marked in the treatment of 0.5 mg/L. With higher concentration than 0.5 mg/L ABA, number of induced SEs were decreased. And the lowest number of SEs were obtained from the treatment of 20 mg/L ABA. Differences of 8 treatments of the nitrogen sources in the medium were also compared. In the experiment of 8 treatments for SEs production, the highest result showed in the treatment of 500 mg/L casamino acid (223/5 mg PEM). In comparison of different kinds/concentrations of osmotica for SEs induction, the best response was obtained from the treatment of 4% sucrose (317/5 mg PEM). In contrast, no SEs were found from the treatments supplemented with any concentrations of maltose.

Development of Liquefied Seasoning Material from Cockle Shell By-Product (새조개 가공부산물을 이용한 액상 조미료 소재 개발)

  • A liquefied seasoning material was manufactured by using the enzymatic hydrolysis for the benefit of highly effective utilization of cockle shell by-product, and their quality was investigated. The weight ratio of by-product to whole cockle shell was 32.7%, and the contents of moisture and crude protein in the raw cockle shell by-product were 83.1% and 10.7%, respectively. The optimal concentrations of protease such as Protease N. P.(Pacific Chemical Co.) and Alcalase(Noo co), used in order to reduced the hydrolysis period, were effective at 4%(w/w), and optimal hydrolyzing time was 8 hours and after 8 hours were little changed. To improve flavor of the liquefied seasoning material, by Maillard reaction used thermal treatment, addition of glucose was very effective. And addition in hydrolysate with 10% glucose, 9% table salt, 2% starch and 0.5% caramel were suitable for promotion of taste. Total nitrogen and amino type nitrogen in the product were 1,607mg% and 1,264mg%, respectively. And the ratio of amino type nitrogen to the total nitrogen was 78.6%. The major free amino acid were glutamic acid, lysine, leucine, valine and aspartic acid, and content of glutamic acid was 1,027.5mg%.
