To determine the preventive effects of selenium and/or vitamin E on placenta retention in Holstein dairy cows. The cows were divided six groups ; control, Se, vit.E 1x, vit.E 3x, Se + vit./E 1x and Se + vit.E 3x in Cooong Nam province. 50mg of Se as sodium selenite and 700 IU of vit. E as dl-${\alpha}$ tocopherol acetate were injected 21 day perior to expected calving date, and 700 IU vit.E was injected 3 times at 21, 14 and 7 days perior to expected calving date in vit.E 3x groups. The cows which did not expelled their placenta until 12 hours post-parturient were termed as cow of placenta retention. The i.m. injection of a single dose of selenium and vit.E has significantly(P<0.05) affected the incidence of retained placenta by 7.0% versus 25.6% of controls, but a single injection of selenium or vit.E and a combination of selenium 1x and vit.E 3x has not significantly affected. But they affected neiher subsequent reproductive performances nor milk yields.
Root extract of Lythrum salicaria reported a hepato-protective effect on $CCl_4$-induced liver toxicity of rat was prepared into fractions such as n-hexane up layer (HA), n-hexane down layer (HB), diethyl ether (E), ethylacetate (EA), n-butanol (B) and water (W). Fractions prepared were tested their activities in vitro and in vivo condition. All of the fractions showed effective antioxidant asctivities on DPPH radical and $CuSO_4$-induced oxidation of human low density lipoprotein and E fraction showed the highest inhibitory effect (98.1% at $50\;{\mu}g/m{\ell}$) on linoleic acid autoxidation at $40^{\circ}C$, which was more effective than $\alpha$-tocopherol (82.4%). Five fractions (H = HA plus HB, E, EA, B, and W, 150 mg/kg/day) were fed into Sprague Dawley, male rats for 4 days, which were intoxicated with intra-peritoneal injection of carbon tetrachloride ($1\;m{\ell}/kg$ in corn oil) at the 4th day and were sacrificed in 24 hrs. Serum tumor necrosis factor-alpha (TNF-$\alpha$), a proinflammatory cytokine, elevated with $CCl_4$-intoxication in negative control group ($83\;pg/m{\ell}$) was significantly decreased in E fraction-supplemented group ($18\;pg/m{\ell}$). Cu, Zn-superoxide dismutase (SOD) activity increased in negative control group (0.12 U/mg protein) was decreased in E fraction (0.07 U/mg protein). From the results, it is suggested that ether fraction from root extract of L. salicaria would be a potent antioxidant candidate for ameliorating liver injury induced by chemical intoxicant.

The fatty acid composition, selected minor components, and the oxidative stability of the mixed edible oil (perilla seed oil and rice bran oil, 3:7 (v/v)) were analyzed. The fatty acid composition of the mixed oil was 32.1% of oleic acid, 30.6% of linoleic acid, 21.4% of linolenic acid, 13.0% of palmitic acid, and 1.7% of stearic acid. The mixed oil contained ${\alpha}$, ${\gamma}$ and ${\delta}$-tocopherols and tocotrienols showing the highest contents of ${\alpha}$-tocopherol. Total amount of tocopherols contained in the mixed oil was 46.63 mg/100 g oil. The composition and content of phytosterols were determined by a GC equipped with a flame ionization detector. Total quantity of phytosterols in the mixed oil was 712.80 mg/100 g oil. The most predominant phytosterol in the mixed oil was ${\beta}$-sitosterol, followed by campesterol and stigmasterol, in a decreasing order. The oxidative stability of the mixed oil was much higher than that of perilla oil, and similar to that of soybean oil, indicating the high oxidative stability of the mixed oil.
Natural compounds which could improve the oxidative stability of fish oil was screened from spices, herbs and naturally occurring antioxidants. Induction period of fish oil determined from oxidation curve by Rancimat ($80^{\circ}C$) was hardly affected with the addition of water-soluble and lipid-soluble fractions of garlic, leek, sesame leave and orange peel, and of organic acids such as citric acid, EDTA and selenium. Caffeic acid, catechin, quercetin and gallic acid laurylester, however, could extend the induction periods by $2.2{\sim}3.8$ times with the addition level of 0.1%(w/w). Rosemary extract and sesamol have a marked effect in retarding oxidation of fish oil. For example, induction periods of the oil samples stabilized with 0.1 rosemary extract and 0.1 sesamol were 16.4 hr and 11.6 hr, respectively, as compared to 4.0 hr of a control. When rosemary extract was used in combination with ascorbic acid (0.02%) or 8-tocopherol (0.2%), induction period could be extended to ca. 28 hr due to the synergism.

The colored rice, particularly black rice, has been well characterized to possess antioxidant properties. Rice bran, a by-product of the rice milling process, contains high levels of several phytochemicals which have antioxidant activities as well as health-beneficial properties. The purposes of this study were to evaluate the antioxidant compounds and antioxidant activities of the methanolic extracts from milling fractions and whole grain of two black rice cultivars. Whole black rice of the two cultivars were milled into rice bran and endosperm using the gradual milling system. These were evaluated for antioxidative activities by DPPH and ABTS radical scavenging activities. To determine the antioxidant compounds in the methanolic extract from the milling fractions, the content of polyphenolics, flavonoids, anthocycanins and ${\gamma}$-oryzanol were measured by spectrophotometric methods and vitamin E analysis was carried out by HPLC. The yield of whole black rice, rice bran, and endosperm were 3.1, 15.3, and 0.9% for Heugjinjubyeo and 2.7, 15.5, and 1.1% for Heugkwangbyeo, respectively. The methanolic extracts from rice bran showed generally higher antioxidant activities than the extracts from whole grain and endosperm. In addition, antioxidant compounds distributed much higher contents in rice bran extract than in the extracts from whole grain and endosperm. A significant correlation was also noted between free radical scavenging activity and polyphenolic compounds. The results of this study show that notable antioxidant activity in black rice bran are considered to have significant health benefits.
Antioxidant vitamin supplementation focuses one's attention on the prevention of age-related diseases. This study was conducted to investigate the antioxidant status and lipid profiles and to look into the antioxidant vitamin supplementation that affects lipid metabolism in 20 elderly non-smoking Korean women (placebo group: n = 6, vitC suppl: n = 7, vitE suppl: n = 7). Age, height, weight, muscle, percent of fat and WHR were not significantly different among the groups, however $\%$ of fat was above $33\%$ and WHR was above 0.9. And blood pressure of the placebo group was 131.7/81.7 (border line hypertension), that of vitamin C supplement was 141.4/87.1 (hypertension) and that of vitamin E supplement was 151.4/92.9 (hypertension). Although nutrient intakes of all groups were poor, antioxidant status (blood vitamins C, E, A, and beta-carotene) and lipid profile (TG, total-cholesterol, VLDL-cholesterol, LDL-cholesterol, HDL-cholesterol) were normal. For nutritional intervention, the vitamin C supplement group received L-ascorbic acid 1,000 mg, and vitamin E supplement group received d-alpha-tocopherol 400IU for 4 weeks, showing the effects of vitamin E supplementation. Response total cholesterol of HDL-cholesterol (T-Chol/HDL) in vitamin E supplement group was significantly decreased from 4.3 to 3.2. And response LDL-cholesterol of HDL-cholesterol (LDL/HDL) in the vitamin E supplement group was also significantly decreased from 2.6 to 1.7. In addition, after the adjustment for plasma lipids (TG, total cholesterol), plasma vitamin A levels in vitamin E supplement group were significantly increased from 7.89 mg/g to 14.91 mg/g. And systolic blood pressure in vitamin E supplement group was significantly reduced. These results suggested that vitamin E supplementation affects the lipid profiles and blood pressure in elderly non-smoking women. So various nutrition programs must be implemented against age-related diseases and further studies are needed regarding sorts and amounts of antioxidant nutrients and supplementation periods.
BACKGROUND/OBJECTIVES: Glutathione S-transferase (GST) forms a multigene family of phase II detoxification enzymes which are involved in the detoxification of xenobiotics by conjugating substances with glutathione. The aim of this study is to assess the antioxidative status and the degree of DNA damage in the subclinical hypertensive patients in Korea using glutathione S-transferase polymorphisms. SUBJECTS/METHODS: We examined whether DNA damage and antioxidative status show a difference between GSTM1 or GSTT1 genotype in 227 newly diagnosed, untreated (systolic blood pressure $(BP){\geq}130mmHg$ or diastolic $BP{\geq}85mmHg$) subclinical hypertensive patients and 130 normotensive subjects (systolic BP < 120 mmHg and diastolic BP < 80 mmHg). From the blood of the subjects, the degree of the DNA damage in lymphocyte, the activities of erythrocyte superoxide dismutase, the catalase, and the glutathione peroxidase, the level of glutathione, plasma total radical-trapping antioxidant potential (TRAP), anti-oxidative vitamins, as well as plasma lipid profiles and conjugated diene (CD) were analyzed. RESULTS: Of the 227 subjects studied, 68.3% were GSTM1 null genotype and 66.5% were GSTT1 null genotype. GSTM1 null genotype had an increased risk of hypertension (OR: 2.104, CI: 1.38-3.35), but no significant association in GSTT1 null genotype (OR 0.982, CI: 0.62-1.55). No difference in erythrocyte activities of superoxide dismutase, catalase, or glutathione peroxidase, and plasma TRAP, CD, lipid profiles, and GSH levels were observed between GSTM1 or GSTT1 genotype. Plasma levels of ${\alpha}-tocopherol$ increased significantly in GSTT1 wild genotype (P < 0.05); however, plasma level of ${\beta}-carotene$ increased significantly in GSTT1 null genotype (P < 0.01). DNA damage assessed by the Comet assay was significantly higher in GSTM1 null genotype than wild genotype (P < 0.05). CONCLUSIONS: These results confirm the association between GSTM1 null genotype and risk of hypertension as they suggest that GSTM1 null genotype leads to an increased oxidative stress compared with wild genotype.
Body antioxidant status is an important factor for the prevention of many chronic diseases in the elderly. This study was done to evaluate the nutritional status of antioxidant vitamins of the elderly by determining their intakes and plasma levels. It was also aimed to compare daily intakes and plasma levels of antioxidant vitamins by sex of age. Subjects were 225 elderly persons aged over 60 years old (63 males, 162 females) living in Ulsan area. Subjects were divided by groups according to age(< 65, 65-74, 75) and sex. Dietary Intakes were assessed by semi-quantitative food frequency questionnaires(FFQ). Plasma Vitamin C level was measured by 2,4-dinitrophenylhydrazine method and plasma levels of vitamin E, A and ${\beta}$-carotene were measured by HFLC. The average intakes of vitamin C were 104.9g(150% of RDA) and 104.4g(149% of RDA) in the elder1y males and females, respectively. Vitamin C intake of the elderly was significantly decreased by aging but not different by sex. The average intakes of vitamin A were 678$\mu\textrm{g}$RE(96.9% of RDA) and 604$\mu\textrm{g}$:RE(86.2% of RDA) in elderly males and females, respectively. The average levels of Plasma vitamin C were 6.22mg/L and 11.45mg/L in the elderly males and females, respectively. Those levels are within normal range(6-20mg/L). However the percentage of the elderly males with deficiency(< 2mg/L) and marginal level(2-4mg/L) of vitamin C were 27.4% and 16.1%, respectively. Plasma retinaol levels were 0.39mg/L for the elderly males and 0.37mg/L for the elderly females, which were within normal range. But the percentage of subjects with marginal level were 28% ill both males and females. Plasma ${\alpha}$-tocopherol levels of the elderly were lower than normal range(5-12mg/L). Plasma levels of vitamin C, E and ${\beta}$-carotene, except retinol, were significantly higher in the elderly females compared to males and showed significant decrease by aging.
This study was worked out to investigate the compounds of antioxidant constituents extracted from Stachys Sieboldii MiQ. and their effects on antioxidant activity by DPPH method, ferric thiocyanate method, and nitrite scavenging ability. Solvents such as methanol, hexane, chloroform, ethyl acetate, butanol, and water were used for this purpose. Total concentrations of polyphenols and flavonoids were measured in the methanol fraction. The ethyl acetate fraction showed the strongest activity by DPPH method, ferric thiocyanate method, and nitrite scavenging ability. The ethyl acetate extract was fractionated on a silica gel column using elution solvent (chloroform: methanol: water = 70 : 30 : 5 lower phase) at a flow rate of 1.0 mL/min. UV-VIS spectral data of each fraction showed adsorption maxima in the range of 284~330 nm. Among fractions, the fraction 1 that has λ$\_$max/ (nm) of 284 nm showed the strongest activity by DPPH method. The UV-VIS spectral data of phenolic compounds were known to lie in the range of 210~290 nm and 300~550 nm. Therefore, the results of our study suggested that Stachys sieboldii MIQ. contains phenolic compounds showing natural antioxidant activity.
This study was carried out to investigate the antioxidative activities of Rosa davurica Pall for the purpose of development of novel antioxidant from natural products. Antioxidant activities of four different parts of Rosa davurica Pall such as fruit, leaf, stem and root were examined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract from the root of Rosa davurica Pall showed the highest antioxidative activity among 16 samples tested. And, we also tested radical scavenging effects of 5 different extract compartments(Hexane, $CHCl_3$, EtOAc, BuOH and $H_2O$ fraction). EtOAc and BuOH fractions from the root of Rosa davurica Pall exhibited antioxidative activities higher to those of natural, ${\alpha}-tocopherol$ or synthetic antioxidants, BHT. The antioxidative substance of EtOAc fraction from the root of Rosa davurica Pall was successively purified with silica gel adsorption column chromatography and Sephadex LH-20 column chromatography. The purified active substance was isolated as crystal and identified as (+)-catechin by $^{l}H-NMR$ and $^{13}C-NMR$. This compound exhibited DPPH radical scavenging activity with the $IC_50$ value of $1.7\;{\mu}g/ml$. In the analysis of catechin content, the leaf extracts contained the highest catechin, and fruit extracts contained the lowest catechin. Considering antioxidative activity on DPPH assay, the extracts of Rosa davurica Pall showed a possibility to be used as a new material for natural antioxidant and functional food.
